文摘The Gaoligong Mountains(GLGM),located in southwestern China, extend north to south along the western border of the Hengduan Mountains, spanning approximately 600 km.In this study,we consolidated findings from 17 bird surveys conducted in the GLGM between 2010 and 2022.We found that the GLGM harbors tremendous bird diversity, with a total of 796 documented bird species in the region.Nearly a quarter(23.0%)of these species are listed as state key protected species or as Chinese and global threatened species.Analysis of species richness at the county level showed a decreasing trend with increasing latitude, with the greatest diversity in Yingjiang(661 species).Observations indicated that the GLGM belongs to the Oriental realm, primarily composed of bird species from southern and southwestern China. The GLGM plays an important role in avian conservation by sheltering exceptional bird diversity, providing corridors and flyways for bird migration and dispersal, and mitigating the effects of climate change. In response to the conservation needs of birds and other wildlife, the Chinese government has established numerous protected areas within the GLGM.Despite these efforts, avian conservation still faces considerable challenges in the GLGM due to limitations in the protected area network, transboundary nature of the regions, and existing gaps in monitoring and research.

文摘The dual impact of climate change and human activities has precipitated a sharp decline in primate biodiversity globally.China is home to the most diverse primate species in the Northern hemisphere, which face severe ecological threats due to the expansion of modern agriculture, extensive exploitation and consumption of natural resources, and excessive land development during its transition from an agricultural to a modern society. In response,China has implemented various ecological conservation measures, including habitat restoration and protection. These efforts have made substantial strides in biodiversity conservation, with certain regions witnessing an increase in primate populations. In the current study,we conducted a systematic review of historical documents and field research data related to Chinese primates, evaluating the endangered status of primate species in China.Despite improvements in the habitats of most primate species and some population growth, many species still face severe threats, including declining and small populations.Species such as the Myanmar snub-nosed monkey(Rhinopithecus strykeri),eastern black crested gibbon(Nomascus nasutus),and Hainan gibbon(N.hainanus)remain particularly vulnerable due to their limited distribution ranges and extremely small populations.Insufficient scientific data, fragmented information, and not enough studies in conservation biology further compound the challenges. Moreover, there is a notable lack of detailed population monitoring data for species such as the Bengal slow loris(Nycticebus bengalensis),pygmy slow loris(N.pygmaeus),Indochinese gray langur(Trachypithecus crepusculus),Shortridge’s langur(T.shortridgei),and capped langur(T.pileatus),which hinders the development of practical and targeted conservation management strategies. Therefore, for national biodiversity conservation, there is an urgent need for specialized primate surveys, enhancing habitat protection and restoration, and increasing focus on cross-border conservation strategies and regional cooperation. There is also a need to establish a comprehensive and systematic research database platform, conduct continuous and in-depth research in primate biology, and actively engage in the scientific assessment of ecotourism.Additionally, strengthening public awareness and education on wildlife conservation remains essential.Such integrated and systematic efforts will provide scientific support for the current and future conservation and management of primate species in China.

文摘Gaoligong Mountain(hereafter,GLGM)is located at the intersection of Myanmar and China’s Yunnan Province and Xizang Zizhiqu, and spans three globally significant biodiversity hotspots:the Himalayas,Indo-Burma,and the Mountains of Southwest China.Although surveys of mammals in this ecologically important region have a long history, there is no comprehensive systematic checklist and distribution account of the mammals of GLGM.Here,we compiled a mammal species checklist of GLGM based on thorough field investigations and literature reviews. We also examined specimen collections and applied camera trapping surveys to explore the region’s mammal diversity and distribution patterns. We recorded 212 mammal species in nine orders,33 families, and 119 genera, which accounts for 30.5%of China’s mammal species, and a high proportion of nationally protected(50)and globally threatened(29)species.Mammal species richness showed a symmetrical unimodal curve along the elevation gradient, peaking at intermediate elevations(2000 to 2500 m above sea level(asl)),and increasing generally from south to north, slightly higher in the east slope than in the west.Cluster analysis and non-metric multidimensional scaling revealed three distinct elevational assemblages(<900 m asl.,900-3500 m asl.,and>3500 m asl)and significant south-to-north variation, but no substantial differences between the east and west slopes. The GLGM present a unique conservation value due to the high proportions of rare and endangered mammal species, complex faunal composition, high endemism, and being the distribution boundary for many species. This study is an important phased account of mammal diversity in GLGM and makes a prospect for future research.

