Preparation of Purified and Inactivated HP-PRRS Vaccine

[Objective] The aim of this study was to improve the purification and protective potency of HP-PRRS inactivated vaccine. [Method] HP-PRRS virus that had been multiplied inside Marc-145 cells was collected and concentrated 50 times and then inactivated. Complete virions were separated and collected by chromatography with Sepharose 4 Fast Flow. Oil adjuvant was added to prepare purified inactivated vaccine. [Result] Viral protein was separated from other proteins by purification and the viral protein contents ranged from 76.7% to 82.4%, and 96% of the expected serum proteins were removed. Protective potency of purified vaccine was above 4/5 and positive conversion rate of antibody was over 86%, both higher than that of unpurified vaccine. The differences were significant. [Conclusion] The experiment il-lustrated that the immune efficacy of vaccine can be enhanced through concentrat- ing and purifying, while the non-viral protein can be removed, so that allergic reaction and stress response cadsed by vaccine inoculation can be avoided. 展开更多

A fuzzy neural network (FNN) model is developed to predict the 4-CBA concentration of the oxidation unit in purified terephthalic acid process. Several technologies are used to deal with the process data before modeling.First,a set of preliminary input variables is selected according to prior knowledge and experience. Secondly,a method based on the maximum correlation coefficient is proposed to detect the dead time between the process variables and response variables. Finally, the fuzzy curve method is used to reduce the unimportant input variables.The simulation results based on industrial data show that the relative error range of the FNN model is narrower than that of the American Oil Company (AMOCO) model. Furthermore, the FNN model can predict the trend of the 4-CBA concentration more accurately. 展开更多

The modei is a semi-empirical structured into two series ideal continuously stirred tank reactor (CSTR) models. The optimal objectives include maximizing the yield or inlet rate and minimizing the concentration of 4-carboxy-benzaldhyde, which is the main undesirable intermediate product in the reaction process. The multi-objective optimization algorithra applied in this study is non-dominated sorting genetic algorithm Ⅱ (NSGA-Ⅱ). Decreasing the acetic acid consumption in purified terephthalic acid (PTA) solvent system has become a hot issue with common concern. In accordance with the technical features, the electrical conductivity is in direct proportion to the acetic acid content. General regression neural network (GRNN) is used to establish the model of electrical conductivity on the basis of mechanism analysis, and then particle swarm optimization (PSO) algorithm with the improvement of inertia weight and population diversity is proposed to regulate the operating conditions. Thus, the method of decreasing the acid loss is derived and applied to PTA solvent system in a chemical plant. Cases studies show that the precision of modeling and optimization are higher. The results also provide the optimal operating conditions, which decrease the cost and improve the profit. 展开更多

The biodegradation and toxicity of the purified terephthalic acid(PTA) processing wastewater was researched at NJYZ pilot with the fusant strain Fhhh in the carrier activated sludge process( CASP). Sludge loading rate(SLR) for Fhhh to COD of the wastewater was 1.09 d^-1 and to PTA in the wastewater was 0.29 d^-1. The results of bioassay at the pilot and calculation with software Ebis3 showed that the 48h-LC50 (median lethal concentration) to Daphnia magna for the PTA concentration in the wastewater was only 1/10 of that for the chemical PTA. There were .5 kinds of benzoate pollutants and their toxicities existing in the wastewater at least. The toxicity parameter value of the pure chemical PTA cannot be used to predicate the PTA wastewater toxicity. The toxicity of the NJYZ PTA wastewater will be discussed in detail in this paper. 展开更多

[Objectives]The antifungal protein HAS1 newly obtained was evaluated for acute toxicity safety in KM mice according to relevant national regulations,so as to eliminate people s concerns about the safety of transgenic plants.[Methods]The acute toxicity of the purified protein HAS1 was observed by intragastric administration of mice,and the poisoning symptoms,poisoning degree,recovery and death were observed.[Results]No abnormal toxicity symptoms were observed in the test group,the vehicle control group and the blank control group.The main tissues and organs were not abnormal in gross anatomy.The average body weight of each group showed an increasing trend compared with before administration at 1,3,7,11 and 14 d after administration.It was found that after giving the purified protein HAS1 to KM mice at a cumulative dose of 64 mg/kg a day,no obvious toxicity was observed in the acute toxicity test,indicating that the test substance was non-toxic by oral administration.[Conclusions]This study provides a basis for further use of the protein and its coding genes. 展开更多

