卵子透明带保存精子在第二代试管婴儿中的应用研究

目的:探讨利用卵子透明带冷冻保存微量精子在第二代试管婴儿助孕中的应用效果。方法:回顾性分析2019年6月—2023年10月接受睾丸活组织检查取精助孕的40对夫妻,共计65个助孕周期的资料,根据患者助孕周期中精子来源分为A组(40个周期)和B组... 目的:探讨利用卵子透明带冷冻保存微量精子在第二代试管婴儿助孕中的应用效果。方法:回顾性分析2019年6月—2023年10月接受睾丸活组织检查取精助孕的40对夫妻,共计65个助孕周期的资料,根据患者助孕周期中精子来源分为A组(40个周期)和B组(25个周期)。A组采用女性取卵当日男方睾丸穿刺活检精子助孕,B组采用女性取卵当日利用卵子透明带冷冻保存的男性精子助孕(B组患者为上述经历了第一次新鲜睾丸精子助孕后,其中25对夫妻又开展了第2次助孕周期,第2次助孕周期时不再进行睾丸活组织检查取精,而是利用冷冻保存复苏后的精子助孕,共25个周期)。比较两组实验室各项指标[成熟卵母细胞百分率(MII卵率)、2原核胚胎受精百分率(2PN受精率)、2原核卵裂胚胎百分率(2PN卵裂率)、卵裂期优质胚胎百分率(D3优胚率)、囊胚形成率、临床妊娠率],25对接受两次助孕史的夫妻依据前后两次助孕的精子来源,分为新鲜睾丸精子和解冻精子两部分,比较其上述实验室各项指标的差异性。结果:两组MII卵率、2PN受精率、2PN卵裂率、D3优胚率、囊胚形成率、临床妊娠率比较,差异均无统计学意义(P>0.05);另外25对夫妻前后接受两种不同来源精子助孕的指标比较,差异也均无统计学意义(P>0.05)。结论:利用卵子透明带冷冻保存微量精子在无精症患者中能够在保证无新鲜睾丸穿刺精子的情况下得到类似的临床结局,同时可减少男性反复睾丸活组织检查的痛苦及创伤,该方法确保极少量精子在冷冻-复苏过程中不会丢失,此方法容易获取高质量解冻精子。 展开更多

外科模板结合Pick-up技术在无牙颌即刻负重种植修复中的临床应用 被引量:7

目的:探讨应用种植外科模板结合Pick-up技术进行无牙颌即刻负重种植修复,评估应用Pick-up技术使预先制作的修复体即刻负重的可行性及临床效果。方法:9例无牙颌患者采用带有预成修复体信息的种植外科模板引导种植体植入,而后应用预成的... 目的:探讨应用种植外科模板结合Pick-up技术进行无牙颌即刻负重种植修复,评估应用Pick-up技术使预先制作的修复体即刻负重的可行性及临床效果。方法:9例无牙颌患者采用带有预成修复体信息的种植外科模板引导种植体植入,而后应用预成的修复体结合Pick-up技术制作种植体支持的临时修复体,完成无牙颌即刻负重修复。随访临时修复体功能状况,总结Pick-up技术的就诊次数、被动就位情况、颌位关系、患者满意度。6个月后进行永久修复,定期随访观察种植体存留情况,种植体周围边缘骨吸收情况和种植体支持的修复体应用状况。结果:9例患者共植入56枚种植体,1颗种植体脱落,53颗种植体即刻负重,完成下颌6个、上颌3个种植体支持的树脂材料临时修复体。随访时间12-60个月(平均36个月),全部即刻负重的种植体稳定无松动,种植体边缘骨吸收0.5±0.24m m,即刻负重种植体存留率100%。全部种植体存留率98.2%,全部永久修复体均正常行使功能。利用种植外科模板通过Pick-up技术进行即刻负重修复治疗时间短,就诊次数少,一次获得被动就位,颌位关系准确,患者满意度高。结论:种植外科模板结合Pick-up技术进行种植体的即刻负重修复,可实现微创准确快速的种植治疗,临床效果满意。 展开更多

