This article presents an innovative approach to automatic rule discovery for data transformation tasks leveraging XGBoost,a machine learning algorithm renowned for its efficiency and performance.The framework proposed herein utilizes the fusion of diversified feature formats,specifically,metadata,textual,and pattern features.The goal is to enhance the system’s ability to discern and generalize transformation rules fromsource to destination formats in varied contexts.Firstly,the article delves into the methodology for extracting these distinct features from raw data and the pre-processing steps undertaken to prepare the data for the model.Subsequent sections expound on the mechanism of feature optimization using Recursive Feature Elimination(RFE)with linear regression,aiming to retain the most contributive features and eliminate redundant or less significant ones.The core of the research revolves around the deployment of the XGBoostmodel for training,using the prepared and optimized feature sets.The article presents a detailed overview of the mathematical model and algorithmic steps behind this procedure.Finally,the process of rule discovery(prediction phase)by the trained XGBoost model is explained,underscoring its role in real-time,automated data transformations.By employingmachine learning and particularly,the XGBoost model in the context of Business Rule Engine(BRE)data transformation,the article underscores a paradigm shift towardsmore scalable,efficient,and less human-dependent data transformation systems.This research opens doors for further exploration into automated rule discovery systems and their applications in various sectors. 展开更多

AIM: To study the hepatoprotective capacity of Sapindus mukorossi (S. mukorossi) and Rheum emodi (R. emodi) extracts in CCl4 treated male rats. METHODS: The dried powder of S. mukorossi and R. emodi was extracted successively with petroleum ether, benzene, chloroform, and ethanol and concentrated in vacuum. Primary rat hepatocyte monolayer cultures were used for in vitro studies. In vivo, the hepatoprotective capacity of the extract of the fruit pericarp of S. mukorossi and the rhizomes of R. emodi was analyzed in liver injured CCl4-treated male rats. RESULTS: In vitro: primary hepatocytes monolayer cultures were treated with CCl4 and extracts of S. mukorossi & R. emodi. A protective activity could be demonstrated in the CCl4 damaged primary monolayer culture. In vivo : extracts of the fruit pericarp of S. mukorossi (2.5 mg/mL) and rhizomes of R. emodi (3.0 mg/mL) were found to have protective properties in rats with CCl4 induced liver damage as judged from serum marker enzyme activities. CONCLUSION: The extracts of S. mukorossi and R. emodi do have a protective capacity both in vitro on primary hepatocytes cultures and in in vivo in a rat model of CCl4 mediated liver injury. 展开更多

A simple and rapid liquid chromatography-tandem mass spectrometric(LC-MS/MS) assay method has been developed and fully validated for simultaneous quantification of pioglitazone and candesartan in human plasma.Irbesartan was used as an internal standard.The analytes were extracted from human plasma samples by solid-phase extraction technique using a Strata-X 33 mm polymeric sorbent.The reconstituted samples were chromatographed on a C18 column by using a 80:20(v/v) mixture of acetonitrile and 0.1% formic acid as the mobile phase at a flow rate of 0.8 mL/min.The calibration curves obtained were linear(rZ0.99) over the concentration range of 15-3000 ng/mL for pioglitazone and 5-608 ng/mL for candesartan.The results of the intra-and inter-day precision and accuracy studies were well within the acceptable limits.A run time of 2.7 min for each sample made it possible to analyze more than 300 plasma samples per day.The proposed method was found to be applicable to clinical studies. 展开更多

A rapid and sensitive liquid chromatography-tandem mass spectrometric(LC-MS/MS) assay method has been developed and fully validated for the simultaneous quantification of pravastatin and aspirin in human plasma.Furosemide was used as an internal standard.Analytes and the internal standard were extracted from human plasma by liquid-liquid extraction technique using methyl tertiary butyl ether.The reconstituted samples were chromatographed on a Zorbax SB-C;8 column by using a mixture of 5 mM ammonium acetate buffer and acetonitrile(20:80,v/v) as the mobile phase at a flow rate of 0.8 mL/min.The calibration curve obtained was linear(r≥0.99) over the concentration range of 0.50-600.29 ng/mL for pravastatin and 20.07-2012.00 ng/mL for aspirin.Method validation was performed as per FDA guidelines and the results met the acceptance criteria.A run time of 2.0 min for each sample made it possible to analyze more than 400 human plasma samples per day.The proposed method was found to be applicable to clinical studies. 展开更多

