Background:Formaldehyde is a chemical substance used to preserve the biochemical and physiological components of a tissue by preventing autolysis and putrefaction,leaving the tissue in a life-like manner.Objective:This study aims to assess the effect of formalin concentrations on the histochemical staining in liver,kidneys and lungs of Wistar rats.Methods:A total of ten(10)Wistar rats were purchased and allowed to acclimatize for a period of 2 weeks in plastic cages with iron netting at the Animal House facility,Babcock University.The animals were placed on standard rat feeds and water ad-libitum.The rats were randomly sacrificed by cervical dislocation,following mild anaesthesia using chloroform inhalation.For histological examination,the liver,lungs,and kidneys of Wistar rats were removed and preserved in neutral buffered formalin at 2%,4%,6%,8%,and 10%concentrations.Sections were stained in Haematoxylin and Eosin(Vector Laboratories,Inc.Newark,US).The quality of the nuclei,cytoplasm and cell membrane were graded on a scale of 1(poor)to 5(excellent).Results:Six percent(6%)formalin was observed to produce excellent results in liver tissues with a total grade of sixteen(16).10%formalin produced excellent results in lung tissues with a total grade or nineteen(19).Interestingly,2%,6%and 10%formalin concentrations produced excellently results in kidney tissues with the highest total grade fourteen(14).Conclusion:This study concludes that 6%formalin produced better section and staining qualities more than the commonly used 10%formalin according to the observations from the stained sections.This implies that the concentration of formalin affects the histochemical staining in liver,kidneys and lungs of Wistar rats. 展开更多

The association between heavy metals in the blood and obesity has been examined in many studies.However,inconsistencies have been observed in the results of these studies.The present study was conducted using data from 119,181 participants of the Korea National Health and Nutrition Examination Survey(KNHANES)for 11 years in 2005 and between 2008 and 2017.The subjects with missing heavy metal blood tests,health interview data,and health examination data were excluded from the study.The study population comprised 1,844 individuals(972 men,and 872 women)who were eligible for inclusion.It was found that obesity and abdominal obesity were associated with an increase in both blood mercury(P<0.001)and alanine aminotransferase(ALT)(P<0.001).After adjusting the confounding factors,those with concurrent high levels of ALT and the highest tertile of mercury showed an increased risk of obesity(odds ratio 4.46,95%confidence interval 2.23-8.90,P<0.001)as well as abdominal obesity(odds ratio 5.36,95%confidence interval 2.57-11.17,P<0.001).The interrelationship of mercury and ALT with the parameters of body mass index(P for interaction=0.009)and waist circumference(P for interaction=0.012),respectively,have been observed to be significant,suggesting that the reciprocal relationship could contribute to obesity and abdominal obesity. 展开更多

Diseases like Alzheimer’s and Parkinson’s diseases are defined by inflammation and the damage neurons undergo due to oxidative stress. A primary reactive oxygen species contributor in the central nervous system, NADPH oxidase 4, is viewed as a potential therapeutic touchstone and indicative marker for these ailments. This in-depth review brings to light distinct features of NADPH oxidase 4, responsible for generating superoxide and hydrogen peroxide, emphasizing its pivotal role in activating glial cells, inciting inflammation, and disturbing neuronal functions. Significantly, malfunctioning astrocytes, forming the majority in the central nervous system, play a part in advancing neurodegenerative diseases, due to their reactive oxygen species and inflammatory factor secretion. Our study reveals that aiming at NADPH oxidase 4 within astrocytes could be a viable treatment pathway to reduce oxidative damage and halt neurodegenerative processes. Adjusting NADPH oxidase 4 activity might influence the neuroinflammatory cytokine levels, including myeloperoxidase and osteopontin, offering better prospects for conditions like Alzheimer’s disease and Parkinson’s disease. This review sheds light on the role of NADPH oxidase 4 in neural degeneration, emphasizing its drug target potential, and paving the path for novel treatment approaches to combat these severe conditions. 展开更多

