Walp.), a staple legume crop in Sub-Saharan Africa, faces challenges due to biotic and abiotic constraints. Induced mutagenesis was deployed to create genetic variation in two cowpea varieties (KVX396-4-5-2D and Moussa local). The radio-sensitivity tests led to determe the lethal dose 50 (LD50) corresponding to 230 Gy and 220 Gy for KVX396-4-5-2D and Moussa local varieties, respectively. Dried seeds (M0) of each variety were gamma-ray irradiated with LD50 − 50, LD50 and LD50 + 50. M1 seeds were advanced to generate M2, M3 and M4 mutants using the single-seed-descent method. M4 mutant lines were evaluated in rain-fed conditions using a randomized complete block design to assess phenotypic differences. Data on seven qualitative and eleven quantitative traits were collected. The results indicated that the mutation induced variability in three qualitative traits: in KVX 396-4-5-2D mutant lines, with flower and seed color frequencies at 2.61% and 0.56% respectively, and pod dehiscence at a frequency of 0.24%. While in Moussa local mutants, a pod color changed at a frequency of 17%. ANOVA results revealed significant differences between mutants of both varieties for all quantitative traits, including photosynthetic parameters. Positive correlations were observed between leaf diameter and 100-seed weight, and between branch number and 100-seed weight. Hierarchical clustering revealed three clusters among KVX 396-4-5-2D mutants and six clusters among Moussa local mutants. Early maturity and high foliage were induced traits in Cluster 3 of KVX 396-4-5-2D mutants while high hundred-seed weight was induced in Cluster 6 of Moussa local mutants. 展开更多

Walp] holds a significant agricultural position in Uganda, ranking fourth among legume crops, following common beans, groundnuts, and soybeans. Known for its versatility, cowpeas are consumable at various developmental stages, from early seedling to maturity. However, the crop faces persistent pest challenges at each stage, leading to substantial yield losses. In Uganda, chemical insecticides are the primary pest control means, but their increased and excessive use raises environmental, health, and economic concerns. This has prompted a quest for alternative and sustainable solutions, prompting an exploration of botanical insecticides. This study, conducted at Makerere University Agricultural Research Institute (MUARIK), aimed to evaluate the effectiveness of three selected botanical insecticides versus four established chemical insecticides for managing cowpea insect pests under field conditions. The treatments included: Carbofuran, Cypermethrin 10% EC, Dimethoate, Pestwin, Pyrethrum ewc , Pyrethrum 5ew, Profenofos 40% Cypermethrin 4% EC mix, and Untreated, arranged in a randomized complete block design with three replications. The significant pests studied were aphids, thrips, pod-sucking bugs, and legume pod borer. Results indicated substantial impacts of the treatments on pest infestation, with Profenofos 40% Cypermethrin 4% EC being the most effective against most pests. The plant parameter, plant height, was significantly affected by treatments in 2016B, while the number of pods was impacted in 2017A. Pestwin, a botanical insecticide blend (containing Azadirachtin indica, Pongamia pinnata, and Ricinus communis extracts) demonstrated superior efficacy against cowpea aphids. Moreover, it positively influenced plant height, number of pods, and pod biomass, surpassing many chemical insecticides. Pestwin’s environmental friendliness positions it as a potential contributor to reducing environmental pollution, making it a promising candidate for inclusion in IPM programs. Overall, the study underscores the importance of exploring botanical alternatives to chemical insecticides for sustainable pest management in cowpea cultivation. 展开更多

