Swingle polysaccharides (FP), we used extraction by hot water (WFP), ultrasonic-assisted treatment (UFP), microwave-assisted treatment (MFP) and ultrasonic/microwave-assisted treatment (UMFP), and then Fourier transform infrared (FT-IR) spectroscopy as well as 1H and 13C nuclear magnetic resonance (NMR) spectroscopy to characterize the structural properties of FP extracted. The molar weight (Mw), polydispersity index (Mw/Mn), root-mean-square (RMS) turning radius (Rg), molar mass distribution and chain conformation of FP were studied systematically using size-exclusion chromatography (SEC), multi-angle laser light-scattering (MALLS) and refractive index (RI). WFP, UFP, MFP and UMFP are all typical carbohydrates according to 1H NMR, laC NMR and FT-IR measurements. The type of glycosidic linkage is mainly a fl-glycosidic bond with a small amount of a-glycosidic bond. The results obtained by the SEC-MALLS-RI system showed the molar masses of WFP and UMFP were distributed mainly in the range of 5.0x10^6-1.0x10^7 g-mo1-1 and they accounted for 57.80% and 56.84% of total FP, respectively. The molar masses of UFP and MFP were distributed mainly in the 1.0x10^6-5.0x10^6 g.mol-1 range, which accounted for 38.24% and 52.39% of FP, respectively. WFP and UMFP in water were uniform spherical polymers; UFP and MFP were typical highly branched polymers and the degree of branching for MFP was higher compared to UFP. These results indicated the ultrasonic- and microwave-assisted extraction methods caused a significant decrease of the molar mass of FP but the ultrasonic/microwave synergistic extraction method had no effect.展开更多

The M. tuberculosis was incubated with various concentrations(50–800 mg/m L) of the extracts for 5 days in the REMA, and for 4 weeks in MGIT 960 system assay. Results: M. azedarach and L. chinensis extracts showed their anti-M. tuberculosis activity by strongly inhibiting the growth of M. tuberculosis in a concentration-dependent manner in the REMA and the MGIT 960 system assay. Particularly, the methanol extract of M. azedarach and n-hexane extract of L. chinensis consistently exhibited their effects by effectively inhibiting the growth of M. tuberculosis in MGIT 960 system for 4 weeks with a single-treatment, indicating higher anti-M. tuberculosis activity than other extracts, and their minimum inhibitory concentrations were measured as 400 mg/m L and 800 mg/m L, respectively. Conclusions: These results demonstrate that M. azedarach and L. chinensis extracts not only have unique anti-M. tuberculosis activity, but also induce the selective antiM. tuberculosis effects by consistently inhibiting or blocking the growth of M. tuberculosis through a new pharmacological action. Therefore, this study suggests the potential of them as effective candidate agents of next-generation for developing a new anti-tuberculosis drug, as well as the advantage for utilizing traditional medicinal plants as one of effective strategies against tuberculosis.展开更多

Merr. 70% ethanol extract with a cellular model of LPS-stimulated RAW264.7 cell. Methods RAW264.7 cells were treated with different concentrations of ethanol fraction (25,50 and 100 g · L^-1 ) and stimu- the conditioned media was collected and analyzed. The quantity of ni- lated with LPS (10 μg· L^-1) for 24 hours, tric oxide (NO) was assayed by Griess reagent. The production of inflammatory mediators was determined by en- zyme-linked immunosorbent assay (ELISA), such as prostaglandin E2 (PGE2), tumor necrosis factor ot (TNF-ot) interleukin- 1 β (IL-1 β) and interleukin 6 (IL-6) in cell supernatant. The concentrations of inflammatory medi- 9 ators were calculated according to the standard curves generated by each of the recombinant cytokines provided with LPS can induce RAW264.7 cells to promote the pro- the ELISA kits. Results Compared with the control group, TNF-α, IL-1β and IL-6. Compared with the duction of inflammatory mediators (P 〈 0.01 ) , including NO, PGE2, model group, ethanol fraction significantly suppressed LPS induced release of inflammatory mediators such as nitric NO, PGE2, TNF-α, IL-1β and IL-6 in a good dose dependent manner (P 〈 0.05, P 〈 0.01 ). Conclusions Eth- anol fraction could significantly inhibit the production of LPS-induced inflammatory response in RAW264.7 cells, and its anti-inflammatory effect may be related to reduce the production of inflammatory mediators NO, PGE2, TNF-α, IL-1 β and IL-6. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense (Lour.) Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases.展开更多

Therefore, this study suggests the potential of them as effective candidate agents of next-generation for developing a new anti-tuberculosis drug, as well as the advantage for utilizing traditional medicinal plants as one of effective strategies against tuberculosis.
文摘Aim To evaluate the anti-inflammatory effects of ethanol fraction prepared from Disporum cantoniense (Lour.) Merr. These results demonstrate that the ethanol fraction is the bioactive component of Disporum can- toniense (Lour.) Merr. , and the ethanol fraction will be further developed as a herbal remedy for preventive and/ or curative purposes in various inflammatory diseases.

文摘Objective:To systematically explore the effect and mechanism of melastomatis dodecandri herba(Melastoma dodecandrum Lour.)in the treatment of hepatitis based on network pharmacology.Method:We evaluated the hepatoprotective effects of M.dodecandrum in concanavalin A(Con A)-induced hepatitis in mice by assessing survival rate, histological analysis, serum transaminases, and related cytokines. Then the mechanism of action was predicted by a network pharmacology-based strategy.Based on the results,we measured the hepatic expression of related genes at mRNA level and proteins related to the phosphoinositide 3-kinase(PI3K)/protein kinase B(Akt)and nuclear factorkappa B(NF-кB)pathways.Results:Our study results clearly demonstrated that M.dodecandrum pretreatment significantly alleviated liver injury. This was demonstrated by an increase in survival rate, decreased severity of liver damage, and reduced serum transaminase levels compared with those in the Con A group.Moreover,M.dodecandrum significantly reduced the serum levels of tumor necrosis factor-a,interleukin-6,and interferon-g and increased the liver levels of superoxide dismutase, which indicated that M.dodecandrum exhibits anti-inflammatory and antioxidant activities. On the basis of network pharmacology,50 nodes were selected as major hubs based on their topological importance.Pathway enrichment analyses indicated that the putative targets of M.dodecandrum mostly participate in various pathways associated with the anti-inflammation response, which implies the underlying mechanism by which M.dodecandrum acts on hepatitis.Real-time fluorescent quantitative PCR analysis showed that M.dodecandrum downregulates the mRNA expression of interleukin-6,Toll-like receptor 7,interleukin-1 receptor-associated kinase-4,NF-кB and tumor necrosis factor-a in liver tissues.Western blotting showed that M.dodecandrum pretreatment protected against inflammation through activating the PI3K-Akt pathway by upregulating phosphorylated Akt(p-Akt)expression and suppressing NF-кB activation by inhibiting the phosphorylation of IKK,IkBa,and p65.Conclusion:The present work demonstrated the hepatoprotective effects of M.dodecandrum by regulating the PI3K/Akt and NF-кB pathways in Con A-induced mice, which provide insights into the treatment of hepatitis using M.dodecandrum.
