[Method] Antibodies against infectious bursal disease virus in chicken serum were detected using test strips developed in our laboratory, and the results were comparad^with that using traditional agar diffusion method. [Result] The comparative study of the two methods showed that the sensitivity of test strips was eight times over agar gel immunodiffusion; test strips showed higher detection rate in the deter- mination test of 216 clinical samples, with high specificity, easy preservation, and simple and rapid operation, thereby being more suitable for the monitoring of clinical antibodies. [Conclusion] Test strips could replace the existing serological methods, having great promotion and application value in antibody monitoring.展开更多
Traditional transgenic detection methods require high test conditions and struggle to be both sensitive and efficient.In this study,a one-tube dual recombinase polymerase amplification(RPA)reaction system for CP4-EPSP...Traditional transgenic detection methods require high test conditions and struggle to be both sensitive and efficient.In this study,a one-tube dual recombinase polymerase amplification(RPA)reaction system for CP4-EPSPS and Cry1Ab/Ac was proposed and combined with a lateral flow immunochromatographic assay,named“Dual-RPA-LFD”,to visualize the dual detection of genetically modified(GM)crops.In which,the herbicide tolerance gene CP4-EPSPS and the insect resistance gene Cry1Ab/Ac were selected as targets taking into account the current status of the most widespread application of insect resistance and herbicide tolerance traits and their stacked traits.Gradient diluted plasmids,transgenic standards,and actual samples were used as templates to conduct sensitivity,specificity,and practicality assays,respectively.The constructed method achieved the visual detection of plasmid at levels as low as 100 copies,demonstrating its high sensitivity.In addition,good applicability to transgenic samples was observed,with no cross-interference between two test lines and no influence from other genes.In conclusion,this strategy achieved the expected purpose of simultaneous detection of the two popular targets in GM crops within 20 min at 37°C in a rapid,equipmentfree field manner,providing a new alternative for rapid screening for transgenic assays in the field.展开更多

I...Traditional methods for detecting lactoperoxidase (LP) are complex and time-consuming, so a test strip was made based on the enzymatic reaction principle to enable quick and convenient detection of LP in raw milk. In this study 0.1 mol/L citric acid (CA)/0.2 mol/L disodium hydrogen phosphate (NAP) buffer solution (pH 5.0), 22 mmol/L 3,3',5,5'-tetramethylbenzidine (TMB), 0.6 mmol/L hydrogen peroxide (H202), and 0.5% Tween-20 or 0.3% cetyltri- methyl ammonium bromide (CTAB) were optimal for preparing a quick, sensitive, and accurate LP test strip. The coefficient of variation (CV) of the estimated LP concentrations ranged from 2.47% to 6.72% and the minimum LP concentration detected by the test strip was 1-2 mg/L. Estimates of active LP in sixteen raw milk samples obtained using the test strip or the TMB method showed a good correlation (c=0.9776). So the test strip provides a quick, convenient, and accurate method for detecting the LP concentration of raw milk.展开更多

In this work,a very simple dual-readout lateral flow test strip(LFTS)platform was developed for sensitive detection of alkaline phosphatase(ALP)based on a portable device.In this assay,quantum dots(QDs)conjugated with...In this work,a very simple dual-readout lateral flow test strip(LFTS)platform was developed for sensitive detection of alkaline phosphatase(ALP)based on a portable device.In this assay,quantum dots(QDs)conjugated with bovine serum albumin(QDs-BSA)were chosen as fluorescence signal labels.In the absence of ALP,MnO_(2)nanosheets aggregate on the test line and exhibit an obvious brown color,which can be observed by naked eyes to realize semi-qualitative analysis.Meanwhile,fluorescence intensity of QDs-BSA can also be effectively quenched by MnO_(2)nanosheets due to inner-filter effect.Correspondingly,in the presence of ALP,ALP can catalyze the hydrolysis of ascorbic acid 2-phosphate(AAP)to generate L-ascorbic acid(AA),which can reduce MnO_(2)into Mn^(2+),accompanying with the obvious fluorescence recovery of the QDs.By simply monitoring the change of colorimetric and fluorescent signal on the test line,trace amount of ALP can be quantitatively detected.Under the optimal conditions,measurable evaluation of ALP was reached in a linear range from 1 U/L to 20 U/L with a detection limit of 0.7 U/L based on fluorescence signal.Furthermore,this colorimetric/fluorescent dual-readout assay was successfully applied to monitor ALP in human serum samples,showing its great potential as a point of care biosensor for clinical diagnosis.展开更多