【目的】探明疣吻沙蚕成熟精子及卵子的超微结构特征,为其繁殖生物学的基础研究提供依据。【方法】利用扫描电镜和透射电镜观察疣吻沙蚕成熟精子及卵子的超微结构。【结果】疣吻沙蚕精子全长为(39.80±1.24)μm,由头部、中段、尾部... 【目的】探明疣吻沙蚕成熟精子及卵子的超微结构特征,为其繁殖生物学的基础研究提供依据。【方法】利用扫描电镜和透射电镜观察疣吻沙蚕成熟精子及卵子的超微结构。【结果】疣吻沙蚕精子全长为(39.80±1.24)μm,由头部、中段、尾部3个部分构成。精子头部主要由顶体和细胞核组成,顶体呈倒“U”形,细胞核为卵圆形。中段较粗,与头部紧紧相连,主要由中心粒和线粒体等构成。线粒体位于宽大的核膜中,围绕在远端中心粒周围,形状为卵圆形,透射电镜下可观测到5个线粒体。尾部为细长鞭毛,轴丝为典型“9+2”结构,外侧有波浪形侧鳍。成熟卵子呈卵圆形或近圆形,直径为(140.63±2.09)μm,透射电镜下可见明显的细胞核,核内有核仁,成熟的卵黄颗粒电子密度深且均匀,卵黄颗粒周围存在大量的脂滴。【结论】明确了疣吻沙蚕成熟精子及卵子的超微结构特征,为理解其生殖细胞发育、受精机制提供了重要的形态学基础。 展开更多

淘汰母梅花鹿再利用模式的初探:体内外胚胎的生产

试验旨在探索体内胚胎生产+活体采卵+离体取卵模式在淘汰母鹿再利用中的可行性。试验以4只淘汰母梅花鹿作为研究对象,先用羊用阴道内孕酮释放装置(CIDR)+促卵泡激素(FSH)+孕马血清促性腺激素(PMSG)方案进行超排,生产体内胚胎。冲胚结束... 试验旨在探索体内胚胎生产+活体采卵+离体取卵模式在淘汰母鹿再利用中的可行性。试验以4只淘汰母梅花鹿作为研究对象,先用羊用阴道内孕酮释放装置(CIDR)+促卵泡激素(FSH)+孕马血清促性腺激素(PMSG)方案进行超排,生产体内胚胎。冲胚结束时,对供体鹿两侧卵巢进行手术法吸卵,获取体内卵母细胞。最后随机选择2只母鹿屠宰获取体外卵母细胞,并利用所获卵母细胞研究DSOF液体系和SOF液体系在梅花鹿体外胚胎生产中的使用效果。结果显示,经超排处理后,所有供体母鹿均出现了发情表现,并有3只配种成功,平均冲胚2.7枚/只,回收率33.3%,平均获可移植胚0.3枚/只,可移植胚率12.5%。对供体鹿进行活体采卵共获卵母细胞6枚,回收率26.1%,仅1枚卵母细胞可用,可用率16.7%。对2只母鹿进行离体取卵共获可用卵丘-卵母细胞复合体(COCs)14枚,将该14枚COCs分别于DSOF液体系、SOFaa+E-DSOF和SOF液体系中受精与培养,卵裂率分别为22.2%(2/9)、50.0%(1/2)和66.7%(2/3)。仅SOF液体系中有一个受精卵发育到了8-cell阶段,其余所有卵裂胚均停滞在2-cell阶段。研究表明,体内胚胎生产+活体采卵+离体取卵模式可用于淘汰母鹿的再利用,但试验中使用技术的效率还有待进一步提高。SOF液体系可用于我国梅花鹿至少8-cell体外胚胎的生产。 展开更多

海参花是雌海参的性腺,在海参加工过程中常被作为下脚料,但它含有丰富的营养成分和多种活性因子,本研究检测了海参花的一般营养物质、元素含量以及氨基酸组成,同时,研究了海参花水提取物和乙醇提取物对人卵巢癌OVCAR-3细胞增殖以及对其... 海参花是雌海参的性腺,在海参加工过程中常被作为下脚料,但它含有丰富的营养成分和多种活性因子,本研究检测了海参花的一般营养物质、元素含量以及氨基酸组成,同时,研究了海参花水提取物和乙醇提取物对人卵巢癌OVCAR-3细胞增殖以及对其雌二醇分泌的影响。结果表明,海参花含水量为86.63%,粗蛋白、粗脂质和粗多糖(干重)占比分别为65.06%、14.82%和1.93%。共检测出17种氨基酸,总氨基酸含量为519.09 mg/kg。其中,必需氨基酸含量为197.04 mg/kg。海参花还含有多种对人体有益的元素。海参花提取物对人卵巢癌OVCAR-3细胞的增殖有明显的抑制作用,海参花水提取物、乙醇提取物均可提高细胞培养液上清液中雌二醇的含量,其中乙醇提取物可显著提高上清液中雌二醇的含量(P<0.01)。以上表明,海参花的营养丰富,其提取物能显著促进人卵巢癌OVCAR-3细胞雌二醇的分泌调节,因此具有很高的开发利用价值。 展开更多

