建鲤(Cyprinus carpio var. Jian)微卫星DNA亲权鉴定

利用16个微卫星座位对建鲤10个全同胞家系647个子代进行亲权鉴定。Cervus3.0分析表明,16个微卫星位点的平均多态信息含量为0.7025,平均等位基因数为6.63,期望杂合度平均为0.7405。当双亲未知时,累积排除概率为0.999225,已知单亲时的累积排除概率为0.999996,置信度为95%。进一步模拟分析表明,要达到亲权鉴定的要求在双亲未知时通常需要8～12个微卫星位点,已知单亲时需要5～8个微卫星位点。在双亲均未知的情况下进行亲权鉴定,94.6%的后裔找到了其父母本,真实鉴定率低于模拟分析预测值,分析可能是与候选亲本间存在亲缘关系、无效等位基因的存在以及分型错误等因素有关。9个建鲤全同胞家系的鉴定,为今后的遗传图谱构建、QTL定位及分子标记辅助育种研究奠定了基础。

周期性饥饿再投喂对建鲤(Cyprinus carpio var.Jian)生长、体组成、消化酶的影响

采用周期性饥饿再投喂的方法,研究周期性饥饿再投喂对建鲤(1.5±0.2g)的生长、体组成、消化酶的影响。试验分为10个组,共进行60d。结果表明,在相同的饥饿天数下,延长再投喂天数可以使建鲤的增重率逐渐增加,而且S1F4、S2F8、S4F16组的增重率与对照组无显著性差异(P>0.05),9个周期性饥饿再投喂组在摄食期间的摄食率和特定生长率均显著性高于对照组(P<0.05),而且饵料系数均低于对照组,说明本试验建鲤的补偿生长是通过提高摄食率和饵料利用率共同实现的;周期性饥饿再投喂对全鱼水分、粗蛋白、粗灰分影响差异不显著(P>0.05),但9个周期性饥饿再投喂组的全鱼粗脂肪含量均低于对照组,而且在相同的饥饿天数下,随着再投喂天数的延长而升高;9个周期性饥饿再投喂组的肠道消化酶(蛋白酶、脂肪酶、淀粉酶)活力均高于对照组。

裂壶藻对津新鲤(Cyprinus carpio var.Jian)生长、血液非特异性免疫指标及抗病力的影响

选用初始体重为(26.77±1.56)g的津新鲤(Cyprinus carpio var.Jian)540尾,随机分为6组,每组3个重复,分别投喂含0、0.40%、0.80%、1.20%、1.60%和2.00%裂壶藻的等氮等能饲料,饲养56 d,探讨饲料中不同水平的裂壶藻对津新鲤生长、血液非特异性免疫指标及抗病力的影响。结果显示,在0.80%裂壶藻水平组中,增重率和特定生长率均显著提高(P<0.05),肥满度则在1.20%水平组中达到最大值(P<0.05),各组间成活率差异不显著(P>0.05)。不同水平的裂壶藻均可以显著提高津新鲤的抗氧化能力,其中过氧化氢酶、总超氧化物歧化酶和谷胱甘肽的含量均呈先上升后下降的趋势,且分别在1.20%、0.80%和1.20%组中达到最高(P<0.05);而丙二醛含量呈先下降后上升的趋势,在0.80%组中达最低(P<0.05)。不同水平的裂壶藻均可以显著提高津新鲤的非特异性免疫力,溶菌酶呈先上升后下降的趋势,其中1.20%组活性最高(P<0.05)。一氧化氮含量随裂壶藻水平增加而增加,各添加组含量均显著高于对照组(P<0.05)。谷草转氨酶/谷丙转氨酶在0.80%组中达到最小值(P<0.05)。随着裂壶藻水平的增加,津新鲤血细胞呼吸爆发活性和免疫保护率得到显著提高,感染嗜水气单胞菌后的累计死亡率则显著降低(P<0.05),其中,以0.80%组最佳。综合生长、血液非特异性免疫指标和抗病力来分析,饲料中裂壶藻的适宜添加水平为0.80%–1.20%。

