Rice(Oryza sativa L.)is one of the most important cereal crops in the world.Bakanae disease is a significant rice disease widely distributed in rice-growing regions worldwide.Therefore,the present investigation aimed ...Rice(Oryza sativa L.)is one of the most important cereal crops in the world.Bakanae disease is a significant rice disease widely distributed in rice-growing regions worldwide.Therefore,the present investigation aimed to assess the optimal concentrations of paclobutrazol(PBZ)as a treatment for rice grains(cv.Sakha 108)to control bakanae disease,also evaluating its impact on grain germination,seedling growth parameters as well as disease index.Paclobutrazol concentrations had no significant impact on seed germination,regardless of whether the seeds were incubated with Fusarium fujikuroi or not.Application of PBZ,either alone or in combination with fungal pathogens,negatively impacted the rice seedlings’height.Paclobutrazol at 25,50 and 100 mg/L,combined with the fungal pathogen positively impacted root length.Paclobutrazol at 3 and 6 mg/L mitigated the adverse impact on chlorophyll pigments content in infected seedlings.The highest proline contents were achieved by 100 mg/L PBZ alone or in combination with fungal pathogens.It has been observed that the application of PBZ,either alone or in combination with a fungal pathogen,leads to the enhancement of catalase,peroxidase,and polyphenol oxidase activities.The median lethal concentration of PBZ was 0.874 mg/L;applying low concentrations of paclobutrazol effectively increased the percentage of fungal growth suppression.Application of PBZ,at higher concentrations(50 and 100 mg/L),decreased infection percentage and disease severity index(DSI)significantly.These findings suggest that PBZ can be an effective treatment for controlling bakanae disease and enhancing resistance in rice plants.展开更多
Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the ...Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the quality traits studied.For every trait under study,the phenotypic coefficient of variation was higher than the correspon-dence genotypic coefficient of variation.Heritability in a broad sense varied from 29.75%for grain length to 98.31%for the elongation trait.Hulling percentage recovery had a highly significant positive correlation with milling and head rice percentage.Consequently,milling percentage had a highly positive correlation with head rice percentage.In amylose percentage,all the genotypes belonged to low amylose except the Hassawi-1 variety,which had intermediate amylose content.Mineral nutrition contents of magnesium(Mg),sodium(Na),potas-sium(K),calcium(Ca),copper(Cu),manganese(Mn),zinc(Zn),iron(Fe),or protein percentage gave different variations for 24 rice genotypes under all the nutritional elements.Among the 24 genotypes,ten rice genotypes–HighNutrient-1,HighNutrient-2,HighNutrient-9,HighNutrient-8,HighNutrient-3,Hassawi-2,HighNutrient-7,HighNutrient-6,Hassawi-1,and HighNutrient-4–had the highest heist value for all nutritional and protein con-tents,and could be used as a donor to improving new varieties.There was a positive and significant correlation between magnesium Mg,K,Zn and Fe.Consequently,K had a positive correlation with zinc Zn,Fe,and protein percentage.Clustering analysis was divided into two groups:thefirst group included all genotypes rich in nutri-ents,while the remaining genotypes with low nutritional content were included in the second group.展开更多

The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharve...The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharvest soft rot(Rhizopus stolonifer)incidence following trimming,and washing,ultraviolet light(UV-C)treatment was used as a main sanitizer for eliminating the soft rot.Exposure of the roots to UV-C(254 nm)was applied in a UV-C room on freshly harvested and cured sweet potato while rolling up on a movable line at 20 cm distance for 1,2,and 3 hr.As combining UV-C treatment with chlorine(200 ppm)on roots,marked and significant reduction of the total microbial load and Rhizopus potential was achieved on root surfaces respectively compared with chlorine alone.It also reduced soft rot percentage to almost 0%infection.After 3 months of cold-storage,quality assessment of sweet potato showed that root characteristics were markedly maintained.The ability of UV-C light to induce phenylalanine ammonia lyase(PAL)enzyme activity in root tissue and maintain the activities of peroxidase and polyphenol oxidase,however with slight increase,was detected.UV-C caused an increase of phenol content in sweet potato tissue that made an activation of defense reaction against the rot causal pathogen.As the exposure time to UV-C light increased,a higher content of phenols occurred.Moreover,UV-C application caused decrease in sugar content of root tissue that is flavored by soft rot-causal pathogen.展开更多
