Afforestation can increase C storage in terrestrial ecosystems, and has been recognized as an important measure to offset CO_2 emissions. In order to analyze the C benefits of planting wolfberry(Lycium barbarum L.) on the secondary saline lands in arid areas, we conducted a case study on the dynamics of biomass carbon(BC) storage and soil organic carbon(SOC) storage in different-aged wolfberry plantations(4-, 7-and 11-year-old) established on a secondary saline land as well as on the influence of wolfberry plantations on C storage in the plant-soil system in an arid irrigated area(Jingtai County) of Gansu Province, China. The C sequestration and its potential in the wolfberry plantations of Gansu Province were also evaluated. An intact secondary saline land was selected as control. Results show that wolfberry planting could decrease soil salinity, and increase BC, SOC and litter C storage of the secondary saline land significantly, especially in the first 4 years after planting. The aboveground and belowground BC storage values in the intact secondary saline land(control) accounted for only 1.0% and 1.2% of those in the wolfberry plantations, respectively. Compared to the intact secondary saline land, the SOC storage values in the 4-, 7-and 11-year-old wolfberry plantations increased by 36.4%, 37.3% and 43.3%, respectively, and the SOC storage in the wolfberry plantations occupied more than 92% of the ecosystem C storage. The average BC and SOC sequestration rates of the wolfberry plantations for the age group of 0–11 years were 0.73 and 3.30 Mg C/(hm^2·a), respectively. There were no significant difference in BC and SOC storage between the 7-year-old and 11-year-old wolfberry plantations, which may be due in part to the large amounts of C offtakes in new branches and fruits. In Gansu Province, the C storage in the wolfberry plantations has reached up to 3.574 Tg in 2013, and the C sequestration potential of the existing wolfberry plantations was 0.134 Tg C/a. These results indicate that wolfberry planting is an ideal agricultural model to restore the degraded saline lands and increase the C sequestration capacity of agricultural lands in arid areas.展开更多

METHODS: The effects of LWB on lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) and interleukin (IL)-6 secretion were assessed in a murine macrophage cell line. in vitro assessment also included characterizing the effects of LWB on the activation of NF-E2 related 2 pathway and inhibition of tumor necrosis factor-α (TNF-α)-induced nuclear factor-κB (NFκB) activation, utilizing reporter cell lines. Following the in vitro assessment, the anti-inflammatory efficacy of an oral intervention with LWB was tested in vivo using a preclinical model of intestinal inflammation. Multiple outcomes including body weight, intestinal histology, colonic cytokine levels and anti-oxidative measures were investigated.RESULTS: LWB reduced the LPS-mediated inductionof ROS production [+LPS vs 1% LWB + LPS, 1590 ± 188.5 relative luminescence units (RLU) vs 389 ± 5.9 RLU, P < 0.001]. LWB was more effective than wolfberry alone in reducing LPS-induced IL-6 secretion in vitro (wolfberry vs 0.5% LWB, 15% ± 7.8% vs 64% ± 5%, P < 0.001). In addition, LWB increased reporter gene expression via the anti-oxidant response element activation (wolfberry vs LWB, 73% ± 6.9% vs 148% ± 28.3%, P < 0.001) and inhibited the TNF-α-induced activation of the NF-κB pathway (milk vs LWB, 10% ± 6.7% vs 35% ± 3.3%, P < 0.05). Furthermore, oral supplementation with LWB resulted in a reduction of macroscopic (-LWB vs +LWB, 5.39 ± 0.61 vs 3.66 ± 0.59, P = 0.0445) and histological scores (-LWB vs +LWB, 5.44 ± 0.32 vs 3.66 ± 0.59, P = 0.0087) in colitic mice. These effects were associated with a significant decrease in levels of inflammatory cytokines such as IL-1β (-LWB vs +LWB, 570 ± 245 μg/L vs 89 ± 38 μg/L, P = 0.0106), keratinocyte-derived chemokine/growth regulated protein-α (-LWB vs +LWB, 184 ± 49 μg/Lvs 75 ± 20 μg/L,P = 0.0244), IL-6 (-LWBvs +LWB, 318 ± 99 μg/L vs 117 ± 18 μg/L, P = 0.0315) and other pro-inflammatory proteins such as cyclooxygenase-2 (-LWB vs +LWB, 0.95 ± 0.12 AU vs 0.36 ± 0.11 AU, P = 0.0036) and phosphorylated signal transducer and activator of transcription-3 (-LWB vs +LWB, 0.51 ± 0.15 AU vs 0.1 ± 0.04 AU, P = 0.057). Moreover, antioxidant biomarkers, including expression of gene encoding for the glutathione peroxidase, in the colon and the plasma anti-oxidant capacity were significantly increased by supplementation with LWB (-LWB vs +LWB, 1.2 ± 0.21 mmol/L vs 2.1 ± 0.19 mmol/L, P = 0.0095).CONCLUSION: These results demonstrate the antiinflammatory properties of LWB and suggest that the underlying mechanism is at least in part due to NF-κB inhibition and improved anti-oxidative capacity.展开更多