There is a need to reduce the burden of child drop-off and pick-up for child-rearing generations, but most studies on the actual situation in Japan are based on survey results. In this study, we analyzed differences in child drop-off and pick-up by employment type and gender, utilizing the “Metropolitan Area Person Trip Survey,” which is a statistical data set. The study targeted households in which both spouses were between 30 and 49 years old, had children under the age of 6, and included the following three groups. 1) Dual-income Group 1 (both spouses employed/on contract/temporary);2) Dual-income Group 2 (husband employed/on contract/temporary, wife part-time);3) Full-time housewife group (husband employed, wife unemployed). The analysis revealed that a) wives are almost always responsible for dropping off and picking up their children;b) husbands drop off and pick up their children less frequently in dual-income households;and c) households with children raising within 10 to 30 km of Tokyo Station have longer commuting times and need to reduce the burden of dropping off and picking up their children. 展开更多

目的探讨辅助生殖技术中棕色卵子对妊娠结局的影响。方法回顾性分析2020年8月至2023年4月在昆明市第一人民医院接受辅助生殖助孕治疗的217例患者的临床资料,根据患者卵子透明带颜色将其分为棕色卵子组(n=69)和正常卵子组(n=148),比较两... 目的探讨辅助生殖技术中棕色卵子对妊娠结局的影响。方法回顾性分析2020年8月至2023年4月在昆明市第一人民医院接受辅助生殖助孕治疗的217例患者的临床资料,根据患者卵子透明带颜色将其分为棕色卵子组(n=69)和正常卵子组(n=148),比较两组患者的基础性激素[卵泡刺激素(FSH)、促黄体生成素(LH)、雌二醇(E2)、孕酮(P)、血清抗苗勒管激素(AMH)]水平、促性腺激素(Gn)使用天数、Gn总量以及获卵数、卵子成熟率(MⅡ)、受精率、卵裂率、D3优胚率、囊胚形成率、体外受精(IVF)/卵胞浆内单精子注射(ICSI)的正常受精率;比较两组患者的临床妊娠率、累积妊娠率、活产率及流产率;比较两组患者的颗粒细胞线粒体DNA含量及线粒体膜电位(MMP)水平。结果棕色卵子组患者AMH为(2.21±1.42)ng/mL,明显低于正常卵子组的(3.34±3.26)ng/mL,Gn用量为(3124±758.52)IU,明显高于正常卵子组的(2563±612.40)IU,差异均有统计学意义(P<0.05),但两组患者的平均年龄、体质量指数(BMI)、基础FSH、LH、E2、P水平及Gn的使用天数比较差异均无统计学意义(P>0.05);棕色卵子组患者的获卵数、D3优胚率、囊胚形成率分别为8.9±1.30、(34.2±12.20)%、(34.6±10.90)%,明显低于正常卵子组的11.2±1.01、(45.1±19.80)%、(48.4±11.80)%,差异均有统计学意义(P<0.05),但两组患者的MⅡ、IVF/ICSI的正常受精率、ICSI退化率、卵裂率比较差异均无统计学意义(P>0.05);棕色卵子组患者的临床妊娠率、累积妊娠率、活产率分别为28.99%、39.13%、21.74%,明显低于正常卵子组的53.38%、58.11%、39.86%,差异均有统计学意义(P<0.05);棕色卵子组患者的线粒体膜电位MMP平均灰度值为15.33±1.20,明显低于正常卵子组的22.33±0.33,差异有统计学意义(P<0.05),但两组患者的mtDNA拷贝数比较差异无统计学意义(P>0.05)。结论人类辅助生殖助孕中棕色卵子形成机制与Gn用量相关,其获卵数、D3优胚率、囊胚形成率均显著降低,膜电位也显著较低,线粒体功能异常,颗粒细胞受损,胚胎质量及发育受影响,因此,应避免移植棕色卵子。 展开更多

Currently, transplanting mechanisms for dryland plug seedlings in China are mainly semiautomatic and have low efficiency. The rotary seedling pick-up mechanism with a planetary gear train for non-uniform intermittent transmission, and a concave and convex locking arc device, has a large rigid impact. To solve these problems, according to the design requirements for a dryland plug seedling transplanting mechanism, a rotary seedling pick-up mechanism of a planetary gear train with combined non-circular gear transmission of incomplete eccentric circular and noncircular gears was proposed. This has the characteristics of two-times greater fluctuation of the transmission ratio in a cycle, and can achieve a non-uniform continuous drive. Through analysis of the working principle of the seedling pick-up mechanism, its kinematics model was established. The human–computer interaction optimization method and self-developed computer-aided analysis and optimization software were used to obtain a set of parameters that satisfy the operation requirements of the seedling pick-up mechanism. According to the optimized parameters, the structure of the seedling pick-up mechanism was designed, a virtual prototype of the mechanism was created, and a physical prototype was manufactured. A virtual motion simulation of the mechanism was performed, high-speed photographic kinematics tests were conducted, and the kinematic properties of the physical prototype were investigated, whereby the correctness of the theoretical model and the optimized design of the mechanism were verified. Further, laboratory seedling pick-up tests were conducted. The success ratio of seedling pick-up was 93.8% when the seedling pick-up efficiency of the mechanism was 60 plants per minute per row, indicating that the mechanism has a high efficiency and success ratio for seedling pick-up and can be applied to a dryland plug seedling transplanter. 展开更多