Sclerotium rolfsii Sacc. is a destructive soilborne fungal pathogen with a wide host range that includes peanuts. Biological control offers an interesting alternative to fungicides for sustainable management of soilborne diseases. The current investigation is aimed at evaluating one potential biocontrol agent Streptomyces sp. RP1 A-12 for growth promotion and the management of peanut stem rot disease caused by S. rolfsii under field conditions. Preliminary studies conducted under in vitro and the greenhouse conditions showed promising results against the stem rot pathogen. Further in vitro and pot experiments conducted to assess Streptomyces sp. RP1 A-12 for its growth promoting abilities using whole organisms have shown an increase in seed germination, root and shoot length. Other parameters like nodule number and plant biomass were also significantly increased over control treatments indicating that the test bioagent possesses growth promoting abilities along with disease suppression capabilities. Subsequently field studies were carried out for two consecutive rainy seasons. The bioagent was applied as whole organism and partially purified crude metabolites. Results indicate the bioagent reduced stem rot disease incidence by 64–67% and 22–49% respectively in two field trials conducted with notable increase in yield. Partially purified Streptomyces sp. RP1 A-12 metabolites exhibited an even greater effect in reducing the incidence and severity of stem rot compared to the pathogen inoculated control. 展开更多

Coronary heart disease (CHD) that is due to atherosclerosis is associated with low-grade systemic inflammation. Congestive cardiac failure and arrhythmias that are responsible for mortality in CHD can be suppressed by appropriate vagal stimulation that is anti-inflammatory in nature. Acetylcholine, the principal vagal neurotransmitter, is a potent anti-inflammatory molecule. Polyunsaturated fatty acids (PUFAs) augment acetylcholine release, while acetylcholine can enhance the formation of prostacyclin, lipoxins, resolvins, protectins and maresins from PUFAs, which are anti-inflammatory and anti-arrhythmic molecules. Furthermore, plasma and tissue levels of PUFAs are low in those with CHD and atherosclerosis. Hence, vagal nerve stimulation is beneficial in the prevention of CHD and cardiac arrhythmias. Thus, measurement of catecholamines, acetylcholine, various PUFAs, and their products lipoxins, resolvins, protectins and maresins in the plasma and peripheral leukocytes, and vagal tone by heart rate variation could be useful in the prediction, prevention and management of CHD and cardiac arrhythmias. 展开更多

Maraging steel (250) and 13-8 Mo stainless steel plates were joined by gas tungsten constricted arc welding(GTCAW) process in similar and dissimilar metal combinations using 13-8 Mo stainless steel filler wire. The similar and dissimilar metal welds made in solutionized condition were subjected to standard post weld hardening treatments direct ageing at 485 ℃, soaking for 31/2 hours followed by air cooling(ageing treatment of maraging steel) and direct ageing at 510 ℃, soaking for 4 h followed by air cooling(ageing treatment of 13-8 Mo stainless steel). The joint characterization studies include microstructure examination, microhardness survey across the weldments and transverse weld tensile test.Similar and dissimilar metal weldments responded to both the post weld ageing treatment. After post weld aging, increase in yield strength, UTS and slight reduction in % elongation of similar and dissimilar metal were observed. The observed tensile properties were correlated with microstructure and hardness distribution across the welds. 展开更多

Non-alcoholic fatty liver disease (NAFLD) is a low-grade systemic inflammatory condition, since liver and adipose tissue tumor necrosis factor-α (TNF-α) and TNF receptor 1 transcripts and serum TNF-α levels are increased and IL-6-/-mice are less prone to NAFLD. Fatty liver damage caused by high-fat diets is associated with the generation of pro-inflammatory prostaglandin E2 (PGE2). A decrease in the levels of arachidonic acid (AA), eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) and the usefulness of EPA and DHA both in the prevention and management of NAFLD has been reported. AA, EPA and DHA and their anti-inflammatory products lipoxins (LXs), resolvins and protectins suppress IL-6 and TNF-α and PGE2 production. These results suggest that the activities of △6 and △5 desaturases are reduced in NAFLD and hence, the dietary essential fatty acids, linoleic acid (LA) and α-linolenic acid (ALA) are not metabolized to their long-chain products AA, EPA and DHA, the precursors of anti-inflammatory molecules, LXs, resolvins and protectins that could prevent NAFLD. This suggests that an imbalance between proand anti-inflammatory bioactive lipids contribute to NAFLD. Hence, it is proposed that plasma and tissue levels of AA, EPA, DHA and LXs, resolvins and protectins could be used as predictors and prognostic biomarkers of NAFLD. It is suggested that the synthesis and use of more stable analogues of LXs, resolvins and protectins need to be explored in the prevention and management of NAFLD. 展开更多

