This study examines the influence of non-ventilation (NV) conditi...The exchange of oxygen (O2) and carbon dioxide (CO2) within an incubator has a significant impact on embryonic development (ED) and hatching processes. This study examines the influence of non-ventilation (NV) conditions during the first ten days of incubation at high altitudes on Leghorn hens hatching eggs. Five hundred four hatching eggs were equally divided into three treatment groups and placed in twelve incubators (R = 4). The first group was subjected to standard ventilated conditions (V) during the setting phase. The ventilation inlet holes of the remaining incubators in the NV treatments were closed with either micropore (M) or polypropylene (P) tape, referred to as NVM and NVP groups, respectively. These two different airtight settings were intended to allow for a gradual rise in CO2 naturally generated by the embryos. Results indicate that carbon dioxide concentration gradually increased during the first half of incubation, reaching 1.42% in the NVM group and 1.20% in the NVP group, while the V condition group remained at 0.15%. From 10 days of incubation onwards, normal V conditions were restored in all incubators. The highest hatchability of fertile eggs (HFE) was shown by the NVP group (55.7%), followed by the V (52.6%) and NVM (38.6%) groups. The NVP group showed a greater yolk-free body mass (YFBM) from 10 days of incubation until the hatch basket transfer. NV conditions during the first 10 days of incubation at high altitude produced higher YFBM with gradually decreasing yolk sac mass. In comparison to the NVM and V conditions, the particular NVP condition showed a beneficial impact on the quality of hatched chicks. Sustaining NVP condition (1.2% of CO2) throughout the first half of incubation at high altitude generated the optimal environment in the incubator ensuring the best hatchability results. This study highlights how important it is for hatchery managers to recognize the influence of low O2 and high levels of CO2 on the development trajectories of Leghorn embryos during early incubation at high altitudes.展开更多
The aim of the present study was to compare the ixodicide efficacy of 1% permethrin applied as a shampoo-bath in single treatment and a combined formulation of 2% permethrin + 1% piperonyl butoxide applied as powder i...The aim of the present study was to compare the ixodicide efficacy of 1% permethrin applied as a shampoo-bath in single treatment and a combined formulation of 2% permethrin + 1% piperonyl butoxide applied as powder in a single treatment against Rhipicephalus sanguineus ticks, in naturally infested dogs. Eighteen adult cross-mixed dogs, harboring to at least 20 ticks/animal, were selected on pre-treatment live tick counts. On day 0, the dogs were then divided in 3 groups of 6 animals each, for treatment: Group 1 (G1) received 1% permethrin applied as shampoo-bath (approximately 10 ml/kg bw), according to label instructions. G2 was treated with 2% permethrin + 1% piperonyl butoxide applied as powder, and rubbed as massage on the body, as recommended by the producer. G3 served as a non-treated control. After the treatment, the dogs were checked out for live ticks counting on days 0, 1, 7, and 14. The ixodicide efficacy was assessed based on the percentage reductions of ticks on the days analyzed post-treatment, with respect to the untreated control. Results showed that permethrin alone removed ticks on 81%, 93.1% and 89.6% for days 1, 7 and 14, respectively. The combined formulation of 2% permethrin + 1% piperonyl butoxide exerted a better efficacy of 98.8, 97.7 and 97.0%, respectively. It is concluded that this combined formulation was highly effective at rapidly repelling and killing Riphicephalus sanguineus ticks on naturally infected dogs, showing a significant effect on days 1, 7 and 14 after tick exposure. The sustained high and quick level of efficacy of this combination may well interfere with the transmission of Riphicephalus sanguineus ticks. It is concluded that the sustained high level of efficacy of the permethrin + piperonyl butoxide combination may well exert an additive effect and could interfere with the transmission of Riphicephalus sanguineus ticks.展开更多

Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. The engineering of a human RPE monolayer with these artificial BM, emulating the in vivo retina, arouse their potential subretinal implantation in patients with wet age-related macular degeneration (AMD) where there is a rupture of the Bruch’s membrane.展开更多

Correlations between fat content, organoleptic characteristics and lipid profiles were also investigated. Methods: A sample of 436 largely unrelated purebred bulls belonging to 15 breeds and reared under comparable management conditions was analyzed. Phenotypic data -including fatness score, fat percentage, individual fatty acids (FA) profiles and sensory panel tests- and genotypic information from 11 polymorphisms was used. Results: The correlation coefficients between muscle total lipid measurements and absolute vs. relative amounts of polyunsaturated FA (PUFA) were in opposite directions. Increasing carcass fat leads to an increasing amount of FAs in triglycerides, but at the same time the relative amount of PUFAs is decreasing, which is in concordance with the negative correlation obtained here between the percentage of PUFA and fat measurements, as well as the weaker correlation between total phospholipids and total lipid muscle content compared with neutral lipids. Concerning organoleptic characteristics, a negative correlation between fiavour scores and the percentage of total PUFA, particularly to n-6 fraction, was found. The correlation between juiciness and texture is higher than with flavour scores. The distribution of SNPs plotted by principal components analysis (PCA) mainly reflects their known trait associations, although influenced by their specific breed allele frequencies. Conclusions: The results presented here help to understand the phenotypic and genotypic background underlying variations in FA composition and sensory parameters between breeds. The wide range of traits and breeds studied, along with the genotypic information on polymorphisms previously associated with different lipid traits, provide a broad characterization of beef meat, which allows giving a better response to the variety of consumers' preferences. Also, the development and implementation of low-density SNP panels with predictive value for economically important traits, such as those summarized here, may be used to improve production efficiency and meat quality in the beef industry.展开更多

Background:Estimating inbreeding,which is omnipresent and inevitable in livestock populations,is a primary goal for management and animal breeding especially for those interested in mitigating the negative consequence...Background:Estimating inbreeding,which is omnipresent and inevitable in livestock populations,is a primary goal for management and animal breeding especially for those interested in mitigating the negative consequences of inbreeding.Inbreeding coefficients have been historically estimated by using pedigree information;however,over the last decade,genome-base inbreeding coefficients have come to the forefront in this field.The Pura Raza Espanola(PRE)horse is an autochthonous Spanish horse breed which has been recognised since 1912.The total PRE population(344,718 horses)was used to estimate Classical(F),Ballou’s ancestral,Kalinowski’s ancestral,Kalinowski’s new and the ancestral history coefficient values.In addition,genotypic data from a selected population of 805 PRE individuals was used to determine the individual inbreeding coefficient using SNP-by-SNP-based techniques(methods of moments-FHOM-,the diagonal elements of the genomic-FG-,and hybrid matrixes-FH-)and ROH measures(FRZ).The analyse of both pedigree and genomic based inbreeding coefficients in a large and robust population such as the PRE horse,with proven parenteral information for the last 40 years and a high degree of completeness(over 90%for the last 70 years)will allow us to understand PRE genetic variability better and the correlations between the estimations will give the data greater reliability.Results:The mean values of the pedigree-based inbreeding coefficients ranged from 0.01(F for the last 3 generations-F3-)to 0.44(ancestral history coefficient)and the mean values of genomic-based inbreeding coefficients varied from 0.05(FRZ for three generations,FH and FHOM)to 0.11(FRZ for nine generations).Significant correlations were also found between pedigree and genomic inbreeding values,which ranged between 0.58(F3 with FHOM)and 0.79(F with FRZ).In addition,the correlations between FRZ estimated for the last 20 generations and the pedigree-based inbreeding highlight the fact that fewer generations of genomic data are required when comparing total inbreeding values,and the opposite when ancient values are calculated.Conclusions:Ultimately,our results show that it is still useful to work with a deep and reliable pedigree in pedigreebased genetic studies with very large effective population sizes.Obtaining a satisfactory parameter will always be desirable,but the approximation obtained with a robust pedigree will allow us to work more efficiently and economically than with massive genotyping.展开更多

