Acaricidal effect of the antimicrobial metabolite xenocoumacin 1 on spider mite control

The two-spotted spider mite,Tetranychus urticae Koch,is one of the most harmful pests in many agroecosystems worldwide.To effectively manage this pest,there is an urgent need to develop novel bio-active acaricides that support integrated pest management strategies targeting T.urticae.In this study,we explored the acaricidal effects of xenocoumacin 1 (Xcn1) on T.urticae and its predator Neoseiulus californicus using the highly puri?ed compound.Xcn1 was extracted and purified from the cell-free supernatant of the Xenorhabdus nematophila CB6 mutant constructed by the easy promoter activated compound identi?cation (easyPACId) method.When the concentration of Xcn1 exceeded 100μg mL~(–1),the survival rate of spider mite adults declined to below 40%and the fecundity was decreased by 80%at six days post-application.At concentrations of 25 and 50μg mL~(–1),Xcn1 signi?cantly impeded spider mite development by inhibiting the molt.However,neither concentration had any adverse effects on the survival or reproduction of the predatory mite N.californicus.The results from laboratory and semi-?eld experiments consistently demonstrated the effectiveness of the antimicrobial metabolite Xcn1 in controlling pest mites at both the molecular and physiological levels.Our study offers a promising possibility that combines the compatible biocontrol agents of Xcn1 and predatory mites for integrated pest mite control. 展开更多

为在烟草(Nicotiana tabacum)上进行IMP(inter-MITE polymorphism,MITE位点间多态性)标记开发,利用美国烟草基因组测序数据和MITE(miniature inverted repeat transposable element,微型反向重复转座元件)结构特征,通过生物信息学方法获... 为在烟草(Nicotiana tabacum)上进行IMP(inter-MITE polymorphism,MITE位点间多态性)标记开发,利用美国烟草基因组测序数据和MITE(miniature inverted repeat transposable element,微型反向重复转座元件)结构特征,通过生物信息学方法获得11 581组具有特异TIR(terminal inverted repeat,末端重复元件)序列的MITE及其多序列联配结果,并根据联配结果设计219条烟草IMP引物;利用9个不同类型烟草代表性品种进行测试,其中119条引物PCR扩增成功,引物开发成功率为54.6%;进一步对一个烤烟遗传群体亲本‘红花大金元’和‘HicksBroadleaf’进行标记筛选,共获得29个多态性标记,群体检测表明有21个标记可以整合定位到一个SSR标记遗传连锁图上.说明IMP标记在烟草中是一种可以利用的分子标记,但跟其他标记一样表现出较低的多态性,需高通量检测平台与之配套来提高其开发效率. 展开更多

Co-occurrence Patterns of Above-ground and Below-ground Mite Communities in Farmland of Sanjiang Plain, Northeast China 被引量:7

One of the fundamental questions in community ecology is whether communities are random or formed by deterministic mechanisms. Although many efforts have been made to verify non-randomness in community structure, little is known with regard to co-occurrence patterns in above-ground and below-ground communities. In this paper, we used a null model to test non-randomness in the structure of the above-ground and below-ground mite communities in farmland of the Sanjiang Plain, Northeast China. Then, we used four tests for non-randomness to recognize species pairs that would be demonstrated as significantly aggregated or segregated co-occurrences of the above-ground and below-ground mite communities. The pattern of the above-ground mite commu- nity was significantly non-random in October, suggesting species segregation and hence interspecific competition. Additionally, species co-occurrence patterns did not differ from randomness in the above-ground mite community in August or in below-ground mite com- munities in August and October. Only one significant species pair was detected in the above-ground mite community in August, while no significant species pairs were recognized in the above-ground mite community in October or in the below-ground mite communities in August and October. The results indicate that non-randomness and significant species pairs may not be the general rule in the above-ground and below-ground mite communities in farmland of the Sanjiang Plain at the fine scale. 展开更多