Walp) is a multi-purpose legume with high quality protein for human consumption and livestock. The objective of this work was to develop near-infrared spectroscopy (NIRS) prediction models to estimate protein content in cowpea. A total of 116 cowpea breeding lines with a wide range of protein contents (19.28 % to 32.04%) were selected to build the model using whole seed and ground seed samples. Partial least-squares discriminant analysis (PLS-DA) regression technique with different pre-treatments (derivatives, standard normal variate, and multiplicative scatter correction) were carried out to develop the protein prediction model. Results showed: 1) spectral plots of both the whole seed and ground seed showed higher spectral scatter at higher wavelengths (>1450 nm), 2) data pre-processing affects prediction accuracy for bot whole seed and ground seed samples, 3) prediction using ground seed samples (0.64 R2 0.85) is better than the whole seed (0.33 R2 0.78), and 4) the data pre-processing second derivative with standard normal variate has the best prediction (R2_whole seed = 0.78, R2_ground seed = 0.85). The results will be of interest in cowpea breeding programs aimed at improving total seed protein content. 展开更多

Walp), particularly in response to insect infestation stress. Over a period spanning 2015 to 2017, forty accessions of cowpeas were evaluated to determine their variability under both insecticide spray and no insecticide spray conditions at the Teachings and Research Farms, Federal University of Agriculture, Abeokuta. The experimental design was a randomized complete block design in three replicates. The accessions were evaluated for plant height, leaf length, leaf width, number of days of 50% flowering, number of pods per plant, pod length, number of seeds per plant, 100-seed weight, and seed yield. Data collected were subjected to principal component and single linkage cluster analyses. Principal axis I (PCA1) accounted for 39% and 35% under insecticide spray and no insecticide spray respectively to the total variation in the accessions. Plant height with a factor score of 0.38, leaf length (0.41), number of leaves (0.37), and 100-seed, weight (0.30) was related to PCAI under insecticide spray while leaf width (0.32). Pod length (0.37) and number of seeds/plant (0.38) were significant to PCA1 under no insecticide spray. Notably, accessions such as SAMPEA6, SAMPEA10, IFE-Brown, and IFE-BPE exhibited consistent performance across both conditions, while others displayed condition-specific attributes. For instance, NGB1063, NGB1152, and NGB1093 demonstrated distinct traits under insecticide spray, while NGB1146 and NGB1124 exhibited notable characteristics under no insecticide spray conditions. Therefore, identifying these forty accessions with desirable traits hold promise for future genetic improvement efforts of cowpea cultivation in Nigeria and beyond. 展开更多

[Objectives]The paper was to screen new varieties of long cowpea that are suitable for autumn cultivation in Hunan,as well as to develop a comprehensive evaluation method to assess their adaptability and performance.[Methods]A total of 48 long cowpea varieties were introduced, and a range of comprehensive evaluation methods was employed to assess these varieties through the collection and analysis of field data.[Results]The square Euclidean distance of 14 allowed for the classification of all varieties into eight distinct groups.Groups II,III,and V belong to the autumn dominant group within this region, while groups I and VIII belong to the intermediate group.Additionally, groups IV,VI,and VII belong to the autumn inferior group in this area.Through a comparative analysis of various comprehensive evaluation methods,it was determined that the common factor comprehensive evaluation, grey correlation method, and fuzzy evaluation method were appropriate for application in the selection of long cowpea varieties.Furthermore, the evaluation outcomes were largely consistent with the cluster pedigree diagram.[Conclusions]Through comprehensive index method, ten varieties demonstrating superior performance in autumn cultivation have been identified, including C20,C42,C29,C40,C3,C14,C18,C25,C15,and C47.The selected varieties exhibit several advantageous traits, such as a reduced growth duration,a lower position of initial flower nodes,a decreased number of branches, predominantly green young pods, elongated pod strips, thicker pod structures,an increased number of pods per plant, and higher overall yields. These characteristics render them particularly valuable for extensive cultivation. 展开更多

