We report a novel double-shelled nanoboxes photocatalyst architecture with tailored interfaces that accelerate quantum efficiency for photocatalytic CO_(2) reduction reaction(CO_(2)RR)via Mo–S bridging bonds sites in S_(v)–In_(2)S_(3)@2H–MoTe_(2).The X-ray absorption near-edge structure shows that the formation of S_(v)–In_(2)S_(3)@2H–MoTe_(2) adjusts the coordination environment via interface engineering and forms Mo–S polarized sites at the interface.The interfacial dynamics and catalytic behavior are clearly revealed by ultrafast femtosecond transient absorption,time-resolved,and in situ diffuse reflectance–Infrared Fourier transform spectroscopy.A tunable electronic structure through steric interaction of Mo–S bridging bonds induces a 1.7-fold enhancement in S_(v)–In_(2)S_(3)@2H–MoTe_(2)(5)photogenerated carrier concentration relative to pristine S_(v)–In_(2)S_(3).Benefiting from lower carrier transport activation energy,an internal quantum efficiency of 94.01%at 380 nm was used for photocatalytic CO_(2)RR.This study proposes a new strategy to design photocatalyst through bridging sites to adjust the selectivity of photocatalytic CO_(2)RR. 展开更多

The present study reports a physicochemical comparison of shade-grown and sun-grown coffee beans, under unripe, rip and roasted-ripe conditions, using electrical conductivity measurements, electron paramagnetic resonance (EPR), infrared spectroscopy (FTIR), and high performance liquid chromatography (HPLC). Moreover, the assessed physicochemical parameters were compared with organoleptic evaluations based on the Coffee Quality Institute protocol. The values found for electrical conductivity, leached potassium, and stable free radicals were respectively 29%, 31%, and 350% higher for shade-grown coffee beans, whereas polyphenol oxidase enzymatic activity was 23% lower. By contrast, FTIR and HPLC measurements identified higher chlorogenic acid and lipid contents in sun-grown coffee beans. Importantly, the sensorial grade attributed to roasted-ripe grains was 12% higher for sun-grown coffee. Our findings suggest that shade-grown coffee beans have undergone microorganismal activity and undesired fermentation during cultivation, which resulted in lower coffee quality. A correlation between a set of selected physicochemical properties and organoleptic properties was robustly established and could be used in the development of future coffee bean quality control protocols. 展开更多

Cannabidiol (CBD), one of the most studied phytocannabinoids, is non-psychotropic and can induce protective effects on the central nervous system against acute and chronic brain injury. Interestingly, CBD inhibits processes relating to amyloid beta (Aβ)-induced neurotoxicity in mouse models of Alzheimer’s disease, though the detailed molecular mechanism underlying the CBD neurotoxicity modulation is not fully understood. In this study, using atomic force microscopy, we find that CBD promotes the aggregation of Aβ peptides, enhancing the formation of Aβ oligomers, also known as Aβ-derived diffusible ligands (ADDLs). The CBD-mediated sequestration of Aβ monomers in soluble ADDLs could reduce neurotoxicity. This study highlights a possible role of CBD in modulating the formation of ADDL aggregates and provides insight into potentially neuroprotective properties of CBD in Alzheimer’s disease. 展开更多

Data gaps and biases are two important issues that affect the quality of biodiversity information and downstream results.Understanding how best to fill existing gaps and account for biases is necessary to improve our current information most effectively.Two current main approaches for obtaining and improving data include(1)curation of biological collections,and(2)fieldwork.However,the comparative effectiveness of these approaches in improving biodiversity data remains little explored.We used the Flora de Bogota project to study the magnitude of change in species richness,spatial coverage,and sample coverage of plant records based on curation versus fieldwork.The process of curation resulted in a decrease in species richness(synonym and error removal),but it significantly increased the number of records per species.Fieldwork contributed to a slight increase in species richness,via accumulation of new records.Additionally,curation led to increases in spatial coverage,species observed by locality,the number of plant records by species,and localities by species compared to fieldwork.Overall,curationwas more efficient in producing new information compared to fieldwork,mainly because of the large number of records available in herbaria.We recommend intensive curatorial work as the first step in increasing biodiversity data quality and quantity,to identify bias and gaps at the regional scale that can then be targeted with fieldwork.The stepwise strategy would enable fieldwork to be planned more costeffectively given the limited resources for biodiversity exploration and characterization. 展开更多

