The treatment of fungi with DNA methyltransferase(DNMT)and/or histone deacetylase(HDAC)inhibitors is a promising way to activate secondary metabolite biosynthetic pathways that are dormant under normal conditions.In this study,we included an HDAC inhibitor,suberoylanilide hydroxamic acid(SBHA),in the culture medium of endophytic fungi isolated from the medicinal plant Datura stramonium L.The production of two compounds was induced in the culture supplemented with SBHA,and their structures were determined to be the fusaric acid derivatives 5-butyl-6-oxo-1,6-dihydropyridine-2-carboxylic acid and 5-(but-9-enyl)-6-oxo-1,6-dihydropyridine-2-carboxylic acid.The result confirmed that the use of chemical epigenetic modifiers is an effective technique for promoting the expression of silent biosynthetic pathways to produce unique secondary metabolites. 展开更多

The recent discovery of cancer cell plasticity,i.e.their ability to reprogram into cancer stem cells(CSCs)either naturally or under chemotherapy and/or radiotherapy,has changed,once again,the way we consider cancer treatment.If cancer stemness is a reversible epigenetic state rather than a genetic identity,opportunities will arise for therapeutic strategies that remodel epigenetic landscapes of CSCs.However,the systematic use of DNA methyltransferase and histone deacetylase inhibitors,alone or in combination,in advanced solid tumors including colorectal cancers,regardless of their molecular subtypes,does not seem to be the best strategy.In this review,we first summarize the knowledge researchers have gathered on the epigenetic signatures of CSCs with the difficulty of isolating rare populations of cells.We raise questions about the relevant use of currently available epigenetic inhibitors(epidrugs)while the expression of numerous cancer stem cell markers are often repressed by epigenetic mechanisms.These markers include the three cluster of differentiation CD133,CD44 and CD166 that have been extensively used for the isolation of colon CSCs.and.Finally,we describe current treatment strategies using epidrugs,and we hypothesize that,using correlation tools comparing associations of relevant CSC markers with chromatin modifier expression,we could identify better candidates for epienzyme targeting. 展开更多

Deflection modification factors of multistory buildings considering soil-structure interaction

In this investigation,attempts are made to examine the effects of soil-structure interaction on the deflection modification factor of multistory buildings and to clarify the role of deformable soil on this factor.A comprehensive study is conducted to investigate the effect of inertial interaction on displacement demand ratios in soil-structure systems with different structural characteristics and various non-dimensional parameters,using a simplified interacting model.A two-step regression analysis of the nonlinear,dynamic computer simulation results obtained,leads to a reliable and accurate formula for predicting deflection modification factors that incorporate foundation flexibility.The results illustrate that the application of deflection modification factors derived from fixed-base structures can lead to unsafe evaluation of the structures on soft soil.The proposed formula provides a useful tool for design engineers to incorporate the effect of soil-structure interaction into the evaluation of structural deformations. 展开更多

In order to look for the best proportion of β-tricalcium phosphate(β-TCP)and poly(lactideco-glycolide)(PLGA)we fabricated porous composites β-TCP/PLGA scaffold using freeze-drying method.Morphologicalcharacterization using scanning electron microscopy showed that the interconnected pore distribution was even and there was no significant difference with the increase of PLGA content.Moreover,the porosity,compressive strength and degradation in vitro were characterized.The fabricated scaffolds with increased PLGA in the composites β-TCP/PLGA scaffolds willget stronger mechanicalproperty and better appearance,furthermore,get suitable environment for cells.According to the evaluation indexes for the tissue engineering scaffold,the group of scaffold(β-TCP/PLGA=6:4)was selected to evaluate the induced celladhesion and proliferative ability of the scaffolds.Then as transplant embed into the bone criticaldefect sites on rats femur.The repairing processes of bone defect sites were characterized by X-ray analysis within 12 weeks.X-ray analysis showed that the bone defect sites alldisplayed the formation of callus obviously,In summary,our data suggest that the scaffold(β-TCP/PLGA=6:4)has a promising clinicalfuture in regeneration of bone criticaldefects. 展开更多

The surface of UF microcapsules was modifi ed by γ-(2,3-epoxypropoxy) propytrimethoxysilane(KH-560). The interfacial interactions between UF microcapsules and KH-560 were studied by Fourier transform infrared spectroscopy(FTIR) and X-ray photoelectron spectrometric analysis(XPS) of microcapsules. The surface topography of microcapsules was characterized by scanning electron microscopy(SEM). The thermal stability and mechanical properties were evaluated. FTIR and XPS results showed that there were physical and chemical combinations between the silicon coupling agent and the microcapsules surface. The thermal stability and mechanical property analysis showed that the addition of KH-560 could greatly improve the thermal stability, tensile property and elastic property. SEM results indicated that the addition of KH-560 could improve the bonding between the surface of microcapsules and resin matrix and improve the ability of self-healing. 展开更多