油酸诱导建鲤(Cyprinus carpio var. Jian)肝细胞脂肪变性模型的建立

通过对油酸诱导时间及有效作用浓度的研究,建立油酸致建鲤肝细胞脂肪变性模型。本研究以胰蛋白酶消化法获得的原代肝细胞作为实验材料,加入不同浓度油酸(0、0.05、0.1、0.2、0.4、0.8 mmol/L)与肝细胞共培养24–48 h,诱导肝细胞脂肪变性,分别收集不同时段的肝细胞及上清液,采用试剂盒测定肝细胞中甘油三酯(TG)、总胆固醇(TC)的含量,同时测定上清培养液中谷丙转氨酶(GPT)、谷草转氨酶(GOT)、γ-谷氨酰转肽酶(γ-GT)、乳酸脱氢酶(LDH)和超氧化物歧化酶(SOD)的活性,MTT法测定肝细胞的存活情况,确定油酸最佳诱导浓度,油红O染色法观察细胞内脂肪滴的形成情况。研究结果显示,0.4 mmol/L油酸与肝细胞共培养48 h,可以显著提高肝细胞内TG、TC含量(P﹤0.05或P﹤0.01),极显著提高上清培养液中GOT、GPT、LDH、γ-GT的活性,显著降低上清培养液中SOD的活性,但与24 h相比有升高趋势,0.4 mmol/L油酸与肝细胞共培养24 h,光镜下可见细胞内有脂肪滴形成,以48 h最明显。综上所述,油酸浓度0.4 mmol/L、诱导时间48 h成功构建了肝细胞脂肪变性模型,为研究鱼类脂肪肝类疾病奠定了基础。

不同脂肪源饲料对津新鲤(Cyprinus carpio var.Jian)生长及脂类代谢的影响

试验选用初始体重为(44.34±1.58)g的津新鲤1 260尾,随机分为6组,每组3个重复,每个重复70尾鱼。6组饲料分别含有豆油、鸡油、玉米油、棕榈油、菜籽油、棉籽油。饲养8周后探讨饲料中不同脂肪源对津新鲤生长、相关脂代谢酶活性及脂肪酸合成酶(FAS)、脂蛋白脂酶(LPL)基因表达的影响。结果表明:玉米油组的相对增重率、特定生长率及蛋白质效率显著高于其它5组(P<0.05),但玉米油组饵料系数则最低。不同脂肪源饲料对肝胰脏及肾脏中LPL、肝脂酶(HL)、总脂酶(GE)、脂肪代谢酶活性影响很大(P<0.05),且玉米油组酶活性均最高;各组肝胰脏及肾脏FAS变化不大(P>0.05)。从基因表达来看,玉米油组更容易诱导和调节肝胰脏和肾脏中FAS及LPL m RNA的表达(P<0.05),再次投喂后6 h,肝胰脏和肾脏中LPL m RNA表达丰度均达到峰值,而肝胰脏中FASm RNA表达丰度达到最低值,肾脏中FAS m RNA表达丰度逐渐下降。总之,玉米油可作为津新鲤优质脂肪源。

Genetic mapping and QTL analysis for body weight in Jian carp (Cyprinus carpio var.Jian) compared with mirror carp (Cyprinus carpio L.)

We report the genetic linkage map of Jian carp(C yprinus carpio var. Jian). An F1 population comprising 94 Jian carp individuals was mapped using 254 microsatellite markers. The genetic map spanned 1 381.592 c M and comprised 44 linkage groups,with an average marker distance of 6.58 c M. We identified eight quantitative trait loci(QTLs) for body weight(BW) in seven linkage groups,explaining 12.6% to 17.3% of the phenotypic variance. Comparative mapping was performed between Jian carp and mirror carp( Cyprinus carpio L.),which both have 50 chromosomes. One hundred and ninety-eight Jian carp marker loci were found in common with the mirror carp map,with 186(93.94%) showing synteny. All 44 Jian carp linkage groups could be one-to-one aligned to the 44 mirror carp linkage groups,mostly sharing two or more common loci. Three QTLs for BW in Jian carp were conserved in mirror carp. QTL comparison suggested that the QTL confidence interval in mirror carp was more precise than the homologous interval in Jian carp,which was contained within the QTL interval in Jian carp. The syntenic relationship and consensus QTLs between the two varieties provide a foundation for genomic research and genetic breeding in common carp.