Anti-ganglioside antibodies are associated with delayed/poor clinical recovery in Guillain-Barrèsyndrome,mostly related to halted axon regeneration.Cross-linking of cell surface gangliosides by anti-ganglioside antibodies triggers inhibition of nerve repair in in vitro and in vivo paradigms of axon regeneration.These effects involve the activation of the small GTPase Rho A/ROCK signaling pathways,which negatively modulate growth cone cytoskeleton,similarly to well stablished inhibitors of axon regeneration described so far.The aim of this work was to perform a proof of concept study to demonstrate the effectiveness of Y-27632,a selective pharmacological inhibitor of ROCK,in a mouse model of axon regeneration of peripheral nerves,where the passive immunization with a monoclonal antibody targeting gangliosides GD1a and GT1b was previously reported to exert a potent inhibitory effect on regeneration of both myelinated and unmyelinated fibers.Our results demonstrate a differential sensitivity of myelinated and unmyelinated axons to the pro-regenerative effect of Y-27632.Treatment with a total dosage of 9 mg/kg of Y-27632 resulted in a complete prevention of anti-GD1a/GT1b monoclonal antibody-mediated inhibition of axon regeneration of unmyelinated fibers to skin and the functional recovery of mechanical cutaneous sensitivity.In contrast,the same dose showed toxic effects on the regeneration of myelinated fibers.Interestingly,scale down of the dosage of Y-27632 to 5 mg/kg resulted in a significant although not complete recovery of regenerated myelinated axons exposed to anti-GD1a/GT1b monoclonal antibody in the absence of toxicity in animals exposed to only Y-27632.Overall,these findings confirm the in vivo participation of Rho A/ROCK signaling pathways in the molecular mechanisms associated with the inhibition of axon regeneration induced by anti-GD1a/GT1b monoclonal antibody.Our findings open the possibility of therapeutic pharmacological intervention targeting Rho A/Rock pathway in immune neuropathies associated with the presence of anti-ganglioside antibodies and delayed or incomplete clinical recovery after injury in the peripheral nervous system. 展开更多

The Effect of Water and Salt Stress on Paspalum dilatatum,a Constituent of Pampas Natural Grasslands

The effects of the salt stress on plant growth are usually increased by the water stress.We studied the impact of both stresses in simultaneous pulses of drought and salinity on Paspalum dilatatum.This forage species is native to South America,spread in grasslands in many tropical,subtropical,and temperate areas of the world,and very common in grasslands of the Flooding Pampas of Argentina.Mimicking what happens in nature.We compared a pot experiment,a non-stressed control against water stress for a month(midpoint between field capacity and wilting point),and two saline stresses(moderate,6 d·Sm^(−1)and strong,12 d·Sm^(−1)),also for a month.Aerial biomass(green leaf;non-leaf green material,and dry material)and roots were harvested,weighed,and analyzed for nitrogen,phosphorus,and cations.The biomass of all components significantly decreased when both stresses were applied.Water plus strong saline stress reduced by half the total biomasses,compared to the control.The proportion of aerial biomass/root biomass ratio as well as aerial green component/dry materials ratio tend to decrease when subjected to both stresses.Nitrogen concentration in plants was not significantly affected,but phosphorus concentration increased in aerial biomass components,from 0.10 to 0.18 mg·kg^(−1)between the extreme treatments,but did not change in roots.Sodium concentration in plants increased(i.e.,in green leave sodium(Na)increased from 0.27 to 2.01 mg·kg^(−1)between the extreme treatments),whereas other cations either did not change or decreased,affecting the ratios between them.Sodium performance allows us to infer that the Na accumulation of P.dilatatum behaves in an intermediate range,compared to very tolerant to salts or non-salt tolerant species of the Paspalum genus.In agreement,when salts were applied in the form of a pulse,P.dilatatum tolerated higher salinity than that found by other authors for the same species,using continuous salinity. 展开更多

The use of biochar can have several effects on plant germination,depending on raw material,preparation method and application dose.However,the molecular mechanisms that lead to those results have yet to be elucidated.The aim of this research was to improve the understanding of these mechanisms by characterizing the metabolic effects of sugarcane bagasse biochar on soybean germination.Three types of biochars were prepared by pyrolysis at 300℃(SCB300),400℃(SCB400)and 600℃(SCB600).Then,each one was mixed into sand at 1%,3%,5%(w/w)dose,respectively.The experiment was performed in 8 days of incubation,when the number of germinated seeds and the average radicle length were determined.To evaluate the metabolome,the dry biomass(DB)was subjected to extraction with a mixture of methanol-d4 and D2O(1:1 v/v).The extracts were submitted to metabolomics analysis by Proton Nuclear Magnetic Resonance.The Relative Germination,Relative Average Radicle Growth and Germination Index increased in all treatments compared to control.On the other hand,the DB increased in all treatments,except for SCB300,at doses of 1%and 3%w/w.Seven metabolites(alanine,asparagine,acetic acid,citric acid,glycerol,fatty acids and sucrose)were identified and quantified in DB extracts as the most influential finding for the separation of treatments.Taken together,these results strongly suggested that biochars accelerated the catabolism of triacylglycerols to sucrose and induced a slight osmotic stress. 展开更多

