Chinese mitten crab (Eriocheir sinensis) has higher commercial value as food source than any other species of Eriocheir in China. To evaluate the germplasm resources and characterize the genetic diversity and population structure of the crabs in different water systems, two stocks and two farming populations were assessed with 25 polymorphic microsallite loci available in public GenBank. Basic statistics showed that the average observed heterozygosity (Ho) amongst populations ranged from 0.5789 to 0.6824. However, a remarkable presence of inbreeding and heterozygote deficiencies were observed. To analyze population structure, pairwise FST coefficients explained only -10.3% variability from the subdivision of mitten crab populations, the remaining variability stems from the subdivision within subpopulations. Although the four populations had slight differentiation, different allelic frequencies resulted in distinct population structures. Two stocks and one farming population were clustered together to the phylogenetic branch of Yangtze crab, with an approximate membership of 95%. Whereas, another farming population was clustered singly to the phylogenetic branch of the Liaohe crab, with a membership of 97.1%. The tests for individual admixture showed that Yangtze crab had probably been contaminated with individuals from other water systems. Genetic relationships between populations also supported the conclusion that Yangtze crab and Liaohe crab had different gene pools in spite of the origins of the same species. 展开更多

In this paper, the numerical model of the net cage with the grid mooring system in waves is set up by the lumped mass method and rigid kinematics theory, and then the motion equations of floating system, net system, mooring system, and floaters are solved by the Runge-Kutta fifth-order method. For the verification of the numerical model, a series of physical model tests have been carried out. According to the comparisons between the simulated and experimental results, it can be found that the simulated and experimental results agree well in each condition. Then, the effects of submerged depth of grid and direction of incident wave propagation on hydrodynamic behaviors of the net cage are analyzed. According to the simulated results, it can be found that with the increase of submerged depth of grid, the forces acting on mooring lines and bridle lines increase, while the forces on grid lines decrease; the horizontal motion amplitudes of floating collar decrease obviously, while the vertical motion amplitudes of floating collar change little. When the direction of incident wave propagation changes, forces on mooring lines and motion of net cage also change accordingly. When the propagation direction of incident wave changes from 0° to 45°, forces on the main ropes and bridle ropes increase, while the forces on the grid ropes decrease. With the increasing propagation direction of incident wave, the horizontal amplitude of the forces collar decreases, while the vertical amplitude of the floating collar has little variation. 展开更多

As the most widely used plasticizers in the world, phthalate esters (PAEs) are potential endocrine disruption compounds (EDCs). In the present study, the toxicity of dimethyl phthalate (DMP), diethyl phthalate (DEP), dibutyl phthalate (DBP), di (2-ethylhexyl) phthalate (DEHP) on embryogenesis and larvae development of the marine univalve Haliotis diversicolor supertexta was examined in laboratory. The results show that the malformation of embryos appeared during the experiment, such as embryos died or lysed, small transparent flocculent rings studded on the periphery of the embryo, and the larvae could failed to hatch. In embryo toxic test, embryos incubated at the highest concentration of DMR DEP and DBP solutions showed significantly high abnormal rate compared with the control, while DEHP solutions displayed no significant difference. In larval toxic test, in all concentrations of DMP, DEP and DBP solutions, larval settlement rates were low significantly than that of the control. Similarly, DEHP solutions show nearly no effect on the larval settlement. The order of toxicity on embryos and larvae is DBP〉DEP〉DMP〉DEHE Being a simple and easy stimulation to indoor spawn, sensitive to environmental factors, and short culture time, the embryos of H. diversicolor supertexta can be used to indicate toxicity of the PAEs. 展开更多

The relationship between microsatellite polymorphism and body weight of captive bred Chinese sea cucumber Apostichopus japonicus was investigated in two local populations in Dalian. Among ten loci discovered, nine show changes except for AJ07 loci. Seven loci were found highly polymorphic in both populations. For each locus in two populations, the average number of alleles is 6.428 6 and 6.285 7, the average observed heterozygosity at 0.225 7 and 0.245 9, the expected heterozygosity at 0.776 8 and 0.748 8, the polymorphism information content (PIC) at 0.709 2 and 0.674 6, respectively. Further analysis show significant correlation between A. japonicus body weight and occurrence markers AJ02 and AJ04. The findings of the relation may be helpful for molecular breeding, as well as the marker-assisted selection of sea cucumbers. 展开更多