Objective: To investigate the anti-inflammatory effects of decursin and decursinol angelate-rich Angelica gigas Nakai(AGNE) on dextran sulfate sodium(DSS)-induced murine ulcerative colitis(UC).Methods: The therapeutic effect of an AGNE was analyzed in a mouse model of UC induced by DSS. Disease activity index values were measured by clinical signs such as a weight loss, stool consistency, rectal bleeding and colon length. A histological analysis was performed using hematoxylin and eosin staining. Key inflammatory cytokines and mediators including IL-6, TNF-a, PGE2, COX-2 and HIF-1 a were assayed by enzymelinked immunosorbent assay or western blotting.Results: Treatment with the AGNE at 10, 20, and 40 mg/kg alleviated weight loss,decreased disease activity index scores, and reduced colon shortening in mice with DSSinduced UC. AGNE inhibited the production of IL-6 and TNF-a in serum and colon tissue. Moreover, AGNE suppressed the increased expression of COX-2 and HIF-1 a and the increased production of PGE2 in colon tissue were observed in mice with DSSinduced UC. Additionally, histological damage was also alleviated by AGNE treatment.Conclusions: The findings of this study verified that AGNE significantly improves clinical symptoms and reduces the activity of various inflammatory mediators. These results indicate the AGNE has the therapeutic potential in mice with DSS-induced UC. 展开更多

Methods: The viability of HT-29 cells was assayed by the 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2 H-tetrazolium(MTS) method. Apoptosis induction and cell cycle inhibition were confirmed by fluorescein isothiocyanate and propidium iodide staining using flow cytometry.Morphological changes in the nucleus were observed, using a fluorescence microscope with4',6-diamidino-2-phenylindole(DAPI) nuclear staining. The expression levels of the upstream and downstream proteins involved in the anti-cancer mechanism were confirmed by Western blotting. Results: After treating HT-29 cells with different concentrations of ethylacetate fraction from O. japonicus, the viability of cells decreased in a concentration-dependent manner,while apoptosis induction and apoptotic body formation increased. Cell cycle analysis showed that the arrest occurred at the sub-G_1 and S phase. Among the upstream and downstream proteins involved in anti-cancer activity, the level of B cell lymphoma-2 decreased, and the bcl-2-associated x protein increased. The level of pro-caspase-3, pro-caspase-8, and pro-caspase-9 decreased, while the level of cleaved-caspase-3, cleaved-caspase-8, and cleaved-caspase-9 increased. Moreover, the phosphorylation, that is, activation of extracellular signal regulated kinase 1/2, Jun-N-terminal kinase, and p38 increased. Conclusions: Combining the above results, it is thought that the survival of HT-29 cells is suppressed by ethylacetate fraction from0. japonicus through mitochondrial regulation-induced caspase cascade activation, induction of apoptosis and cell cycle arrest. 展开更多

Human life span has dramatically increased over several decades,and the quality of life has been considered to be equally important.However,diabetes mellitus(DM) characterized by problems related to insulin secretion and recognition has become a serious health problem in recent years that threatens human health by causing decline in brain functions and finally leading to neurodegenerative diseases.Exercise is recognized as an effective therapy for DM without medication administration.Exercise studiesusing experimental animals are a suitable option to overcome this drawback,and animal studies have improved continuously according to the needs of the experimenters.Since brain health is the most significant factor in human life,it is very important to assess brain functions according to the different exercise conditions using experimental animal models.Generally,there are two types of DM; insulin-dependent type 1 DM and an insulin-independent type 2 DM(T2DM); however,the author will mostly discuss brain functions in T2 DM animal models in this review.Additionally,many physiopathologic alterations are caused in the brain by DM such as increased adiposity,inflammation,hormonal dysregulation,uncontrolled hyperphagia,insulin and leptin resistance,and dysregulation of neurotransmitters and declined neurogenesis in the hippocampus and we describe how exercise corrects these alterations in animal models.The results of changes in the brain environment differ according to voluntary,involuntary running exercises and resistance exercise,and gender in the animal studies.These factors have been mentioned in this review,and this review will be a good reference for studying how exercise can be used with therapy for treating DM. 展开更多

Effect of Varicocelectomy on Semen Parameters of Men Seeking Infertility Treatment in Tamale, Ghana 被引量:1

