The selective killing effect of adenoviral mediated HSV-tk/GCV suicide gene system controlled by hTERT promoter on bladder cancer cells

To investigate the selective killing effect of adenoviral mediated herpes simplex virus-thymidine kinase/ganciclovir (HSV-tk/GCV) suicide gene system controlled by human telomerase reverse transcriptase (hTERT) promoter on bladder cancer cells in vitro.Methods Bladder cancer cell line 253J and human fibroblast cell line MRC-5 were transfected by the recombinant adenovirus of different multiplicities of infection (MOI),and the infection rate was measured by observing the expression of enhanced green fluorescent protein (EGFP) under the fluorescent microscopy.Ad-hTERT-HSV/tk and Ad-CMV-SHV/tk were transduced into 253J and MRC-5,followed by GCV treatment.The relative survival rate of cells in presence of prodrug GCV was measured with MTT method.Results Recombinant adenovirus Ad-hTERT-EGFP could selectively infect 253J cells,with the infection rate associated with the increasing MOI of recombinant adenovirus (P<0.01).When MOI was 1,the infection rate was 8.3%;when MOI was 1000,the infection rate was 100.0%.GCV significantly inhibited the growth of both cell lines (253J and MRC-5) infected with Ad-CMV-HSV/tk and the growth of only 253J infected with Ad-hTERT-HSV/tk (P<0.001).The survival rate of 253J was correlated with the concentration of both the prodrugs and MOI of recombinant adenovirus (P<0.01).When MOI was 1,with 1 μmol/L GCV,the survival rate of 253J was 95.4%;when MOI was 100,with 1000 μmol/L GCV,the survival rate of 253J was 6.1%.Conclusion Recombinant adenovirus vector containing EGFP can report the transfection rate accurately and easily.Adenoviral vector containing hTERT promoter for the HSV/tk plus GCV treatment appears to be highly selective in killing bladder cancer cell line in vitro.7 refs.2 tabs.

Study on a New Type of All-Solid-StateReference Electrode

A new type of all-solid-state Ag/AgCl reference electrode was constructed by using urea-formaldehyde resin as the frame material and KCl powder as the active component. The properties of the electrode were studied in detail. Preliminary experience on laparoscopically assisted treatment for high imperforate anus, a report of 6 cases

Objective To evaluate laparoscopically assisted anorectal pull-through for the treatment of high imperforate anus. Methods Six patients aged at 2 days to 17 years of high imperforate anus were enrolled. Among them, 2 were with globular shape colon and rectourethral fistula, one with cloaca and globular shape colon. The rectum was exposed via laparoscopy down to the rectourethral fistula or vaginal fistula, which was ligated and dissected, and then, the pelvic floor musculature was assessed and the center of the levator muscle identified.

Diagnostic value of special examination of thumb syndactyly

Objective To investigate the value of special examination in the diagnosis of thumb syndactyly. Methods Fourteen cases of thumb deformities were detected preoperatively by X-ray plain film, angiography and ultrasound examination. The deformities of bone, hemokinesis of thumbs and its blood supply were analysed. The results were compared with their pathological changes during the operation. The deformity was corrected by operations of bone resection, reconstruction of tendon insertion site and preservation of main arteries. Results There were bone deformities 13 cases, abnormal or absent tendon insertion of the main thumb 14 cases. The nutrient of the deformity thumb was supplied by main trunk artery in 11 cases. Conclusion Angiography and ultrasonic examination play an important role in the diagnosis of thumb syndactyly. It can help determine appropriate surgical procedure so that secondary deformities can be prevented after operation. 3 refs,5 figs.

Cellular compatibilty of the nano-hap/collagen and bone marrow stem cell

To assess the compatibility of the nano-hydroxyapatite/collagen composite and human bone marrow stem cell.Methods The biocompatibility of BMSC was studied by co-culturing NHAC with BMSC in vitro.Results The BMSC could adhere to,proliferate and grow normally and on the surface and holes of NHAC.The cellular function and alkaline phosphatase activity were not affected by NHAC.Conclusion NHAC is biocompatible and can be used as ideal biomaterial in bone tissue engineering.8 refs,8 figs.