Monogenic lines,which carried 23 genes for blast resistance were tested and used donors to transfer resistance genes by crossing method.The results under blast nursery revealed that 9 genes from 23 genes were suscepti...Monogenic lines,which carried 23 genes for blast resistance were tested and used donors to transfer resistance genes by crossing method.The results under blast nursery revealed that 9 genes from 23 genes were susceptible to highly susceptible under the three locations(Sakha,Gemmeza,and Zarzoura in Egypt);Pia,Pik,Pik-p,Piz-t,Pita,Pi b,Pi,Pi 19 and Pi 20.While,the genes Pii,Pik-s,Pik-h,Pi z,Piz-5,Pi sh,Pi 3,Pi 1,Pi 5,Pi 7,Pi 9,Pi 12,Pikm and Pita-2 were highly resistant at the same locations.Clustering analysis confirmed the results,which divided into two groups;the first one included all the susceptible genes,while the second one included the resistance genes.In the greenhouse test,the reaction pattern of five races produced 100%resistance under artificial inoculation with eight genes showing complete resistance to all isolates.The completely resistant genes:Pii,Pik-s,Piz,Piz-5(=bi2)(t),Pita(=Pi4)(t),Pita,Pi b and Pi1 as well as clustering analysis confirmed the results.In the F1 crosses,the results showed all the 25 crosses were resistant for leaf blast disease under field conditions.While,the results in F2 population showed seven crosses with segregation ratio of 15(R):1(S),two cross gave segregated ratio of 3 R:1 S and one gave 13:3.For the identification of blast resistance genes in the parental lines,the marker K3959,linked to Pik-s gene and the variety IRBLKS-F5 carry this gene,which was from the monogenic line.The results showed that four genotypes;Sakha 105,Sakha 103,Sakha 106 and IRBLKS-F5 were carrying Pik-s gene,while was absent in the Sakha 101,Sakha 104,IRBL5-M,IRBL9-W,IRBLTACP1 and IRBL9-W(R)genotypes.As for Pi 5 gene,the results showed that it was present in Sakha 103 and Sakha 104 varieties and absent in the rest of the genotypes.In addition,Pita-Pita-2 gene was found in the three Egyptian genotypes(Sakha 105,Sakha 101 and Sakha 104)plus IRBLTACP1 monogenetic.In F2 generation,six populations were used to study the inheritance of blast resistance and specific primers to confirm the ratio and identify the resistance genes.However,the ratios in molecular markers were the same of the ratio under field evaluation in the most population studies.These findings would facilitate in breeding programs for gene pyramiding and gene accumulation to produce durable resistance for blast using those genotypes.展开更多
Production of peaches(Prunus persica(L.)Batsch)for both local market and export is increasing each year in Egypt.Brown rot disease,caused by Monilinia laxa and Monilinia fructigena,is considered one of the most import...Production of peaches(Prunus persica(L.)Batsch)for both local market and export is increasing each year in Egypt.Brown rot disease,caused by Monilinia laxa and Monilinia fructigena,is considered one of the most important postharvest rots affecting peaches in Egypt and economic losses are increasing.Antifungal activity of glycyrrhizic acid nanoparticles(GA-NPs)and glycyrrhizic acid(GA)at 0.2 and 0.4 mmol/L was investigated as a control for both these brown rot pathogens on peach fruits in both in vitro and in vivo studies.In the in vitro studies,GA-NPs were the most effective as shown by the ability to decrease linear growth of both brown rot pathogens in potato dextrose agar(PDA)amended with 0.4 mmol/L GA-NPs.Micrographs of M.fructigena exposed to 0.4 mmol/LGA showed mycelial deformations,nodule formation,detachment of the cell wall,shrinkage and inhomogeneous cytoplasmic materials with large vacuoles.Mycelium of M.laxa exposed to 0.4 mmol/LGA-NPs resulted in thinner and distorted hyphae,nodule formation,cell wall thinning,and swellings.The GANPs and GA treatments improved fruit quality by maintaining firmness and total soluble solids(TSS).GA-NPs were more effective in decreasing decay incidence than their bulk material.The 0.4 mmol/L GA-NPs completely inhibited the disease on naturally infected peach fruits for both seasons of 2018 and 2019.Furthermore,0.4 mmol/L GA-NPs reduced the disease incidence in inoculated fruits by 95(M.laxa)and 88%(M.fructigena)in 2018 season and 96(M.laxa)and 85%(M.fructigena)in 2019 season.In conclusion,GA-NPs could enhance the resistance of peaches against brown rot caused by M.laxa and M.fructigena.展开更多