文摘Genetic load and inbreeding are recognized as important factors to be considered in conservation programs.Elevated levels of both can increase the risk of population extinction by negatively impacting fitness-related characters in many species of plants and animals, including humans(inbreeding depression).Genomic tech-niques are increasingly used in measuring and understanding genetic load and inbreeding and their importance in evolution and conservation. We used whole genome resequencing data from two sibling grouse species in subarctic Eurasia to quantify both.We found a large range of inbreeding measured as FROH(fraction of runs of homozygosity)in individuals from different populations of Chinese Grouse(Tetrastes sewerzowi)and Hazel Grouse(T.bonasia).FROH estimated from genome-wide runs of homozygosity(ROH)ranged from 0.02 to 0.24 among Chinese Grouse populations and from 0.01 to 0.44 in Hazel Grouse.Individuals from a population of Chinese Grouse residing in the Qilian mountains and from the European populations of Hazel Grouse(including samples from Sweden,Germany and Northeast Poland)were the most inbred(FROH ranged from 0.10 to 0.23 and 0.11 to 0.44,respectively).These levels are comparable to other highly inbred populations of birds.Hazel Grouse from northern China and Chinese Grouse residing in the Qinghai-Tibetan Plateau showed relatively lower inbreeding levels.Comparisons of the ratio between deleterious missense mutations and synonymous mutations revealed higher levels in Chinese Grouse as compared to Hazel Grouse. These results are possibly explained by higher fixation rates, mutational melt down,in the range-restricted Chinese Grouse compared to the wide-ranging Hazel Grouse. However, when we compared the relatively more severe class of loss-of-function muta-tions,Hazel Grouse had slightly higher levels than Chinese Grouse,a result which may indicate that purifying selection(purging)has been more efficient in Chinese Grouse on this class of mutations.

文摘Background:Pig organ xenotransplantation is a potential solution for the severe organ shortage in clinic, while immunogenic genes need to be eliminated to improve the immune compatibility between humans and pigs.Current knockout strategies are mainly aimed at the genes causing hyperacute immune rejection(HAR)that occurs in the first few hours while adaptive immune reactions orchestrated by CD4 T cell thereafter also cause graft failure,in which process the MHCⅡmolecule plays critical roles. Methods:Thus,we generate a 4-gene(GGTA1,CMAH,β4GalNT2,and CIITA)knockout pig by CRISPR/Cas9 and somatic cell nuclear transfer to compromise HAR and CD4 T cell reactions simultaneously. Results:We successfully obtained 4KO piglets with deficiency in all alleles of genes, and at cellular and tissue levels.Additionally, the safety of our animals after gene editing was verified by using whole-genome sequencing and karyotyping.Piglets have survived for more than one year in the barrier, and also survived for more than 3 months in the conventional environment, suggesting that the piglets without MHCⅡcan be raised in the barrier and then gradually mated in the conventional environment.Conclusions:4KO piglets have lower immunogenicity, are safe in genomic level, and are easier to breed than the model with both MHCⅠandⅡdeletion.

文摘In the intricate skeletal muscle tissue, the symbiotic relationship between myotubes and their supporting vasculature is pivotal in delivering essential oxygen and nutrients. This study explored the complex interplay between skeletal muscle and endothelial cells in the vascularization ofmuscle tissue.By harnessing the capabilities of three-dimensional(3D)bioprinting and modeling,we developed a novel approach involving the co-construction of endothelial and muscle cells, followed by their subsequent differentiation. Our findings highlight the importance of the interaction dynamics between these two cell types.Notably, introducing endothelial cells during the advanced phases of muscle differentiation enhanced myotube assembly. Moreover,it stimulated the development of the vascular network, paving the way for the early stages of vascularized skeletal muscle development. The methodology proposed in this study indicates the potential for constructing large-scale,physiologically aligned skeletal muscle.Additionally,it highlights the need for exploring the delicate equilibrium and mutual interactions between muscle and endothelial cells.Based on the multicell-type interaction model,we can predict promising pathways for constructing even more intricate tissues or organs.