Fish specific genome duplication (FSGD) may have occurred before the radiation of the teleosts. Common carp (Cyprinus carpio L.) has been considered to be a tetraploid species, because of its chromosome numbers (2n=100) and its high DNA content. Using 69 microsatellite primer pairs, the variations were studied to better understand the genome evolution (genome duplication and diploidization) of common carp from a gynogenetic family. About 48% of primer pairs were estimated to amplify duplicates based on the number of PCR amplification per individual. Segregation patterns in the family suggested a partially duplicated genome structure and disomic inheritance. This indicates that the common carp is tetraploid and polyploidy occurred by allotetraploidy. Two primer pairs (HLJ021 and HLJ332) were estimated to amplify reduction based on the number of PCR amplification per individual. One allele in HLJ002 locus and HLJ332 locus was clearly lost in the gynogenetic family and the same as in six wild populations. Segregation patterns in the family suggested a partially diplodization genome structure. A hypothesis transition (dynamic) and equilibrium (static) were proposed to explain the common carp genome evolution between genome duplication and diploidization. 展开更多

As water depth increases, the structural safety and reliability of a system become more and more important and challenging. Therefore, the structural reliability method must be applied in ocean engineering design such as offshore platform design. If the performance function is known in structural reliability analysis, the first-order second-moment method is often used. If the performance function could not be definitely expressed, the response surface method is always used because it has a very clear train of thought and simple programming. However, the traditional response surface method fits the response surface of quadratic polynomials where the problem of accuracy could not be solved, because the true limit state surface can be fitted well only in the area near the checking point. In this paper, an intelligent computing method based on the whole response surface is proposed, which can be used for the situation where the performance function could not be definitely expressed in structural reliability analysis. In this method, a response surface of the fuzzy neural network for the whole area should be constructed first, and then the structural reliability can be calculated by the genetic algorithm. In the proposed method, all the sample points for the training network come from the whole area, so the true limit state surface in the whole area can be fitted. Through calculational examples and comparative analysis, it can be known that the proposed method is much better than the traditional response surface method of quadratic polynomials, because, the amount of calculation of finite element analysis is largely reduced, the accuracy of calculation is improved, and the true limit state surface can be fitted very well in the whole area. So, the method proposed in this paper is suitable for engineering application. 展开更多

A stowage planning for container ship made at one port must take account of the influence on subsequent ports. So the complexity of stowage planning problem increases due to its multi-ports nature. This problem is NP-hard problem. In order to reduce the computational complexity, the problem is decomposed into two sub-problems in this paper. First, container ship stowage problem (CSSP) is regarded as 'packing problem', ship-bays on the board of vessel are regarded as bins, the number of slots at each bay are taken as capacities of bins, and containers with different characteristics (homogeneous containers group) are treated as items packed. At this stage, there are two objective functions, one is to minimize the number of bays packed by containers and the other is to minimize the number of overstows. Secondly, containers assigned to each bays at first stage are allocate to special slot, the objective functions are to minimize the metacentric height, heel and overstows.The taboo search heuristics algorithm are used to solve the subproblem. The main focus of this paper is on the first subproblem. A case certifies the feasibility of the model and algorithm. 展开更多

The manipulation of the chromosome set for commercially valuable marine animals is important for enhancing aquacultural production. In this study, triploid and tetraploid sea cucumber Apostichopus japonicus were induced by hydrostatic pressure shock, and the conditions of appropriate induction were tested with different starting times, and hydrostatic pressure intensities and durations. The highest rate of triploid induction reached 20% and that of tetraploid was 60%. In consideration of the survival rate and hatch rate, the appropriate treatment for triploid was 55 Mpa of hydrostatic pressure for 5 rain at 55 min after fertilization (a.f.), while for tetraploid it was 60 Mpa for 5 rain at 61 min a.f. The triploid of the sea cucumber could survive through the pelagic larval stage and attachment stage, and develop like the control group of the experiment. The tetraploid, however, could not survive the attachment stage. 展开更多

