Purpose: This paper aims to explore the genetic correlation between osteosarcoma (OS) and Ewing’s sarcoma (EWS) by bioinformatics, and to find the common differentially expressed genes between the two in order to pro...Purpose: This paper aims to explore the genetic correlation between osteosarcoma (OS) and Ewing’s sarcoma (EWS) by bioinformatics, and to find the common differentially expressed genes between the two in order to provide reference for early clinical diagnosis. Method: The GEO gene chip public database in NCBI was used for data retrieval, and the chip data GSE17674 and GSE16088 were selected as the analysis objects. DEmRNAs were screened by R language limma kit, and the data were standardized. The common differentially expressed genes were screened by Venn diagram. The R language clusterProfiler package was used to perform GO function and KEGG pathway enrichment analysis on the common differentially expressed genes. The String database was used for PPI analysis, and the results were imported into Cytoscape software to obtain PPI interaction map, core module and Hub gene. Result: In this study, 1482 differentially expressed genes were screened from GSE17674 and 933 differentially expressed genes were screened from GSE16088. The Wayne diagram analyzed 335 common differentially expressed genes. GO/KEGG analysis suggested that the above common differentially expressed genes were mainly involved in cell cycle, ECM receptor interaction, sister chromatid separation, ossification, etc. Five core genes NCAPG, MAD2L1, CDK1, RRM2 and RFC4 were screened from the PPI network. The five genes were highly expressed in sarcoma. Conclusion: The five core common differentially expressed genes and related signaling pathways screened by bioinformatics analysis are helpful to understand the molecular mechanism of OS and ES pathogenesis, and are related to the prognosis of patients. They may become potential biomarkers for future research on OS comorbid ES, provide a basis for early diagnosis of OS combined with ES, and provide new ideas for clinical drug treatment research.展开更多

Means: The GSE17674 dataset was selected for analysis, obtained by data retrieval based on the GEO public database. The R language limma toolkit was used to screen DEmRNAs. After the data were normalized, the Metascape online analysis software and the R language clusterProfiler package were used to analyze the GO function and KEGG pathway enrichment of DEmRNAs lines, respectively. The string database was selected for PPI analysis, and the results were imported into Cytoscape software to derive the core modules and predicted core genes. The genes selected above were analyzed for tissue localization specificity. Results: Through the analysis of GSE17674, differentially expressed genes were screened out, and GO and KEGG analyses were performed on the differentially expressed genes. The GO functional enrichment analysis was mainly enriched in the process of muscle system, muscle contraction, myocyte development, contractile fibers, myogenic fibers, myofibers, myofibrillar segments, actin binding, structural composition of muscle, and actin filament binding. KEGG pathway analysis showed that the core pathways associated with the development of ES were the core genes for myocardial contraction, congestive cardiomyopathy, and hypertrophic cardiomyopathy. Five Hub genes were obtained based on Cytoscape prediction. Tissue localization specificity analysis of Hub genes was performed, and a total of 2 Hub genes with tissue specificity were screened;MYH6 was specifically expressed in cardiac cells and MYL1 was specifically expressed in skeletal muscle cells. Conclusions: The differential genes screened will help to understand the molecular mechanisms underlying the highly invasive and metastasis-prone biological characteristics of ES, as well as provide new ideas for clinical drug-targeted treatment of ES.展开更多

The spatial distributions of daintain in 66 breast tumor specimens were investigated with immuno-histochemistry method. Reverse transcription polymerase chain reaction (RT-PCR) and in situ hybridization inspection system were also used to detect daintain in 45 cases of malignant breast tumors. The final results show that 93% high positive responses to daintain on breast cancer tumors. RT-PCR demonstrated that, no smear of daintain transcripted in benign tissues was found, and light smear in peri-cancer tissue was observed. Distribution of daintain was distinguishable among benign tissues, hyperplasia tissues, immature hyperplasia and invasive breast cancer, which can be used to mark the progression of the malignant lesion development. We conclude that the expression of daintain is up-regulated in breast cancers, which indicates that the peptide is closely associated with the disease progression. So daintain could be used as the biomarker for detecting breast cancer.展开更多

Using the equation of motion for Keldysh nonequilibrium Green function, we study the spin accumulation and polarization for the system. Through analytical analysis and a few numerical examples, it is demonstrated that fairly large spin accumulation and polarization can be produced due to the breaking symmetry of the chemical potential for different electron spin in the leads. Moreover, the direction and the strength of the spin polarization can be conveniently controlled and tuned by varying the charge bias or the gate voltage.展开更多

