Water transport at the root/soil interface of 1 year old Pinus sylvestris Linn. var. sylvestriformis (Takenouchi) Cheng et C. D. Chu seedlings under CO 2 doubling was studied by measuring soil electric conductance to survey soil water profiles and comparing it with root distribution surveyed by soil coring and root harvesting in Changbai Mountain in 1999. The results were: (1) The profiles of soil water content were adjusted by root activity. The water content of the soil layer with abundant roots was higher. (2) When CO 2 concentration was doubled, water transport was more active at the root/soil interface and the roots were distributed into deeper layer. It was shown in this work that the method of measuring electric conductance is an inexpensive, non_destructive and relatively sensitive way for underground water transport process. 展开更多

The gas_well system permanently installed in the soil was adopted for studying the dynamic relationship between CO 2 profiles and seedling root growth of Pinus sylvestris var. sylvestriformis (Takenouchi) Cheng et C. D. Chu. The study was conducted in the Open Research Station of Changbai Mountain Forest Ecological System, The Chinese Academy of Sciences from 1999 to 2001. Four treatments were arranged in the rectangular open_top chambers (OTCs): ambient CO 2+no_seedling, 700 μmol/mol CO 2+no_seedling, ambient CO 2 +seedlings, 700 μmol/mol CO 2+seedlings. By collecting and analyzing soil gas synchronously, it was found that the dynamics of CO 2 profiles were related to the biological activity of seedlings. There were more roots distributed in the top soil and the boundary layer across soil and sand, which made more contributions to the CO 2 profiles due to respiration root. Compared with the ambient CO 2, elevated CO 2 led to the peak of CO 2 concentration distribution shifted from soil surface layer to the boundary layer as seasonally growing of seedling roots. It is suggested the gas_well system is an inexpensive, non_destructive and relatively sensitive method for study of soil CO 2 concentration profiles. 展开更多

Four-year-old Pinus sylvestriformis were exposed for four growing seasons in open top chambers to ambient CO2 concentration (approx. 350 μmol·mol-1) and high CO2 concentrations (500 and 700 μmol·mol-1) at Research Station of Changbai Mountain Forest Ecosystems, Chinese Academy of Sciences at Antu Town, Jilin Province, China (42oN, 128oE). Stomatal response to elevated CO2 concentrations was examined by stomatal conductance (gs), ratio of intercellular to ambient CO2 concentration (ci/ca) and stomatal number. Reciprocal transfer experiments of stomatal conductance showed that stomatal conductance in high-[CO2]-grown plants increased in comparison with ambient-[CO2]-grown plants when measured at their respective growth CO2 concentration and at the same measurement CO2 concentration (except a reduction in 700 μmol·mol-1 CO2 grown plants compared with plants on unchambered field when measured at growth CO2 concentration and 350 μmol·mol-1CO2). High-[CO2]-grown plants exhibited lower ci/ca ratios than ambient-[CO2]-grown plants when measured at their respective growth CO2 concentration. However, ci/ca ratios increased for plants grown in high CO2 concentrations compared with control plants when measured at the same CO2 concentration. There was no significant difference in stomatal number per unit long needle between elevated and ambient CO2. However, elevated CO2 concentrations reduced the total stomatal number of whole needle by the decline of stomatal line and changed the allocation pattern of stomata between upper and lower surface of needle. 展开更多

The photosynthetic response of 12-year old Pinus sylvestriformis to elevated CO2 and its influential factors were tested and analyzed in the forest region of Changbai Mountain in 1999. Thees grown at the natural condition were controlled at three levels of CO2 concentration (350μL·L-1, 500 μL·L -1 and 700μL·L-1) by CO2 rich settlement designed by us. Net photosynthetic rates (NPR), temperature, relative humidity, stomatal conduc- tance, intercellular CO2 concentration and photosynthetic active radiation (PAR) were measured at 6:00, 8:00, 10:00, 14:00, 16:00 and 18:00 hours a day. Experimental results showed that the NPR of Pinus sylvestriformis increased by 32.6% and 123.0% at 500 μL·L-1 and 700 μL·L-1 CO2 concentration respectively, compared to am- bient atmospheric CO2 concentration (350 μL·L-1). The relations betWeen NPR and influential factors, including temperature, relative humidity, intercellular CO2 concentration and photosynthetic active radiation, were analyzed respectively by regression analysis at different CO2 concentrations. 展开更多