Di...Infections produced by feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV), two of the most prevalent pathogens in cats, range from passing unnoticed to presenting a wide variety of clinical signs. Different epidemiological, clinical, hematological and virological parameters were analyzed in 78 FIV- and/or FeLV-infected cats. FeLV-infected (FeLV+) cats were considerably younger than FIV-infected (FIV+) cats, and in general were seen to have a more severe disease than FIV+ cats. Around one third of the cats presented anemia, and neutropenia was also frequently observed. Though a higher percentage of FIV+ than FeLV+ cats had altered leukocyte counts, FeLV+ cats had altered counts of both neutrophils and lymphocytes more frequently than FIV+, which usually presented only either altered neutrophils or lymphocyte counts. Virological markers were only detected in FeLV+cats, either as mono- or dual-infection, and correlated with the severity of the disease, but not in FIV+ cats. In conclusion, these results suggest that FeLV affects more blood cell types and provokes death of affected animals at a much earlier age than FIV and that the severity of the disease seemed to depend on the viral status of the cat.展开更多

Some plant extracts, such as soluble wheat extract, have been shown to be effective in inhibiting or destroying the biofilm of certain micro-organisms under specific conditions. The objective of this study is to evaluate the capacity of the pathogen to form biofilm on different surfaces used in livestock, as well as to evaluate the anti-biofilm capacity of the soluble wheat extract against <i>S. aureus</i> on these surfaces. The inhibition potential of inhibition or destruction of biofilm was tested in vitro. Wheat extract at a concentration of 0.29 mg/100mL showed anti-biofilm activity on <i>S. aureus</i>, inhibiting its formation, as well as destroying it greatly after a contact time of 24 hours, on those surfaces where the microorganism presents more adhesion capacity.展开更多
Verticillium wilt is one of the most significant agricultural diseases in the world, in view of the fact that not only does it affec olive groves but also a wide variety of fruit, vegetable and ornamental plants. Curr...Verticillium wilt is one of the most significant agricultural diseases in the world, in view of the fact that not only does it affec olive groves but also a wide variety of fruit, vegetable and ornamental plants. Currently, the most efficient and economical method of control involves the use of plant species that are resistant to the disease. However, as there are few varieties of olive tree with this characteristic, early diagnosis and the replacement of affected trees are key strategies to combat the disease. The present research proposes the use of a canine unit that is specifically trained to detect Verticillium dahliae as an early detection method for the fungus in olive groves. For the odorous samples are produced in the laboratory from V. dahliae, it is calculated that the dog dis-plays a level of sensitivity of 97% and specificity of 95%, and an initial field assay shows that the sensitivity and specificity are constant when working with infected olive trees. In view of the fact that there is currently no curative treatment for this disease and the affected plants are completely lost, fast on-site detection provides significant advantages in terms of preventing the transmission of the disease. Moreover, it also enables focalized treatment to be provided on plots of land and plants and, in combination with the latest prevention methods, real-time detection of affected olive trees enables a reduction in the costs involved in vaccination, thereby contributing overall to a reduction in the financial losses caused by verticillium wilt.展开更多
The aim of the present study was to evaluate the impact of the technological level in small-scale dual-purpose cow-calf <em>Bos indicus</em> female operations on the resumption of the ovarian activity post...The aim of the present study was to evaluate the impact of the technological level in small-scale dual-purpose cow-calf <em>Bos indicus</em> female operations on the resumption of the ovarian activity post-partum. A total of 13 small scale dual-purpose <em>Bos indicus</em> × <em>Bos taurus</em> operations were included in this experiment and they were classified according to their technological status as: high technological status (HT), medium technological status (MT) and low technological status (LT). Ninety-three mature cows were treated with either a CIDR (controlled internal drug release device) with estradiol benzoate (CIDR+EB) or alone (CIDR). At day 9 all animals received intravaginally a CIDR for nine days. At day 0, the CIDR was withdrawn and 24 hours later one dose of estradiol benzoate was administered to 51 cows (17 in HT, 17 in MT and 17 in LT), the remaining animals (n = 42) did not receive the estradiol benzoate administration (14 in HT, 13 in MT and 15 in LT). Comparisons were performed on serum progesterone concentrations after CIDR treatment. By day 7 after implant withdrawal, the proportion of cows that resume ovarian activity detected by progesterone concentration increased in all technological level farms, no statistical main effect was observed within technological level. By day 17 there was a significant main effect of technological level (P = 0.05) on the proportion of animal that resumed the estrous cycles, but neither a main effect of treatment (P = 0.97) nor an interaction between technological level and treatment (P = 0.98). Furthermore, technological level of the farm showed a tendency (P = 0.07) to affect that resume the estrous activity, and a significant effect of treatment was observed (P = 0.005) but no interaction between technological level and treatment. The proportion of cows that showed estrus was different across technological level (P = 0.02), the highest proportion of cows showing estrus regardless of treatment was in the HT: 90% (40% CIDR and 50% CIDR+EB), MT: 50% (13.3% CIDR and 36.7% CIDR+EB) and in the LT: 50% (18.8% CIDR and 31.3% CIDR+EB). In conclusion, the combination of a progestogen and estradiol benzoate resulted in a high proportion of cows that were induced to resume the ovarian activity and this treatment was particularly beneficial in the medium and low technological status of the farms.展开更多

The serological profile of the goat population, from nine...Mycoplasma mycoides subsp. capri is the causative agent of severe and acute respiratory problems in goats, which spreads rapidly and represents high mortality. The serological profile of the goat population, from nine regions in seven states of Mexico, was screened by the Complement Fixation test (CF) in sera from asymptomatic goats and animals with mild respiratory symptoms. Sera and nasal swabs of 827 goats were collected for the isolation of the organism. An antiserum was prepared against a previously isolated field strain of Mycoplasma mycoides subsp. capri. CF Antibody titers were associated with the results of the isolates to determine the cutoff point. The CF was considered as positive if its result was ≥1/16. The CF registered 251 positive goats (30.35%) and 576 (69.65%) negative;the test showed high sensitivity (93.33%) and specificity (72.27%). In the specific case of diagnosis for mycoplasmosis associated with respiratory problems in goats, the CF proved to be a good diagnosis test, this study determined that 30% of the goat population showed antibody titers against Mycoplasma mycoides subsp. capri and revealed those animals who have had contact with this microorganism during their lives regardless of the presence or absence of respiratory symptoms.展开更多