高油酸花生因其优质的油脂组成,越来越受到食品、油脂加工、种业等领域的青睐。然而,目前我国育成的高油酸品种遗传背景狭窄,拓宽遗传背景、开发相应的分子标记,有利于高效选育性状优异、适应性广的高油酸花生新品种。本研究针对具有F45... 高油酸花生因其优质的油脂组成,越来越受到食品、油脂加工、种业等领域的青睐。然而,目前我国育成的高油酸品种遗传背景狭窄,拓宽遗传背景、开发相应的分子标记,有利于高效选育性状优异、适应性广的高油酸花生新品种。本研究针对具有F458背景的花生AhFAD2B基因的MITE插入突变,开发了用以辅助鉴定真杂种及后代材料的MITE分子标记。该方法运用1轮常规PCR反应和普通琼脂糖凝胶电泳就可以区分出3种基因型,简便易行、成本低、稳定性好,显著提高了目标性状选择效率。同时,以耐盐性强、油食兼用型北方大花生品种宇花2号(YH2)做母本,高油酸花生潍花25号(WH25)作父本,配制杂交组合,利用系谱法逐代进行田间农艺性状筛选,并运用MITE标记辅助进行F1真杂种和后代单株基因型鉴别,获得了5个半直立型和1个直立型高油酸花生新品系。并且,对这些品系主要种子品质性状和耐盐性进行了调查分析。 展开更多

微型反向重复转座基因(minialure inverted repeat transposable elements,MITEs)的演化可能是造成枣树基因组及品种多样性的重要原因.为研究MITEs在枣树全基因组中的分布、种类及演化,使用MITE预测软件(MITEDigger)识别枣树全基因组中... 微型反向重复转座基因(minialure inverted repeat transposable elements,MITEs)的演化可能是造成枣树基因组及品种多样性的重要原因.为研究MITEs在枣树全基因组中的分布、种类及演化,使用MITE预测软件(MITEDigger)识别枣树全基因组中的MITEs序列,在植物MITEs数据库中进行分类注释,并用MEGA 6.0构建枣树基因组中MITEs的系统进化树.结果显示,MITE-Digger检测出MITEs总共3 230条,碱基数为1 104 679 bp,占基因组比例为0.34%;其中共有948条在植物MITEs数据库中比对到相似序列,这些MITEs可以归类到5个家族:h AT,PIF/Harbinger,CACTA,Mutator和Tc1/Mariner,MITEs系统进化树显示:枣树基因组中MITEs来源于少数几个共同祖先,在长期的进化中有明显的扩增和突变,这可能与枣树品种多样性有关.研究结果将为枣树的遗传资源及育种研究提供重要参考. 展开更多

实验以日本引进粳稻品种松前为受体材料,以水稻的近缘属菰为供体,利用花粉管通道法导入受体水稻菰的基因组DNA。经过两个世代的培养,T1世代表型(株形、穗形、籽粒特征)变异不明显,但统计分析结果表明生育期及有效分蘖数与母本有显著差... 实验以日本引进粳稻品种松前为受体材料,以水稻的近缘属菰为供体,利用花粉管通道法导入受体水稻菰的基因组DNA。经过两个世代的培养,T1世代表型(株形、穗形、籽粒特征)变异不明显,但统计分析结果表明生育期及有效分蘖数与母本有显著差异。在T2世代发现明显的表型变异株,并且此变异后代类型丰富。随机选取22个表型变异株进行Southern杂交和转座子分析,发现MITE类转座子mPing发生了转座激活。根据日本晴的基因组序列设计了含有mPing的特异位点引物以及用TAIL-PCR方法检测到了T2世代中mPing的跳出及插入频繁。实验获得了水稻的优良变异后代,扩大了遗传资源,丰富了生物多样性。研究结果为远缘杂交在基因组进化和作物遗传改良中的可能作用提供了新的理论依据。 展开更多

This paper deals with the validation and theoretical analyses of a simple method for calculating the intrinsic rate of increase, rm, of aphids and mites. This method does not require a detailed fecundity table data. The value of rm can be estimated by the simple equation: rm=0.74ln (Md)/d, where d is the time from birth to first reproduction; Md is the number of female offspring produced per original female in 2d. This method was developed by Wyatt and White. We reached the following conclusions from our study: When the parameter, Md is less than 1, this equation is not appropriate for estimating rm of populations; When the parameter, Md is larger than 1 and about 70% or more of the reproductive contribution to the rm is achieved in 2d, the equation is appropriate for calculatng the rm of aphids and mites.As Md decreases, the required reproductive contribution to the rm in 2d corresponding to the constant 0.74 will increase. However, whether 70% or more of reproductive contribution to rm is achieved in 2d still is a problem for whole taxa of aphids and mites. Therefore, further siudy on reproductive distribution will be required for practical application of this method. 展开更多