CD133+ prostate cancer stem cells (PCSCs) have recently been identified in human prostate cancer tissues. The present study reports the integrin profile of prostate cancer progenitor cells and the role of α1 and β1 integrins in the homing and differentiation of PCSCs in vitro. PCSCs were isolated from the tissue specimens of patients with prostate cancer and the expression of surface integrins and adhesion patterns were determined. Our analysis of the expression of surface integrins and their adhesion patterns of prostate cancer stem cells derived from prostate cancer tissues revealed that the levels of β1 and α2β1 integrins were significantly higher (P 〈 0.05) than those of the other integrins. By contrast, peripheral blood-derived CD 133+ cells from prostate cancer patients showed a high level of expression (P 〈 0.01) of α2β1,αvβ3, αvβ5, β1 and α1 integrins and a minimal expression of α4β1 integrins. Moreover, CD 133+ cells derived from both prostate cancer tissues and peripheral blood exhibited an increased degree of attachment to extraceUular matrix proteins (P 〈 0.001) and a high expression level of α2β1 integrin. In vitro experiments using blocking antibodies indicated that α1 and β1 integrins have a role in the homing and differentiation of PCSCs. This is the first report to suggest the importance of integrins in mediating the homing and differentiation of PCSCs. 展开更多

The current communication reports in vitro characterization of the optimized hollow floating microballoons of amoxicillin on the basis of micromeritic properties and in vitro minimum inhibitory concentration(MIC).Amoxicillin loaded hollow microballoons were prepared by emulsion solvent diffusion method.The morphological characterization was done on the basis of scanning electron microscopy(SEM).Fourier Transformed Infrared Spectroscopy(FTIR)was used to investigate drugepolymer interactions.The correlation between the in vitro buoyancy of microballoons and their physical properties,e.g.density and porosity were elucidated.The results of FTIR spectroscopy revealed the absence of any drugepolymer interactions.The porosity values of more than 69%and diameter to thickness ratio greater than 2.90,proved a high cavity volume within the microballoons in all size ranges.The spherical shape of microballoons with hollow internal cavity was confirmed from SEM photomicrographs.The in vitro MIC results showed a sustained drug effect from the microballoons.In conclusion,it can be said that the developed microballoons can be used for the effective treatment of Helicobacter pylori induced gastric ulcer. 展开更多

A high throughput ultra pressure liquid chromatography-mass spectrometry (UPLC-MS/MS) method with good sensitivity and selectivity has been developed and validated for simultaneous quantification of esomeprazole, rabeprazole and levosulpiride in human plasma using lansoprazole as internal standard (IS). The extraction method based on liquid-liquid extraction technique was used to extract the analytes and IS from of 50 μL of human plasma using methyl tert-butyl ether:ethyl acetate (80:20, v/v), which offers a high recovery. Chromatographic separation of analytes and IS was achieved on a Hypersil gold C18 column using gradient mobile phase consisting of 2 mM ammonium formate/acetonitrile. The flow rate was set at 0.5 mL/min to elute all the analytes and IS within 1.00 min runtime. Detection of target compounds was performed on a triple quadruple mass spectrometer by multiple reaction monitoring (MRM) mode via positive electrospray ionization (ESI). Method validation results demonstrated that the developed method has good precision and accuracy over the concentration ranges of 0.1-2000 ng/mL for each analyte. Stability of compounds was established in a battery of stability studies, i.e., bench top, autosampler, dry extract and long-term storage stability as well as freeze-thaw cycles. The validated method has been successfully applied to analyze human plasma samples for application in pharmaco- kinetic studies. 展开更多