Citrus Tristeza Virus(CTV),usually occurs in nature as a mixture of genotypes.Six naturally infected citrus(Citrus sinensis)trees grafted on sour orange rootstock were collected from three citrus growing governorates ...Citrus Tristeza Virus(CTV),usually occurs in nature as a mixture of genotypes.Six naturally infected citrus(Citrus sinensis)trees grafted on sour orange rootstock were collected from three citrus growing governorates in Egypt(Sharqia,Qalyubia and Garbia).In this study,RT-PCR,Single-Strand Conformation Polymorphism(SSCP)and nucleotide sequence analysis were used for four independent CTV genomic regions(p65,p18,p20,and p23)to detect and assess the sequence and genetic variabilities among CTV Egyptian isolates.RTPCR products(650 bp)for the CTV p23 gene obtained from the selected isolates were used for the SSCP analysis and DNA sequencing.SSCP patterns of p23 gene for individual isolates yielded different complex haplotype patterns.Nucleotide sequence analysis of p23 region amplified from six isolates under study revealed that p23 shared high nucleotide identity 98.7%with T36 isolate from USA,Florida.Phylogenetic analysis of p23 gene indicated a close evolutionary relationship between all examined isolates and Qaha isolate(T36 isolate group),suggesting that they may have originated from closely related ancestors.Nucleotide sequence analysis of the three genes located on CTV 3′-coterminal overhang,p18,p20 and p65,amplified from isolate A3,Sharqia governorate,revealed that the p18,p65,and p20 genes were related to the T3-KB isolate from South Africa with 99%–100%sequence homology.Phylogenetic relationship analysis for p65,p18 and p20 ORFs clustered the current A3 isolate with T3 genotype group.The recombination analysis identified three of six isolates from Sharqia,and Garbia as potential recombinant for p23 gene.The isolates T36 and T3 were identified as major donors for recombination events in isolate A3.Our results concluded that p23 ORF likely to be as a hotspot region for recombination and originated through recombination event.The current study indicated that recombination is an important factor for the origin of CTV strains in Egypt.展开更多
Deficiencies of essential vitamins,iron(Fe),and zinc(Zn)affect over one-half of the world’s population.A significant progress has been made to control micronutrient deficiencies through supplementation,but new approa...Deficiencies of essential vitamins,iron(Fe),and zinc(Zn)affect over one-half of the world’s population.A significant progress has been made to control micronutrient deficiencies through supplementation,but new approaches are needed,especially to reach the rural poor.Agronomic biofortification of pulses with Zn,Fe,and boron(B)offers a pragmatic solution to combat hidden hunger instead of food fortification and supplementation.Moreover,it also has positive effects on crop production as well.Therefore,we conducted three separate field experiments for two consecutive years to evaluate the impact of soil and foliar application of the aforementioned nutrients on the yield and seed biofortification of mungbean.Soil application of Zn at 0,4.125,8.25,Fe at 0,2.5,5.0 and B at 0,0.55,1.1 kg ha−1 was done in the first,second and third experiment,respectively.Foliar application in these experiments was done at 0.3%Zn,0.2%Fe and 0.1%B respectively one week after flowering initiation.Data revealed that soil-applied Zn,Fe and B at 8.25,5.0 and 1.1 kg ha−1,respectively,enhanced the grain yield of mungbean;however,this increase in yield was statistically similar to that recorded with Zn,Fe and B at 4.125,2.5 and 0.55 kg ha−1,respectively.Foliar application of these nutrients at flower initiation significantly enhanced the Zn contents by 28%and 31%,Fe contents by 80%and 78%,while B contents by 98%and 116%over control during 2019 and 2020,respectively.It was concluded from the results that soil application of Zn,Fe,and B enhanced the yield performance of mungbean;while significant improvements in seed Zn,Fe,and B contents were recorded with foliar application of these nutrients.展开更多

In this study we investigated whether root colonization of tomato (Solanum lycopersicum) infected by Trichoderma harzianum leads to alterations in the biosynthesis of secondary plant metabolites including phytohormones and osmolyte proline under drought stress. Exposure of tomato to drought caused a drastic decline in plant growth and physiological parameters. Tomato inoculated with T. harzianum showed increased root and shoot growth and chlorophyll pigments as compared to uninoculated controls as well as drought stressed plants. Proline and total soluble protein content was increased in plants inoculated with T. harzianum under both normal as well as drought conditions. An obvious increase in phenol and flavonoid content was observed due to T. haczianum. In addition, T. hat-zianum inoculated plants maintained higher levels of growth regulators indole acetic acid, indole butyric acid, and gibberellic acid under drought stress. Improved secondary metabolites which play an important role in plant stress tolerance by T. hat-zianum may have coordinately worked for bringing the growth regulation by protecting membranes from reactive oxygen species (ROS) and enhance plant growth through accessing more nutrients by root system.展开更多