微型颠倒重复序列(miniature inverted repeat transposable elements,MITEs)是一类对基因组进化和基因表达有重要调节作用的转座子。为分析MITEs在毛竹基因组中的分布特性,借鉴Fast Isolation by AFLP of Sequences Containing repeats... 微型颠倒重复序列(miniature inverted repeat transposable elements,MITEs)是一类对基因组进化和基因表达有重要调节作用的转座子。为分析MITEs在毛竹基因组中的分布特性,借鉴Fast Isolation by AFLP of Sequences Containing repeats(FIASCO)方法,首次构建Stowaway-like MITEs富集文库。随机挑取21个克隆,测序发现2个含有Stowaway-like MITEs序列(Stow-Ph1和Stow-Ph2),阳性率为9.52%。Stow-Ph1和Stow-Ph2的长度分别为260和258bp,具有Stowaway-like MITEs典型的末端颠倒重复序列(terminal inverted repeats,TIRs)和靶位点重复序列(target site duplication,TSD)。Stow-Ph1和Stow-Ph2与水稻的Stow-Os8(FJ266024)的同源性分别为46.4%和52.6%。Stow-Ph1和Stow-Ph2与水稻16类Stowaway-like MITEs的TIRs前20bp的相似性均高于50%,表明毛竹的Stowaway-like MITEs在TIRs上是高度保守的。 展开更多

We found a 276-bp tourist-like MITE insertion in the 3′-UTR of a 16.9 k Da small heat shock protein gene(TaH SP16.9-3A) on chromosome 3A of common wheat. Haplotype analysis revealed two haplotypes, s HSP-W(wild type without MITE insertion) and s HSP-M(mutant with MITE insertion), present in wheat germplasm. Both semiquantitative PCR and quantitative real-time PCR analyses showed increased transcription levels of TaH SP16.9-3A in s HSP-M compared with those of s HSP-W after heat treatment at 42 °C. It appeared that the MITE insertion into the 3′-UTR enhances the transcription of TaH SP16.9-3A. 展开更多

Methods Seventy houses were enrolled for a mite indoor environment study. Dust samples were obtained from two sites of each patients’ house: bed and floor. Derp1 and Blot5 levels were quantified by a two-site monoclonal antibody-based ELISA technique. Results The levels of Derp1 allergens found in bed (geometric mean (GM) 3.43 μg/g of dust; 95%CI, 1.89-4.96 μg/g) and on the floor (GM 1.12 μg/g of dust; 95%CI, 0.71-1.53 μg/g) indicated significant differences (P = 0.005). However, the levels of Blot5 allergens found in bed (GM 19.00 μg/g of dust; 95%CI, 0.89-38.90 μg/g) and on the floor (GM 6.14 μg/g of dust; 95%CI, 0.40-11.90 μg/g) showed no statistically significant difference. In addition, in regards to the exposure index for Derp1 and Blot5 allergens found in bed and on the floor, 17.6% in bed and 8.6% on the floor had levels of Blot5 ≥ 10 μg/g of dust, higher than those obtained for Derp1 (7.2% and 0% in bed and on the floor respectively, P < 0.05); higher percentages in bed and on the floor (25.0% and 35.7%) were observed for levels of Blot5 = 0 μg/g of dust as compared with Derp1 in bed and on the floor (4.3% and 14.5% respectively, P < 0.05). Conclusions Derp1 and Blot5 are the major allergens found in this regional study, Blot5 is a more potent allergen in Hongkong, probably reflecting the high level of exposure to Blomia tropicalis (Bt). Bt and Dermatophagoides pteronyssinus(Dp) allergens should be included for precise diagnosis and effective immuno-therapeutic treatment of mite allergy in Hong-kong. 展开更多