Thus, flagellin prote... The Gram-negative Vibrio parahaemolyticus is a common pathogen in humans and marine animals. Bacteria flagellins play an important role during infection and induction of the host immune response. Thus, flagellin proteins are an ideal target for vaccines. We amplified the complete flagellin subunit gene (tTaA) from V. parahaemolyticus ATCC 17802. We then cloned and expressed the gene into Escherichia coli BL21 (DE3) cells. The gene coded for a protein that was 62.78 kDa. We purified and characterized the protein using Ni-NTA affinity chromatography and Anti-His antibody Western blotting, respectively. Our results provide a basis for further studies into the utility of the FlaA protein as a vaccine candidate against infection by Vibrio parahaemolyticus. In addition, the purified FlaA protein can he used for further functional and structural studies. 展开更多

The effect of the bacterial strain C14 on the germination of spores from the green alga Ulva pertusa was assayed and it was found that the bacterial biofilm and cell-free supernatant strongly inhibited spore germination. In attempts to define the chemical nature of she antifouling substance in the supernatant of C14, the culture supernatants were tested for activity after heat treatment, enzymatic treatments, size fractionation, and separation into aqueous and organic fractions. Results suggest that this bacterium produces an extracellular component with specific activity toward algal spores that was heat-sensitive and between 3 and 10 kDa in molecular size. The exposure of the organic phase fraction to spores showed inhibitive effect on spore germination. Pronase and carboxypeptidase y did not significantly affect the activity of inhibitory component, suggesting that the component was not a protein or a peptide. The bacterium C14 was identified as Pseudoalteromonas haloplanktis based on the phenotypic characters and 16S rRNA gene analvsis. 展开更多

Survival, growth and immune response of the scallop, Chlamys farreri, cultured in lantern nets at five different depths (2, 5, 10, 15, and 20 m below the sea surface) were studied in Haizhou Bay during the hot season (summer and autumn) of 2007. Survival and growth rates were quantified bimonthly. Immune activities in hemolymph (superoxide dismutase (SOD) and acid phosphatase (ACP)) were measured to evaluate the health of scallops at the end of the study. Environmental parameters at the five depths were also monitored during the experiment. Mortalities mainly occurred during summer. Survival of scallops suspended at 15 m (78.0%) and 20 m (86.7%) was significantly higher than at 2 m (62.9%), 5 m (60.8%) or 10 m (66.8%) at the end of the study. Mean shell height grew significantly faster at 10 m (205.0 μm/d) and 20 m (236.9 μm/d) than at 2, 5 or 15 m in summer (July 9 to September 1); however, shell growth rate at 20 m was significantly lower than at the other four depths in autumn (September 2 to November 6). In contrast to summer, scallops at 5 m grew faster (262.9 μm/d) during autumn. The growth of soft tissue at different depths showed a similar trend to the shell. Growth rates of shell height and soft tissue were faster in autumn than in summer, with the exception of shell height at 20 m. SOD activity of scallops increased with depth, and ACP activity was significantly higher at 15 and 20 m than at other depths, which suggests that scallops were healthier near the bottom. Factors explaining the depth-related mortality and growth of scallops are also discussed. We conclude that the mass mortality of scallop, C. farreri, during summer can be prevented by moving the culture area to deeper water and yield can be maximized by suspending the scallops in deep water during summer and then transferring them to shallow water in autumn. 展开更多

The results indicated that in the conditions of salinity ranging from 1.36 to 20 g/L, total alkalinity changing from 2.4 to 7.2 mmol/L and pH 8–9, zooplankton in saline-alkaline ponds was composed of freshwater salt-tolerated species or halophile species, some of which are halobiont species and usually occurs in freshwater. In our study, copepods were predominant in many fish-culture ponds and all control ponds without fishes in spring, late autumn and early winter. Dominant species of copepods were Sinocalanus tenellus, Cyclops vicinus, Thermocyclops taiho- kuensis. The biomass of copepods in the control ponds without fishes was higher than that of the fish-culture ponds. 展开更多