Without AGPs, there has been a rise in diseases that were traditionally controlled by subtherapeutic levels of antibiotics in the diet. This has impacted the health of commercial poultry and has been a significant cost to poultry producers. To mitigate this, the industry has started to investigate alternatives to antibiotics to treat these forthcoming health issues, such as necrotic enteritis (NE). NE is an enteric disease caused by an over proliferation of toxigenic Clostridium perfringens (CP) in the gastrointestinal tract. Although CP is a commensal in the avian intestinal tract, dysbiosis caused by inflammation and impaired intestinal integrity facilitates uncontrolled replication of CP. Infectious agents, such as Eimeria maxima, appear to be a predominant predisposing factor that promotes NE. However, non-infectious stressors, including dietary changes, have also been associated with NE to some degree. As a result of increased pressure to restrict the use of antibiotics, there is a need for research evaluating the efficacy of alternatives, such as plant-derived essential oils, as potential tools to mitigate NE in commercial poultry flocks. The aim of this study is to review the effects of essential oils as an alternative to antibiotics to reduce the incidence and severity of necrotic enteritis in broiler chickens.展开更多

The work was carried out at the FMVZ-BUAP Gen...The purpose of the study was to evaluate the sperm viability of semen infected with PRRSV viral particles, observing the effect of the Virus on the motility of boar sperm. The work was carried out at the FMVZ-BUAP Genetics and Reproduction Laboratory. 5 stallions were used. Each sample contained 1 × 10<sup>6</sup> sperm, the PRRS virus strain was ATCC-VR-2332 (0, 10<sup>2</sup>, 10<sup>4</sup> and 10<sup>6</sup> copies of RNA/mL in triplicate), it was observed daily at the CASA;Hamilton Thorne<sup>®</sup>. Cells with MT (P < 0.05) on days 1, 3, 5, 7 and 10 of evaluation with 201 ± 7.3, 167 ± 10.1, 165 ± 14.6, 134 ± 8.2 and 120 ± 8.8, respectively. The % MP between control and virus concentrations (P ≥ 0.05). The LCV on day 1 and 7 PI at 10X<sup>2</sup> and 10X<sup>6</sup> (P < 0.05) vs control. In the Correlation Matrix, where it is observed that there is a correlation between VSL and VAP, VSL and VCL, VCL and ALH, VAP with ALH. There is a correlation of VSL and ALH, STR and ALH. In this study there were (P ≤ 0.01) in the VCL, in the concentrations (10<sup>2</sup>) 162.81 ± 10.65 and (10<sup>6</sup>) 177.12 ± 5.77 vs 193.04 ± 4.62 of control. This indicates that altering these parameters would be related to fertility and the PRRS virus affects the LCV. Regarding the VSL, it was observed that the sperm infected with viruses 10<sup>2</sup>, 10<sup>4</sup> and 10<sup>6</sup> of 48.00 ± 3.38, 49.88 ± 1.83 and 50.55 ± 2.24 Vs. 56.66 ± 1.68 of control respectively, the control would have greater possibilities of fertilizing the oocyte. In this study, it was found (P ≤ 0.01) in the VAP with 102 of 77.26 ± 5.16, 10<sup>4</sup> with 83.35 ± 2.41 and 10<sup>6</sup> with 81.29 ± 3.14 vs the control with 90.56 ± 2.07. Regarding the ALH there is (P < 0.05) a 10<sup>4</sup> with 8.70 ± .26 and 10<sup>6</sup> with 9.64 ± 0.23 vs control 8.50 ± 0.27. The presence of different concentrations of PRRSV in boar semen induces changes in different types of sperm motility. Infection of ejaculates with the PRRS virus affects sperm motility on days 1, 3, 5, 7, and 10 post-infections.展开更多

These parasites target the gastrointestinal tract in different types of hosts, causing sores in the intestinal lining. The presence of these lesions reduces the animal’s ability to digest and absorb nutrients, significantly impacting their overall performance. The current study aimed to explore the potential correlation between seasonal variations and the incidence of Eimeria spp-induced lesions in broiler chickens’ gastrointestinal tracts in Brazil from 2012 to 2018. A total of 8,607 broiler chickens, aged 14 to 42 days, were sampled from 103 poultry integrated companies in Brazil to conduct intestinal health examinations. The sampling process involved selecting 3 to 6 chickens from each poultry house for examination. The assessment included various abnormalities such as shedding of intestinal cells and excessive fluid and mucus presence, thickening and tension of the intestines, food movement, roughened mucosal surface resembling a Turkish towel in the small intestine, tissue death, duodenal inflammation, intestinal inflammation, gizzard erosion, presence of worms and bedding material, and ingestion of mealworms. During the seasons, winter exhibited the highest average occurrence of Eimeria maxima microorganisms at 52.83%, with E. acervulina following closely at 26.42% in second place. In spring, E. maxima had an occurrence of 11.31%, while in fall, E. tenella had the lowest occurrence at 6.74%. When analyzing the seasonal occurrence of Eimeria, it was observed that E. maxima micro was more common during winter compared to summer (P = 0.0491). However, no discernible variation was observed in the occurrence of the remaining species across different seasons. Research findings suggest that subclinical coccidiosis is most prevalent during the winter season in Brazil. Likewise, clinical disease caused by E. acervulina is also prevalent during this time. In contrast, E. maxima is more likely to cause clinical disease in the spring, whereas E. tenella is more commonly associated with clinical disease in the fall. Lesions induced by Eimeria spp. are associated with factors influencing the overall health of broiler intestines. These findings allow for the utilization of seasonal metrics in disease management, thereby reducing economic losses associated with the condition.展开更多

The objective of this study was to evaluate differences in ruminal bacteria, biopolymer hydrolyzing enzyme activities, and overall performance between the most-and the leastefficient dairy cows during the peripartal period. Twenty multiparous Holstein dairy cows with daily ad libitum access to a total mixed ration from d-10 to d 60 relative to the calving date were used. Cows were classified into most-efficient(i.e. with low RFI, n = 10) and least-efficient(i.e. with high RFI, n = 10) based on a linear regression model involving dry matter intake(DMI), fat-corrected milk(FCM), changes in body weight(BW), and metabolic BW.Results: The most-efficient cows had ~ 2.6 kg/d lower DMI at wk 4, 6, 7, and 8 compared with the least-efficient cows. In addition, the most-efficient cows had greater relative abundance of total ruminal bacterial community during the peripartal period. Compared with the least-efficient cows, the most-efficient cows had 4-fold greater relative abundance of Succinivibrio dextrinosolvens at d-10 and d 10 around parturition and tended to have greater abundance of Fibrobacter succinogenes and Megaspheara elsdenii. In contrast, the relative abundance of Butyrivibrio proteoclasticus and Streptococcus bovis was lower and Succinimonas amylolytica and Prevotella bryantii tended to be lower in the most-efficient cows around calving. During the peripartal period, the most-efficient cows had lower enzymatic activities of cellulase, amylase, and protease compared with the least-efficient cows.Conclusions: The results suggest that shifts in ruminal bacteria and digestive enzyme activities during the peripartal period could, at least in part, be part of the mechanism associated with better feed efficiency in dairy cows.展开更多