Four isolates of Bacillus subtilis coded,B4,B7,B8 and B10 were examined as biocontrol agents for their abilities and antagonistic effect on the in vitro growth of certain phytopathogenic fungi of peanut,Rhizoctonia so...Four isolates of Bacillus subtilis coded,B4,B7,B8 and B10 were examined as biocontrol agents for their abilities and antagonistic effect on the in vitro growth of certain phytopathogenic fungi of peanut,Rhizoctonia solani and Sclerotium rolfsii.Bacillus subtilis isolate B4(GenBank accession no.EF150884)was the highly effective one for inhibiting the fungal mycelial growth.Batch fermentation of B.subtilis isolate B4 was carried out and the maximum biomass achieved was 4.53 g L-1 at 11 h.Bacillus subtilis isolate B4 was formulated and evaluated as a biofungicide to reduce peanut soil-borne diseases under greenhouse and field conditions at the side of Rizolex-T(fungicide)as standard.Treatments by formulated plant growth-promoting rhizobacteria(PGPR)B.subtilis B4 and Rizolex-T in a soil infested with R.solani,S.rolfsii and mixture of them were more effective in decreasing percentage of damping-off,root and pod rot disease incidence(%)in greenhouse and open field environment during the two seasons 2015 and 2016.Treatments by PGPR gave highly dry weight and number of healthy pods compared to control of fungi treatment which was nearby to dry weights of healthy pods achieved by treatments by Rizolex-T in a soil infested with S.rolfsii,R.solani and mixture of them.Formulated PGPR B.subtilis B4 gave higher increasing of yield percentage than treatment by Rizolex-T in the two evaluated seasons 2015 and 2016.It can conclude that the produced bioforumlated agent was more efficient as fungicide when compared with the other chemical synthesized fungicides,safe for human and the environment and economy.展开更多
Gibberellins(GAs)are an important hormone in regulating plant growth and development,and DELLA protein is an essential negative regulator of GA signal transduction.The aim of the study was to clone a GAinhibiting prot...Gibberellins(GAs)are an important hormone in regulating plant growth and development,and DELLA protein is an essential negative regulator of GA signal transduction.The aim of the study was to clone a GAinhibiting protein DELLA from trifoliate orange(Poncirus trifoliata L.Raf.)and to analyze the bioinformations and expression patterns of the protein gene in tissues and in response to drought stress.A DELLA protein was isolated from trifoliate orange and named as PtGai(Genebank number:MZ170959).The PtGai protein had 1731 bp open reading frames,along with 576 amino acid codes,and also grouped with sweet orange(XM_006430552.4).The PtGai protein sequence was 65%homology with the sequences of DELLA proteins in other plant families.PtGai protein existed in the nucleus based on the prediction of subcellular localization.PtGai protein could be expressed in roots,stems,and leaves,along with the highest expression in stems.PtGai was upregulated by drought stress in leaves and roots,along with the decrease of root total GA concentration and the inhibition of shoot and root biomass production.It indicated the characteristics of PtGai protein and the roles of PtGai in GA synthesis and plant growth.展开更多

Ea...Many analytic strategies have emerged to estimate plant responses to Fusarium wilt. The demand for fast and reliable method (diagnosis, prediction) to determine isolate strength accurately is not established yet. Early determination of pathogen strength helps in plant medication. The aim of this study was to develop a faster strategy and method for early determination of fungal isolates strength in correlation to plant response. Till now, the scientists have no consensus on the most correlated parameters that could express wilt precisely. In this study, 30 isolates of Fusarium oxysporum isolated from Lupinus termis L. were used to provide an explicit image about the real strength of Fusarium isolates and its impact on the plant. Wilting percentage ranged from 26.67% to 93.33% of the infected plants depending on isolate virulence. Some of cellular, morphological and physical measurements were conducted on 8 out of 30 isolates, including root (length, fresh weight (FW) and dry weight (DW)), nodules (water content (WC), FW, DW), stem (height, WC, FW, DW), total leaves/plant (WC, FW, DW) and the fourth leaf (WC, FW, DW, leaf area, epidermal cell area, epidermal cell number, succulence). Hierarchical clustering was used to determine the variance between the isolates. Detrended correspondence analysis (DCA) and canonical correspondence analysis (CCA) were used to determine the most important growth parameters that could express wilting accurately. The CCA results showed that most of the measured parameters on the fourth leaf, except for leaf epidermal cell number, were highly and positively correlated to wilt. That makes these specific parameters valuable and sensitive for any changes in isolates strength. Accordingly, a mathematical model was created to be helpful in the quick determination of isolate strength and precise medication.展开更多