Recently, researchers explored garlic and its derivatives as a scientific strategy in veterinary practices for diverse purposes, such as improving poultry production characteristics and acting as antibiotic growth promoters. This comprehensive review delves into garlic and its derivatives as preventive and corrective treatments for viral diseases in laying hens and broilers. BACKGROUND Patients with hepatitis C virus(HCV) and hepatocellular carcinoma(HCC) may or not develop iron overload(IO),which is associated with worst prognosis,because can cause serious damage to organs.HFE gene controls the iron uptake from gut,particularly in patients with hereditary hemochromatosis(HH).AIM To identify associations between HFE coding region in patients exhibiting hereditary hemochromatosis and in diseases associated with acquired IO.METHODS We sequenced exons 2 to 5 and boundary introns of HFE gene,evaluating all polymorphic sites in patients presenting hereditary(hemochromatosis) or acquired iron overload HCV and HCC) and in healthy controls,using Sanger sequencing.We also determined the ensemble of extended haplotype in healthy control individuals,including several major histocompatibility complex loci,using sequence specific probes.Haplotype reconstruction was performed using the Arlequin and Phase softwares,and linkage disequilibrium(LD) between histocompatibility loci and HFE gene was performed using the Haploview software.RESULTS The HFE*003 allele was overrepresented(f = 71%) and HFE*001 allele was underrepresented(f = 14%) in HH patients compared to all groups.A strong linkage disequilibrium was observed among the H63 D-G,IVS2(+4)-C and C282 YG gene variants,particularly in HH;however,the mutation IVS2(+4)T>C was not directly associated with HH susceptibility.The HFE*001/HFE*002 genotype conferred susceptibility to HCC in HCV patients exhibiting IO(P = 0.02,OR =14.14).Although HFE is telomeric to other histocompatibility genes,the H63 DG/IVS2(+4)-C(P ≤ 0.00001/P ≤ 0.0057) combination was in LD with HLA-B*44 allele group in healthy controls.No LD was observed between HFE alleles and other major histocompatibility loci.CONCLUSION A differential HFE association was observed for HH and for diseases associated with acquired IO(HCV,HCC).Since HFE is very distant from other histocompatibility loci,only weak associations were observed with these alleles. 展开更多

An analytical methodology based on an O-[2-(methacryloyloxy)-ethylcarbamoyl]-10,11-dihydroquinidine(MQD)-silica hybrid monolithic column was developed for the enantioseparation of 9-fluorenylmethoxycarbonyl(FMOC)derivatized amino acids by nano-liquid chromatography.The mobile phase was optimized including the apparent pH,content of ACN,and concentration of the buffer to obtain a satisfactory enantioresolution performance.27 FMOC derivatized amino acids including 19 protein and 8 non-protein amino acids were tested,and 19 out of them were enantiomerically discriminated obtaining baseline separation for 11 of them.Analytical characteristics of the method were evaluated for norvaline and tryptophan in terms of linearity,precision,accuracy,limits of detection(LOD)and quantitation(LOQ)showing good performance to be applied to the enantiomeric determination of these amino acids in dietary supplements.LOD and LOQ values were 9.3 and 31 mM for norvaline enantiomers and 7.5 and 25 mM for tryptophan enantiomers,respectively.The contents of D-norvaline and D-tryptophan were below their respective LODs in all the analyzed samples.Quantitation of L-tryptophan and L-norvaline showed good agreement with the labeled contents except for one sample which did not show presence of L-norvaline,contrary to the label indication. 展开更多

The reactive Ga species in vapor the phase was formed with NH4Cl and gallium. The unidirectional growth was catalyzed with gold nanoparticles formed onto the substrate prior to the CVD reaction in order to induce a vapor-liquid-solid (VLS) mechanism. However, this method of synthesis seems to be influenced by other growth mechanisms which formed additional depositions of GaN with different morphology than the rods catalyzed by gold nanoparticles. The moieties of GaN that grew in the absence of gold formed branches in the rods or increased the lateral growth of rods resulting in larger diameters than the size of the gold particle that guided the growth. 展开更多