The antifungal protein HAS1 is a new antifungal protein isolated from the genome of Bacillus subtilis HAS,which has a good inhibitory effect on various pathogenic fungi in sugarcane.This study aimed at evaluating the immunological transfer reaction of the test sample through repeated skin contact by observing whether the cavy skin repeatedly exposed to purified protein HAS1 suffers from allergic reaction and how strong is the allergic reaction.The results showed that the test group and the vehicle control group exhibited no allergic reaction in the skin immediately and at 24,48 and 72 h and had an mean reaction score and a sensitization rate both of 0,so the results of the naked eye observation were both nonallergenic to cavy skin;and in the positive control group,the cavies were observed to be highly sensitized immediately after removing the drug,mildly sensitized at 24 h,and not sensitized at 48 and 72 h,which meant the skin allergy was alleviated with the observation time.It is suggested that the purified protein HAS1 is negative for cavy skin anaphylaxis test (nonallergenic to cavy skin),which provides an experimental basis for further utilization of the protein and its coding gene. 展开更多

The current work seeks firstly the integration of a rainwater reuse system that will be of use in the watering of the green spaces, and secondly the re-use of purified water in the irrigation of agriculture spaces, which are in the nearby of wastewater treatment plant. The results show that 41% of the green spaces in the city of Fez require a renovation and installation of adequate watering systems taking into account the specificity of the city which is crossed by several rivers badly exploited for watering. The purified wastewater is of much better quality than the Moroccan reuse standards and is discharged with a flow that is large enough for the irrigation water requirements of the fields close to wastewater treatment plant. 展开更多

Recalcitrant warts can accurately be defined as warts that persist after six months of conventional therapy. Up to one-third of non-genital warts, especially periungual and plantar warts, become recalcitrant. Traditional treatment options for warts include topical salicylic acid, cryotherapy, and electrocautery;however, patients with recalcitrant warts remain a major therapeutic challenge. There is evidence that immunotherapy can clear recalcitrant warts if traditional treatment fails. Given this, clinical studies published in PubMed and Google Scholar that used Bacillus Calmette-Guerin (BCG), Mycobacterium Indicus Pranii (Mw vaccine), and purified protein derivative (PPD) as immunotherapy for wart, were reviewed in this study. Neither of these treatments has been subjected to a randomized controlled trial, thus to date, there are no standardized protocols to use them. Our review highlights the scientific facts in the clinical applications of the previous options to treat recalcitrant warts and investigate the differences among them, concerning efficacy, adverse effects, dosage, and route of administration. 展开更多

Emblica officinalis (E. oJficinalis) dried fruits were evaluated for its antitrypanosomal activity and cytotoxic effects. Vero cell line maintained in DMEM (Dubecco's Modified Eagle Medium) and incubated with Trypanosoma evansi for more than 12 h. MPE was added to the Vero cell culture medium at different concentrations (250-1,000 μg/mL) with trypanosomes concentration (1 × 106 trypanosomes/mL in each ELISA plate well) and incubated at appropriate conditions for 72 h. In-vitro cytotoxieity of MPE of E. officinalis was determined on Vero cells at concentrations ((1.56-100 ~tg/mL). Acute toxicity and in-vivo infectivity tests were done in mice. Obtained MPE ofE. officinalis underwent process of purification via column chromatography, preparative chromatography and HPLC (higher performance liquid chromatography) with bioassay at different strata on Alsever's medium. In-vivo assay for trypanocidal activity, MPE and PPFs (partially purified fractions) of E. officinalis with two sets of mice, each mouse was inoculated with 1 × 104/mL oftrypanosomes and treated (48 h post inoculation) at concentrations (12.5, 25, 50, 100 and 200 mg/kg body weight) were administered at dose rate of 100 [tL per mouse via intraperitoneal route (in treating parassitemic mice) to different groups of mice, 6 mice per concentration. HPLC of partially purified fractions ofE. officinalis was carried out with mobile phase ofacetonitdle: water (40:60) in gradient mode. In vitro, MPE induced immobilization and killing of the parasites in concentration-time dependent manner. Significant reduction of trypanosomes counts from concentration of 250μg/mL and complete killing of trypanosomes at 5th hour of observation, which was statistically equivalent to 4th hour of Diminazine Aceturate (Berenil), standard reference drug used. HPLC of the partially purified fractions revealed two major prominent peaks at retention time of 1-4 min. In vivo, both MPE and PPFs of test material did prolong lives of mice by 6-9 days but could not cure them. At concentration of 2,000 kg/kg body weight of MPE in acute test, all mice survived. For in-vivo infectivity test, mice injected with immobilized trypanosomes developed parasitemia and died while, the other group survived. MPE, PPFs and Diminazine Aceturate were toxic to Vero cells at all concentrations exception of 1.56, 1.56-3.13 and 1.56-6.25 μg/mL, respectively. From this report, PPFs ofE. officinalis dried fruits demonstrated potential pathway for a new development oftrypanocide in near future if additional investigations are put in place. 展开更多