Studies by using scanning electron microscopy (SEM) confirmed the importance of the seed coat, and seed cells as infection sites as well as location of the mycelium of the investigated fungus. Taxonomic names are key links between various databases that store information on different organisms.Several global fungal nomenclural and taxonomic databases(notably Index Fungorum,Species Fungorum and MycoBank)can ...Taxonomic names are key links between various databases that store information on different organisms.Several global fungal nomenclural and taxonomic databases(notably Index Fungorum,Species Fungorum and MycoBank)can be sourced to find taxonomic details about fungi,while DNA sequence data can be sourced from NCBI,EBI and UNITE databases.Although the sequence data may be linked to a name,the quality of the metadata is variable and generally there is no corresponding link to images,descriptions or herbarium material.There is generally no way to establish the accuracy of the names in these genomic databases,other than whether the submission is from a reputable source.To tackle this problem,a new database(FacesofFungi),accessible at www.facesoffungi.org(FoF)has been established.This fungal database allows deposition of taxonomic data,phenotypic details and other useful data,which will enhance our current taxonomic understanding and ultimately enable mycologists to gain better and updated insights into the current fungal classification system.In addition,the database will also allow access to comprehensive metadata including descriptions of voucher and type specimens.This database is user-friendly,providing links and easy access between taxonomic ranks,with the classification system based primarily on molecular data(from the literature and via updated web-based phylogenetic trees),and to a lesser extent on morphological data when molecular data are unavailable.In FoF species are not only linked to the closest phylogenetic representatives,but also relevant data is provided,wherever available,on various applied aspects,such as ecological,industrial,quarantine and chemical uses.The data include the three main fungal groups(Ascomycota,Basidiomycota,Basal fungi)and fungus-like organisms.The FoF webpage is an output funded by the Mushroom Research Foundation which is an NGO with seven directors with mycological expertise.The webpage has 76 curators,and with the help of these specialists,FoF will provide an updated natural classification of the fungi,with illustrated accounts of species linked to molecular data.The present paper introduces the FoF database to the scientific community and briefly reviews some of the problems associated with classification and identification of the main fungal groups.The structure and use of the database is then explained.We would like to invite all mycologists to contribute to these web pages.展开更多

文摘Twenty-four rice genotypes were examined to assess genetic variability,heritability,and correlations for seven-grain quality traits,eight nutritional elements,and protein.ANOVA revealed significant differences for the quality traits studied.For every trait under study,the phenotypic coefficient of variation was higher than the correspon-dence genotypic coefficient of variation.Heritability in a broad sense varied from 29.75%for grain length to 98.31%for the elongation trait.Hulling percentage recovery had a highly significant positive correlation with milling and head rice percentage.Consequently,milling percentage had a highly positive correlation with head rice percentage.In amylose percentage,all the genotypes belonged to low amylose except the Hassawi-1 variety,which had intermediate amylose content.Mineral nutrition contents of magnesium(Mg),sodium(Na),potas-sium(K),calcium(Ca),copper(Cu),manganese(Mn),zinc(Zn),iron(Fe),or protein percentage gave different variations for 24 rice genotypes under all the nutritional elements.Among the 24 genotypes,ten rice genotypes–HighNutrient-1,HighNutrient-2,HighNutrient-9,HighNutrient-8,HighNutrient-3,Hassawi-2,HighNutrient-7,HighNutrient-6,Hassawi-1,and HighNutrient-4–had the highest heist value for all nutritional and protein con-tents,and could be used as a donor to improving new varieties.There was a positive and significant correlation between magnesium Mg,K,Zn and Fe.Consequently,K had a positive correlation with zinc Zn,Fe,and protein percentage.Clustering analysis was divided into two groups:thefirst group included all genotypes rich in nutri-ents,while the remaining genotypes with low nutritional content were included in the second group.