Effect of epsilon-aminocaproic acid on fibrinolysis in patients undergoing open heart surgery with cardiopulmonary bypass

To investigate the dynamic changes in the fibrinolytic system during and after open heart surgery performed with cardiopulmonary bypass (CPB) and the effect of epsilon-aminocaproic acid (EACA) on fibrinolysis,postoperative blood loss and transfusion requirements.Methods Forty patients (24 males,16 females )aged 17～43 yr undergoing valve replacement with CPB were randomized to receive EACA 200 mg·kg -1 (group A) or normal saline (group B) added to the priming solution.The patients were premedicated with intramuscular morphine 0.2 mg·kg -1 and scopolamine 0.3 mg.Anesthesia was induced with midazolam 0.2 mg·kg,fetanyl 10 μg·kg -1 and vecuronium 0.1 mg·kg -1 and maintained with isoflurane inhalation and intermittent i.v. boluses of fentanyl and vecuroniu.The patients were mechanically ventilated (VT=10 mg·kg -1 ,RR=10～12 bpm,I∶E=1∶2) after tracheal intubation.Blood samples were taken before skin incision (baseline),8 and 30 min on CPB,10 min after protamine administration and 2 h after operation for determination of tissue type plasminogen activator (t-PA) activity,plasminogen activator inhibitor (PAI) activity,plasmin (Plm) activity and D-dimer.The amount of blood drained from chest-and bank blood infused within 24 h after operation were recorded.Results (1) Group B (control group):PAI activity was maintained stable perioperatively.The t-PA activity and t-PA/PAI ratio increased significantly during operation but returned to the baseline value 2 h after operation.D-dimer level also significantly increased during and after operation.The Plm activity increased significantly at 8 h on CPB and 10 min after protamine administration compared to the baseline value.(2) The t-PA activity and t-PA/PAI ratio were significantly lower in group A than in group B.The D-dimer lever and Plm activity were also significantly lower in group A than in group B during and after operation.The amount of blood loss and bank blood infused within 24 h after operation was 40% and 37% less than that in group B.Conclusion Fibrinolytic system is activated during CPB as shown by the increase in plasminogen and t-PA/PAI ratio.EACA in effective in reducing postoperative blood loss and blood transfusion through inhibition of the activation of fibrinolytic system.8 refs,2 tabs.

Usage of buttress plate internal fixation associated with autografting of fibula and iliac bone for the treatment of distal femoral C_3 type fracture

To assess the effect of using buttress plate associated with antografting of fibula and iliac bone for the treatment of distal femoral C3 type fracture.Methods Seventeen cases of distal femoral C3 type fracture using buttress plate associated with antografting of fibula and iliac bone were analyzed retrospectively.Results All cases were followed up for an average of 24 months（8～55 months）.The average time of octets bridge forming were 4 months（3～5 months） while the average time for bone union were 8 months （6～14 months）.According to Shelbourne rating system,result of all 18 cases were excellent and no malunion,infection were found.Conclusion Buttress plate associated with antografting of fibula and iliac bone is an effective alternative for the treatment of distal femoral C3 type fracture.It can provide more stable fixation to the bone and earlier functional exercises can be achieved.5 refs,3 figs,1 tab.

Follicular fluid levels of prostaglandin E2 and the effect of prostaglandin E2 on steroidogenesis in granulosa-lutein cells in women with moderate and severe endometriosis undergoing in vitro fertilization and embryo transfer

Background The mechanisms of endometriosis with infertility have not been fully studied. The present study aimed to assess the follicular fluid （FF） levels of prostaglandin E2 （PGE2）, which plays a critical role within the ovary, and to investigate the effect of PGE2 on steroidogenesis in granulosa-lutein cells （GLCs） from women with and without endometriosis. Methods Thirty-three women with laparoscopically documented endometriosis and 40 controls undergoing in vitro fertilization （IVF） were studied. We assayed the concentrations of PGE2 in FF, the production of E2 and progesterone in FF and in culture medium, and the expression of steroidogenic acute regulatory protein （STAR） and CYP19A1 in GLCs with the intervention of PGE2. Results PGE2 and progesterone concentrations were increased and displayed positive correlation in endometriotic FE PGE2 induced the expression of STAR and the production of progesterone in GLCs from women with endometriosis, and the expression of STAR and the production of progesterone were increased in GLCs from women with endometriosis. However, there were no significant effects of PGE2 on promoting the production of E2 or the expression of CYP19A1 in GLCs. Moreover, the production of E2 and the expression of CYP19A1 in GLCs from women with endometriosis were significantly decreased compared to the controls. Conclusions PGE2 concentrations are increased in endometriotic FF, along with concomitant increases in progesterone and STAR. In contrast, the E2 and CYP19A1 are decreased in GLCs, which may delay the development of the follicles and cause an imbalance in the follicular steroid hormone levels. These changes may have close relationship with endometriosis-associated infertility.