文摘Excessive amounts of nitrogen(N)fertilizers are applied during wolfberry production, resulting in some soil problems as well as potential environmental risks in the Qinghai-Tibet Plateau. In this study, organic fertilizers were used to replace part of the N fertilizer in wolfberry fields with different fertility levels.N fertilizer rates had 0,50,100,150,200,and 250 g N/plant.Organic fertilizer rates had 0,2,4,6,8,and 10 kg organic fertilizer/plant.The experimental treatments included 6 combinations of N0M10,N50M8,N100M6,N150M4,N200M2,and control was N250M0.The results showed that in the high-fertility soils, combinations of N150M4,N100M6 and N50M8 treatments were increased in yields, fruit shape index, flavonoid content, total phenol content, mineral nutrient content, and antioxidant activity of wolfberry fruits. Also they were improved in soil fertility and decreased in residual nitrate through the soil depth of 0-300 cm.In the soil with less fertility, fruit yield, amino acid contents, flavonoids, total phenols, mineral nutrients and antioxidant activity of fruits were increased by the N200M2,N150M4 and N100M6 treatments and soil fertility was improved as well. Also more residual nitrate was found in the depth of 0-100 cm of soil with both chemical and organic fertilizer compared with the control. Therefore,in the Qinghai-Tibet Plateau, combining decreased N fertilizer with organic fertilizer rather than chemical fertilizer alone could help farmers achieve satisfactory yields and quality of wolfberry fruits and reduce the risk of nitrate leaching. In conclusion,50-150 g/plant of N fertilizer combined with 4-8 kg/plant of organic fertilizer in high-fertility gardens and 100-200 g/plant of N fertilizer combined with 2-6 kg/plant of organic fertilizer in low-fertility gardens are recommended for wolfberry cultivation.

文摘Carbon(C) storage has received significant attention for its relevance to agricultural security and climate change. These results indicate that wolfberry planting is an ideal agricultural model to restore the degraded saline lands and increase the C sequestration capacity of agricultural lands in arid areas.