文摘Awareness policy intended to contribute to changing rural women realities to urgent needs of information and gain knowledge was to be demonstrated through in-depth information and communication technology-based(ICTs-based)training program that focused on the importance of advanced agricultural technologies in the production chain in developing countries like Egypt through access and use of the ICTs.Women are becoming well trained on the detailed steps of improved technologies applied in supply chain.Their increased awareness of the necessity of quality management to be followed during their work in the postharvest handling system helped them to produce high-quality products to meet the export requirements of foreign markets and add value to the export quality.Women have been able to reduce the extremely high losses that occurred due to improper handling in particular.The outcomes of proper and healthy procedures,precautions and personal protection were gained by rural women and technicians working in the supply chain.Moreover,women themselves became more confident in their know-how and more comfortable in transgressing cultural norms that inhibited their progress.
文摘The whole cold-chain for exporting sweet potato(native variety“Abees”),to foreign market included immediate curing operation directly after harvest helped in healing skin texture,however,in order to reduce postharvest soft rot(Rhizopus stolonifer)incidence following trimming,and washing,ultraviolet light(UV-C)treatment was used as a main sanitizer for eliminating the soft rot.Exposure of the roots to UV-C(254 nm)was applied in a UV-C room on freshly harvested and cured sweet potato while rolling up on a movable line at 20 cm distance for 1,2,and 3 hr.As combining UV-C treatment with chlorine(200 ppm)on roots,marked and significant reduction of the total microbial load and Rhizopus potential was achieved on root surfaces respectively compared with chlorine alone.It also reduced soft rot percentage to almost 0%infection.After 3 months of cold-storage,quality assessment of sweet potato showed that root characteristics were markedly maintained.The ability of UV-C light to induce phenylalanine ammonia lyase(PAL)enzyme activity in root tissue and maintain the activities of peroxidase and polyphenol oxidase,however with slight increase,was detected.UV-C caused an increase of phenol content in sweet potato tissue that made an activation of defense reaction against the rot causal pathogen.As the exposure time to UV-C light increased,a higher content of phenols occurred.Moreover,UV-C application caused decrease in sugar content of root tissue that is flavored by soft rot-causal pathogen.

文摘Monogenic lines,which carried 23 genes for blast resistance were tested and used donors to transfer resistance genes by crossing method.The results under blast nursery revealed that 9 genes from 23 genes were susceptible to highly susceptible under the three locations(Sakha,Gemmeza,and Zarzoura in Egypt);Pia,Pik,Pik-p,Piz-t,Pita,Pi b,Pi,Pi 19 and Pi 20.While,the genes Pii,Pik-s,Pik-h,Pi z,Piz-5,Pi sh,Pi 3,Pi 1,Pi 5,Pi 7,Pi 9,Pi 12,Pikm and Pita-2 were highly resistant at the same locations.Clustering analysis confirmed the results,which divided into two groups;the first one included all the susceptible genes,while the second one included the resistance genes.In the greenhouse test,the reaction pattern of five races produced 100%resistance under artificial inoculation with eight genes showing complete resistance to all isolates.The completely resistant genes:Pii,Pik-s,Piz,Piz-5(=bi2)(t),Pita(=Pi4)(t),Pita,Pi b and Pi1 as well as clustering analysis confirmed the results.In the F1 crosses,the results showed all the 25 crosses were resistant for leaf blast disease under field conditions.While,the results in F2 population showed seven crosses with segregation ratio of 15(R):1(S),two cross gave segregated ratio of 3 R:1 S and one gave 13:3.For the identification of blast resistance genes in the parental lines,the marker K3959,linked to Pik-s gene and the variety IRBLKS-F5 carry this gene,which was from the monogenic line.The results showed that four genotypes;Sakha 105,Sakha 103,Sakha 106 and IRBLKS-F5 were carrying Pik-s gene,while was absent in the Sakha 101,Sakha 104,IRBL5-M,IRBL9-W,IRBLTACP1 and IRBL9-W(R)genotypes.As for Pi 5 gene,the results showed that it was present in Sakha 103 and Sakha 104 varieties and absent in the rest of the genotypes.In addition,Pita-Pita-2 gene was found in the three Egyptian genotypes(Sakha 105,Sakha 101 and Sakha 104)plus IRBLTACP1 monogenetic.In F2 generation,six populations were used to study the inheritance of blast resistance and specific primers to confirm the ratio and identify the resistance genes.However,the ratios in molecular markers were the same of the ratio under field evaluation in the most population studies.These findings would facilitate in breeding programs for gene pyramiding and gene accumulation to produce durable resistance for blast using those genotypes.