The study aimed to determine the effect of microsurgical sub-inguinal varicocelectomy on semen parameters among men seeking infertility treatment in Ghana. This was an intervention study conducted at Tamale Teaching Hospital in the Tamale Metropolis from September 2017 to August 2021. The study involves two groups;the surgery group (n = 75) and the observed group (n = 63). Duplicate semen samples (mean values adopted) were collected at the onset and assessed according to the criteria established by World Health Organization (WHO), 2010. Varicocelectomy was performed for the surgery group and no intervention was given to the observed group. The two groups were followed for 180 days and repeated semen samples were collected and analyzed. The data was computed using GraphPad Prism (v8.0) at an alpha of 0.05. All the men had varicocele and were aged between 46.0 and 67.0 years old. There was no difference between semen parameters among the two groups before the surgery. However, after 180 days of follow-up, all of the semen parameters significantly improved in the surgery group (p < 0.0001), while sperm concentration (p = 0.0068), progressive motility (p = 0.0281), and normal sperm morphology (p = 0.0015) decreased in the observed group. The surgery group had an overall percent increase in total sperm count (840.7%;p = 0.0197), sperm concentration (582.1%;p = 0.0125), total viable sperms (155.2%;p < 0.0001), and normal sperm morphology (110.9%;p < 0.0001) while immotile sperms (-51.71%;p < 0.0001) reduced. A pregnancy rate of 25.3% (19/75) was reported among the surgery group but none was reported among the observed group after 180 days. Microsurgical sub-inguinal varicocelectomy improves semen parameters and hence effective treatment of infertile men with a clinically palpable varicocele. It is recommended to use this choice for similar patients, however, further studies with a larger sample size are needed to provide more evidence to recommend this therapy. 展开更多

To evaluate the effect of methotrexate on collagen-induced arthritis,micro-computed tomography(micro-CT)and histopathological analyses were used in male Wistar rats.Rats were divided randomly into three groups.Group 1 was treated with 0.9% saline,and groups 2 and 3 were boosted with type II collagen.From day 21 to 42,groups 1 and 2 were orally treated with 0.9% saline and group 3 was orally treated with 1.5 mg/kg methotrexate.All rats were sacrificed at day 42 after the first collagen treatment.Micro-CT analyses showed bony parameters,such as bone volume and trabecular number,were decreased in group 2 compared to group 1,and these parameters were recovered in group 3.Histopathological examination and pathological parameter scoring showed that the knee joints of rats in group 2 had severe joint destruction,showing cartilage and bone erosion,enlarged cavities with inflammatory cell infiltration and activation of synovial fibroblasts.By contrast,these changes were reduced in group 3.Taken together,methotrexate treatment showed therapeutic potential in male rat collageninduced arthritis model,and micro-CT analysis and histopathological tools could be integrated to assess the quantification/qualification of arthritic lesions. 展开更多

Antibody is an important part of adaptive immune system and is produced only by B cells.There are five main classes(IgM,IgD,IgG,IgA,IgE)and some subclasses in antibodies.IgM and IgD are produced by mature naïve B cells.On the other hand,IgG,IgA and IgE are produced by activated antigen-specific B cells via class switch recombination(CSR).CSR is the irreversible DNA rearrangement from upstream to downstream classes in immunoglobulin heavy chain genes.Co-stimulations of CD40 ligand(CD40L)and cytokines are required for induction of CSR by activating several transcription factors.These signal transduction pathways involve many protein phosphorylation.Phosphorylation or dephosphorylation of cellular protein is an important kind of post-translational protein modification in intracellular signal transduction.In the fact,more than one third of the intracellular proteins are said to be transiently phosphorylated in human.A protein kinase is an enzyme that catalyzes the addition of phosphate to substrate protein.Whereas,a protein phosphatase catalyzes the removal of phosphate from the substrate.This review focuses on the mechanism of CSR controlled by protein phosphorylation and dephosphorylation.We provide the role of protein kinase and phosphatase in the regulation of class switch recombination. 展开更多

Investigating Spatio-Temporal Pattern of Relative Risk of Tuberculosis in Kenya Using Bayesian Hierarchical Approaches