Methods and Results: To determine the optimal levels of incorporation of several cry oprotectants (skim milk, sucrose and trehalose) in the freeze-drying process of Lactobacillus plantarum, a range of experiments based on a Rotational Central Composite design (CCD) were conducted. The results were adjusted to a quadratic model, resulting in the presence of interaction between the different variables. Solving a regression equation, we obtained the optimum concentrations of cryoprotective agents: 24.06% milk powder, 6.22% sucrose, 5.63% trehalose. To visualize the interactions between the three variables involved in the study, Design Expert? software was used. Conclusions: The analysis reveals that while trehalose has a direct effect on the viability of L. plantarum, skim milk and sucrose exert quadratic effects. There are also interactions between cryoprotectants, which emphasize the synergies produced between milk and sucrose and between sucrose and trehalose, which allows maintaining the viability of L. plantarum. Significance and Impact of the Study: The addition of new oligosaccharides as trehalose in premixtures for functional feed can maintain the viability of L. plantarum during longer periods of time, ensuring the proper administration of probiotics to their destinations.展开更多

In addition, the antioxidant activity was assessed by the CUPRAC method and the free-radical scavenging activity was determined using the antioxidant-promoting decay of the stable free-radical DPPH. Caffeine and the most important catechins were identified using RP-HPLC previously validated method. Results from this study suggest that teas, even though they content similar amounts of caffeine, differ considerably in polyphenolic content and therefore in antioxidant and free-radical scavenging activities, depending on the part of the plant used and the process applied to the material for the preparation of the final tea presentation. Correlations among Folin-Ciocalteau’s reactivity of tea samples, the neocuproine reactivity, the DPPH decolourization and the HPLC analysis suggest that the antioxidant activity is due essentially to polyphenolic compounds present in teas, mainly EGCG. The best extraction method of tea powder was the one indicated by dealers. In general, the richest samples belong to green teas and the poorest samples belong to the black teas.展开更多
The objective of this work was to determine the effect of transport time (up to 11 hours) on animal welfare. 540 animals (cows, bulls and calves), three transport times of 2, 4 to 6, and 10 to 11 hours, and two sp...The objective of this work was to determine the effect of transport time (up to 11 hours) on animal welfare. 540 animals (cows, bulls and calves), three transport times of 2, 4 to 6, and 10 to 11 hours, and two space allowances (2 m2 and 1.5 m2 per animal for cows and bulls; and 1.2 m2 and 0.8 m2 per animal for calves) were used for the experiment during transport from farms to the abattoir in Uppsala, Sweden. Measurements were made on five animals on each trip. Stress response parameters considered were cortisol, glucose, lactate, creatine kinase and heart rate. Blood samples were taken before and after transport. Heart rate sensors were mounted on the animals at least 30 minutes before loading and measurements were made continuously from farms to the abattoirs until stunning. The results of heart rate measurement indicated that loading, un-loading and forced movement in the stunning box were the most stressful events. However, the results of statistical analysis confirmed that transport time doesn't have significant effect (P 〈 0.05) on heart rate. Concentration level of cortisol increased by 10 folds during short transport. However, cortisol concentration decreased with an increase of transport times (P 〈 0.01). Glucose concentrations increased with transport time in all animal categories (P ≤ 0.01). Transport time has significant effect on concentration levels of glucose (P ≤ 0.01), creatine kinase (P 〈 0.001) and lactate (P 〈 0.01) particularly after 6 hours journey time.展开更多