文摘Production of peaches(Prunus persica(L.)Batsch)for both local market and export is increasing each year in Egypt.Brown rot disease,caused by Monilinia laxa and Monilinia fructigena,is considered one of the most important postharvest rots affecting peaches in Egypt and economic losses are increasing.Antifungal activity of glycyrrhizic acid nanoparticles(GA-NPs)and glycyrrhizic acid(GA)at 0.2 and 0.4 mmol/L was investigated as a control for both these brown rot pathogens on peach fruits in both in vitro and in vivo studies.In the in vitro studies,GA-NPs were the most effective as shown by the ability to decrease linear growth of both brown rot pathogens in potato dextrose agar(PDA)amended with 0.4 mmol/L GA-NPs.Micrographs of M.fructigena exposed to 0.4 mmol/LGA showed mycelial deformations,nodule formation,detachment of the cell wall,shrinkage and inhomogeneous cytoplasmic materials with large vacuoles.Mycelium of M.laxa exposed to 0.4 mmol/LGA-NPs resulted in thinner and distorted hyphae,nodule formation,cell wall thinning,and swellings.The GANPs and GA treatments improved fruit quality by maintaining firmness and total soluble solids(TSS).GA-NPs were more effective in decreasing decay incidence than their bulk material.The 0.4 mmol/L GA-NPs completely inhibited the disease on naturally infected peach fruits for both seasons of 2018 and 2019.Furthermore,0.4 mmol/L GA-NPs reduced the disease incidence in inoculated fruits by 95(M.laxa)and 88%(M.fructigena)in 2018 season and 96(M.laxa)and 85%(M.fructigena)in 2019 season.In conclusion,GA-NPs could enhance the resistance of peaches against brown rot caused by M.laxa and M.fructigena.

文摘Citrus Tristeza Virus(CTV),usually occurs in nature as a mixture of genotypes.Six naturally infected citrus(Citrus sinensis)trees grafted on sour orange rootstock were collected from three citrus growing governorates in Egypt(Sharqia,Qalyubia and Garbia).In this study,RT-PCR,Single-Strand Conformation Polymorphism(SSCP)and nucleotide sequence analysis were used for four independent CTV genomic regions(p65,p18,p20,and p23)to detect and assess the sequence and genetic variabilities among CTV Egyptian isolates.RTPCR products(650 bp)for the CTV p23 gene obtained from the selected isolates were used for the SSCP analysis and DNA sequencing.SSCP patterns of p23 gene for individual isolates yielded different complex haplotype patterns.Nucleotide sequence analysis of p23 region amplified from six isolates under study revealed that p23 shared high nucleotide identity 98.7%with T36 isolate from USA,Florida.Phylogenetic analysis of p23 gene indicated a close evolutionary relationship between all examined isolates and Qaha isolate(T36 isolate group),suggesting that they may have originated from closely related ancestors.Nucleotide sequence analysis of the three genes located on CTV 3′-coterminal overhang,p18,p20 and p65,amplified from isolate A3,Sharqia governorate,revealed that the p18,p65,and p20 genes were related to the T3-KB isolate from South Africa with 99%–100%sequence homology.Phylogenetic relationship analysis for p65,p18 and p20 ORFs clustered the current A3 isolate with T3 genotype group.The recombination analysis identified three of six isolates from Sharqia,and Garbia as potential recombinant for p23 gene.The isolates T36 and T3 were identified as major donors for recombination events in isolate A3.Our results concluded that p23 ORF likely to be as a hotspot region for recombination and originated through recombination event.The current study indicated that recombination is an important factor for the origin of CTV strains in Egypt.