为了提升天然植物染料栀子黄在纤维素纤维上的染色性能,设计合成了一系列以3-氯-2-羟丙基为反应基团、季铵盐为阳离子基团的阳离子改性剂,通过亲核加成反应在纤维素纤维上引入季铵基团,提升栀子黄染料对阳离子改性纤维素纤维的染色性能... 为了提升天然植物染料栀子黄在纤维素纤维上的染色性能,设计合成了一系列以3-氯-2-羟丙基为反应基团、季铵盐为阳离子基团的阳离子改性剂,通过亲核加成反应在纤维素纤维上引入季铵基团,提升栀子黄染料对阳离子改性纤维素纤维的染色性能。借助电喷雾电离-串联质谱法(ESI-MS)、核磁共振氢谱(~1H NMR)对合成的不同烷基链长的季铵盐改性剂分子结构进行表征。通过红外光谱图和X射线光电子能谱图证实季铵盐化合物以醚键形式成功接枝在棉织物上。采用栀子黄染料对改性棉织物染色,比较不同烷基链长改性剂改性棉织物的染色K/S值发现,改性剂烷基链长为12时最佳,上染率为85.46%,K/S值为4.33。进一步研究染色工艺条件对染色织物K/S值、上染百分率以及色牢度的影响,结果表明,在染色温度为80℃,pH值为9,时间为60 min时,经3-氯-2-羟丙基二甲基辛烷氯化铵(CH-12)改性棉织物染色性能最佳,其上染率为88.07%,K/S值为4.71,耐皂洗牢度及耐摩擦色牢度均可达到3级以上。研究结果表明,CH-12改性棉织物可实现无盐染色,且改性织物的上染率、固色率、耐皂洗和耐摩擦牢度均优于未改性织物的传统有盐染色。 展开更多

混凝调理是一种成本低廉、操作方便的污泥调理方法.而传统无机盐混凝剂和合成有机高分子混凝剂在使用过程中存在金属离子及高毒性单体残留等问题.本文以自然界中来源广泛的一种可再生资源——木质素为原材,以丙烯酰胺和甲基丙烯酰氧乙... 混凝调理是一种成本低廉、操作方便的污泥调理方法.而传统无机盐混凝剂和合成有机高分子混凝剂在使用过程中存在金属离子及高毒性单体残留等问题.本文以自然界中来源广泛的一种可再生资源——木质素为原材,以丙烯酰胺和甲基丙烯酰氧乙基三甲基氯化铵为改性单体,采用接枝共聚技术,制得一系列电荷密度(CD)不同的阳离子改性木质素混凝剂(LN-CD).以南京仙林某市政污水处理厂活性污泥为研究对象,测定了活性污泥经系列LN-CD调理后的泥饼含水率(FCMC)、污泥比阻(SRF)、毛细吸水时间(CST)和压缩系数(s),系统考察了LN-CD污泥调理性能;结合污泥絮体结构、泥饼表面形貌和污泥中胞外聚合物(EPS)不同形态及成分含量与分布变化情况,详细讨论了LN-CD混凝剂的污泥脱水机理.研究结果表明,LN-CD具有良好的污泥调理效果,CD是影响LN-CD脱水性能的重要因素,CD越高污泥脱水效果越佳,其中经CD最高的LN-CD1调理后,FCMC、SRF、CST和s分别为:68.91%、0.89×10^(12)m·kg^(−1)、15.0 s及0.81;且获得的污泥絮体尺寸最大结构也最为紧密,污泥中不同形态EPS含量及蛋白质与多糖组分也均有效下降.这是由于LN-CD通过电中和及粘结架桥作用有效聚集污泥颗粒,对EPS结构造成破坏,释放出结合水;此外,木质素本身特有的芳环结构不仅可利用其刚性结构特征起到骨料支撑作用有利于改善泥饼可压缩性,增强其泥饼渗透性,同时其疏水性能还可通过疏水缔合作用与EPS中疏水片段结合有效压缩EPS,从而进一步提高其污泥脱水性能.综上所述,LN-CD制备简单,绿色环保,污泥调理性能优良,其在污泥脱水中应具有良好的应用前景. 展开更多

本研究利用阳离子改性纤维具有正电荷和高保水值的特点,与漂白木浆复合抄造制备加热不燃烧烟草薄片基片,以提升基片的吸收性能。结果表明,阳离子改性纤维的添加能有效降低基片的紧度,提升其对涂布液的吸收性能,增加加热不燃烧烟草薄片... 本研究利用阳离子改性纤维具有正电荷和高保水值的特点,与漂白木浆复合抄造制备加热不燃烧烟草薄片基片,以提升基片的吸收性能。结果表明,阳离子改性纤维的添加能有效降低基片的紧度,提升其对涂布液的吸收性能,增加加热不燃烧烟草薄片对涂布液的负载率。未添加阳离子改性纤维的加热不燃烧烟草薄片涂布负载率达121.69%,而阳离子改性纤维添加量为100%的加热不燃烧烟草薄片涂布负载率达215.58%,与前者相比增加了93.89个百分点。选择添加量为60%(相对于绝干木浆总质量)的阳离子改性纤维用于加热不燃烧烟草薄片基片制备,其基片紧度为0.45 g/cm^(3),孔隙率达70.90%,透气度17.92μm/(Pa·s),制备的加热不燃烧烟草薄片对涂布液的负载率达154.67%,并具有良好的吸食口感。 展开更多