[Walp.]) in one of the main grain legumes contributing to food security and poverty alleviation in Sub-Saharan Africa. To control the highly damaging legume pod borer Maruca vitrata F., transgenic cowpea lines expressing the insecticidal Cry1Ab Bt protein were developed. In this study, we evaluated the impact of Cry1Ab transgene expression on the susceptibility of four cowpea lines (named IT97K-T, IT98K-T, Gourgou-T and Nafi-T) and their respective non-transgenic near isogenic lines (IT97K, IT98K, Gourgou and Nafi) to Cowpea aphid-borne mosaic virus (CABMV) in greenhouse conditions. In a preliminary quality control test by enzyme-linked immunosorbent assay, the presence of Cry1Ab protein in transgenic seed lots ranged from 59% to 72%, with no significant differences among the lines (χ2 = 3.26;p = 0.35). Upon virus inoculation, all cowpea lines exhibited mosaic symptoms with similar severity between 7- and 11-day post-inoculation. No significant differences were observed in symptom severity. Significant differences were found between cowpea lines for time of symptom onset, virus accumulation in plants and days to 50% flowering. However, while comparing pairs of transgenic lines and corresponding non-transgenic lines, virus accumulation showed not significant differences whatever the pair. Time of symptom onset and days to 50% flowering did not also differ significantly between pairs of cowpea lines except Nafi/Nafi-T in which transgenic Nafi-T showed earlier symptoms (7.4 ± 0.7 vs. 8.9 ± 0.8 days post-inoculation) and shorter flowering time (37.3 ± 0.6 vs. 42 ± 1.7 days after sowing). Overall, these findings improve our understanding of the effects of Cry1Ab gene mediated genetic modification on cowpea infection by Cowpea aphid-borne mosaic virus, with potential implications for environmental safety assessment. 展开更多

Before the advent of cheap, synthetic fertilizers, legumes were commonly used as green manure crops for their ability to fix atmospheric nitrogen (N). A three-year study at Overton, TX examined legume integration into high-biomass sorghum (Sorghum bicolor L.) production systems on a Lilbert loamy fine sand recently cultivated after a fertilized bermudagrass [Cynodon dactylon (L.) Pers.] pasture. In this split-split plot design, ‘Dixie’ crimson clover (Trifolium incarnatum L.) and ‘Iron and Clay’ cowpea (Vigna unguiculata L.) were integrated into a high-biomass sorghum production system to evaluate impacts on N concentration, C concentration, and yield of high-biomass sorghum and their impacts on soil total N and soil organic carbon (SOC). Main plots were split into crimson clover green manure (CLGM) and winter fallow (FALL) followed by three sub-plots split into warm-season crop rotations: cowpea green manure (CPGM), cowpea-sorghum intercrop (CPSR), and sorghum monocrop (SORG). Three N fertilizer treatments (0, 45, 90 kg N∙ha−1) were randomized and applied as sub-sub plots. The CLGM increased (P sorghum biomass yield (16.5 t DM∙ha−1) 28% in year three but had no effect in the first two years. The CPSR treatment reduced sorghum yield up to 62% compared to SORG;whereas CPGM increased sorghum yield 56% and 18% the two years following cowpea incorporation. Rate of N fertilizer had no effect on sorghum biomass yield. Decrease in SOC and soil N over time indicated mineralization of organic N and may explain why no N fertilizer response was observed in sorghum biomass yield. Cowpea showed strong potential as a green manure crop but proved to be too competitive for successful intercropping in high-biomass sorghum production systems. 展开更多

When contributing to participatory research, farmers usually appreciate the performance of cowpea varieties using qualitative scores. The score they attribute to each variety are based on local knowledge. The specific criteria they individually use to attribute a score are not well described. The objectives of this work were to: 1) identify and describe exhaustively the local criteria used by farmers to measure the agronomic performance of cowpea;2) assess the variability and statistical structure of these farmer criteria across local contexts;3) and analyze the association between these farmer criteria and the classical agronomic measurement. To achieve these objectives, an augmented block design was implemented across fifteen locations in the regions of Maradi, Dosso and Tillabéri, representing a diversity of local contexts. From a set of 36 cowpea varieties, fifteen varieties were sown per location, including five varieties (controls) common to all locations. In each location, two replicates were sown in randomized Fisher’s blocks. After agronomic measurement and participatory evaluation (scoring of varieties by farmers), a group survey (focus group) was conducted in each location to identify the criteria considered by farmers to found their discretional scoring of varieties during the participatory evaluation. The analysis of the data identified, across locations, thirteen criteria defined by farmers to characterize the agronomic performance of cowpea. Some of these criteria were different according to location. Farmers ranked the three varieties with the best performance for each agronomical trait (Top 3 varieties). A comparison of the farmer ranking with the ranking based on agronomic measurements revealed similarity and complementary between both methods. This study highlighted the importance of considering both local and scientific knowledge in local varietal evaluations. 展开更多