Treatments consisted o...One hundred ninety-two crossbred steers (229 kg) were used to evaluate the influence of metabolizable amino acid intake on growth performance and health during the initial 42-d receiving period. Treatments consisted of four levels of metabolizable lysine (23, 24, 25 and 26 g/kg diet DM). Morbidity averaged 36%, and was not affected (P > 0.20) by treatments. No steers died during the study. Increasing the metabolizable lysine supply increased DMI, ADG, gain efficiency, and dietary NE (linear effect, P < 0.01). Metabolizable amino acid supply of the basal diet was determined using 6 steers (214 kg) with cannulas in the rumen and proximal duodenum. Metabolizable amino acid supply of the diet was in close agreement with expected (R2 = 0.99;P < 0.0001) based on NRC (2000) [12] Level 1 model. The metabolizable amino acid supplies for treatments in Trial 1 were estimated by adding tabular metabolizable amino acid values for the respective supplemental proteins (cottonseed meal and fish meal) to the observed metabolizable amino acid supply of the basal diet. Treatment effects on metabolizable lysine supply explained 99% of the variation (P < 0.01) in ADG, and 91% of the variation in observed versus expected dietary NE. The biological value for the intestinal chyme was determined based on chemical score technique, using bovine tissue as the reference protein. Accordingly, methionine and lysine were closely co-limiting amino acids having ratios of 77% and 79%, respectively. We conclude that current NRC standards reliably predict both requirements and supplies of metabolizable amino acids for feedlot calves. Diet formulations that do not meet the metabolizable amino acid requirements may depress both ADG and the partial efficiency of utilization of metabolizable energy for maintenance and gain.展开更多
文摘The exchange of oxygen (O2) and carbon dioxide (CO2) within an incubator has a significant impact on embryonic development (ED) and hatching processes. This study examines the influence of non-ventilation (NV) conditions during the first ten days of incubation at high altitudes on Leghorn hens hatching eggs. Five hundred four hatching eggs were equally divided into three treatment groups and placed in twelve incubators (R = 4). The first group was subjected to standard ventilated conditions (V) during the setting phase. The ventilation inlet holes of the remaining incubators in the NV treatments were closed with either micropore (M) or polypropylene (P) tape, referred to as NVM and NVP groups, respectively. These two different airtight settings were intended to allow for a gradual rise in CO2 naturally generated by the embryos. Results indicate that carbon dioxide concentration gradually increased during the first half of incubation, reaching 1.42% in the NVM group and 1.20% in the NVP group, while the V condition group remained at 0.15%. From 10 days of incubation onwards, normal V conditions were restored in all incubators. The highest hatchability of fertile eggs (HFE) was shown by the NVP group (55.7%), followed by the V (52.6%) and NVM (38.6%) groups. The NVP group showed a greater yolk-free body mass (YFBM) from 10 days of incubation until the hatch basket transfer. NV conditions during the first 10 days of incubation at high altitude produced higher YFBM with gradually decreasing yolk sac mass. In comparison to the NVM and V conditions, the particular NVP condition showed a beneficial impact on the quality of hatched chicks. Sustaining NVP condition (1.2% of CO2) throughout the first half of incubation at high altitude generated the optimal environment in the incubator ensuring the best hatchability results. This study highlights how important it is for hatchery managers to recognize the influence of low O2 and high levels of CO2 on the development trajectories of Leghorn embryos during early incubation at high altitudes.
文摘The aim of the present study was to compare the ixodicide efficacy of 1% permethrin applied as a shampoo-bath in single treatment and a combined formulation of 2% permethrin + 1% piperonyl butoxide applied as powder in a single treatment against Rhipicephalus sanguineus ticks, in naturally infested dogs. Eighteen adult cross-mixed dogs, harboring to at least 20 ticks/animal, were selected on pre-treatment live tick counts. On day 0, the dogs were then divided in 3 groups of 6 animals each, for treatment: Group 1 (G1) received 1% permethrin applied as shampoo-bath (approximately 10 ml/kg bw), according to label instructions. G2 was treated with 2% permethrin + 1% piperonyl butoxide applied as powder, and rubbed as massage on the body, as recommended by the producer. G3 served as a non-treated control. After the treatment, the dogs were checked out for live ticks counting on days 0, 1, 7, and 14. The ixodicide efficacy was assessed based on the percentage reductions of ticks on the days analyzed post-treatment, with respect to the untreated control. Results showed that permethrin alone removed ticks on 81%, 93.1% and 89.6% for days 1, 7 and 14, respectively. The combined formulation of 2% permethrin + 1% piperonyl butoxide exerted a better efficacy of 98.8, 97.7 and 97.0%, respectively. It is concluded that this combined formulation was highly effective at rapidly repelling and killing Riphicephalus sanguineus ticks on naturally infected dogs, showing a significant effect on days 1, 7 and 14 after tick exposure. The sustained high and quick level of efficacy of this combination may well interfere with the transmission of Riphicephalus sanguineus ticks. It is concluded that the sustained high level of efficacy of the permethrin + piperonyl butoxide combination may well exert an additive effect and could interfere with the transmission of Riphicephalus sanguineus ticks.
文摘Purpose: The primary aim of this paper is to design, create, and improve functional and artificial Bruch’s membranes (BM) using bioengineering techniques, which can be applied in the treatment of maculopathies by supporting the growth and maintenance of retinal pigment epithelium (RPE) cells, thereby potentially enabling subretinal implantation in patients. Methods: We fabricated by electrospinning ultrathin 3D nanofibrous membranes from Polycaprolactone (PCL), and different concentrations of gelatin (5%, 15% and 30%). ARPE-19 cells were seeded onto these artificial membranes. MTT assays were performed in order to evaluate ARPE-19 cell viability and cytotoxicity. IF assays were realized to observe the ARPE-19 cells onto each membrane. Ultrastructure of the modified Bruch’s membrane and ARPE-19 morphology after 25 days of culture were studied with transmission and scanning electron microscopy. To evaluate expression changes in markers of ARPE-19 (RPE65 and ZO-1) qRT-PCR assays were realized. Data from three independent experiments were pooled and expressed as the mean SD. A confidence level of P 0.05 was considered to be statistically significant. Results ARPE-19 cells grew on PCL/Gelatin membranes mainly in PCL/30% gelatin combination, which had not cytotoxic effect. RPE65/cytokeratin-18/ and actin-positive ARPE-19 cells formed a correctly orientated monolayer of polygonal cells with morphological polarity. The apical cell surfaces exhibited abundant protruding microfolds. Moreover, zones of polygonal border look as if ARPE-19 cells fused among, suggesting the presence of tight junctions. The expression of RPE65 and ZO-1 was unchanged. Conclusions: PCL/30% Gelatine membranes may imitate the natural BM to such extent that they support RPE-cells and exhibited RPE-like morphology. The engineering of a human RPE monolayer with these artificial BM, emulating the in vivo retina, arouse their potential subretinal implantation in patients with wet age-related macular degeneration (AMD) where there is a rupture of the Bruch’s membrane.
文摘Background: Consuming moderate amounts of lean red meat as part of a balanced diet valuably contributes to intakes of essential nutrients, in this study, we merged phenotypic and genotypic information to characterize the variation in lipid profile and sensory parameters and to represent the diversity among 15 cattle populations. Correlations between fat content, organoleptic characteristics and lipid profiles were also investigated. Methods: A sample of 436 largely unrelated purebred bulls belonging to 15 breeds and reared under comparable management conditions was analyzed. Phenotypic data -including fatness score, fat percentage, individual fatty acids (FA) profiles and sensory panel tests- and genotypic information from 11 polymorphisms was used. Results: The correlation coefficients between muscle total lipid measurements and absolute vs. relative amounts of polyunsaturated FA (PUFA) were in opposite directions. Increasing carcass fat leads to an increasing amount of FAs in triglycerides, but at the same time the relative amount of PUFAs is decreasing, which is in concordance with the negative correlation obtained here between the percentage of PUFA and fat measurements, as well as the weaker correlation between total phospholipids and total lipid muscle content compared with neutral lipids. Concerning organoleptic characteristics, a negative correlation between fiavour scores and the percentage of total PUFA, particularly to n-6 fraction, was found. The correlation between juiciness and texture is higher than with flavour scores. The distribution of SNPs plotted by principal components analysis (PCA) mainly reflects their known trait associations, although influenced by their specific breed allele frequencies. Conclusions: The results presented here help to understand the phenotypic and genotypic background underlying variations in FA composition and sensory parameters between breeds. The wide range of traits and breeds studied, along with the genotypic information on polymorphisms previously associated with different lipid traits, provide a broad characterization of beef meat, which allows giving a better response to the variety of consumers' preferences. Also, the development and implementation of low-density SNP panels with predictive value for economically important traits, such as those summarized here, may be used to improve production efficiency and meat quality in the beef industry.

Hence, research to improve production has been centered in technology that possible is not adequate for this region. The present review aims to utilize subjects such as Genetics, Reproduction, Nutrition, Management and Animal Health as examples of research endeavors illustrating the scarcely bond existing between research and adoption of technology. A final section is an attempt to illustrate the possible reasons why the transfer of technology is failing to be of any consequence at least under the conditions of developing countries.