The aim of the study was to develop a method for large-batch detection of PCV2 infection. Colloidal gold-labeled staphylococcal protein A (SPA) was sprayed on glass fibers to prepare a conjugate pad. The recombinant ORF2 protein of PCV2 was blotted on the test line of the nitrocellulose (NC) membrane, and pig IgG was streaked on the control line of the NC membrane. Then, the immunochromatographic strip was used for detection of antibodies against PCV2. The results show that the strip test was simple and the results could be determined within 10 min with naked eyes. The test strip was highly specific for pig serum against PCV2 and no cross-reaction with pig serum against other pathogens was observed. The test strip had close similarity with ELISA. Storage at RT for 6 months did not affect the specificity and sensitivity obviously. A total of 324 clinical pig sera were detected by both ELISA and the developed test strip, and the coincidence was 98.77%. Therefore, the developed immunochromatographic strip is specific, sensitive, stable, fast and simple, and it is suitable for on-site detection of antibodies against PCV2.展开更多
Objective: This study compared the performance of the immunochromatographic strip (ICS) to the Venereal Disease Research Laboratory (VDRL) test and Treponema pallidum haemagglutination assay (TPHA) at a primary...Objective: This study compared the performance of the immunochromatographic strip (ICS) to the Venereal Disease Research Laboratory (VDRL) test and Treponema pallidum haemagglutination assay (TPHA) at a primary health care setting. Methods: The study group was comprised of 150 females randomly drawn from a population of pregnant women attending their first antenatal visit or follow-up visits at West Maternity Hospital in Eldoret Kenya, but without a previous syphilis test during that pregnancy. On-site VDRL, ICS and TPHA tests were performed and immediate treatment provided where appropriate. The performance of the three tests was compared, Results: The sero-prevalence of syphilis as determined by the VDRL test was 3%. There was no significant difference between the ICS and the VDRL test (P 〉 0.05). The sensitivity and specificity of the ICS test were 80% and 98.6% respectively, while the negative predictive value (NPV) and positive predictive value (PPV) were both 100%. On the other hand, the sensitivity and specificity of the VDRL test were 66.7% and 99.3%, while the NPV and PPV were 80% and 98.6% respectively. The Treponema pallidum haemagglutination assay was used as a reference test and had sensitivity, specificity, NPV and PPV of 100%. Conclusion: The diagnostic accuracy of the ICS compared favorably with theVDRL gold standard. The use of the ICS in Kenya can improve the diagnosis of syphilis in health facilities both with and without laboratories and allow community health care workers to make a rapid diagnosis of the disease, and consequently make immediate therapeutic decisions.展开更多

A new experimental method of soil characterizati...One of the important issues for geotechnical engineers is the characterization of soil properties such as cohesion and internal friction angle by means of soil testing. A new experimental method of soil characterization based on the surface displacement of strip loaded soils is proposed. The theory to relate the soil deformation/displacement to soil strength properties is presented and compared with a series of conventional soil characterization techniques with direct shear tests. The proposed/developed strip loading tests provide reasonably accurate results compared with traditional direct shear tests. The new strip loading physical simulation and testing devices are helpful for understanding soil strength concepts and also provide an effective bridge connecting with engineering mechanics and foundation engineering courses instructions wherein derivation of bearing capacity theory equations is based on the same MohrCoulomb soil strength parameters. The advantages, limitations, and use of the strip loading modeling/testing technique in engineering education and further more in depth researches are discussed in the concluding remarks part.展开更多

Four different tool surfaces w...Current work is focused on the influence of friction in deep drawing process. Friction measurements were also conducted using a modified tribotester based on strip sliding between tools. Four different tool surfaces were tested under similar contact conditions regarding contact area, normal pressure, sliding speed, lubricant and surface characteristics to calculate the friction coefficient between the tool surface and a high strength low alloy steel sheet HSLA 380. The results showed that friction coefficient varies over a wide range with different lubricating conditions and different sliding velocities. For some sliding velocities, the coefficient of friction is stable and low, while for others it is unstable and higher. Results of the experiments reveal that this novel tribotester is a very useful tool to evaluate and compare the friction between steel sheet and tool surfaces in alloyed steel for cold working applications. The outcomes have only small dispersion within the different test series, which indicates a stable process with good repeatability. The test method enables comparison of different surface finishes and treatments, lubricants and coatings in terms of friction and galling under conditions similar to those found in sheet metal forming processes. The four different types of surfaces considered for this study were grinded, polished, nitrided and quenched/tempered. The main difference among the tested tools in this work was the surface roughness, which was found to have a strong influence on friction.展开更多
To investigate the seismic response of the steel-strip reinforced soil retaining wall with fullheight rigid facing in terms of the acceleration in the backfill, dynamic earth pressure in the backfill, the displacement...To investigate the seismic response of the steel-strip reinforced soil retaining wall with fullheight rigid facing in terms of the acceleration in the backfill, dynamic earth pressure in the backfill, the displacements on the facing and the dynamic reinforcement strain distribution under different peak acceleration, a large 1-g shaking table test was performed on a reduced-scale reinforced-earth retaining wall model. It was observed that the acceleration response in non-strip region is greater than that in potential fracture region which is similar with the stability region under small earthquake,while the acceleration response in potential fracture region is greater than that in stability region in middle-upper of the wall under moderately strong earthquakes. The potential failure model of the rigid wall is rotating around the wall toe. It also was discovered that the Fourier spectra produced by the inputting white noises after seismic wave presents double peaks, rather than original single peak, and the frequency of the second peak trends to increase with increasing the PGA(peak ground amplitude) of the excitation which is greater than 0.4 g. Additionally,the non-liner distribution of strip strain along the strips was observed, and the distribution trend was not constant in different row. Soil pressure peak value in stability region is larger than that in potential fracture region. The wall was effective under 0.1 g-0.3 g seismic wave according to the analyses of the facing displacement and relative density. Also, it was discovered that the potential failure surface is corresponds to that in design code, but the area is larger. The results from the study can provide guidance for a more rational design of reinforced earth retaining walls with full-height rigid facing in the earthquake zone.展开更多

Soil pressure peak value in stability region is larger than that in potential fracture region. The wall was effective under 0.1 g-0.3 g seismic wave according to the analyses of the facing displacement and relative density. Also, it was discovered that the potential failure surface is corresponds to that in design code, but the area is larger. The results from the study can provide guidance for a more rational design of reinforced earth retaining walls with full-height rigid facing in the earthquake zone.
文摘Friction spun core yarn has two components: filament core and staple fiber sheath. Under axial rubbing action, the failure mode of the core yarn is the stripping of the sheath from the core. This paper introduces a method to test the anti - stripping property of the core yarn. With a modified Universal Testing Machine, the stripping resistance of friction spun core yarn can be continuously measured. Some factors Influencing the measurements are discussed in detail. The testing results are compared with those from a Y731 Yarn Abrasion Tester and fur - ther confirmed by weaving practice.