Targeting of the synthesized polypeptide in the cells is an important research field in modern cell biology. Cowpea trypsin inhibitor (cpti) gene has been modified and a fusion protein gene (sck) was produced by fusing a signal peptide sequence at cpti 5' end and an endoplasm reticulum (ER) retention signal peptide at cpti3' end respectively. The signal peptide can direct the newly synthesized polypeptide into ER, while ER retention signal can make the protein retained in the ER and its derivative protein body. ELISA test indicated that the accumulation level of foreign CpTI protein in sck transgenic tobacco (Nicotiana tabacum L.) was two times higher than cpti transgenic tobaccos and some individuals were four times higher. At the same time, sck transgenic tobacco has a high resistance to Lepidoptera pest due to the increased accumulation level of foreign CpTI protein. The strategy of foreign protein targeting can be used to increase the accumulation level of foreign protein in transgenic plants and can be widely applied to other related research field in plant genetic engineering. 展开更多

This study aimed to investigate the effects of different precooling treatments(0 ℃ ice water, 4 ℃ cold water and 4 ℃ refrigerator) on the physiological characteristics of post-harvest cowpea. The results showed that precooling treatments could effectively maintain the sensory quality and slow the loss of nutrients of cowpea. Among the treatments, cold water precooling showed the best freshkeeping effect on post-harvest cowpea. It slowed the decline of sensory quality of post-harvest cowpea, inhibited the increase of weight loss rate, ethylene release amount, relative conductivity and malondialdehyde(MDA) content, and the degradation of total soluble solids(TSS), vitamin C, chlorophyll and soluble protein, and improved the activities of catalase(CAT) and peroxidase(POD). 展开更多

Both non-transgenic hybrid triploid poplars [ (Populus tomentosa×P.bolleana)×P.tomentosa ] and transgenic ones expressing cowpea trypsin inhibitor were cut at the base of the stem to produce auxoblasts, and used as source of leaves for insect feeding trials performed on 3 major insect species of poplar, the forest tent caterpillar ( Malacosoma disstria L.), gypsy moth ( Lymantria dispar L.) and willow moth ( Stilpnotia candida Staudinger). The height and basal diameter of trees were measured by the end of that year (2000). The results indicated that the growth elements of transgenic poplars were not interfered by the incorporation of the CpTI gene. Intriguingly, the height and basal diameter of the clone TG04 were much greater than that of the control. The transgenic foliage consumed by insects induced the increase of larval mortality, and decrease of larval wet weight gain, faecal output, pupal weight and egg deposition. Among them 3 transgenic clones, TG04, TG07 and TG71 received special attention for their outstanding insect resistance compared with other transgenic clones, which showed that the CpTI gene in them was expressed more actively and stably than in others. 展开更多