OBJECTIVE: To analyze the pathways for IFNγ to promote the differentiation of primarily cultured cholinergic neurons in rat embryonic basal forebrain/septal nuclei through culture in different conditioned medium. DESIGN: A controlled experiment taking cells as the observational target. SETTINGS: Department of Biochemistry and Molecular Biology, Youjiang Medical College for Nationalities; Department of Cell Biology, Beijing University Health Science Center. MATERIALS: Sixty-four pregnant Wistar rats for 16 days (250-350 g) and 84 Wistar rats (either male or female, 5-7 g) of 0-1 day after birth were provided by the experimental animal department of Beijing University Health Science Center. Rat IFNγ were provided by Gibco Company; Glial fibrillary acidic protein by Huamei Company. METHODS: The experiments were carried out in the Department of Cell Biology, Beijing University Health Science Center and Daheng Image Company of Chinese Academy of Science from July 1995 to December 2002. ① Interventions: The nerve cells in the basal forebrain/septal nuclei of the pregnant Wistar rats for 16 days were primarily cultured, and then divided into four groups: Blank control group (not any supernatant and medium was added); Control group (added by mixed glial cell or astrocyte conditioned medium); IFNγ group (added by mixed glial cell or astrocyte conditioned medium+IFNγ). Antibody group (added by mixed glial cell or astrocyte conditioned medium+IFNγ+Ab-IFNγ). Mixed glial cell or astrocyte conditioned medium was prepared using cerebral cortex of Wistar rats of 0-1 day after birth. ② Evaluation: The immunohistochemical method was used to perform the choline acetyltransferase (ChAT) staining of cholinergic neurons. The ChAT positive cells were counted. MAIN OUTCOME MEASURES: Comparison of ChAT positive cells in rat basal forebrain and septal nuclei in different conditioned medium. RESULTS: ① ChAT positive cells in mixed glial cell conditioned medium: The ChAT positive cells in the IFNγ group and antibody group were significantly more than those in the control group (P 〈 0.01). ② ChAT positive cells in astrocyte conditioned medium: The ChAT positive cells in the IFNγ group were significantly more than those in the control group, but there was no significant difference between the antibody group and control group (P 〉 0.05). CONCLUSION: IFNγ cannot directly promote the differentiation of cholinergic neurons, but plays a role through activating glial cells (except astrocytes) to produce IFNγ like molecules.展开更多

[ Method] The fresh pig manure were as materials, and the effect of temperature (6 ℃, 12℃, 18 ℃, 24 ℃, 30 ℃) on fermentation start-up and period of biogas production were studied for 180 days of anaerobic fermentation. [ Result] The results showed that fermentation could be started in all temper- ature levels, the higher the temperature, the more biogas production, and the fermented period reduced with the temperature going up. The period was 7 months at low temperature ( below 20 ℃), 4 months at middle temperature (20 - 45 ℃). The aniseed was changed twice a year in February to March and July to August, which took 20 -30 days. It had the best biogas production period in 24 ℃ when the biogas production volume was high and stable. [ Conclusion] The best temperature for pig manure fermentation was 24 ℃.展开更多

The important effect of IFN-γ on the development and differentiation of neurons has been found by some scholars. OBJ EClIVE:To investigate whether IFN-γ has differentiational effect on cholinergic neurons in basal forebrain and septal nuclei, and make clear that the increased number of cholinergic neurons is resulted by cell differentiation or cell proliferation. DESIGN : Controlled observation trial SETTING: Department of Cell Biology, Medical School, Beijing University MATERIALS: Sixty-eight female Wistar rats at embryonic 16 days, weighing 250 to 350 g, were enrolled in this study, and they were provided by the Experimental Animal Center, Medical School, Beijing University. IFN-γ was the product of Gibco Company. METHODS: This study was carried out in the Department of Cell Biology, Medical School, Beijing University and Daheng Image Company of Chinese Academy of Sciences during September 1995 to December 2002. The female Wistar rats at embryonic 16 days were sacrificed, and their fetuses were taken out. Primary culture of the isolated basal forebrain and septal nuclei was performed. The cultured nerve cells were assigned into 3 groups: control group (nothing added), IFN-γ group(1×10^5 U/L interferon), IFN-γ+ IFN-γ antibody group (1 ×10^5 U/L IFN-γ + IFN-γ antibody). The specific marker enzyme (choline acetyl transferase) of cholinergic neuron was stained with immunohistochemical method. Choline acetyl transferase positive cells were counted, and ^14C-acetyl CoA was used as substrate to detect the activity of choline acetyl transferase, so as to reflect the differentiational effect of IFN-γ on cholinergic neuron in basal forebrain and septal nuclei. Flow cytometry was used to analyze cell circle and detect the proliferation of nerve cells. Nerve cells were marked with MAP2 and counted to evaluate the neuronal proliferation in basal forebrain and septal nuclei. MAIN OUTCOME MEASURES: Effect of interferon-γ on the number and activity of choline acetyl transferase-positive ceils in basal forebrain and septal nuclei as well as the effect on neuronal proliferation. RESULTS : ① Nerve cells in the basal forebrain and septal nuclei of IFN-γ group grew well compared with control group.②The differentiation of cholinergic neurons: The number and activity of choline acetyl transferase positive cells in IFN-γ group were significantly higher than those in the control group [(49.30 ±4.92) /100 cells vs (7.50±1.58) /100 cells; (2 049.00±12.30) min^-1 vs (1 227.30±12.59) min^-1, p 〈 0.01], while there was no significant difference in the number and activity of choline acetyl transferase positive cells between IFN-γ + IFN-γ antibody group and control group(P 〉 0.05). ③The proliferation of cholinergic neurons: Cell percentage was 17.2% and 19.8% at S-stage, 6.2% and 6.1% at G2+M stage in the control group and IFN-γ group respectively, without significant difference (P 〉 0.05). CONCLUSION : IFN-γ does not promote the neuronal proliferation in basal forebrain and septal nuclei, and the increased expression of cholinergic neurons is not resulted by the increase in the number of neurons, but its differentiation.展开更多