HiTIP-seq profiles epigenomic reprogramming of patient-derived diffuse midline glioma stem cells to epigenetic therapy

Diffuse midline glioma(DMG),H3K27-altered,is lethal pediatric-type,high-grade,localized to the midline region of the central nervous system.Effective treatment guidelines are absent,and clinical trials are preferred for primary or recur-rent DMG patients.Recently,epigenetic agent-based immunotherapy has exhibited promising therapeutic effects in the clinical setting.However,the underlying mechanisms remain a mystery.The rare DMG tumor samples from biopsy or resection largely impede basic research,by using patient-derived tumor cells which better recapitulate the parental tu-mor’s heterogeneity compared to established cell lines.As an epigenetic reprogramming disease,DMG exhibits a global loss of H3K27 trimethylation(H3K27me3)and a gain of H3K27 acetylation(H3K27ac).Analysis of multiple epige-netic marks is fundamentally necessary.However,traditional techniques cannot allow ultra-low input and high-throughput.Herein we have developed a new method called high-throughput in situ tagged immunoprecipitation sequencing(HiTIP-seq),which uses an integrated superhydrophobic microwell array technology(InSMART).We were able to perform 100 parallel assays from as few as 100 cells per microwell on a single chip.We applied the tech-nology to profile epigenetic alterations of three-dimensional(3D)cell cultures derived from DMG patients.Our HiTIP-seq integrated with RNA sequencing(RNA-seq)analysis revealed that the combination of epigenetic agents(panobino-stat and tazemetostat),reprogrammed histone modifications and drove transcriptome changes.Among them,Wnt inhibitory factor 1(WIF1)has a gain of H3K27ac and a loss of H3K27me3,which leads to the upregulated expression.Altogether,HiTIP-seq is a versatile method for high-throughput analysis of histone modifications,suitable for both DMG research and studying rare 3D models. 展开更多

In order to improve the substrate diffusion properties and stability of an immobilized enzyme alginate microgels modified with TiO2 nanoparticles were employed as the enzyme immobilizing support.Ionotropic gelation was applied for the preparation of hybrid gels while Ca2+ Ce3+ Ni2+Cu2+and Fe3+were employed as the crosslinkers.Papain was selected as the model enzyme. UV-Vis spectroscopy was employed to investigate the activity of papain to evaluate kinetics and stability.Analysis results show that the highest affinity the lowest Michaelis-Menten constant Km =11.0 mg/mL and the highest stability are obtained when using Cu2+as the crosslinker.The effect of the mass ratio of TiO2 to papain on the stability and leakage of papain is also investigated and the results show that 10∶1 TiO2∶papain is optimal because the proper use of TiO2 can reduce enzyme leakage and ensure enzyme stability.Preparing Cu/alginate/TiO2 hybrid gels via ionotropic gelation can provide a satisfactory diffusion capability and enzyme stability. 展开更多

Applying conventional analytical methods,the following results have been found:the satura-tion value of acid--modified multifunction copolyest fibre( acid-modified polyester fibre)is1.74,then this kind of fibre is good for being dyed medium and light colours with cationicdyes;its level of erystalization is lower than of PET fibre;the third monomer(SIPM)con-tent in the fibre will be reduced by about 8% by the "alkali treatment"in dyeing and finishingprocess:through measuring and analysing the dye-up-take curves of time and temperature ofdyeing the fibre with cationic dyes,we indicate the key factors in dyeing are the 100℃ dyeingtemperature and the time for heat preservation;moveover,adding sodium sulphate will help leveldyeing but lower the dye-uptake. 展开更多

以阳离子改性淀粉作为絮凝剂,在天津某饮用水源地一蓝藻富集区进行原位处置,对比分析水体在投加淀粉前后pH值、透明度、总氮、总磷、浮游植物丰度及种群结构的变化。结果表明,阳离子改性淀粉能使水体表层藻类迅速絮凝成团下沉,水体透明... 以阳离子改性淀粉作为絮凝剂,在天津某饮用水源地一蓝藻富集区进行原位处置,对比分析水体在投加淀粉前后pH值、透明度、总氮、总磷、浮游植物丰度及种群结构的变化。结果表明,阳离子改性淀粉能使水体表层藻类迅速絮凝成团下沉,水体透明度显著提升,水中氮磷浓度明显降低。与对照区相比,试验区表层浮游植物的丰度显著下降,前6 d浮游植物去除率为86%~95%,整个试验周期平均去除率为78%,其中原优势种铜绿微囊藻下降最为突出,该絮凝剂对球状藻的去除和控制效果优于丝状藻。 展开更多