Proper understanding of global distribution of infectious diseases is an important part of disease management and policy making. However, data are subject to complexities caused by heterogeneities across host classes and space-time epidemic processes. This paper seeks to suggest or propose Bayesian spatio-temporal model for modeling and mapping tuberculosis relative risks in space and time as well identify risks factors associated with the tuberculosis and counties in Kenya with high tuberculosis relative risks. In this paper, we used spatio-temporal Bayesian hierarchical models to study the pattern of tuberculosis relative risks in Kenya. The Markov Chain Monte Carlo method via WinBUGS and R packages were used for simulations and estimation of the parameter estimates. The best fitting model is selected using the Deviance Information Criterion proposed by Spiegelhalter and colleagues. Among the spatio-temporal models used, the Knorr-Held model with space-time interaction type III and IV fit the data well but type IV appears better than type III. Variation in tuberculosis risk is observed among Kenya counties and clustering among counties with high tuberculosis relative risks. The prevalence of HIV is identified as the determinant of TB. We found clustering and heterogeneity of TB risk among high rate counties and the overall tuberculosis risk is slightly decreasing from 2002-2009. We proposed that the Knorr-Held model with interaction type IV should be used to model and map Kenyan tuberculosis relative risks. Interaction of TB relative risk in space and time increases among rural counties that share boundaries with urban counties with high tuberculosis risk. This is due to the ability of models to borrow strength from neighboring counties, such that nearby counties have similar risk. Although the approaches are less than ideal, we hope that our study provide a useful stepping stone in the development of spatial and spatio-temporal methodology for the statistical analysis of risk from tuberculosis in Kenya. 展开更多

Classical regression equations of spirometric parameters are not applicable for diagnosing spirometric abnormalities in adipotic adults

A remarkable increase in the adipotic population urgently needs developing the regression equations for predicting spirometric parameters (SPs), which are specifically applicable to adipotic adults.Unfortunately, however, the reliable equations suitable for adipotic adults have not been reported to date. Recently, Yamaguchi et al have proposed the quantitative method to estimate the effects of adiposity on deciding the SPs incorporating with age-specific contributions of various explanatory,independent variables such as age (A), standing height (H), body weight (BW),and fat fraction of body mass(F). Extending the method proposed by Yamaguchi et al, we attempted to elaborate the novel regression equations applicable for diagnosing the spirometric abnormality in adipotic adults. For accomplishing this purpose, never-smoking, adipotic adults with body mass index (BMI) over 25 kg/m^2 and no respiratory illness were recruited from the general population in Japan (n = 3696, including men: 1890 and women: 1806). Introducing the four explanatory variables of A, H, BW, and F, gender-specific and age-dependent regression equations that allowed for prescribing the SPs in adipotic adults were constructed. Comparing the results obtained for non-adipotic adults (i.e., those with normal BMI), the negative or positive impact of height on SPs was preserved in adipotic adults, as well. However, the negative impact of age on SPs was blunted in adipotic men and the positive effect of BW on SPs was impeded in adipotic men and women. The fat fraction of body mass-elicited negative impact on SPs vanished in adipotic women. These results indicate that the regression equations of SPs for adipotic adults differ significantly from those for nonadipotic adults, leading to the conclusion that the regression equations for nonadipotic adults should not be used while judging the spirometric abnormalities in adipotic adults. 展开更多