Sertoli cells are indispensable for guaranteeing normal spermatogenesis and male fertility.Although a huge number of long non-coding RNAs(lncRNAs)are identified from developing porcine testicular tissues and have been...Sertoli cells are indispensable for guaranteeing normal spermatogenesis and male fertility.Although a huge number of long non-coding RNAs(lncRNAs)are identified from developing porcine testicular tissues and have been predicted with crucial regulatory roles in spermatogenesis,their functions and regulatory mechanisms are still in infancy.Herein,we mainly explored the regulatory and functional roles of lncFPFSC in proliferation and apoptosis of immature porcine Sertoli cells.The results demonstrated that lncFPFSC was predominantly located in the cytoplasm of immature porcine Sertoli cells.lncFPFSC overexpression promoted cell cycle progression and cell proliferation,as well as inhibited cell apoptosis,whereas siRNA-induced lncFPFSC knockdown resulted in the opposite effects.Mechanistically,lncFPFSC acted as a sponge for miR-326.Overexpression of miR-326 inhibited cell proliferation and induced cell apoptosis,which further abolished the effects of lncFPFSC overexpression.The euchromatic histone-lysine N-methyltransferase 2(EHMT2)gene was directly targeted by miR-326,and its mRNA and protein expressions were both negatively regulated by miR-326 in immature porcine Sertoli cells.Then,siRNA-induced EHMT2 knockdown resulted a similar effect of miR-326 inhibition.Collectively,lncFPFSC promoted proliferation and inhibited apoptosis in immature porcine Sertoli cells through modulating the miR-326/EHMT2 axis.This study expanded our understanding of non-coding RNAs in participating porcine spermatogenesis through deciding the fate of Sertoli cells,and the competing endogenous RNA(ceRNA)network,and provided new molecular markers to treat Sertoli cell disorder inducing male infertility.展开更多

In the miscible two-component condensate, the formation of the Kármán vortex street is observed in one component, while ‘the half-quantum vortex street' is observed in the other component. Other patterns of vortex shedding, such as oblique vortex dipoles, V-shaped vortex pairs, irregular turbulence, and combined modes of various wakes, can also be found. The ratio of inter-vortex spacing in one row to the distance between vortex rows is approximately0.18, which is less than the stability condition 0.28 of classical fluid. The drag force acting on the obstacle potential is simulated. The parametric regions of Kármán vortex street and other vortex patterns are calculated. The range of Kármán vortex street is surrounded by the region of combined modes. In addition, spin–orbit coupling disrupts the symmetry of the system and the gain-loss affects the local particle distribution of the system, which leads to the local symmetry breaking of the system, and finally influences the stability of the Kármán vortex street. Finally, we propose an experimental protocol to realize the Kármán vortex street in a system.展开更多