The male gamete undergoes continuous morphological and biochemical modifications during spermdevelopment in the testis (spermatogenesis), maturation in the epididymis, and capacitation in the female reproductivetract. Only the capacitated spermatozoa are able to recognize and bind to the bioactive glycan residue(s) on the ovum'sextracellular coat, the zona pellucida (ZP). Sperm-zona binding in the mouse and several other species is believed totake place in two stages. First, capacitated (acrosome-intact) spermatozoa loosely and reversibly adhere to the zona-in-tact ovum. In the second stage tight irreversible binding occurs. Both types of bindings are attributed to the presence ofglycan- binding proteins (receptors) on the sperm plasma membrane and their complementary bioactive glycan units(ligands) on the surface of the ZP. The carbohydrate-mediated adhesion event initiates a signal transduction cascade re-sulting in the exocytosis of acrosomal contents. This step is believed to be prerequisite which allows the hyperactivatedacrosome-reacted spermatozoa to penetrate the ZP and fertilize the ovum. This review focuses on the role of carbohy-drate residues in sperm-ovum interaction, and triggering of the acrosome reaction. I have attempted to discuss extensiveprogress that has been made to enhance our understanding of the well programmed multiple molecular events necessaryfor successful fertilization. This review will identify these events, and discuss the functional significance of carbohy-drates in these events. 展开更多

This article discusses the development of astronomical tables in ancient China based on the calendrical chapter titled“Lü-li zhi”律曆志(Monograph on harmonics and calendrical astronomy)in official histories.After surveying various types of astral scientific tables in ancient China and their layouts,this paper discusses the characteristics and adoption of“licheng”立成tables,a specific kind of pick-up table that seems to have come into use in the Sui period(581–618)and to have been widespread from the Tang period(618–907)onward.The emergence of licheng tables relates largely to the internal development of ancient Chinese astronomy,but they were also probably inspired by auspicial tables and foreign astronomical tables.By comparing tables recorded in the“Lü-li zhi”and the existing licheng tables,we find that most licheng were deleted during the compilation of“Lü-li zhi”to reduce the number of volumes.Moreover,this paper discusses several common solutions used to compress the size of tables in“Lü-li zhi.”The adoption of licheng tables into the ancient Chinese astral sciences and the reformatting of them in“Lü-li zhi”give us a different perspective for understanding the development of ancient Chinese astronomical tables and the compilation of the calendrical portion of official histories. 展开更多

Therefore,wavelet analysis based on wavelet entropy measure can signify the complexity of non-steady signal or system in both time and frequency domain.In this paper,in order to meet the requirements of post-analysis on abundant wavelet transform result data and the need of information mergence,the basic definition of wavelet entropy measure is proposed,corresponding algorithms of several wavelet entropies,such as wavelet average entropy,wavelet time-frequency entropy,wavelet distance entropy, etc.are put forward,and the physical meanings of these entropies are analyzed as well.The application principle of wavelet entropy measure in ElectroEncephaloGraphy (EEG) signal analysis,mechanical fault diagnosis,fault detection and classification in power system are analyzed.Finally,take the transmission line fault detection in power system for example,simulations in two different systems,a 10kV automatic blocking and continuous power transmission line and a 500kV Extra High Voltage (EHV) transmission line,are carried out,and the two methods,wavelet entropy and wavelet modulus maxima,are compared,the results show feasibility and application prospect of the six wavelet entro- pies. 展开更多

Since the furnace rollers and the strips contact frequently, alloy elements on the roller surface will form hard protuberance. We call this protuberance "pick-up". In addition, with the increase of the amount of dust in the furnace atmosphere, the occurrence of furnace roller pick-up will also increase. Through chemical reaction and physical accumulation analysis combined with practical experience, the causes of pick-up in different furnace sections were analyzed. Methods of preventing furnace roller pick-up in the high strength steel sheet processing line were discussed and solutions to improving the strip surface quality in the continous annealing furnace were provided. 展开更多