文摘Background:Estimating inbreeding,which is omnipresent and inevitable in livestock populations,is a primary goal for management and animal breeding especially for those interested in mitigating the negative consequences of inbreeding.Inbreeding coefficients have been historically estimated by using pedigree information;however,over the last decade,genome-base inbreeding coefficients have come to the forefront in this field.The Pura Raza Espanola(PRE)horse is an autochthonous Spanish horse breed which has been recognised since 1912.The total PRE population(344,718 horses)was used to estimate Classical(F),Ballou’s ancestral,Kalinowski’s ancestral,Kalinowski’s new and the ancestral history coefficient values.In addition,genotypic data from a selected population of 805 PRE individuals was used to determine the individual inbreeding coefficient using SNP-by-SNP-based techniques(methods of moments-FHOM-,the diagonal elements of the genomic-FG-,and hybrid matrixes-FH-)and ROH measures(FRZ).The analyse of both pedigree and genomic based inbreeding coefficients in a large and robust population such as the PRE horse,with proven parenteral information for the last 40 years and a high degree of completeness(over 90%for the last 70 years)will allow us to understand PRE genetic variability better and the correlations between the estimations will give the data greater reliability.Results:The mean values of the pedigree-based inbreeding coefficients ranged from 0.01(F for the last 3 generations-F3-)to 0.44(ancestral history coefficient)and the mean values of genomic-based inbreeding coefficients varied from 0.05(FRZ for three generations,FH and FHOM)to 0.11(FRZ for nine generations).Significant correlations were also found between pedigree and genomic inbreeding values,which ranged between 0.58(F3 with FHOM)and 0.79(F with FRZ).In addition,the correlations between FRZ estimated for the last 20 generations and the pedigree-based inbreeding highlight the fact that fewer generations of genomic data are required when comparing total inbreeding values,and the opposite when ancient values are calculated.Conclusions:Ultimately,our results show that it is still useful to work with a deep and reliable pedigree in pedigreebased genetic studies with very large effective population sizes.Obtaining a satisfactory parameter will always be desirable,but the approximation obtained with a robust pedigree will allow us to work more efficiently and economically than with massive genotyping.
文摘Infections produced by feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV), two of the most prevalent pathogens in cats, range from passing unnoticed to presenting a wide variety of clinical signs. Different epidemiological, clinical, hematological and virological parameters were analyzed in 78 FIV- and/or FeLV-infected cats. FeLV-infected (FeLV+) cats were considerably younger than FIV-infected (FIV+) cats, and in general were seen to have a more severe disease than FIV+ cats. Around one third of the cats presented anemia, and neutropenia was also frequently observed. Though a higher percentage of FIV+ than FeLV+ cats had altered leukocyte counts, FeLV+ cats had altered counts of both neutrophils and lymphocytes more frequently than FIV+, which usually presented only either altered neutrophils or lymphocyte counts. Virological markers were only detected in FeLV+cats, either as mono- or dual-infection, and correlated with the severity of the disease, but not in FIV+ cats. In conclusion, these results suggest that FeLV affects more blood cell types and provokes death of affected animals at a much earlier age than FIV and that the severity of the disease seemed to depend on the viral status of the cat.
文摘The formation of biofilm by pathogenic microorganisms has become a problem in the livestock industry since it is considered a potential source of infection for farm animals while increasing microbial resistance to physical and chemical agents. Some plant extracts, such as soluble wheat extract, have been shown to be effective in inhibiting or destroying the biofilm of certain micro-organisms under specific conditions. The objective of this study is to evaluate the capacity of the pathogen to form biofilm on different surfaces used in livestock, as well as to evaluate the anti-biofilm capacity of the soluble wheat extract against <i>S. aureus</i> on these surfaces. The inhibition potential of inhibition or destruction of biofilm was tested in vitro. Wheat extract at a concentration of 0.29 mg/100mL showed anti-biofilm activity on <i>S. aureus</i>, inhibiting its formation, as well as destroying it greatly after a contact time of 24 hours, on those surfaces where the microorganism presents more adhesion capacity.
文摘Verticillium wilt is one of the most significant agricultural diseases in the world, in view of the fact that not only does it affec olive groves but also a wide variety of fruit, vegetable and ornamental plants. Currently, the most efficient and economical method of control involves the use of plant species that are resistant to the disease. However, as there are few varieties of olive tree with this characteristic, early diagnosis and the replacement of affected trees are key strategies to combat the disease. The present research proposes the use of a canine unit that is specifically trained to detect Verticillium dahliae as an early detection method for the fungus in olive groves. For the odorous samples are produced in the laboratory from V. dahliae, it is calculated that the dog dis-plays a level of sensitivity of 97% and specificity of 95%, and an initial field assay shows that the sensitivity and specificity are constant when working with infected olive trees. In view of the fact that there is currently no curative treatment for this disease and the affected plants are completely lost, fast on-site detection provides significant advantages in terms of preventing the transmission of the disease. Moreover, it also enables focalized treatment to be provided on plots of land and plants and, in combination with the latest prevention methods, real-time detection of affected olive trees enables a reduction in the costs involved in vaccination, thereby contributing overall to a reduction in the financial losses caused by verticillium wilt.
文摘The aim of the present study was to evaluate the impact of the technological level in small-scale dual-purpose cow-calf <em>Bos indicus</em> female operations on the resumption of the ovarian activity post-partum. A total of 13 small scale dual-purpose <em>Bos indicus</em> × <em>Bos taurus</em> operations were included in this experiment and they were classified according to their technological status as: high technological status (HT), medium technological status (MT) and low technological status (LT). Ninety-three mature cows were treated with either a CIDR (controlled internal drug release device) with estradiol benzoate (CIDR+EB) or alone (CIDR). At day 9 all animals received intravaginally a CIDR for nine days. At day 0, the CIDR was withdrawn and 24 hours later one dose of estradiol benzoate was administered to 51 cows (17 in HT, 17 in MT and 17 in LT), the remaining animals (n = 42) did not receive the estradiol benzoate administration (14 in HT, 13 in MT and 15 in LT). Comparisons were performed on serum progesterone concentrations after CIDR treatment. By day 7 after implant withdrawal, the proportion of cows that resume ovarian activity detected by progesterone concentration increased in all technological level farms, no statistical main effect was observed within technological level. By day 17 there was a significant main effect of technological level (P = 0.05) on the proportion of animal that resumed the estrous cycles, but neither a main effect of treatment (P = 0.97) nor an interaction between technological level and treatment (P = 0.98). Furthermore, technological level of the farm showed a tendency (P = 0.07) to affect that resume the estrous activity, and a significant effect of treatment was observed (P = 0.005) but no interaction between technological level and treatment. The proportion of cows that showed estrus was different across technological level (P = 0.02), the highest proportion of cows showing estrus regardless of treatment was in the HT: 90% (40% CIDR and 50% CIDR+EB), MT: 50% (13.3% CIDR and 36.7% CIDR+EB) and in the LT: 50% (18.8% CIDR and 31.3% CIDR+EB). In conclusion, the combination of a progestogen and estradiol benzoate resulted in a high proportion of cows that were induced to resume the ovarian activity and this treatment was particularly beneficial in the medium and low technological status of the farms.