Objective: To compare the effectiveness and safety of two surgical methods for lumbar degenerative diseases;the combination of the concept of accelerated rehabilitation with the assistance of Tianji Robotics and the c...Objective: To compare the effectiveness and safety of two surgical methods for lumbar degenerative diseases;the combination of the concept of accelerated rehabilitation with the assistance of Tianji Robotics and the concept of accelerated rehabilitation combined with manual pedicle screw placement assisted by conventional C-arm fluoroscopy. Methods: A retrospective analysis was performed on 70 patients who received the concept of accelerated rehabilitation combined with spinal surgery for lumbar degenerative diseases in Baise People’s Hospital from January 2022 to January 2024. Among them, 35 patients in the robot group received accelerated rehabilitation concept combined with robot-assisted surgery;In the conventional C-arm group, 35 patients received the accelerated rehabilitation concept combined with manual pedicle screw placement assisted by conventional C-arm fluoroscopy. VAS score (preoperative/postoperative), ODI score (preoperative/postoperative), intraoperative bleeding volume, postoperative hospital stay, postoperative complications and the accuracy rate of screw placement were compared between the two groups. Result: There was no statistically significant difference in preoperative VAS scores between the robot group and the conventional C-arm group (6.45 ± 0.82 VS 6.63 ± 0.81, P = 0.6600). The postoperative VAS score of the robot group was better than that of the conventional C-arm group (1.69 ± 0.80 VS 2.45 ± 0.85, P = 0.0000*). There was no statistically significant difference in preoperative ODI scores between the robot group and the conventional C-arm group (32.11 ± 3.18 VS 31.66 ± 2.25, P = 0.4900). The postoperative ODI score of the robot group was better than that of the conventional C-arm group (22.68 ± 1.94 VS 24.57 ± 2.25, P = 0.0000*). The postoperative complications in the robot group were less than those in the conventional C-arm group (2.7778% VS 28.5724%, P = 0.0030*). The intraoperative bleeding in the robot group was lower than that in the conventional C-arm group (320.85 ± 276.28 VS 490.00 ± 395.34, P = 0.0420*). The postoperative hospital stay of the robot group was shorter than that of the conventional C-arm group (10.00 ± 9.32 VS 14.49 ± 7.55, P = 0.0300*). The screw placement inaccuracy score of the robot group was lower than that of the conventional C-arm group (0.17 ± 0.51 VS 1.45 ± 1.46, P = 0.0000*). Conclusion: The combination of the concept of accelerated rehabilitation and Tianji Orthopedic robot-assisted surgery is more effective and safer in posterior lumbar decompression and internal fixation surgery with a screw rod system, and is worthy of promotion and application.展开更多
Background: Although socioeconomic support is recommended for frailtymanagement, its association with the prognosis of frailty is unclear.Methods: Using data from participants aged ≥65 years in the ChineseLongitudina...Background: Although socioeconomic support is recommended for frailtymanagement, its association with the prognosis of frailty is unclear.Methods: Using data from participants aged ≥65 years in the ChineseLongitudinal Healthy Longevity Survey (2008–2018), the associations betweensocioeconomic support (source of income, medical insurance, communitysupport, living status), onset of prefrailty/frailty, and worsening of prefrailty,were analyzed using multinominal logistic regression models. The associationsbetween self-reported low quality of life (QoL) and reversion of prefrailty/frailty were analyzed using multivariate logistic regression models. Associationswith mortality risk were analyzed using Cox proportional hazardregression models.Results: A total of 13,859 participants (mean age: 85.8 ± 11.1 years) containing2056 centenarians were included. Financial dependence was a risk factor for lowQoL among prefrail/frail individuals, but not among robust individuals. Havingcommercial or other insurance, and receiving social support from the communitywere protective factors for low QoL among prefrail/frail individuals and for theworsening of prefrailty. Continuing to work was a risk factor for low QoL, but aprotective factor for worsening of prefrailty. A negative association betweencontinuing to work and mortality existed in prefrail individuals aged <85 yearsand ≥85 years. Living alone was a risk factor for low QoL, but was notsignificantly associated with frailty prognosis.Conclusions: Prefrail and frail individuals were vulnerable to changes insocioeconomic support and more sensitive to it compared with robustindividuals. Preferential policies regarding financial support, social support,and medical insurance should be developed for individuals with frailty.展开更多
Objective:To investigate the effect of mir-3168 on the malignant transformation and cisplatin resistance of AGS and AGS/DDP gastric cancer cells,and to verify its target gene.Methods:The expression of mir-3168 in AGS ...Objective:To investigate the effect of mir-3168 on the malignant transformation and cisplatin resistance of AGS and AGS/DDP gastric cancer cells,and to verify its target gene.Methods:The expression of mir-3168 in AGS and AGS/DDP gastric cancer cells was detected by qPCR,and mir-3168 mimic,inhibitor and negative control were synthesized.They were transfected into AGS and AGS/DDP gastric cancer cells,respectively.The expression of mir-3168 and TP53 mRNA was detected by qPCR.Cell viability was detected by CCK8 under gradient cisplatin treatment and non treatment,apoptosis was detected by flow cytometry,cell invasion was detected by Transwell,and TP53 protein expression was detected by western blot,The database predicted the binding sites of mir-3168 and TP53.According to the binding sites,the double luciferase experiment was used to verify the binding of mir-3168 and TP53.Results:Compared with cisplatin sensitive gastric cancer cell AGS,mir-3168 was significantly overexpressed in cisplatin resistant gastric cancer cell AGS/DDP;mir-3168 mimic promotes cisplatin resistance,proliferation and invasion of AGS and AGS/DDP gastric cancer cells,and inhibits apoptosis of AGS and AGS/DDP gastric cancer cells;mir-3168 inhibitor inhibits cisplatin resistance,proliferation and invasion of AGS and AGS/DDP gastric cancer cells,and promotes apoptosis of AGS and AGS/DDP gastric cancer cells;mir-3168 mimic inhibits the expression of TP53 mRNA and protein,and mir-3168 inhibitor promotes the expression of TP53 mRNA and protein;Targetscan database predicted that there was a binding point between mir-3168 and TP53,and the double luciferase experiment suggested that mir-3168 was bound to TP53 through the predicted binding site.Conclusion:mir-3168 may promote the malignant transformation of AGS and AGS/DDP gastric cancer cells and cisplatin resistance by targeting TP53.展开更多
AIM: To investigate the inhibitory effects of emodin, baicalin, etc.on the hefA gene of multidrug resistance(MDR) in Helicobacter pylori(H.pylori).METHODS: The double dilution method was used to screen MDR H.pylori st...AIM: To investigate the inhibitory effects of emodin, baicalin, etc.on the hefA gene of multidrug resistance(MDR) in Helicobacter pylori(H.pylori).METHODS: The double dilution method was used to screen MDR H.pylori strains and determine the minimum inhibitory concentrations(MICs) of emodin, baicalin, schizandrin, berberine, clarithromycin, metronidazole, tetracycline, amoxicillin and levofloxacin against H.pylori strains.After the screened MDR stains were treated with emodin, baicalin, schizandrin or berberine at a 1/2 MIC concentration for 48 h, changes in MICs of amoxicillin, tetracycline, levofloxacin, metronidazole and clarithromycin were determined.MDR strains with reduced MICs of amoxicillin were selected to detect the hefA mR NA expression by realtime quantitative PCR.RESULTS: A total of four MDR H.pylori strains were screened.Treatment with emodin, baicalin, schizandrin and berberine significantly decreased the MICs of amoxicillin and tetracycline against some strains, decreased by 1 to 2 times, but did not significantly change the MICs of clarithromycin, levofloxacin, and metronidazole against MDR strains.In the majority of strains with reduced MICs of amoxicillin, hef A m RNA expression was decreased; one-way ANOVA(SPSS 12.0) used for comparative analysis, P < 0.05.CONCLUSION: Emodin, baicalin, schizandrin and berberine significantly decreased the MICs of amoxicillin and tetracycline against some H.pylori strains, possibly by mechanisms associated with decreasing hefA mR NA expression.展开更多

