The offspring of super-multiparous sows face problems such as decreased growth performance,poor meat quality and even diseases in animal husbandry.Gama-aminobutyric acid(GABA)has long been known to promote growth and ...The offspring of super-multiparous sows face problems such as decreased growth performance,poor meat quality and even diseases in animal husbandry.Gama-aminobutyric acid(GABA)has long been known to promote growth and suppress inflammation,but little is known about the mechanisms.A total of 72 growing-finishing pigs from the 8th generation were randomly allotted to 2 groups with 6 replicates per treatment to receive a cornesoybean basal diet or the basal diet supplemented 20 mg/kg GABA for 60 d.After the animal-trial period,samples of serum and liver were collected for further analysis.Additionally,a lipopolysaccharide(LPS)-induced inflammatory model using HepG2 cells was established to explore the role of GABA on regulating hepatic inflammation.The results indicated that inflammatory cell infiltration occurs in the liver of progeny of super-multiparous sows,and dietary supplementation with GABA influenced liver morphology,increased activities of antioxidant enzymes and decreased the expression abundance of pro-inflammatory cytokines,including tumor necrosis factor-α(TNFα)and interleukin(IL)-1β,in the liver of growing-finishing pigs(P<0.05).In addition,GABA supplementation increased mRNA expressions of peroxisome proliferator-activated receptor g(PPARg)and GABA receptors(GABARs),and reduced the expression of toll-like receptor 4(TLR4)/nuclear factor-kB(NF-kB)signaling(P<0.05).Additionally,an in vitro experiment demonstrated that GABA decreased the expressions of hepatic TLR4/NF-kB signaling via activating GABARs under LPS-stress(P<0.05).In summary,liver injury may affect the growth performance of growing-finishing pigs by changing hepatic mitochondrial metabolism,the expression of pro-inflammatory cytokines and TLR4/NF-kB pathway and that GABA supplementation has a restorative effect by acting on GABARs.展开更多

After the freeze slice of cerebrum was made,in situ hybridization was used to detect the mRNA of GABAA receptor subunits α1, β1, and γ2 in rat cerebral cortex, basal nuclei, substantia nigra and hippocampi. Image data were collected and analyzed quantitatively by QWin550CWmodel image signal gather and analysis system. RESULTS: In rats with hepatic encephalopathy, mRNA expression levels of GABAA receptor subunits α1, β1 increased significantly in basal nuclei, substantia nigra pars compacta, substantia nigra pars reticularis and hippocampi (144.7±15.67/184.14±4.41, 60.61±33.66/113.07±32.44,87.71± 21.25/128.40±18.85, 122.34±5.56/161.60±4.56,123.29±5.21/140.65±4.15, 123.40±4.42/140.09±4.52,124.76±4.18/140.09±4.12, 141.62±15.09/182.80 ±5.20,69.13±30.74/134.21±43.76, 87.87±25.16/151.01±19.49,122.14±6.30/162.33±3.92, 122.81±5.09/137.19±7.12,123.00±4.63/138.11±5.92, 125.75 ±2.43/138.81±6.10,P<0.01), but did not change in the cerebral cortex compared to the control group. Similar changes were found in the mRNA expression levels of GABAA receptor subunit γ2,which increased significantly in basal nuclei, substantia nigra pars compacta, substantia nigra pars reticularis (136.81±26.41/167.97±16.23, 51.00±36.14/113.18±36.52, 86.35±20.30/126.90±19.74, P<0.01), CA1 of hippocampal (162.15±9.05/178.62±6.45, P<0.05), and no changes were found in the cerebral cortex and CA2, CA3, CA4 of hippocampi.CONCLUSION: In rats with hepatic encephalopathy, mRNA expression levels of GABAA receptor subunits α1,β1, γ2 increase significantly in basal nuclei, substantia nigra and hippocampi, suggesting that the changes of mRNA expression levels in GABAA receptor subunits may contribute to the pathogenesis of hepatic encephalopathy.展开更多

Increasing Cu from 0.25 to 500 ?mol/L significantly enhanced levels of GABA and histidine (His), but considerably decreased levels of aspartate (Asp) and glutamate (Glu) in the leaves. The leaf Asp level negatively correlated with leaf Cu level, while leaf GABA level positively correlated with leaf Cu level. The GABA immunoreaction was first demonstrated by chromogen DAB, then the Gin immunoreaction was demonstrated by molybdic acid-TAB method. After being stabilized by DAB-cobalt chloride. the sections were processed for electron microscopic embedding. Under electron microscope, there were many Gin immunoreaction-positive neurons in the pyramidal layer of hippocampal CA1 area and some GABA immunoreaction-positive neurons with pyramidal or polygonal perikarya in the pyramidal, polymorphic and radiant layer of CA1 area. There were also symmetric dendro-axonic synapses formed by GABA-positive dendrites and Glu-positive a-cons in the polymorphic layer and symmetric axo-dendritic synapses formed by GABA-positive axons and Glu-positive dendrites in the radiant layer. In addition, there were symmetric autoregulatory axo-dendritic synapses be- tween Gin-positive axons and dendrites and autoregulatory axo-axonic synapses (both symmetric and asymmetric) between GABA-positive axons. Above mentioned results, for the first time, showed that there were complex synaptic regulatory relationships between excitatory Glu neurons and inhibitory GABA neurons in the hippocampal CA1 area, thereby, providing ultrastructural evidence for different neurotransmitters participating in epileptic pathogenesis.展开更多
