Methods: The amount of 3H-TdR (3H-thymidine) admixed in cultured rat VSMC was measured and mitogen-activated protein kinase (MAPK) activity and lipid peroxidation end product malondialdehyde (MDA) content of the VSMC were assayed. Results: 1×10?9, 1×10?8, 1×10?7 mol/L LPA in a concentration dependent manner, induced the amount of 3H-TdR admixed, MAP kinase activity, and MDA content of the cultured rat VSMC to increase. However, 5%, 10%, and 15% Yangxueqingnao serum preincubation resulted in a decrease of 23.0%, 42.0%, and 52.0% (P<0.01) respectively in the amount of 3H-TdR admixed, a decline in VSMC MAP kinase activity of 13.9% (P<0.05), 29.6% (P<0.01), and 48.9% (P<0.01) respectively, and also, a decrease in MDA content of VSMC of 19.4%, 24.7%, and 43.2% (P<0.01) respectively, in the 1×10?7 mol/L LPA-treated VSMC. Conclusions: LPA activates the proliferation and lipid peroxidation of VSMC in a concentration dependent manner. The LPA-induced VSMC proliferation is related to the activity of MAP kinases, enzymes involved in an intracellular signalling pathway. The results of the present study showed that Yangxueqingnao particles can effectively inhibit LPA-induced VSMC proliferation, MAP kinase activation, and reduce lipid peroxidative lesion.展开更多
目的 探讨养血清脑颗粒对肾性高血压大鼠 (Renovascularhypertension rat,RH R )的降压作用及对内皮素(Endothelin,ET)、一氧化氮 (Nitric O xide,N O )、降钙素基因相关肽 (Calcitonin gene related peptide,CG RP)等的影响。方法 采...目的 探讨养血清脑颗粒对肾性高血压大鼠 (Renovascularhypertension rat,RH R )的降压作用及对内皮素(Endothelin,ET)、一氧化氮 (Nitric O xide,N O )、降钙素基因相关肽 (Calcitonin gene related peptide,CG RP)等的影响。方法 采用改进的 Goldblatt肾性高血压大鼠模型。造模成功后大鼠共分 6组:模型组、养血清脑颗粒高、中、低 3个剂量组、卡托普利组、牛黄降压丸组,每组 10只,每日灌胃 1次,正常组和假手术组灌服等量蒸馏水。分别在第 1、2 4周的周五、六、日测量大鼠尾动脉收缩压。4周后取血,检测血中 ET、NO 、CGR P 的浓度。 、结果 RH R 模型大鼠血浆中 ET 含量增多,NO 、CG RP 含量降低,养血清脑颗粒各剂量降低血压同时能够降低 ET含量,增加 N O、CG RP 含量,以高剂量组为优 (P <0.05)。结论 养血清脑颗粒能够降低 R H R 血压,并对 ET、NO 、CGRP 等产生一定影响,这可能是降压作用的机制之一。展开更多