文摘AIM: To investigate the anti-inflammatory properties of Lacto-Wolfberry (LWB), bothin vitro and using a mouse model of experimental colitis. METHODS: The effects of LWB on lipopolysaccharide (LPS)-induced reactive oxygen species (ROS) and interleukin (IL)-6 secretion were assessed in a murine macrophage cell line. in vitro assessment also included characterizing the effects of LWB on the activation of NF-E2 related 2 pathway and inhibition of tumor necrosis factor-α (TNF-α)-induced nuclear factor-κB (NFκB) activation, utilizing reporter cell lines. Following the in vitro assessment, the anti-inflammatory efficacy of an oral intervention with LWB was tested in vivo using a preclinical model of intestinal inflammation. Multiple outcomes including body weight, intestinal histology, colonic cytokine levels and anti-oxidative measures were investigated.RESULTS: LWB reduced the LPS-mediated inductionof ROS production [+LPS vs 1% LWB + LPS, 1590 ± 188.5 relative luminescence units (RLU) vs 389 ± 5.9 RLU, P < 0.001]. LWB was more effective than wolfberry alone in reducing LPS-induced IL-6 secretion in vitro (wolfberry vs 0.5% LWB, 15% ± 7.8% vs 64% ± 5%, P < 0.001). In addition, LWB increased reporter gene expression via the anti-oxidant response element activation (wolfberry vs LWB, 73% ± 6.9% vs 148% ± 28.3%, P < 0.001) and inhibited the TNF-α-induced activation of the NF-κB pathway (milk vs LWB, 10% ± 6.7% vs 35% ± 3.3%, P < 0.05). Furthermore, oral supplementation with LWB resulted in a reduction of macroscopic (-LWB vs +LWB, 5.39 ± 0.61 vs 3.66 ± 0.59, P = 0.0445) and histological scores (-LWB vs +LWB, 5.44 ± 0.32 vs 3.66 ± 0.59, P = 0.0087) in colitic mice. These effects were associated with a significant decrease in levels of inflammatory cytokines such as IL-1β (-LWB vs +LWB, 570 ± 245 μg/L vs 89 ± 38 μg/L, P = 0.0106), keratinocyte-derived chemokine/growth regulated protein-α (-LWB vs +LWB, 184 ± 49 μg/Lvs 75 ± 20 μg/L,P = 0.0244), IL-6 (-LWBvs +LWB, 318 ± 99 μg/L vs 117 ± 18 μg/L, P = 0.0315) and other pro-inflammatory proteins such as cyclooxygenase-2 (-LWB vs +LWB, 0.95 ± 0.12 AU vs 0.36 ± 0.11 AU, P = 0.0036) and phosphorylated signal transducer and activator of transcription-3 (-LWB vs +LWB, 0.51 ± 0.15 AU vs 0.1 ± 0.04 AU, P = 0.057). Moreover, antioxidant biomarkers, including expression of gene encoding for the glutathione peroxidase, in the colon and the plasma anti-oxidant capacity were significantly increased by supplementation with LWB (-LWB vs +LWB, 1.2 ± 0.21 mmol/L vs 2.1 ± 0.19 mmol/L, P = 0.0095).CONCLUSION: These results demonstrate the antiinflammatory properties of LWB and suggest that the underlying mechanism is at least in part due to NF-κB inhibition and improved anti-oxidative capacity.

文摘In order to realize the efficient and high-quality mechanical picking for Chinese wolfberry, firstly, the forced reciprocating vibration picking principle of the Chinese wolfberry branch was studied, and the mechanical model of vibration picking was established based on the simplified cantilever model, and the response analysis and solution of all positions for the branch were carried out.At the same time, the critical mechanical model of fruit detachment under the condition of fruit hanging on branches was established, and the theoretical values of inertia force for each component of the branch were obtained.Secondly, through actual measurement and finite element modeling, the natural frequency and forced vibration response simulation for each component of the branch of Chinese wolfberry terminal branch model were both studied, and the relationship between single-point periodic excitation force and high-quality fruit shedding parameters was obtained.Thirdly, according to the conclusion of the picking model,a test bench with many groups of adjustable parameters was built. Finally, the last branch of fruit-hanging Chinese wolf berry for Ningqi No.1 was taken as the experimental object,a four-level orthogonal experiment was designed with three factors:frequency, amplitude and entrance angle.Meanwhile, the net picking rate, damage rate and false picking rate were taken as the evaluating indicators, referring to the comprehensive scores of the three factors. It was concluded that the primary and secondary relations of factors affecting the picking effect are frequency, amplitude and entrance angle, and the best operation parameters are frequency of 20 Hz,amplitude of 15 mm,and entrance angle of 45°,then,a hand-held vibration picker with setting parameters was trial-produced,and the optimal parameter combination was verified in the Chinese wolfberry planting base of the National Chinese wolfberry Engineering and Technology Research Center. The results showed that the net picking rate of ripe Chinese wolfberry was 96.13%,the damage rate of fruit was 1.13%,and the false picking rate was 3.23%,mechanized picking efficiency was 30.28 kg/h,which is 6.65 times that of manual picking. The experimental results are consistent with the simulation results. The research results can provide an important basis for the creation and operation standards of large-scale Chinese wolfberry vibration harvesting equipment.