The application potential of cellulosic materials in natural composites and other fields needs to be explored to develop innovative, sustainable, lightweight, functional biomass materials that are also environmentally friendly. This study investigated Typha angustifolia (Typha sp.) as a potential new raw material for extracting cellulose nanocrystals (CNCs) for application in wastewater treatment composites. Alkaline treatments and bleaching were used to remove cellulose from the stem fibres. The CNCs were then isolated from the recovered cellulose using acid hydrolysis. The study showed a few distinct functional groups (O-H, -C-H, =C-H and C-O, and C-O-C) in the Fourier Transform Infrared (FTIR) spectra. A scanning electron microscope (SEM) revealed the smooth surface of CPC and CNCs, which resulted from removing lignin and hemicellulose from powdered Typha angustifolia. Based on the crystalline index, the powdered Typha angustifolia, CPC, and CNCs were 42.86%, 66.94% and 77.41%. The loss of the amorphous section of the Typha sp. fibre resulted in a decrease in particle size. It may be inferred from the features of a Typha sp. CNC that CNCs may be employed as reinforcement in composites for wastewater treatment. 展开更多

Abstract Objectives To identify the effects of tuberculin purified protein derivative (PPD) sensitization on attenuating pulmonary T helper 2 (Th2) reaction and eosinophil infiltration in ovalbumin sensitized mice, and to search for the possibility of its clinical use in the management of asthma in a new way. Methods Sixty C57BL/6 mice were sensitized with PPD and then with ovalbumin and aluminum hydroxide, and randomized into 4 groups: ovalbumin (OVA), pre PPD, post PPD and control groups. Aerosol PPD were administered 3 h before or after ovalbumin challenge in the pre PPD and post PPD groups respectively, and control group received aerosol PPD only. IL 4, IL 5 expression was detected by immunocytochemistry in situ hybridization. Lung slides were stained with eosin and hemotoxylin, and pathological changes were observed. Results Ovalbumin aerosol inhalation caused a mixed inflammatory infiltration dominated by CD4 + T lymphocytes and eosinophils in the lung of sensitized mice. 87.5%-89.7% and 89.0%-89.2% of the CD4 + T lymphocytes were IL 4 mRNA + and IL 5 mRNA + respectively. 88.7%-91.2% of IL 4 mRNA + cells and 89.8%-90.6% of IL 5 mRNA + cells were CD4 + T lymphocytes in OVA group. Aerosol administration of PPD markedly suppressed IL 4 and IL 5 expression, and lung eosinophil infiltration. It was more effective in pre PPD group. 76.6%-78.0% of IL 4 mRNA + and 73.8%-79.7% of IL 5 mRNA + cells were CD4 + and 78.1%-84.9% and 78.4%-85.3% of the CD4 + cells were IL 4 mRNA + or IL 5 mRNA + respectively in pre PPD group, both were markedly lower than that of OVA group. CD4 + percentage of IL 4 mRNA + and IL 5 mRNA + cells were 80.7%- 82.0% and 78.0%-83.9% in post PPD group, which were markedly lower than that of OVA group. Conclusions Sensitization with PPD by intraperitoneal injection and then challenged by PPD inhalation markedly suppressed IL 4, IL 5 expression and eosinophil infiltration, and attenuated pulmonary Th2 reaction in ovalbumin sensitized mice. This induces Th1 type reaction and inhibits Th2 cell differentiation. It may be beneficial for glucocorticoids dependent or resistant patients. 展开更多