文摘Mycoplasma mycoides subsp. capri is the causative agent of severe and acute respiratory problems in goats, which spreads rapidly and represents high mortality. The serological profile of the goat population, from nine regions in seven states of Mexico, was screened by the Complement Fixation test (CF) in sera from asymptomatic goats and animals with mild respiratory symptoms. Sera and nasal swabs of 827 goats were collected for the isolation of the organism. An antiserum was prepared against a previously isolated field strain of Mycoplasma mycoides subsp. capri. CF Antibody titers were associated with the results of the isolates to determine the cutoff point. The CF was considered as positive if its result was ≥1/16. The CF registered 251 positive goats (30.35%) and 576 (69.65%) negative;the test showed high sensitivity (93.33%) and specificity (72.27%). In the specific case of diagnosis for mycoplasmosis associated with respiratory problems in goats, the CF proved to be a good diagnosis test, this study determined that 30% of the goat population showed antibody titers against Mycoplasma mycoides subsp. capri and revealed those animals who have had contact with this microorganism during their lives regardless of the presence or absence of respiratory symptoms.
文摘Due to the removal of antibiotic growth promoters (AGPs) and consumer pressure for antibiotic-free (ABF) or no antibiotics ever (NAE) poultry production, there is a need for sustainable alternatives to prevent disease in commercial poultry operations. Without AGPs, there has been a rise in diseases that were traditionally controlled by subtherapeutic levels of antibiotics in the diet. This has impacted the health of commercial poultry and has been a significant cost to poultry producers. To mitigate this, the industry has started to investigate alternatives to antibiotics to treat these forthcoming health issues, such as necrotic enteritis (NE). NE is an enteric disease caused by an over proliferation of toxigenic Clostridium perfringens (CP) in the gastrointestinal tract. Although CP is a commensal in the avian intestinal tract, dysbiosis caused by inflammation and impaired intestinal integrity facilitates uncontrolled replication of CP. Infectious agents, such as Eimeria maxima, appear to be a predominant predisposing factor that promotes NE. However, non-infectious stressors, including dietary changes, have also been associated with NE to some degree. As a result of increased pressure to restrict the use of antibiotics, there is a need for research evaluating the efficacy of alternatives, such as plant-derived essential oils, as potential tools to mitigate NE in commercial poultry flocks. The aim of this study is to review the effects of essential oils as an alternative to antibiotics to reduce the incidence and severity of necrotic enteritis in broiler chickens.
文摘The purpose of the study was to evaluate the sperm viability of semen infected with PRRSV viral particles, observing the effect of the Virus on the motility of boar sperm. The work was carried out at the FMVZ-BUAP Genetics and Reproduction Laboratory. 5 stallions were used. Each sample contained 1 × 10<sup>6</sup> sperm, the PRRS virus strain was ATCC-VR-2332 (0, 10<sup>2</sup>, 10<sup>4</sup> and 10<sup>6</sup> copies of RNA/mL in triplicate), it was observed daily at the CASA;Hamilton Thorne<sup>®</sup>. Cells with MT (P < 0.05) on days 1, 3, 5, 7 and 10 of evaluation with 201 ± 7.3, 167 ± 10.1, 165 ± 14.6, 134 ± 8.2 and 120 ± 8.8, respectively. The % MP between control and virus concentrations (P ≥ 0.05). The LCV on day 1 and 7 PI at 10X<sup>2</sup> and 10X<sup>6</sup> (P < 0.05) vs control. In the Correlation Matrix, where it is observed that there is a correlation between VSL and VAP, VSL and VCL, VCL and ALH, VAP with ALH. There is a correlation of VSL and ALH, STR and ALH. In this study there were (P ≤ 0.01) in the VCL, in the concentrations (10<sup>2</sup>) 162.81 ± 10.65 and (10<sup>6</sup>) 177.12 ± 5.77 vs 193.04 ± 4.62 of control. This indicates that altering these parameters would be related to fertility and the PRRS virus affects the LCV. Regarding the VSL, it was observed that the sperm infected with viruses 10<sup>2</sup>, 10<sup>4</sup> and 10<sup>6</sup> of 48.00 ± 3.38, 49.88 ± 1.83 and 50.55 ± 2.24 Vs. 56.66 ± 1.68 of control respectively, the control would have greater possibilities of fertilizing the oocyte. In this study, it was found (P ≤ 0.01) in the VAP with 102 of 77.26 ± 5.16, 10<sup>4</sup> with 83.35 ± 2.41 and 10<sup>6</sup> with 81.29 ± 3.14 vs the control with 90.56 ± 2.07. Regarding the ALH there is (P < 0.05) a 10<sup>4</sup> with 8.70 ± .26 and 10<sup>6</sup> with 9.64 ± 0.23 vs control 8.50 ± 0.27. The presence of different concentrations of PRRSV in boar semen induces changes in different types of sperm motility. Infection of ejaculates with the PRRS virus affects sperm motility on days 1, 3, 5, 7, and 10 post-infections.
文摘Coccidiosis is a disease caused by intracellular protozoan parasites, specifically belonging to the genus Eimeria. These parasites target the gastrointestinal tract in different types of hosts, causing sores in the intestinal lining. The presence of these lesions reduces the animal’s ability to digest and absorb nutrients, significantly impacting their overall performance. The current study aimed to explore the potential correlation between seasonal variations and the incidence of Eimeria spp-induced lesions in broiler chickens’ gastrointestinal tracts in Brazil from 2012 to 2018. A total of 8,607 broiler chickens, aged 14 to 42 days, were sampled from 103 poultry integrated companies in Brazil to conduct intestinal health examinations. The sampling process involved selecting 3 to 6 chickens from each poultry house for examination. The assessment included various abnormalities such as shedding of intestinal cells and excessive fluid and mucus presence, thickening and tension of the intestines, food movement, roughened mucosal surface resembling a Turkish towel in the small intestine, tissue death, duodenal inflammation, intestinal inflammation, gizzard erosion, presence of worms and bedding material, and ingestion of mealworms. During the seasons, winter exhibited the highest average occurrence of Eimeria maxima microorganisms at 52.83%, with E. acervulina following closely at 26.42% in second place. In spring, E. maxima had an occurrence of 11.31%, while in fall, E. tenella had the lowest occurrence at 6.74%. When analyzing the seasonal occurrence of Eimeria, it was observed that E. maxima micro was more common during winter compared to summer (P = 0.0491). However, no discernible variation was observed in the occurrence of the remaining species across different seasons. Research findings suggest that subclinical coccidiosis is most prevalent during the winter season in Brazil. Likewise, clinical disease caused by E. acervulina is also prevalent during this time. In contrast, E. maxima is more likely to cause clinical disease in the spring, whereas E. tenella is more commonly associated with clinical disease in the fall. Lesions induced by Eimeria spp. are associated with factors influencing the overall health of broiler intestines. These findings allow for the utilization of seasonal metrics in disease management, thereby reducing economic losses associated with the condition.
文摘Plant-based antimicrobial agents are readily available, cost-effective, and exhibit low toxicity, making them promising alternatives in combatting microbial infections. Among these plants, garlic (Allium sativum) stands out for its traditional medicinal use in effectively combating various microorganisms. In the poultry industry, preventing avian virus and bacterial infections is paramount for chicken husbandry. However, using conventional drugs poses potential risks to human health. Garlic, a widely used Asian plant in traditional medicine for various pathologies, has shown potential as an herbal prophylactic remedy against viral and bacterial infections. Recently, researchers explored garlic and its derivatives as a scientific strategy in veterinary practices for diverse purposes, such as improving poultry production characteristics and acting as antibiotic growth promoters. This comprehensive review delves into garlic and its derivatives as preventive and corrective treatments for viral diseases in laying hens and broilers. The paper highlights their potential effectiveness and safety as a natural means to enhance poultry health and welfare while mitigating the risks associated with conventional drug usage in the food industry.