The impacts of climate change on crop yields are receiving renewed interest, with focus on cereals and staple crops at the regional and national scales.Yet,the impacts of climate change on the yields of leguminous crops in the local context has not been explored. Thus,an in-depth understanding of climate change in the local context may support the design of locally relevant adaptation responses to current and future climate risks. This study examined the impacts of climate variables(annual rainfall, annual average temperature, rainfall indices(rainfall onset, rainfall cessation, and the length of rainy days),and the number of dry days)on the yields of leguminous crops(groundnuts, cowpeas, and soybeans)in the Guinea Savanna agroecological zone of Ghana during the period of 1989-2020.The data were analysed using Mann-Kendall’s trend,Sen’s slope test, correlation analysis, and Multiple Regression Analysis(MRA).The findings revealed that annual rainfall, annual average temperature, rainfall onset, rainfall cessation, and the length of rainy days, and the number of dry days all showed varied impacts on the yields of groundnuts, cowpeas, and soybeans. The trend analysis detected a marginal decrease in the amount of rainfall, rainfall onset, and the number of dry days from 1989 to 2020(P>0.050).Annual average temperature and the length of rainy days substantially varied(P<0.050)from 1989 to 2020,showing an increasing trend. The findings also showed a marked upward trend for the yields of groundnuts, cowpeas, and soybeans during 2005-2020.The climate variables analysed above increased the yields of groundnuts, cowpeas, and soybeans by 49.0%,55.0%,and 69.0%,respectively.The yields of groundnuts, cowpeas, and soybeans fluctuated with the variability of 30.0%,28.0%,and 27.0%from 2005 to 2020,respectively.The three leguminous crops under study demonstrated unpredictable yields due to the variations of annual rainfall, annual average temperature, rainfall onset, rainfall cessation, the length of rainy days, and the number of dry days, which stressed the need for agricultural diversification, changing planting dates, using improved seed variety, and irrigation to respond to climate change. The results of this study implied that climate change considerably impacts crop production in the Guinea Savanna agroecological zone of Ghana, emphasizing the urgency of locally based and farmer-induced adaptation measures for food security and resilient agricultural systems. 展开更多

Information about the type of gene action governing the inheritance of cowpea seed flavonoid content and antioxidant activity is prerequisite for starting an effective breeding program for developing improved varieties. For this purpose, half-diallel crosses among seven diverse parents were made. The homozygous parents and 21 F1 hybrids were evaluated at Maroua in the Sudano-Sahelian zone of Cameroon using a randomized complete block design with three replicates. Flour samples produced from decorticated seeds were used for biochemical analysis. Analysis of variance showed significant differences(P < 0.001) among genotypes for the studied traits with ranges of 363.6–453.9 mg rutin equivalent per 100 g dry weight(DW) for total flavonoids, 13.38–30.73 mg ascorbic acid equivalent per 1 g DW for ferric iron reducing activity, 70.98–266.93 mg trolox equivalent per 100 g DW for 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity, and 90.93–370.62 mg trolox equivalent per 100 g DW for 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) free radical scavenging activity. Both additive and non-additive gene effects were significant in the genetic control of these traits, but dominance variance was greater than additive variance. The traits were mainly controlled by overdominance model suggesting a selection in the delayed generations.Broad- and narrow-sense heritability estimates varied from 0.90 to 0.99 and from 0.12 to 0.45,respectively. The variances due to both general and specific combining ability were highly significant for all studied traits. Recessive alleles had positive effects on DPPH and ABTS scavenging activities, whereas dominant alleles had positive effects on flavonoid content and ferric iron reducing activity. These results could help cowpea breeders to improve the antioxidant potential of cowpea seeds by appropriate selection. 展开更多