To establish the chemical profile, and to evaluate the antibacterial and modulatory activity of the ethanolic extracts of the stalk’s inner bark and heartwood of Secondatia floribunda.MethodsQuantification of total phenols and flavonoids was determined by the Folin-Ciocalteu method and aluminum chloride, respectively. Phenolic compounds were identified and quantified by HPLC-DAD (High Performance Liquid Chromatography-Diodearray Detector) and the Infrared Spectroscopy was performed using the measure by Attenuated Total Reflectance with Fourier Transform (ATR-FTIR). Antibacterial assays for determination of the Minimum Inhibitory Concentration (MIC) and modification of aminoglycosides were performed by microdilution.ResultsInfrared spectra showed similar characteristics, having among its main absorption bands hydroxyl group (OH). The antibacterial activity showed clinically significant results for the strains of Staphylococcus aureus and Escherichia coli. In modulation assay, synergic and antagonistic effect for both extracts was observed. Heartwood extract in combination with antibiotics showed a significant MIC reduction at 19.8% (P < 0.000 1)-79.3% (P < 0.01).ConclusionsThis study is the first report of chemical and biological information of Secondatia floribunda suggesting that it is clinically relevant source of a new antibacterial therapy, especially due to the presence of significant levels of phenolic compounds. 展开更多

The possibility of commercialization of Moro blood oranges in tropical countries such as Brazil was evaluated to verify whether post-harvest management through storage at low temperatures for a period of 60 days can improve the bioactive properties and quality parameters. Moro blood oranges cultivated in Brazil did not contain significant amounts of anthocyanins at the time of harvesting, but these compounds were activated by post-harvest management through storage at low temperatures (4&deg;C and 8&deg;C) for a period of 60 days. The emergence of the anthocyanins in the juices occurred within a few weeks of storage, but the maximum levels were attained after 60 days and at the temperature of 8&deg;C. Cold storage positively influenced other bioactive compounds such as total phenolic compounds, individual phenolic compounds, β-carotene and the antioxidant activity determined by the sequestration of DPPH free radicals. It did not influence the vitamin C content. In addition, storage significantly altered the color, total acidity and pH of the fruits, but it did not prevent its commercial use. The remaining quality parameters were not influenced. It is possible to commercialize these oranges in Brazil through post-harvest management. 展开更多

The main objective of this work was to use reline deep eutectic solvent,containing Al(III)ions,for the electrochemical study of the nucleation and growth of aluminum onto a glassy carbon electrode at different temperatures and angular speeds(ω)of the working electrode.In order to fulfill this,electrochemical and surface characterization techniques were used.It was found that as temperature increased,the onset of the Al(III)DES reduction occurred at less negative potentials while the current peak of the voltammograms increased.These indicate that Al deposition thermodynamics and kinetics were favored.Practically,no anodic current was detected due to Al passivation by Al(OH)_(3)(s)andγ-Al_(2)O_(3)(s).Atω=0 r/min,the Al deposition chronoamperograms were analyzed by a theoretical model comprising Al 3D diffusion-controlled nucleation and growth and residual water reduction.However,those recorded at different angular speeds were analyzed with a theoretical model where adsorption−desorption and diffusion-controlled nucleation−growth occurred simultaneously.The deposits were characterized by SEM,EDX,XPS and XRD.Atω=0 r/min,formation of well distributed nanoparticles((78.1±9.5)nm)was observed,while atω=900 r/min the deposit was formed by multiple 10μm diameter leaf-like flat microstructures,composed by Al,Al(OH)_(3)(s)andγ-A2O3(s). 展开更多

The need for renewable energy sources makes microalgae an essential feedstock for biofuels production.The molecular aspects and the response to nitrogen(N)-limited conditions with a phytohormone stimulus in microalgae have been slightly explored.In this work,Scenedesmus obliquus was used as a study model to analyze the effect of benzyl amino purine(BAP)and gibberellic acid(GA)coupled to nitrogen limitation on cell growth,biomass and fatty acids.The selected 10^(-5)M BAP increased the biomass by 1.44-fold,and 10^(-6)M GA by 1.35-fold.The total lipids also increased by 2.8 and 1.11-fold,respectively.The 10^(-5)M BAP and 10^(-6)M GA addition to S.obliquus cultures at different initial nitrogen percentages(N-0,N-25,and N-50)showed a significant increase in cell growth and biomass productivity compared to the unstimulated cultures.BAP N-0 and GA N-0 produced the highest lipid yields with 55%and 50%,respectively.The lipid profile analysis revealed an increase,particularly in C18:1 and C16:0 fatty acids.Gene expression analysis showed an over-expression of acyl carrying protein(ACP),stearoyl-ACP desaturase(SAD),fatty acid acyl-ACP thioesterase(FATA),and diacylglycerol acyltransferase(DGAT)genes,which were mainly induced by nitrogen limitation.Furthermore,BAP and GA produced a significant over-expression on these genes in the N-replete cultures.This study shows that BAP and GA,coupled to N limitation stress,can be used to increase the biomass and lipid production in S.obliquus for sustainable biofuels. 展开更多