Evidence has linked erectile dysfunc... Background: Diabetes is amongst the most researched diseases worldwide due to its debilitating effects on patients. Diabetics have a higher risk of developing erectile dysfunction. Evidence has linked erectile dysfunction severity to increased age, duration of diabetes, the metabolic syndrome and hypogonadism. However, the role of inflammatory markers in the pathogenesis of SD is still unclear. The contributions of hormonal parameters and adipokines to both macro and micro vascular complications are still poorly understood. This study was conducted to provide some evidence of an association between SD, the metabolic syndrome and inflammatory hormones. Method: A total of 130 diabetic males participated in this study. All participants were at least 18 years and were actively engaged in a stable heterosexual relationship for a minimum of two years before they were recruited for this study. The Diabetic men were evaluated using the Golombok Rust Inventory of Sexual Satisfaction for males (GRISS-M). The NCEP-ATP III, IDF and WHO criteria were used to assess the metabolic syndrome. Blood samples were taken for biochemical and hormonal assays. Results: Participants with raised Triglyceride levels showed significantly higher leptin levels for NCEP-ATP III and IDF criteria. Participants with impotence showed significantly lower leptin/adiponectin ratio (p = 0.0184) whilst participant with non-sensuality showed significantly higher values of leptin/adiponectin ratio (p = 0.0103). A significantly higher levels of insulin (p = 0.0265) was recorded in participants with Dissatisfaction problems. Participants with Non communication problems showed significantly higher levels of insulin (p = 0.0419) and lower levels of adiponectin (p = 0.0438) whilst participants with infrequency showed significantly lower levels of adiponectin (p = 0.0042). Conclusion: The relative balance between pro inflammatory and anti-inflammatory mediators of endothelial dysfunction could possibly be essential for the progression of diabetes to SD and the MetS thus a longitudinal study which measures a range of both pro and anti-inflammatory hormones and adipokines as risk factors for SD and the MetS could potentially aid in improvements in assessing the risk of developing these conditions among diabetics. 展开更多

HIV/AIDS spread has opened a Pandora of medical and health problems apart from creating serious socio-psychological trauma for the victims and their families. Despite the governmental effort to curve the growing infection, very little success has been achieved. The northeastern region of India presents a very peculiar situation due to recurrent insurgency in all its states for long time and thus it became instrumental in paralyzing the social and health machineries to an extent, which still deteriorated the futile national effort to check the deadly infection. The study tried to understand the level of awareness and correct knowledge among the underprivileged scheduled tribes and scheduled castes women in the age group 15 -44 by exploring data from district level of Rapid Household Reproductive and Child Health Project Phase I (1998) and Phase II (1999) for Assam, Manipur and Nagaland and suggested a strategy to control the infection in the region. It is found that HIV/AIDS awareness was 59% among the women in all the three states with very few among them who had correct understanding of its prevention. A pronounced difference in the level of awareness was observed with respect to age, economy and residential status of the respondents. 展开更多

This study aims to determine the effect of microsurgical sub-inguinal varicocelectomy on gonadal function among men reporting sexual dysfunction in Ghana. Methods: This was an intervention study conducted at the Tamale Teaching Hospital from September 2017 to August 2021. A total of 103 participants were randomized into two groups;the surgery group (n = 52) and the observed group (n = 51). Venous blood samples were collected at baseline, varicocelectomy was performed for the surgery group, and no intervention was given to the other. Blood samples were subsequently collected at 12-, 24-, 36-, and 48-month intervals for assay of serum total testosterone, FSH, and LH. The data were analyzed in GraphPad Prism (v8.0) at an alpha value of 0.05. Results: All the participants had varicocele and were aged between 55.0 to 69.0 years old. Numerous reports have shown the close relationship between salivary levels of Streptococcus mutans and dental caries. As S. mutans, is considered to be the principle etiological agent of dental caries, the development of a quick and convenient method for detection and quantification of these bacteria from patient saliva samples would simplify diagnosis and treatment. The purpose of this study was to compare a new means of quantifying bacteria using FTATM Elute cards and Real-Time Polymerase Chain Reaction to a conventional culture-based assay using oral S. mutans as a model sample. A total of 60 different saliva samples were investigated. The results show a significant negative correlation between the two methods, with a correlation coefficient of -0.577 (Spearman’s Correlation) and p < 0.01. The method demonstrates a high sensitivity, specificity and reliable quantitative results, covering a large range of bacterial concentrations. 展开更多