文摘Background: Residual feed intake(RFI) in dairy cattle typically calculated at peak lactation is a measure of feed efficiency independent of milk production level. The objective of this study was to evaluate differences in ruminal bacteria, biopolymer hydrolyzing enzyme activities, and overall performance between the most-and the leastefficient dairy cows during the peripartal period. Twenty multiparous Holstein dairy cows with daily ad libitum access to a total mixed ration from d-10 to d 60 relative to the calving date were used. Cows were classified into most-efficient(i.e. with low RFI, n = 10) and least-efficient(i.e. with high RFI, n = 10) based on a linear regression model involving dry matter intake(DMI), fat-corrected milk(FCM), changes in body weight(BW), and metabolic BW.Results: The most-efficient cows had ~ 2.6 kg/d lower DMI at wk 4, 6, 7, and 8 compared with the least-efficient cows. In addition, the most-efficient cows had greater relative abundance of total ruminal bacterial community during the peripartal period. Compared with the least-efficient cows, the most-efficient cows had 4-fold greater relative abundance of Succinivibrio dextrinosolvens at d-10 and d 10 around parturition and tended to have greater abundance of Fibrobacter succinogenes and Megaspheara elsdenii. In contrast, the relative abundance of Butyrivibrio proteoclasticus and Streptococcus bovis was lower and Succinimonas amylolytica and Prevotella bryantii tended to be lower in the most-efficient cows around calving. During the peripartal period, the most-efficient cows had lower enzymatic activities of cellulase, amylase, and protease compared with the least-efficient cows.Conclusions: The results suggest that shifts in ruminal bacteria and digestive enzyme activities during the peripartal period could, at least in part, be part of the mechanism associated with better feed efficiency in dairy cows.
文摘Objective: The Response Surface Methodology (RSM) is a commonly used system to optimize cell viability of probiotic strains when they are subjected to different preservation and storage processes. Methods and Results: To determine the optimal levels of incorporation of several cry oprotectants (skim milk, sucrose and trehalose) in the freeze-drying process of Lactobacillus plantarum, a range of experiments based on a Rotational Central Composite design (CCD) were conducted. The results were adjusted to a quadratic model, resulting in the presence of interaction between the different variables. Solving a regression equation, we obtained the optimum concentrations of cryoprotective agents: 24.06% milk powder, 6.22% sucrose, 5.63% trehalose. To visualize the interactions between the three variables involved in the study, Design Expert? software was used. Conclusions: The analysis reveals that while trehalose has a direct effect on the viability of L. plantarum, skim milk and sucrose exert quadratic effects. There are also interactions between cryoprotectants, which emphasize the synergies produced between milk and sucrose and between sucrose and trehalose, which allows maintaining the viability of L. plantarum. Significance and Impact of the Study: The addition of new oligosaccharides as trehalose in premixtures for functional feed can maintain the viability of L. plantarum during longer periods of time, ensuring the proper administration of probiotics to their destinations.
文摘Extracts of 40 green, black or oolong high quality tea samples from China, Japan or India among others, have been prepared according to the manufacturer instructions and assayed for total polyphenols content using the Folin-Ciocalteu phenol reagent. In addition, the antioxidant activity was assessed by the CUPRAC method and the free-radical scavenging activity was determined using the antioxidant-promoting decay of the stable free-radical DPPH. Caffeine and the most important catechins were identified using RP-HPLC previously validated method. Results from this study suggest that teas, even though they content similar amounts of caffeine, differ considerably in polyphenolic content and therefore in antioxidant and free-radical scavenging activities, depending on the part of the plant used and the process applied to the material for the preparation of the final tea presentation. Correlations among Folin-Ciocalteau’s reactivity of tea samples, the neocuproine reactivity, the DPPH decolourization and the HPLC analysis suggest that the antioxidant activity is due essentially to polyphenolic compounds present in teas, mainly EGCG. The best extraction method of tea powder was the one indicated by dealers. In general, the richest samples belong to green teas and the poorest samples belong to the black teas.
文摘The objective of this work was to determine the effect of transport time (up to 11 hours) on animal welfare. 540 animals (cows, bulls and calves), three transport times of 2, 4 to 6, and 10 to 11 hours, and two space allowances (2 m2 and 1.5 m2 per animal for cows and bulls; and 1.2 m2 and 0.8 m2 per animal for calves) were used for the experiment during transport from farms to the abattoir in Uppsala, Sweden. Measurements were made on five animals on each trip. Stress response parameters considered were cortisol, glucose, lactate, creatine kinase and heart rate. Blood samples were taken before and after transport. Heart rate sensors were mounted on the animals at least 30 minutes before loading and measurements were made continuously from farms to the abattoirs until stunning. The results of heart rate measurement indicated that loading, un-loading and forced movement in the stunning box were the most stressful events. However, the results of statistical analysis confirmed that transport time doesn't have significant effect (P 〈 0.05) on heart rate. Concentration level of cortisol increased by 10 folds during short transport. However, cortisol concentration decreased with an increase of transport times (P 〈 0.01). Glucose concentrations increased with transport time in all animal categories (P ≤ 0.01). Transport time has significant effect on concentration levels of glucose (P ≤ 0.01), creatine kinase (P 〈 0.001) and lactate (P 〈 0.01) particularly after 6 hours journey time.
文摘One hundred ninety-two crossbred steers (229 kg) were used to evaluate the influence of metabolizable amino acid intake on growth performance and health during the initial 42-d receiving period. Treatments consisted of four levels of metabolizable lysine (23, 24, 25 and 26 g/kg diet DM). Morbidity averaged 36%, and was not affected (P > 0.20) by treatments. No steers died during the study. Increasing the metabolizable lysine supply increased DMI, ADG, gain efficiency, and dietary NE (linear effect, P < 0.01). Metabolizable amino acid supply of the basal diet was determined using 6 steers (214 kg) with cannulas in the rumen and proximal duodenum. Metabolizable amino acid supply of the diet was in close agreement with expected (R2 = 0.99;P < 0.0001) based on NRC (2000) [12] Level 1 model. The metabolizable amino acid supplies for treatments in Trial 1 were estimated by adding tabular metabolizable amino acid values for the respective supplemental proteins (cottonseed meal and fish meal) to the observed metabolizable amino acid supply of the basal diet. Treatment effects on metabolizable lysine supply explained 99% of the variation (P < 0.01) in ADG, and 91% of the variation in observed versus expected dietary NE. The biological value for the intestinal chyme was determined based on chemical score technique, using bovine tissue as the reference protein. Accordingly, methionine and lysine were closely co-limiting amino acids having ratios of 77% and 79%, respectively. We conclude that current NRC standards reliably predict both requirements and supplies of metabolizable amino acids for feedlot calves. Diet formulations that do not meet the metabolizable amino acid requirements may depress both ADG and the partial efficiency of utilization of metabolizable energy for maintenance and gain.