In Mexico,there is a need to produce large quantities of plantlets for the establishment and replanting of blue(cv.azul)agave production areas.Most of these plots are within the origin denomination area(DOT,Spanish acronym)of the distilled product of this plant,known as tequila.The objective of this study was to develop an in vitropropagation protocol for Agave tequilana Weber cv.azul using segmented stems in both:solid and liquid media.A disinfection and in vitro technique were developed to obtain shoots,through plantlets collected in commercial plots,which attained 100%surface-disinfection and budding rate.At the multiplication stage,the effects of 6-Benzylaminopurine(BA)(0.0,4.4 and 13.2μM)and kinetin(0.0,9.4,18.8 and 37.6μM)were evaluated on lateralshoot production of segmented sagittal stems.These were cultivated on Murashige&Skoog(MS)medium,with the addition of 3.0%sucrose and 8 g L−1 agar.It was observed that BA and kinetin increased the number of shoots per explant,obtaining up to 18 and 26,respectively.Furthermore,it was found that just the sagittal segmentation of explants increased axillary budding.On the other hand,segmented-stem bases were grown in MS liquid medium with 3.0%sucrose,inside a RITAsystem,programmed by a 5 min immersion step with a frequency of every 4 h.The effect of Indole−3-Acetic acid(IAA)(0.57,2.9,5.7μM)was evaluated,while maintaining a concentration of BA(13.2μM).It was observed that the greatest concentration of IAA led to the formation of more than 20 buds per explant.These results offer a new methodology to increase the efficiency of A.tequilana Weber cv.azul-in vitro multiplication by sagittal segmentation of stems and the addition of BA and/or IAA. 展开更多

The present work evaluates the feasibility of using the raw material collected from discarded zinc-carbon batteries as heterogeneous catalyst to degrade the dye Indigo Carmine in an aqueous solution. Besides the evident environmental application, this work also presents an economic alternative for the production of new catalysts used to remediate polluted waters. For this, discarded carbon-zinc batteries were gathered, disassembled and their anodic paste collected. After acidic treatment and calcination at 500°C, characterization measurements, i.e. flame atomic absorption spectroscopy (FAAS), nitrogen sorption, X-ray diffraction (XRD) and scanning electron microscopy (SEM), revealed that the so-obtained material consisted mainly of ZnMn2O4. This material acts as a heterogeneous catalyst in a Fenton-like process that degrades the dye Indigo Carmine in water. That is probably due to the presence of Mn(III) (manganese in the +3 oxidation state) in this material that triggers the decomposition of hydrogen peroxide (H2O2) to yield hydroxyl radicals (HO&middot;). Moreover, direct infusion electrospray ionization coupled to high resolution mass spectrometry (ESI-HRMS) was employed to characterize the main by-products resulting from such degradation process. These initial results thus indicate that raw materials from waste batteries can therefore be potentially employed as efficient Fenton-like catalysts to degrade organic pollutants in an aqueous solution. 展开更多

The Stability Geometrical Indicator for short times is calculated to locate regular and chaotic trajectories as the relative extrema of this indicator. Only trajectories with initial conditions at the boundary of the Hill’s region are considered to characterize the dynamics of the system. A square wave voltammetry (DPV) method for trans-Pt[Cl2(Dimethylamine)(isopropylamine)] determination is developed. To this end, all the chemical and instrumental variables affecting the determination of trans-Pt[Cl2(Dimethylamine) (isopropylamine)] are optimized. From studies of the mechanisms governing the electrochemical response of trans-Pt[Cl2(Dimethylamine)(isopropylamine)], it was concluded that it was an electrochemically reversible system with an adsorptive oxidation phenomenon. Under optimal conditions, the variation of analytical signal (Ip) with trans-Pt[Cl2(Dimethylamine)(isopropylamine)] concentration was linear in the 0.05 μg·mL-1 to 10 μg·mL-1 range, with a LOD 91 μg·mL-1 of and a LOQ of 303 μg·mL-1, a RSD 1.10% and Er 0.72%. The optimized method was applied to the determination of trans-Pt[Cl2(Dimethylamine)(isopropylamine)] in biological fluids, in human urine and synthetic urine. 展开更多