The results showed that doubling CO2 concentration was benefit tb seedling growth of the species (500 μL- L1 was better than 700 μLL’ L-1 ) and the biomass production was increased in both aboveground and underground parts of seedlings. Changbai pine (Pinus sylvestris var.Sylvestriformis) is an endemic and important tree species in Changbai Mountain. There were only 63 plant species in Changbai pine plantation, where hemicryptophyte was dominant(39.6... Changbai pine (Pinus sylvestris var.Sylvestriformis) is an endemic and important tree species in Changbai Mountain. There were only 63 plant species in Changbai pine plantation, where hemicryptophyte was dominant(39.68%). Simpson diversity index was 0.87, Shannon-Wiener diversity index was 2.96, and evenness index was 0.82. Community structure were divided into three layers: tree layer, shrub layer and herb layer. The total biomass and net production were 111.982 t/hm2 and 8942.80 kg/(hm2 a) respectively. The total biomass for tree, shrub and herb layers were 106.150, 2.230, 2.264 t/hm2, accounting for 94.79%, 1.99%, and 2.02%, respectively, and net production for those were 7465, 223, and 1182 kg/(hm2 a), accounting for 83.47%, 2.49%, and 13.22% of the total respectively. The nutrient content in various organs is in the order of needle> branch> root> bark> trunk, For the assimilated organ, the nutrient content is in the order of N> K> Ca> P> Mg, and that in absorption organ is in the order of Ca> N> K> P> Mg. For the whole plantation ecosystem, nutrient content is in the order of soil> litter> herb layer> shrub layer> tree layer. Nutrient storage and its accumulation rate in tree layer take up 88.79% and 76.43% of the total, respectively. 展开更多

The water use efficiency (WUE) of this species (11 -year old ) was studied on response to elevated Co, concentration at 500±μLL' L-1 by directly injecting CO2 into the canopy under natural condition in 1998-1999. The results showed that the elevated Co, concentration reduced averagely stomatal opening, stomatal conductance and stomatal density to 78%, 80% and 87% respectively, as compared to normal ambient. The elevated Co, reduced the transpiration and enhances the water use efficiency (WUE) of plant. 展开更多

The long-term effect of elevated CO2 concentrations on needle dark respiration of two coniferous spe- cies—Pinus koraiensis and Pinus sylvestriformis on the Changbai Mountain was investigated using open-top chambers. P. koraiensis and P. sylvestriformis were exposed to 700, 500 μmol·mol-1 CO2 and ambient CO2 (approx. 350 μmol·mol-1) for four growing seasons. Needle dark respiration was meas- ured during the second, third and fourth growing seasons’ exposure to elevated CO2. The results showed that needle dark respiration rate increased for P. koraiensis and P. sylvestriformis grown at elevated CO2 concentrations during the second growing season, could be attributed to the change of carbohydrate and/or nitrogen content of needles. Needle dark respiration of P. koraiensis was stimu- lated and that of P. sylvestriformis was inhibited by elevated CO2 concentrations during the third growing season. Different response of the two tree species to elevated CO2 mainly resulted from the difference in the growth rate. Elevated CO2 concentrations inhibited needle dark respiration of both P. koraiensis and P. sylvestriformis during the fourth growing season. There was consistent trend be- tween the short-term effect and the long-term effect of elevated CO2 on needle dark respiration in P. sylvestriformis during the third growing season by changing measurement CO2 concentrations. How- ever, the short-term effect was different from the long-term effect for P. koraiensis. Response of dark respiration of P. koraiensis and P. sylvestriformis to elevated CO2 concentrations was related to the treatment time of CO2 and the stage of growth and development of plant. The change of dark respiration for the two tree species was determined by the direct effect of CO2 and long-term acclimation. The prediction of the long-term response of needle dark respiration to elevated CO2 concentration based on the short-term response is in dispute. 展开更多

Microorganism DNA of rhizosphere soil from Pinus koraiensis and Pinus sylvestriformis were extracted by proteinase K based on SDS method, CTAB method, PVP (polyvinylpolypyrrolidone) method, and freezing and thawing method and the crude DNA from rhizosphere soil were purified by dialysis method, silver beads absorption method, and squeezing DNA gel method. The results of different extracting and purifying methods were compared and evaluated. Results indicated that the best method of extraction for microorganism DNA in rhizosphere soil was proteinse K based on SDS method with high salt concentration of 1.0% (w/v) NaCl, which could effectively eliminate humic acids and other impurities. The dialysis method was suitable to purify DNA from rhizosphere soil because of effectively removing brown matters and humic acids and the purified products were suited to PCR amplification. Squeezing DNA gel method was also a good purification method with the advantage of inexpensive in cost and efficient in use. 展开更多

To confirm a hybrid swarm population of Pinus densiflora × P. sylvestris in Jilin, China, we used needles and seeds from P. densiflora, P. sylvestris, and P. densiflora × P. sylvestris collected from natural stands or experimental stations to study whether shoot apex morphology of 4-year old seedlings can be correlated with the sequence of a chloroplast DNA simple sequence repeat marker (cpDNA SSRs). Total genomic DNA was extracted and subjected to sequence analysis of the pine cpDNA SSR marker Pt15169. Results show that morphological characters from 4-year old seedlings did not correlate with sequence variants of this marker. Marker haplotypes from all P. sylvestris trees had a CTAT element that was absent from all sampled P. densiflora trees. However, both haplotype classes involving this insertion/deletion element were found in a P. densiflora × P. sylvestris population and its seedling progeny. It was concluded that the P. densiflora × P. sylvestris accessions sampled from Jilin, China resulted from bi-directional crosses, as evidenced by both species’ cpDNA haplotypes within the hybrid swarm population. 展开更多