文摘Aiming at the triangular fuzzy(TF)multi-attribute decision making(MADM)problem with a preference for the distribution density of attribute(DDA),a decision making method with TF number two-dimensional density(TFTD)operator is proposed based on the density operator theory for the decision maker(DM).Firstly,a simple TF vector clustering method is proposed,which considers the feature of TF number and the geometric distance of vectors.Secondly,the least deviation sum of squares method is used in the program model to obtain the density weight vector.Then,two TFTD operators are defined,and the MADM method based on the TFTD operator is proposed.Finally,a numerical example is given to illustrate the superiority of this method,which can not only solve the TF MADM problem with a preference for the DDA but also help the DM make an overall comparison.
文摘Purpose: This paper aims to explore the genetic correlation between osteosarcoma (OS) and Ewing’s sarcoma (EWS) by bioinformatics, and to find the common differentially expressed genes between the two in order to provide reference for early clinical diagnosis. Method: The GEO gene chip public database in NCBI was used for data retrieval, and the chip data GSE17674 and GSE16088 were selected as the analysis objects. DEmRNAs were screened by R language limma kit, and the data were standardized. The common differentially expressed genes were screened by Venn diagram. The R language clusterProfiler package was used to perform GO function and KEGG pathway enrichment analysis on the common differentially expressed genes. The String database was used for PPI analysis, and the results were imported into Cytoscape software to obtain PPI interaction map, core module and Hub gene. Result: In this study, 1482 differentially expressed genes were screened from GSE17674 and 933 differentially expressed genes were screened from GSE16088. The Wayne diagram analyzed 335 common differentially expressed genes. GO/KEGG analysis suggested that the above common differentially expressed genes were mainly involved in cell cycle, ECM receptor interaction, sister chromatid separation, ossification, etc. Five core genes NCAPG, MAD2L1, CDK1, RRM2 and RFC4 were screened from the PPI network. The five genes were highly expressed in sarcoma. Conclusion: The five core common differentially expressed genes and related signaling pathways screened by bioinformatics analysis are helpful to understand the molecular mechanism of OS and ES pathogenesis, and are related to the prognosis of patients. They may become potential biomarkers for future research on OS comorbid ES, provide a basis for early diagnosis of OS combined with ES, and provide new ideas for clinical drug treatment research.
文摘Purpose: Bioinformatics-based approach to screen and analyze differentially expressed genes associated with the biological characteristics of Ewing sarcoma. Means: The GSE17674 dataset was selected for analysis, obtained by data retrieval based on the GEO public database. The R language limma toolkit was used to screen DEmRNAs. After the data were normalized, the Metascape online analysis software and the R language clusterProfiler package were used to analyze the GO function and KEGG pathway enrichment of DEmRNAs lines, respectively. The string database was selected for PPI analysis, and the results were imported into Cytoscape software to derive the core modules and predicted core genes. The genes selected above were analyzed for tissue localization specificity. Results: Through the analysis of GSE17674, differentially expressed genes were screened out, and GO and KEGG analyses were performed on the differentially expressed genes. The GO functional enrichment analysis was mainly enriched in the process of muscle system, muscle contraction, myocyte development, contractile fibers, myogenic fibers, myofibers, myofibrillar segments, actin binding, structural composition of muscle, and actin filament binding. KEGG pathway analysis showed that the core pathways associated with the development of ES were the core genes for myocardial contraction, congestive cardiomyopathy, and hypertrophic cardiomyopathy. Five Hub genes were obtained based on Cytoscape prediction. Tissue localization specificity analysis of Hub genes was performed, and a total of 2 Hub genes with tissue specificity were screened;MYH6 was specifically expressed in cardiac cells and MYL1 was specifically expressed in skeletal muscle cells. Conclusions: The differential genes screened will help to understand the molecular mechanisms underlying the highly invasive and metastasis-prone biological characteristics of ES, as well as provide new ideas for clinical drug-targeted treatment of ES.

文摘Daintain, a novel bioactive peptide produced and secreted by macrophages, was expressed in breast tumor tissues. The spatial distributions of daintain in 66 breast tumor specimens were investigated with immuno-histochemistry method. Reverse transcription polymerase chain reaction (RT-PCR) and in situ hybridization inspection system were also used to detect daintain in 45 cases of malignant breast tumors. The final results show that 93% high positive responses to daintain on breast cancer tumors. RT-PCR demonstrated that, no smear of daintain transcripted in benign tissues was found, and light smear in peri-cancer tissue was observed. Distribution of daintain was distinguishable among benign tissues, hyperplasia tissues, immature hyperplasia and invasive breast cancer, which can be used to mark the progression of the malignant lesion development. We conclude that the expression of daintain is up-regulated in breast cancers, which indicates that the peptide is closely associated with the disease progression. So daintain could be used as the biomarker for detecting breast cancer.

文摘We propose a scheme for the effective polarization and manipulation of electron spin by using a quantum dot with both charge and spin bias. Using the equation of motion for Keldysh nonequilibrium Green function, we study the spin accumulation and polarization for the system. Through analytical analysis and a few numerical examples, it is demonstrated that fairly large spin accumulation and polarization can be produced due to the breaking symmetry of the chemical potential for different electron spin in the leads. Moreover, the direction and the strength of the spin polarization can be conveniently controlled and tuned by varying the charge bias or the gate voltage.