Systems pharmacology approach reveals protective mechanisms of Jian-Pi Qing-Chang decoction on ulcerative colitis

BACKGROUND Given the complex pathogenesis of ulcerative colitis (UC), the conventional therapeutic methods are not fully curative. As a sort of systematic complementary and alternative medicine, traditional Chinese medicine (TCM) provides new options for the standard therapy. Nevertheless, there are still numerous problems with the promotion of TCM attributed to its complexity, and consequently, new research approaches are urgently needed. Thus, we explored the protective effects of Jian-Pi Qing-Chang (JPQC) decoction on UC based on systems pharmacology approach, which might fill the current innovation gap in drug discovery and clinical practice pertaining to TCM. AIM To investigate the protective mechanisms of JPQC decoction on UC based on systems pharmacology approach. METHODS We performed systems pharmacology to predict the active ingredients, the matched targets, and the potential pharmacological mechanism of JPQC on UC. In vivo, we explored the effects of JPQC in a colitis model induced by dextran sulfate sodium. In vitro, we adopted the bone marrow-derived macrophages (BMDMs) as well as BMDMs co-cultured with Caco2 cells to verify the underlying mechanisms and effects of JPQC on UC under TNF-α stimulation. RESULTS Systems pharmacology revealed 170 targets for the 107 active ingredients of JPQC and 112 candidate targets of UC. Protein-protein interaction networks were established to identify the underlying therapeutic targets of JPQC on UC. Based on enrichment analyses, we proposed our hypothesis that JPQC might have a protective effect on UC via the NF-κB/HIF-1α signalling pathway. Subsequent experimental validation revealed that treatment with TNFα activated the NF-κB/HIF-1α signalling pathway in BMDMs, thereby damaging the epithelial barrier permeability in co-cultured Caco2 cells, while JPQC rescued this situation. The findings were also confirmed in a dextran sulfate sodium-induced colitis model. CONCLUSION JPQC could improve the mucosal inflammatory response and intestinal epithelial barrier function via the NF-κB/HIF-1α signalling pathway, which provides new perspectives on the pharmaceutical development and clinical practice of TCM.

Efficacy of Jian’ganle(健肝乐) versus Hugan Pian(护肝片),Glucuronolactone and Reduced Glutathione in Prevention of Antituberculosis Drug-induced Liver Injury

Summary： Evidence-based medicine is advocated by WHO and adopted by developed countries for many years. In China, however, the selection of essential medicine and various medical insurance reimbursement schemes medicine is usually based on experts＇ experience of prescription practice which is under heavy critics resulting from the lack of related comparative efficacy and evidence-based research. The efficacy of Jian＇ganle in prevention of drug-induced liver injury （DILI） caused by antituberculotics was evaluated in this study by comparison with Hugan Pian, glucuronolactone and reduced glutathione. Evidence was provided for relevant sectors such as Ministry for Human Resources and Social Security of the People＇s Republic of China and National Health and Family Planning Commission of the Peo- ple＇s Republic of China to select and renew the Essential Medicine List （EML）, the new rural cooperative medical scheme in China （NRCMS） list or the reimbursement list of industrial injury insurance. A total of 189 patients with initial pulmonary tuberculosis were divided into four groups who took antituberculotics combined with Jian＇ganle, Hugan Pian, glucuronolactone and reduced glutathione respectively. Their liver function profile including alanine aminotransferase （ALT）, aspartate aminotransferase （AST）, total bilirubin （TBIL）, direct bilirubin （DBIL）, total protein （TP）, albumin （A） and globulin （G） were detected at admission as baseline and after treatment. The Jian＇ganle group was compared with the three others by chi-square tests. In an aspect of maintaining bilirubin indexes normal, Jian＇ganle was more efficacious than glucuronolactone. And Jian＇ganle had a little more efficacy than reduced glutathione to maintain protein indexes normal as well. And the therapeutic regimen of antituberculotics combined with Jian＇ganle was the best in treating tuberculosis and preventing DILI at the same time. The study showed that among the four hepatinicas which demonstrated similar prevention of DILI caused by antituberculotics, Jian＇ganle has more advantages over the three others to some extent, which provides a reliable basis for health sectors to select and renew the EML, NRCMS List or the reimbursement list of industrial injury insurance.