Walp.) seeds is important for their acceptability to consumers. The objective of this study was to evaluate the genetic control of soluble sugar content in cowpea using diallel analysis. For this purpose,half-diallel crosses were made among seven diverse parents. The amount of total soluble sugar was estimated by the phenol sulfuric acid reagent method and reducing sugar were measured by the DNS(3,5-dinitrosalicylic acid) reagent method. The total soluble sugar ranged from 11.12(VYA) to 40.79 mg g^(-1)(IT93 K-693-2) with a mean of 22.31 mg g^(-1).Reducing soluble sugar showed a mean value of 9.11 mg g^(-1) and ranged from 13.61(TVx3236) to 4.98 mg g^(-1)(Lori niebe). Diallel analysis showed that both additive and non-additive gene effects were significant in the genetic control of these traits. However, dominance variance was greater than additive variance for soluble sugar. The traits were controlled mainly by an overdominance model, suggesting the advantage of delaying selection to later generations. Soluble sugar content was found to be highly heritable, with broad-sense heritability ranging from 0.987 to 0.976. Narrow-sense heritability ranged from0.154 to 0.422 and revealed the lower importance of additive variance. Parents and F_1 hybrids differed significantly on the basis of their general and specific combining abilities, respectively. Recessive alleles had positive effects on total soluble sugar and reducing soluble sugar. These results could help cowpea breeders improve the acceptability of cowpea seeds and cowpea-based foods by appropriate selection. 展开更多

[Objectives ] The paper was to explore enzyme inhibition rate method for rapid detection of organophosphorus and carbamate pesticides in cowpea. [ Methods ] Acetylcholinesterase (ACHE) was added to cowpea extract, to determine the inhibition rate of extract against enzyme. The influences of different sampiing methods and sampling parts on detection results were compared. [ Results] The positive rate of standard sampling was 18.18% higher than that of non-stand- ard sampling, and the positive rate of samples collected from cowpea tail was 16.67% higher than that collected from other parts. [ Condmions] Enzyme inhibi- tion rate method is suitable for rapid detection of organophosphorus and carbamate pesticides in cowpea. 展开更多

This study determined the effects of genotype-by-environment(G × E) interaction and stability of yield among elite cowpea(Vigna unguiculata L.) selections derived by gamma irradiation. The study was conducted in Namibia at three selected sites: Bagani, Mannheim, and Omahenene, during 2014/2015 and 2015/2016. Thirty-four newly developed mutant genotypes and three local checks were evaluated using a randomized complete block design with three replications. Grain yield data were analyzed using the additive main effects and multiplicative interaction(AMMI) and the genotype main effect plus genotype-by-environment interaction(GGE) biplot methods. The AMMI and GGE biplot models explained 77.49% and 75.57% of total observed genotypic variation, respectively.Bagani and Omahenene were the environments best discriminating the test genotypes during 2014/2015 and 2015/2016, respectively. Four promising mutant genotypes: G9(Sh L3 P74), G10(Sh R3 P4), G12(Sh R9 P5), and G4(Sh L2 P4), showed wide adaptation and grain yields of 2.83, 2.06, 1.99, and 1.95 t ha^(-1), respectively. The novel mutant lines are useful genetic resources for production or future cowpea breeding programs in Namibia or similar environments. 展开更多

Symbiotic and phenotypic characteristics of thirty rhiobial isolates obtained from root nodules of two cowpea (Vigna unguiculata L. Walp) cultivars that grown in different sites of Fezzan (Southern part of Libya) were studied. Cultural characteristics and cross-nodulation with Arachis hypogega and Faidherbia albida showed that they were slow-growing rhizobia. Each isolate was found to coexist with non-symbiotic bacteria similar in their cultural characteristics to fast-growing rhizobia. All isolates formed symbiosis with the test plants, but different in their nitrogen-fixation efficiency. Numerical analysis of phenotypic characteristics showed that at boundary level of 70% average similarity, the isolates formed four distinguished groups and two isolates remained separate. Most isolates exhibited wide tolerance to acidity, alkalinity and extreme temperatures. They also resistant to some heavy metals such as mercury, copper, zinc, lead, cadmium and aluminum at low concentrations and antibiotics like polymyxin, colistin, bacitracin and nalidixic acid. Isolates displayed different response to salinity ranging from sensitive, which unable to grow in 1% NaCI to resistant and grow at 2% NaCl or above. Urea was hydrolyzed by most of them and carbohydrates utilizations were different. Sucrose and maltose were metabolized by most of the test isolates, whereas, monosaccharide and sugar alcohols were poorly utilized. 展开更多