文摘BACKGROUND: The supernatant of interferon-gamma (IFNγ) co-cultured with neonatal rat cortical glia can promote the cells in embryonic basal forebrain/septal nuclei to differentiate into cholinergic neurons, but the mechanism is still unclear. OBJECTIVE: To analyze the pathways for IFNγ to promote the differentiation of primarily cultured cholinergic neurons in rat embryonic basal forebrain/septal nuclei through culture in different conditioned medium. DESIGN: A controlled experiment taking cells as the observational target. SETTINGS: Department of Biochemistry and Molecular Biology, Youjiang Medical College for Nationalities; Department of Cell Biology, Beijing University Health Science Center. MATERIALS: Sixty-four pregnant Wistar rats for 16 days (250-350 g) and 84 Wistar rats (either male or female, 5-7 g) of 0-1 day after birth were provided by the experimental animal department of Beijing University Health Science Center. Rat IFNγ were provided by Gibco Company; Glial fibrillary acidic protein by Huamei Company. METHODS: The experiments were carried out in the Department of Cell Biology, Beijing University Health Science Center and Daheng Image Company of Chinese Academy of Science from July 1995 to December 2002. ① Interventions: The nerve cells in the basal forebrain/septal nuclei of the pregnant Wistar rats for 16 days were primarily cultured, and then divided into four groups: Blank control group (not any supernatant and medium was added); Control group (added by mixed glial cell or astrocyte conditioned medium); IFNγ group (added by mixed glial cell or astrocyte conditioned medium+IFNγ). Antibody group (added by mixed glial cell or astrocyte conditioned medium+IFNγ+Ab-IFNγ). Mixed glial cell or astrocyte conditioned medium was prepared using cerebral cortex of Wistar rats of 0-1 day after birth. ② Evaluation: The immunohistochemical method was used to perform the choline acetyltransferase (ChAT) staining of cholinergic neurons. The ChAT positive cells were counted. MAIN OUTCOME MEASURES: Comparison of ChAT positive cells in rat basal forebrain and septal nuclei in different conditioned medium. RESULTS: ① ChAT positive cells in mixed glial cell conditioned medium: The ChAT positive cells in the IFNγ group and antibody group were significantly more than those in the control group (P 〈 0.01). ② ChAT positive cells in astrocyte conditioned medium: The ChAT positive cells in the IFNγ group were significantly more than those in the control group, but there was no significant difference between the antibody group and control group (P 〉 0.05). CONCLUSION: IFNγ cannot directly promote the differentiation of cholinergic neurons, but plays a role through activating glial cells (except astrocytes) to produce IFNγ like molecules.

文摘Abstract [ Objective] This paper aimed to study the effect of temperature on gas production volume and period of biogas fermentation. [ Method] The fresh pig manure were as materials, and the effect of temperature (6 ℃, 12℃, 18 ℃, 24 ℃, 30 ℃) on fermentation start-up and period of biogas production were studied for 180 days of anaerobic fermentation. [ Result] The results showed that fermentation could be started in all temper- ature levels, the higher the temperature, the more biogas production, and the fermented period reduced with the temperature going up. The period was 7 months at low temperature ( below 20 ℃), 4 months at middle temperature (20 - 45 ℃). The aniseed was changed twice a year in February to March and July to August, which took 20 -30 days. It had the best biogas production period in 24 ℃ when the biogas production volume was high and stable. [ Conclusion] The best temperature for pig manure fermentation was 24 ℃.

文摘Electron–electron correlation plays an important role in the underlying dynamics in physics and chemistry.Helium is the simplest and most fundamental two-electron system.The dynamic process of helium in a strong laser field is still a challenging issue because of the large calculation cost.In this study,a graphic processing unit(GPU)open ACC based ab initio numerical simulations package He TDSE is developed to solve the full-dimensional time-dependent Schrodinger equation of helium subjected to a strong laser pulse.He TDSE uses B-spline basis sets expansion method to construct the radial part of the wavefunction,and the spherical harmonic functions is used to express for the angular part.Adams algorithm is employed for the time propagation.Our example shows that He TDSE running on an NVIDIA Kepler K20 GPU can outperform the one on an Intel E5-2640 single CPU core by a factor of 147.He TDSE code package can be obtained from the author or from the author's personal website(doi:10.13140/RG.2.2.15334.45128)directly under the GPL license,so He TDSE can be downloaded,used and modified freely.

The important effect of IFN-γ on the development and differentiation of neurons has been found by some scholars. OBJ EClIVE:To investigate whether IFN-γ has differentiational effect on cholinergic neurons in basal forebrain and septal nuclei, and make clear that the increased number of cholinergic neurons is resulted by cell differentiation or cell proliferation. DESIGN : Controlled observation trial SETTING: Department of Cell Biology, Medical School, Beijing University MATERIALS: Sixty-eight female Wistar rats at embryonic 16 days, weighing 250 to 350 g, were enrolled in this study, and they were provided by the Experimental Animal Center, Medical School, Beijing University. IFN-γ was the product of Gibco Company. METHODS: This study was carried out in the Department of Cell Biology, Medical School, Beijing University and Daheng Image Company of Chinese Academy of Sciences during September 1995 to December 2002. The female Wistar rats at embryonic 16 days were sacrificed, and their fetuses were taken out. Primary culture of the isolated basal forebrain and septal nuclei was performed. The cultured nerve cells were assigned into 3 groups: control group (nothing added), IFN-γ group(1×10^5 U/L interferon), IFN-γ+ IFN-γ antibody group (1 ×10^5 U/L IFN-γ + IFN-γ antibody). The specific marker enzyme (choline acetyl transferase) of cholinergic neuron was stained with immunohistochemical method. Choline acetyl transferase positive cells were counted, and ^14C-acetyl CoA was used as substrate to detect the activity of choline acetyl transferase, so as to reflect the differentiational effect of IFN-γ on cholinergic neuron in basal forebrain and septal nuclei. Flow cytometry was used to analyze cell circle and detect the proliferation of nerve cells. Nerve cells were marked with MAP2 and counted to evaluate the neuronal proliferation in basal forebrain and septal nuclei. MAIN OUTCOME MEASURES: Effect of interferon-γ on the number and activity of choline acetyl transferase-positive ceils in basal forebrain and septal nuclei as well as the effect on neuronal proliferation. RESULTS : ① Nerve cells in the basal forebrain and septal nuclei of IFN-γ group grew well compared with control group.②The differentiation of cholinergic neurons: The number and activity of choline acetyl transferase positive cells in IFN-γ group were significantly higher than those in the control group [(49.30 ±4.92) /100 cells vs (7.50±1.58) /100 cells; (2 049.00±12.30) min^-1 vs (1 227.30±12.59) min^-1, p 〈 0.01], while there was no significant difference in the number and activity of choline acetyl transferase positive cells between IFN-γ + IFN-γ antibody group and control group(P 〉 0.05). ③The proliferation of cholinergic neurons: Cell percentage was 17.2% and 19.8% at S-stage, 6.2% and 6.1% at G2+M stage in the control group and IFN-γ group respectively, without significant difference (P 〉 0.05). CONCLUSION : IFN-γ does not promote the neuronal proliferation in basal forebrain and septal nuclei, and the increased expression of cholinergic neurons is not resulted by the increase in the number of neurons, but its differentiation.