黑龙江野鲤(Cyprinus haematopteru)与建鲤(Cyprinus carpio var.Jian)正反交与自交子代消化酶活力及其分布比较分析

在网箱养殖45天后,采用比较分析黑龙江野鲤与建鲤正反交与自交子代消化酶活力的方法,研究由杂交引起的遗传背景改变对消化酶活力及其分布的影响。结果表明,两杂交群体肝胰腺蛋白酶活力显著高于建鲤自交子代,后肠则显著低于后者,后肠脂肪酶活力显著高于黑龙江野鲤自交子代;黑龙江野鲤♂×建鲤♀子代各消化器官淀粉酶活力均显著高于两自交群体。两杂交群体除前、中肠淀粉酶活力差异显著度与建鲤自交子代不同外,3种消化酶活力分布及不同器官间的差异程度和两自交群体均相同;四个群体脂肪酶和淀粉酶活力在前、中肠及肝胰腺均表现出互补现象,后肠均有较高活力不互补。

Crises of Socialism in China and Chinese Rock in 1980s： The Case of CUI Jian

This paper focuses on CUI Jian and his rock music to examine the social complexity of the 1980s in China, which is much about the destructions brought by the Cultural Revolution and the economic reformation since 1978, as well as to investigate an alternative discourse that is different from mainstream in understanding the Chinese rock. Also, the paper through CUI＇s works tries to inquire the questions of how the ideal of socialism transforms after the fail of the revolution and how the disillusion connects the country＇s pursuit of the western modernization in music. Moreover, the paper asks how and by which CUI＇s songs associate themselves to the social transformation and in what way they root in the ＂socialistic personality＂ shaped by the socialist revolution since 1950s.

Effect of Bijia Jian Pill on inflammatory factors and liver fibrosis indexes in patients with chronic hepatitis B complicated with cirrhosis

Objective: To detect the effect of BiJia Jian Pill on inflammatory factors and liver fibrosis indexes in patients with chronic hepatitis B complicated with cirrhosis. Methods: A total of 94 patients with chronic hepatitis B combined with cirrhosis admitted in the hospital from January 2017 to December 2017 were selected as the subjects, and they were randomly divided into 47 cases in the observation group and 47 cases in the control group. The control group was given Adefovir Dipivoxil Tablets, and the observation group was treated with Bijia Jian pills on the basis of control group. The changes of liver function, inflammatory factors and liver fibrosis indexes were compared between the two groups before and after treatment. Results Before treatment, there was no significant difference in AST, ALT, TBIL and ALB between the two groups. After treatment, the AST, ALT and TBIL of the two groups were all decreased, and the AST, ALT and TBIL in the observation group were lower than those of the control group;the ALB of the two groups increased, and the ALB of the observation group was higher than that of the control group. Before treatment, there was no significant difference in TNF-α, IL-1 βand IL-10 between the two groups. After treatment, the TNF-α and IL-1β of the two groups were decreased, and the TNF-α and IL-1β in the observation group were lower than those of the control group;the IL-10 of the two groups increased, and the IL-10 in the observation group was higher than that in the control group. Before treatment, there was no significant difference in HA, LN, IV-C and PCIII between the two groups. After treatment, the HA, LN, IV-C and PCIII of the two groups were all decreased, and the HA, LN, IV-C and PCIII of the observation group were lower than those of the control group. Conclusion Bijia Jian pills can effectively improve the inflammatory factors and liver fibrosis in patients with chronic hepatitis B complicated with cirrhosis.

Jian Nao Ning for Treatment of Memory Impairment in Patients with Mild or Moderate Multi-infarct Dementia

Forty patients with multi-infarct dementia (MID) were randomly assigned to the treatment group (25 cases) treated with Jian Nao Ning (健脑宁JNN) and the duxil control group (15 cases). Memory function were assessed at baseline and endpoint using memory subscales of a battery of New Psychometric Tests (Chinese version) including mini-mental state examination (MMSE), verbal memory, and non-verbal memory, etc. After treatment, the mean scores of verbal memory in the Hopkins Verbal Learning Test (P<0.05) and total memory scores of memory items (P<0.001) in JNN group increased significantly; and improvement in episodic memory function including story recall (immediate and delayed), delayed word recall, verbal learning and verbal recognition and visual recognition in the JNN group was better than that in the duxil control group, suggesting that JNN can obviously improve memory function for the patients with mild or moderate multi-infarct dementia.