The antioxidant and antitumor effects as well as the immunomodulatory activities of crude and purified polyphenol extract from blueberries were investigated. The antioxidant and antitumor effects of the polyphenol extract were measured both in vitro and in vivo, and their effect on the immune systems of CD-1 tumor-bearing mice were also analyzed. In vitro assays demonstrated that blueberry purified polyphenol extract (BBPP) exhibited higher antioxidant activities than blueberry crude polyphenol extract (BBCP), but the opposite effect was observed in vivo. Both the in vitro and in vivo antitumor activity and the immunity assay showed that BBCP not only inhibited tumor growth, but also significantly improved the immunity of the mice. According to physical and histological studies, the CD-1 tumor-bearing mice treated with the polyphenol extract, especially high doses of BBCP experienced a higher quality of life than the positive control group (treated with cyclophosphamide). These results indicate that BBCP has significant antioxidant and antitumor activities and that it can enhance the immunity of CD-1 tumor-bearing mice. 展开更多

Non-dispersive solvent extraction (NDSE) with p-xylene as extractant was employed as a novel separation methodto recover both ρ-toluic (PT) acid and water from purified terephthalic acid (PTA) wastewater. The mass transport behavior ofPT acid from aqueous solution to ρ-xylene was investigated by experiments and numerical simulation. Experiments showed thatNDSE is feasible and effective. Residual PT acid in the raffinate can be reduced to lower than the permitted limit of wastewaterre-use (100 g/m^3) with extraction time longer than 60 s in industrial conditions. A mathematical model of PT acid mass transportwas developed to optimize the membrane module performance. The model was validated with the experimental results withrelative errors of less than 6%. Numerical analysis for mass transfer through the lumen side, the porous membrane layer, and theshell side showed that PT acid transport in the aqueous solution is the rate determining step. The effects of the membrane andoperating parameters on membrane module performance were investigated by means of computational simulations. The keyparameters suggested for industrial NDSE design are: fiber inner radius r1=200-250 μm, extraction time tc=50-60 s, aqueous/organic volumetric ratio a/o=9.0, and temperature T-318 K. 展开更多

Objective To investigate the difference in the outcome of IVF-ET between the patients using recombinant FSH versus highly purified urinary FSH for controlled ovarian stimulation (COH). Methods A comparative retrospective analysis was performed between the patients undergoing IVF using recombinant FSH (rFSH) 562 cycles (rFSH group) versus using highly purified urinary FSH(uFSH) 555 cycles (uFSH group) for COH from August 2009 to July 2010. Main outcome measures included: duration of stimulation, total amount of FSH, E2 level on hCG injection day, number of oocytes collected, average number of frozen embryos, and the rate of cancellation, implantation, clinical pregnancy and miscarriage rate. Results uFSH group required a significantly higher total amount of FSH and a longer duration of stimulation than thouse rFSH group (19.3 ± 7.6 vs 18.3 ± 7.0 ampoules, P=0.02, and 9.1 ± 1.6 d vs 8.9 ± 1.5 d, P=0.04, respectively). There was no statistically significant difference in the implantation rate, clinical pregnancy rate and miscarriage rate between uFSH and rFSH groups (22.57% vs 20.16%, 38.15% vs 35.18%, and 10.6% vs 13.45%, respectively, P〉0.05). The other measures including E2 level on hCG injection day, cancellation, number of oocytes collected, fertilization, average number of frozen embryos showed no difference between the two groups. Conclusion There is no significant difference in the outcome of IVF-ET between two groups in effects of recombinant FSH versus highly purified urinary FSH for COH. 展开更多