文摘Sertoli cells are indispensable for guaranteeing normal spermatogenesis and male fertility.Although a huge number of long non-coding RNAs(lncRNAs)are identified from developing porcine testicular tissues and have been predicted with crucial regulatory roles in spermatogenesis,their functions and regulatory mechanisms are still in infancy.Herein,we mainly explored the regulatory and functional roles of lncFPFSC in proliferation and apoptosis of immature porcine Sertoli cells.The results demonstrated that lncFPFSC was predominantly located in the cytoplasm of immature porcine Sertoli cells.lncFPFSC overexpression promoted cell cycle progression and cell proliferation,as well as inhibited cell apoptosis,whereas siRNA-induced lncFPFSC knockdown resulted in the opposite effects.Mechanistically,lncFPFSC acted as a sponge for miR-326.Overexpression of miR-326 inhibited cell proliferation and induced cell apoptosis,which further abolished the effects of lncFPFSC overexpression.The euchromatic histone-lysine N-methyltransferase 2(EHMT2)gene was directly targeted by miR-326,and its mRNA and protein expressions were both negatively regulated by miR-326 in immature porcine Sertoli cells.Then,siRNA-induced EHMT2 knockdown resulted a similar effect of miR-326 inhibition.Collectively,lncFPFSC promoted proliferation and inhibited apoptosis in immature porcine Sertoli cells through modulating the miR-326/EHMT2 axis.This study expanded our understanding of non-coding RNAs in participating porcine spermatogenesis through deciding the fate of Sertoli cells,and the competing endogenous RNA(ceRNA)network,and provided new molecular markers to treat Sertoli cell disorder inducing male infertility.

文摘The dynamics of spin–orbit-coupled Bose–Einstein condensate with parity-time symmetry through a moving obstacle potential is simulated numerically. In the miscible two-component condensate, the formation of the Kármán vortex street is observed in one component, while ‘the half-quantum vortex street' is observed in the other component. Other patterns of vortex shedding, such as oblique vortex dipoles, V-shaped vortex pairs, irregular turbulence, and combined modes of various wakes, can also be found. The ratio of inter-vortex spacing in one row to the distance between vortex rows is approximately0.18, which is less than the stability condition 0.28 of classical fluid. The drag force acting on the obstacle potential is simulated. The parametric regions of Kármán vortex street and other vortex patterns are calculated. The range of Kármán vortex street is surrounded by the region of combined modes. In addition, spin–orbit coupling disrupts the symmetry of the system and the gain-loss affects the local particle distribution of the system, which leads to the local symmetry breaking of the system, and finally influences the stability of the Kármán vortex street. Finally, we propose an experimental protocol to realize the Kármán vortex street in a system.