In the present study, we investigated the effects of hypothyroidism on the morphology of astrocytes and microglia in the hippocampus of Zucker diabetic fatty rats and Zucker lean control rats. To induce hypothyroidism, Zucker lean control and Zucker diabetic fatty rats at 7 weeks of age orally received the vehicle or methimazole, an anti-thyroid drug, treatment for 5 weeks and were sacrificed at 12 weeks of age in all groups for blood chemistry and immunohistochemical staining. In the me- thimazole-treated Zucker lean control and Zucker diabetic fatty rats, the serum circulating triiodo- thyronine (T3) and thyroxine ('I"4) levels were significantly decreased compared to levels observed in the vehicle-treated Zucker lean control or Zucker diabetic fatty rats. This reduction was more prominent in the methimazole-treated Zucker diabetic fatty group. Glial fibrillary acidic protein im- munoreactive astrocytes and ionized calcium-binding adapter molecule 1 (Iba-1)-immunoreactive microglia in the Zucker lean control and Zucker diabetic fatty group were diffusely detected in the hippocampal CA1 region and dentate gyrus. There were no significant differences in the glial fibril- lary acidic protein and Iba-1 immunoreactivity in the CA1 region and dentate gyrus between Zucker lean control and Zucker diabetic fatty groups. However, in the methimazole-treated Zucker lean control and Zucker diabetic fatty groups, the processes of glial fibrillary acidic protein immunoreac- tive astrocytes and Iba-1 immunoreactive microglia, were significantly decreased in both the CA1 region and dentate gyrus compared to that in the vehicle-treated Zucker lean control and Zucker diabetic fatty groups. In this study, we investigated the effects of streptozotocin-induced type 1 diabetes on antioxi- dant-like protein-1 immunoreactivity, protein carbonyl levels, and malondialdehyde formation, a marker for lipid peroxidation, in the hippocampus. For this study, streptozotocin (75 mg/kg) was intraperitoneally injected into adult rats to induce type 1 diabetes. The three experimental pa- rameters were determined at 2, 3, 4 weeks after streptozotocin treatment. Fasting blood glucose levels significantly increased by 20.7-21.9 mM after streptozotocin treatment. The number of antioxidant-like protein-1 immunoreactive neurons significantly decreased in the hippocampal CA1 region, but not the dentate gyrus, 3 weeks after streptozotocin treatment compared to the control group. Malondialdehyde and protein carbonyl levels, which are modified by oxidative stress, significantly increased with a peak at 3 weeks after malondialdehyde treatment, and then decreased 4 weeks after malondialdehyde treatment. These results suggest that neurons in the hippocampal CA1 region, but not the dentate gyrus, are susceptible to oxidative stress 3 weeks after malondialdehyde treatment. 展开更多

Background: Some evidence has shown that the prevalence of hypoactive sexual desire and erectile dysfunction (ED) is associated with testosterone levels whilst higher levels of testosterone have been reported to increase the frequency of intercourse. Available evidence points towards an etiologic role for hypogonadism in the causation of diabetes, metabolic syndrome (Mets) and sexual dysfunction (SD) even though the exact pathophysiological linkage is yet to be fully elucidated. This study therefore sought to evaluate the impact of testosterone as well as its bioactive components on both the MetS and SD. Method: Diabetic men engaged in a stable heterosexual relationship for at least 2 years were recruited for this study. Participants were at least 18 years and provided an informed and signed consent to partake in this study. Fasting blood samples were taken from the participants for biochemical and hormonal assay. The participants were then evaluated using the Golombok Rust Inventory of Sexual Satisfaction for males (GRISS-M). Metabolic syndrome was assessed using the NCEP-ATP III, IDF and WHO criteria. All data analyses were performed using the SPSS software, version 11.0 systat, Inc. Germany and GraphPad Prism, version 5.0, San Diego California, USA. Results: The mean total, free and bioavailable testosterone as well as SHBG recorded among the participants was 7.10 ± 1.23 ng/ml, 0.10 ± 0.01 ng/ml, 7.01 ± 1.41 ng/ml and 4.33 ± 1.12 nmol/l respectively. Subjects with the MetS showed significantly lower SHBG levels in comparison with subjects without the MetS. Participants with raised triglyceride levels showed significantly lower levels of total, free and bioavailable testosterone when compared to participant without raised triglycerides. Participants with problems of avoidance and infrequency of sexual activity showed significantly lower levels of total, free and bioavailable testosterone when compared to participants without avoidance or infrequency problems. The +SD/+MetS group recorded the lowest SHBG levels. Conclusion: Testosterone and its bioactive components are associated with male libido as well as the metabolic syndrome. Low levels of free and bioavailable testosterone are true determinants of the MetS in men. 展开更多