Three factors and three levels were molecular weight of PEG: 8 000, 6 000, 4 000 mol ·g^-1; PEG concentration: 45%, 50%, 55%; treatment duration time: 1, 2, 3 min, respectively. The results showed that the optimal pattern of three factors and three levels on inducing tetraploid of Haliotis discus hannai at two-cell stage through using PEG treatment were: molecular weight 4 000, concentration 55%, treatment duration time: 2 min. The highest tetraploid induction rate was 10.8% at embryo period. The three factors treatment sequence was treatment duration time→concentration→molecular weight. 展开更多

Genomic DNA was extracted from hypnospores of Perkinsus-like parasite of Manila clam Ruditapes philippinarum collected at the fishing grounds in Huanghai Sea coast Shicheng Island and East China Sea coast Ningbo, China. The internal transcribed spacer(ITS) in rDNA was PCR-amplified, cloned, sequenced, and compared with that of five Perkinsus species in GenBank. The fragment amplified from DNA of parasite of either Shicheng Island or Ningbo contained 649 bp, including partial ssrRNA(51 bp) and ITS(+5.8 S) (598 bp) regions. The ITS(+5.SS) sequences of Perkinsus-like parasite of both Shicheng Island and Ningbo were all 99% identical to those ofPerkinsis atlanticus, and were not more than 95% identical to those of other four Perkinsus species including P. marinus, P. andrewsi, P. qugwadi and P. medierraneus.The ITS (+5.8S) sequence of Perkinsus-like parasite of Shicheng Island was 99% identical to that of Ningbo. These facts about nucleotide sequences suggested that the Perkinsus-like parasite in Manila clam, Ruditapes philippinarum collected from either the Huanghai Sea coast or the East China Sea coast was P. atlanticus, and might reflect P. atlanticus strains of distinct geographic distribution. 展开更多

Some natural products, such as traditional Chinese medicines(TCMs), contain compounds with anticancer activity and have attracted a great interest in recent years as alternative anticancer therapies. A quick and convenient assay for screening antimicrotubule compounds in which in vitro microdialysis/high-performance liquid chromatography (HPLC) is used to monitor the binding of the compounds extracted from TCM Taxus cuspidata Siebold & Zucc(Taxus) to microtubules is reported. It was observed that the extract of Taxus contains at least five compounds which have affinity interaction with microtubules by biological fingerprinting analysis, and they were identified as the taxoids of taxol, baccatin III, 10-deacetylbaccatin Ⅲ(10-DAB), cephalomannine and 7-epi-10-deacetyltaxol (7-epi-10-DAT) based on the comparison of their high-performance liquid chromatographic/mass spectrometric and UV spectra with those of the standard samples, both assembly-promoting and disassembly-inhibiting characteristics of those compounds were evaluated. It was observed that baccatin Ⅲ and 10-DAB bound to microtubules and the binding degrees were influenced by GTP. The molecular dynamics (MD) method is used to simulate the interactions of energetic C20 clusters with the dense plasma targets within the framework of the linear Vlasov-Poisson theory. The influences of various clusters (H2, N2, C20 and C60 respectively) on stopping power are discussed. The simulation results show that the vicinage effects in the Coulomb explosion dynamics and the stopping power are strongly affected by the variations in the cluster speed and the plasma parameters. Coulomb explosions are found to proceed faster for higher speeds, lower plasma densities and higher electron temperatures. In addition, the cluster stopping power is strongly enhanced in the early stages of Coulomb explosions due to the vicinage effect, but this enhancement eventually diminishes, after the cluster constituent ions are sufficiently separated. For the large and heavy clusters, the stopping power ratio reaches much higher values in the early stage of Coulomb explosion owing to the constructive interferences in the vicinage effect. 展开更多