文摘Beta diversity, the variation of community composition among sites, bridges alpha and gamma diversity and can reveal the mechanisms of community assembly through applying distance-decay models and/or partitioning beta diversity into turnover and nestedness components from functional and phylogenetic perspectives.Mountains as the most natural experiment system provide good opportunities for exploring beta diversity patterns and the underlying ecological processes. Here,we simultaneously consider distance-decay models and multiple di-mensions of beta diversity to examine spatial variations of bird communities, and to evaluate the relative importance of niche-based and neutral community assembly mechanisms along a 3600-m elevational gradient in the central Himalayas,China. Our results showed that species turnover dominates taxonomic, functional, and phylogenetic beta diversity. We observed strongest evidence of spatial distance decays in taxonomic similarities of birds, followed by its phylogenetic and functional analogues.Turnover component was highest in taxonomic beta diversity, while nestedness component was highest in functional beta diversity.Further, all correlations of assemblage similarity with climatic distance were higher than that with spatial distances.Standardized values of overall taxonomic, functional, and phylogenetic beta diversity and their turnover components increase with increasing elevational distance, while the standardized values of taxonomic and phylogenetic nestedness decreased with increasing elevational distance. Our results highlighted the niche-based deterministic processes in shaping elevational bird diversity patterns that were determined by the relative roles of decreasing trend of environmental filtering and increasing trend of limiting similarity along elevation distances.

文摘The gut microbiota plays a pivotal role in the maintenance of health for amphibians, and it has been fully recognized, but the effectiveness of various influencing factors has not yet been fully clarified.Although this association should be considered while the amphibian order Caudata is facing a severe situation of population decline and extinction, there is little understanding of the association between diets and the diversity of gut microbiota in the amphibian order Caudata. Here,we conducted an extensive analysis of the gut microbiota of Cynops orientalis fed different diets using functional prediction and 16S rRNA amplicon sequencing techniques.First,we found that wild individuals had greater gut microbial diversity and richness in comparison to captive individuals.Second,we identified the bacterial taxa associated with diets and observed differences in the relative abundance of gut microbiota among people on various diets. Finally,we have a predictive comprehension of the selection and adaptative significance of shared core ASVs in the gut microbiota in maintaining the healthy survival of C.orientalis in a large-scale spatiotemporal map.Our study emphasizes how diets alter the gut microbiota of Caudata and offers fresh perspectives on the conservation and captive management of species in Caudata.

文摘BACKGROUND Aberrant methylation is common during the initiation and progression of colorectal cancer(CRC),and detecting these changes that occur during early adenoma(ADE)formation and CRC progression has clinical value.AIM To identify potential DNA methylation markers specific to ADE and CRC.METHODS Here,we performed SeqCap targeted bisulfite sequencing and RNA-seq analysis of colorectal ADE and CRC samples to profile the epigenomic-transcriptomic landscape.RESULTS Comparing 22 CRC and 25 ADE samples, global methylation was higher in the former, but both showed similar methylation patterns regarding differentially methylated gene positions, chromatin signatures, and repeated elements.High-grade CRC tended to exhibit elevated methylation levels in gene promoter regions compared to those in low-grade CRC.Combined with RNA-seq gene expression data,we identified 14 methylation-regulated differentially expressed genes,of which only AGTR1 and NECAB1 methylation had prognostic significance.CONCLUSION Our results suggest that genome-wide alterations in DNA methylation occur during the early stages of CRC and demonstrate the methylation signatures associated with colorectal ADEs and CRC,suggesting prognostic biomarkers for CRC.

文摘Most described Mesozoic ants belong to stem groups that existed only during the Cretaceous period.Previously, the earliest known crown ants were dated to the Turonian(Late Cretaceous,ca.94-90 million years ago(Ma))deposits found in the USA,Kazakhstan, and Botswana. However, the recent discovery of an alate male ant in Kachin amber from the earliest Cenomanian(ca.99 Ma),representing a new genus and species,Antiquiformica alata, revises the narrative on ant diversification.Antiquiformica can be distinctly differentiated from all known male stem ants by its geniculate antennae with elongated scape, extending far beyond the occipital margin of the head and half the length of the funiculus,as well as its partly reduced forewing venation.Furthermore, the combination of a one-segmented waist with a well-developed node, elongated scape extending beyond the occipital margin, and reduced forewing venation, particularly the completely reduced m-cu and rs-m crossveins and absence of rm and mcu closed cells, firmly places the fossil within the extant subfamily Formicinae.Fourier transform infrared spectroscopy(FTIR)confirmed that the amber containing Antiquiformica alata originated from the Kachin mines in Myanmar. This discovery significantly revises our understanding of the early evolution of Formicinae. The presence of Antiquiformica in Cenomanian amber indicates that the subfamily Formicinae emerged at least by the start of the Late Cretaceous, with crown ants likely originating earlier during the earliest Cretaceous or possibly the Late Jurassic, although paleontological evidence is lacking to support the latter hypothesis.