文摘Objective: To compare the effectiveness and safety of two surgical methods for lumbar degenerative diseases;the combination of the concept of accelerated rehabilitation with the assistance of Tianji Robotics and the concept of accelerated rehabilitation combined with manual pedicle screw placement assisted by conventional C-arm fluoroscopy. Methods: A retrospective analysis was performed on 70 patients who received the concept of accelerated rehabilitation combined with spinal surgery for lumbar degenerative diseases in Baise People’s Hospital from January 2022 to January 2024. Among them, 35 patients in the robot group received accelerated rehabilitation concept combined with robot-assisted surgery;In the conventional C-arm group, 35 patients received the accelerated rehabilitation concept combined with manual pedicle screw placement assisted by conventional C-arm fluoroscopy. VAS score (preoperative/postoperative), ODI score (preoperative/postoperative), intraoperative bleeding volume, postoperative hospital stay, postoperative complications and the accuracy rate of screw placement were compared between the two groups. Result: There was no statistically significant difference in preoperative VAS scores between the robot group and the conventional C-arm group (6.45 ± 0.82 VS 6.63 ± 0.81, P = 0.6600). The postoperative VAS score of the robot group was better than that of the conventional C-arm group (1.69 ± 0.80 VS 2.45 ± 0.85, P = 0.0000*). There was no statistically significant difference in preoperative ODI scores between the robot group and the conventional C-arm group (32.11 ± 3.18 VS 31.66 ± 2.25, P = 0.4900). The postoperative ODI score of the robot group was better than that of the conventional C-arm group (22.68 ± 1.94 VS 24.57 ± 2.25, P = 0.0000*). The postoperative complications in the robot group were less than those in the conventional C-arm group (2.7778% VS 28.5724%, P = 0.0030*). The intraoperative bleeding in the robot group was lower than that in the conventional C-arm group (320.85 ± 276.28 VS 490.00 ± 395.34, P = 0.0420*). The postoperative hospital stay of the robot group was shorter than that of the conventional C-arm group (10.00 ± 9.32 VS 14.49 ± 7.55, P = 0.0300*). The screw placement inaccuracy score of the robot group was lower than that of the conventional C-arm group (0.17 ± 0.51 VS 1.45 ± 1.46, P = 0.0000*). Conclusion: The combination of the concept of accelerated rehabilitation and Tianji Orthopedic robot-assisted surgery is more effective and safer in posterior lumbar decompression and internal fixation surgery with a screw rod system, and is worthy of promotion and application.
文摘Background: Although socioeconomic support is recommended for frailtymanagement, its association with the prognosis of frailty is unclear.Methods: Using data from participants aged ≥65 years in the ChineseLongitudinal Healthy Longevity Survey (2008–2018), the associations betweensocioeconomic support (source of income, medical insurance, communitysupport, living status), onset of prefrailty/frailty, and worsening of prefrailty,were analyzed using multinominal logistic regression models. The associationsbetween self-reported low quality of life (QoL) and reversion of prefrailty/frailty were analyzed using multivariate logistic regression models. Associationswith mortality risk were analyzed using Cox proportional hazardregression models.Results: A total of 13,859 participants (mean age: 85.8 ± 11.1 years) containing2056 centenarians were included. Financial dependence was a risk factor for lowQoL among prefrail/frail individuals, but not among robust individuals. Havingcommercial or other insurance, and receiving social support from the communitywere protective factors for low QoL among prefrail/frail individuals and for theworsening of prefrailty. Continuing to work was a risk factor for low QoL, but aprotective factor for worsening of prefrailty. A negative association betweencontinuing to work and mortality existed in prefrail individuals aged <85 yearsand ≥85 years. Living alone was a risk factor for low QoL, but was notsignificantly associated with frailty prognosis.Conclusions: Prefrail and frail individuals were vulnerable to changes insocioeconomic support and more sensitive to it compared with robustindividuals. Preferential policies regarding financial support, social support,and medical insurance should be developed for individuals with frailty.

文摘Objective:To investigate the effect of mir-3168 on the malignant transformation and cisplatin resistance of AGS and AGS/DDP gastric cancer cells,and to verify its target gene.Methods:The expression of mir-3168 in AGS and AGS/DDP gastric cancer cells was detected by qPCR,and mir-3168 mimic,inhibitor and negative control were synthesized.They were transfected into AGS and AGS/DDP gastric cancer cells,respectively.The expression of mir-3168 and TP53 mRNA was detected by qPCR.Cell viability was detected by CCK8 under gradient cisplatin treatment and non treatment,apoptosis was detected by flow cytometry,cell invasion was detected by Transwell,and TP53 protein expression was detected by western blot,The database predicted the binding sites of mir-3168 and TP53.According to the binding sites,the double luciferase experiment was used to verify the binding of mir-3168 and TP53.Results:Compared with cisplatin sensitive gastric cancer cell AGS,mir-3168 was significantly overexpressed in cisplatin resistant gastric cancer cell AGS/DDP;mir-3168 mimic promotes cisplatin resistance,proliferation and invasion of AGS and AGS/DDP gastric cancer cells,and inhibits apoptosis of AGS and AGS/DDP gastric cancer cells;mir-3168 inhibitor inhibits cisplatin resistance,proliferation and invasion of AGS and AGS/DDP gastric cancer cells,and promotes apoptosis of AGS and AGS/DDP gastric cancer cells;mir-3168 mimic inhibits the expression of TP53 mRNA and protein,and mir-3168 inhibitor promotes the expression of TP53 mRNA and protein;Targetscan database predicted that there was a binding point between mir-3168 and TP53,and the double luciferase experiment suggested that mir-3168 was bound to TP53 through the predicted binding site.Conclusion:mir-3168 may promote the malignant transformation of AGS and AGS/DDP gastric cancer cells and cisplatin resistance by targeting TP53.

文摘AIM: To investigate the inhibitory effects of emodin, baicalin, etc.on the hefA gene of multidrug resistance(MDR) in Helicobacter pylori(H.pylori).METHODS: The double dilution method was used to screen MDR H.pylori strains and determine the minimum inhibitory concentrations(MICs) of emodin, baicalin, schizandrin, berberine, clarithromycin, metronidazole, tetracycline, amoxicillin and levofloxacin against H.pylori strains.After the screened MDR stains were treated with emodin, baicalin, schizandrin or berberine at a 1/2 MIC concentration for 48 h, changes in MICs of amoxicillin, tetracycline, levofloxacin, metronidazole and clarithromycin were determined.MDR strains with reduced MICs of amoxicillin were selected to detect the hefA mR NA expression by realtime quantitative PCR.RESULTS: A total of four MDR H.pylori strains were screened.Treatment with emodin, baicalin, schizandrin and berberine significantly decreased the MICs of amoxicillin and tetracycline against some strains, decreased by 1 to 2 times, but did not significantly change the MICs of clarithromycin, levofloxacin, and metronidazole against MDR strains.In the majority of strains with reduced MICs of amoxicillin, hef A m RNA expression was decreased; one-way ANOVA(SPSS 12.0) used for comparative analysis, P < 0.05.CONCLUSION: Emodin, baicalin, schizandrin and berberine significantly decreased the MICs of amoxicillin and tetracycline against some H.pylori strains, possibly by mechanisms associated with decreasing hefA mR NA expression.