The acaricidal activities of fourteen essential oils and fourteen of their major monoterpenoids were tested against house dust mites Dermatophagoides pteronyssinus. Five concentrations were used over two different time intervals 24 and 48 h under laboratory conditions. In general, it was noticed that the acaricidal effect based on LC50 of either essential oils or monoterpenoids against the mite was time dependant. The LCso values were decreased by increasing of exposure time. Clove, matrecary, chenopodium, rosemary, eucalyptus and caraway oils were shown to have high activity. As for the monoterpenoids, cinnamaldehyde and chlorothymol were found to be the most effective followed by citronellol. This study suggests the use of the essential oils and their major constituents as ecofriendly biodegradable agents for the control of house dust mite, D. pteronyssinus. 展开更多

It has been proposed that activation of Toll-like receptors (TLRs) plays crucial roles in the polarization of adaptive immune responses. A synthetic Toll-like receptor 2 (TLR2) ligand, Pam3CSK4, has been reported to modulate the balance of Th1/Th2 responses. We evaluated the modulation effect of Pam3CSK4 on allergic immune response in a mouse rhinitis model sensitized to house dust mite allergen (HDM). Mice were sensitized and challenged with Dermatophagoidesfarinae allergen (Der f), and then the allergic mice were treated by Pam3CSK4. Nasal allergic symptoms and eosinophils were scored. Der f-specific cytokine responses were examined in the splenocytes and bronchoalveolar lavage fluid (BALF). Serum level of total IgE was also detected. After establishing a mouse allergic rhinitis model with HDM, we have showed that Pam3CSK4 treatment not only ameliorated the nasal allergic symptoms remarkably but also decreased the eosinophils and total inflammation cells in BALF significantly. Analysis of cytokine profile found that IFN-7 released from either BALF or stimulated splenocytes increased markedly in Pam3CSK4-treated mice, while IL-13 decreased significantly. Moreover, serum level of total IgE was significantly lower in Pam3CSK4-treated mice than in the untreated. Thus, in an allergic rhinitis mouse model developed with HDM, Pam3CSK4 was shown to exhibit an antiallergic effect, indicating its potential application in allergic diseases. 展开更多

Objective:To examine the acaricidal effects of the essential oil of Cymbopogon citratits leaf extract(lemongrass)and ethanolic Azadirachta indica leaf extract(neem)against house dust mites Dermatophagoides farinae(D.farinae)and Dermalophagoides pteronyssinus(D.pteronyssinus).Methods:Twenty-five adults mites were placed onto treated filter paper that is soaked with plant extract and been tested at different concentrations(50.00%,25.00%,12.50%,6.25%and 3.13%)and exposure times(24hrs,48hrs,72hrs and 96 hrs).All treatments were replicated 7 times,and the experiment repeated once.The topical and contact activities of the two herbs were investigated.Results:Mortalities from lemongrass extract were higher than neem for both topical and contact activities.At 50%concentration,both 24 hrs topical and contact exposures to lemongrass resulted in more than 91%mortalities for both species of inites.At the same concentration and exposure time,neem resulted in topical mortalities of 40.3%and 15.7%against D.pteronyssinus and D.farinae respectively;contact mortalities were 8.0%and 8.9%against the 2 mites,respectively.There was no difference in topical mortalities of D.pteronyssinus from exposure to concentrations oflemongrass and neem up to 12.50%;lemongrass was more effective than neem at the higher concentrations.Conclusions:Generally,topical mortalities of D.farinae due to lemongrass are higher than that due to neem.Contact mortalities of lemongrass are always higher that neem against both species of mites. 展开更多

An experiment was conducted to investigate the prevalence of the red mite (Dermanyssus gallinae) in layer farms in four districts of northern West Bank, Palestine. The four districts were: Nablus, Tulkarm, Tubas, and Jenin. A total of 225 layer farms were investigated for infection with the parasite during the period from September 2009 to August 2010. The highest prevalence of the red mite was observed in the district of Tulkarm (18.3%, 25.5%, 32.7% and 47.3%, for the districts of Nablus, Jenin, Tubas, and Tulkarm, respectively) which can be explained by the hot humid climate in the district and insufficient cleaning. Infection with the red mite was highest in summer (43.8%) and lowest in winter (12%). The high infection rates observed in this study reflect the poor management and housing conditions, characteristic of layer farms in the study region. 展开更多