Objective:To investigate the analgesic activity of methanolic extract of Amaranthus viridis(A. viridis),Amaranthus caudatus(A.caudatus) and Amaranthus spinosus(A.spinosus).Methods:In this study,the analgesic activity of methanol extracts of all three plants at doses of 200 and 400 mg/kg were investigated by acetic acid-induced writhings test,hot plate test and tail immersion test for mice.Results:It was found that all the three plants showed significant pain management effect(P【0.01) at a dose of 400 mg/kg,but showed a less significant effect at a dose of 20 mg/kg in the entire tests used for evaluation of analgesic activities(P【0.05).Conclusions:Methanol extracts of A.viridis,A.caudatus and A.spinosus show potent analgesic activities,and this study provides the scientific proof for their traditional claims. 展开更多

Firstly Mg(x)Fe(1–x)O (magnesiwuestite) nano powder samples were synthesised by low temperature chemical combustion method. Secondly the as synthesised Mg(x)Fe(1–x)O nanoparticles are used to synthesis magnetic core-shell Nano particles byusing 2-propanol, poly ethylene glycol (PEG), ammonia solution 30 wt%, tetraethyl orthosilicate (TEOS). Separation of the core-shell magnetic nanoparticles from the aqueous suspension using a centrifuge. The synthesised MNPs and core shell MNP were characterized by X-ray diffraction (XRD), Thermal gravimetric-differential thermal analyzer (TG-DTA), Transmission electron microscopy (TEM), scanning electron microscopy (SEM), (EDAX) for structural, thermal and morphological respectively. The most suitable bio-analytical method based on liquid liquid extraction has been developed and validated for quantification of Rasagiline in human plasma. Rasagiline-13C3 mesylate was used as an internal standard for Rasagiline. Zorbax Eclipse Plus C18 (2.1 mm × 50 mm, 3.5 um) column provided chromatographic separation of analyte followed by detection with mass spectrometry. The method involved simple isocratic chromatographic condition and mass spectrometric detection in the positive ionization mode using an API-4000 system. The lotal run time was 3.0 min. The proposed method has been validated with the linear range of 5 12000 pg/mL for Rasagiline. The intra-run and inter-run precision values were within 1.3% 2.9% and 1.6% 2.2% respectively for Rasagiline. The overall recovery for Rasagiline and Rasagiline-13C3 mesylate analog was 96.9% and 96.7% respectively. This validated method was successfully applied to the bioequivalence and pharmacokinetic study of human volunteers under fasting condition. 展开更多

It labels the speech... In audio stream containing multiple speakers, speaker diarization aids in ascertaining "who speak when". This is an unsupervised task as there is no prior information about the speakers. It labels the speech signal conforming to the identity of the speaker, namely, input audio stream is partitioned into homogeneous segments. In this work, we present a novel speaker diarization system using the Tangent weighted Mel frequency cepstral coefficient(TMFCC) as the feature parameter and Lion algorithm for the clustering of the voice activity detected audio streams into particular speaker groups. Thus the two main tasks of the speaker indexing, i.e., speaker segmentation and speaker clustering, are improved. The TMFCC makes use of the low energy frame as well as the high energy frame with more effect, improving the performance of the proposed system. The experiments using the audio signal from the ELSDSR corpus datasets having three speakers, four speakers and five speakers are analyzed for the proposed system. The evaluation of the proposed speaker diarization system based on the tracking distance, tracking time as the evaluation metrics is done and the experimental results show that the speaker diarization system with the TMFCC parameterization and Lion based clustering is found to be superior over existing diarization systems with 95% tracking accuracy. 展开更多

A packed column supercritical fluid chromatography (SFC) method for the separation of clopidogrel enantiomers on a chiral stationary phase and CO2 with modifier as mobile phase has been developed at an analytical scale. Among 11 different 2 stationary phases the Chiral cel OD-H column showed by far the best separation properties. The influence of different modifiers, injection solvents, temperature, and pressure, and density of the fluid, respectively, on the separation behaviour has been studied. It was found that the separation behaviour strongly depends on the type of modifier and the modifier content. Temperature and pressure are of less influence. 展开更多