文摘Deficiencies of essential vitamins,iron(Fe),and zinc(Zn)affect over one-half of the world’s population.A significant progress has been made to control micronutrient deficiencies through supplementation,but new approaches are needed,especially to reach the rural poor.Agronomic biofortification of pulses with Zn,Fe,and boron(B)offers a pragmatic solution to combat hidden hunger instead of food fortification and supplementation.Moreover,it also has positive effects on crop production as well.Therefore,we conducted three separate field experiments for two consecutive years to evaluate the impact of soil and foliar application of the aforementioned nutrients on the yield and seed biofortification of mungbean.Soil application of Zn at 0,4.125,8.25,Fe at 0,2.5,5.0 and B at 0,0.55,1.1 kg ha−1 was done in the first,second and third experiment,respectively.Foliar application in these experiments was done at 0.3%Zn,0.2%Fe and 0.1%B respectively one week after flowering initiation.Data revealed that soil-applied Zn,Fe and B at 8.25,5.0 and 1.1 kg ha−1,respectively,enhanced the grain yield of mungbean;however,this increase in yield was statistically similar to that recorded with Zn,Fe and B at 4.125,2.5 and 0.55 kg ha−1,respectively.Foliar application of these nutrients at flower initiation significantly enhanced the Zn contents by 28%and 31%,Fe contents by 80%and 78%,while B contents by 98%and 116%over control during 2019 and 2020,respectively.It was concluded from the results that soil application of Zn,Fe,and B enhanced the yield performance of mungbean;while significant improvements in seed Zn,Fe,and B contents were recorded with foliar application of these nutrients.

文摘Plant secondary metabolites play vital role in plant stress response. In this study we investigated whether root colonization of tomato (Solanum lycopersicum) infected by Trichoderma harzianum leads to alterations in the biosynthesis of secondary plant metabolites including phytohormones and osmolyte proline under drought stress. Exposure of tomato to drought caused a drastic decline in plant growth and physiological parameters. Tomato inoculated with T. harzianum showed increased root and shoot growth and chlorophyll pigments as compared to uninoculated controls as well as drought stressed plants. Proline and total soluble protein content was increased in plants inoculated with T. harzianum under both normal as well as drought conditions. An obvious increase in phenol and flavonoid content was observed due to T. haczianum. In addition, T. hat-zianum inoculated plants maintained higher levels of growth regulators indole acetic acid, indole butyric acid, and gibberellic acid under drought stress. Improved secondary metabolites which play an important role in plant stress tolerance by T. hat-zianum may have coordinately worked for bringing the growth regulation by protecting membranes from reactive oxygen species (ROS) and enhance plant growth through accessing more nutrients by root system.
文摘Four isolates of Bacillus subtilis coded,B4,B7,B8 and B10 were examined as biocontrol agents for their abilities and antagonistic effect on the in vitro growth of certain phytopathogenic fungi of peanut,Rhizoctonia solani and Sclerotium rolfsii.Bacillus subtilis isolate B4(GenBank accession no.EF150884)was the highly effective one for inhibiting the fungal mycelial growth.Batch fermentation of B.subtilis isolate B4 was carried out and the maximum biomass achieved was 4.53 g L-1 at 11 h.Bacillus subtilis isolate B4 was formulated and evaluated as a biofungicide to reduce peanut soil-borne diseases under greenhouse and field conditions at the side of Rizolex-T(fungicide)as standard.Treatments by formulated plant growth-promoting rhizobacteria(PGPR)B.subtilis B4 and Rizolex-T in a soil infested with R.solani,S.rolfsii and mixture of them were more effective in decreasing percentage of damping-off,root and pod rot disease incidence(%)in greenhouse and open field environment during the two seasons 2015 and 2016.Treatments by PGPR gave highly dry weight and number of healthy pods compared to control of fungi treatment which was nearby to dry weights of healthy pods achieved by treatments by Rizolex-T in a soil infested with S.rolfsii,R.solani and mixture of them.Formulated PGPR B.subtilis B4 gave higher increasing of yield percentage than treatment by Rizolex-T in the two evaluated seasons 2015 and 2016.It can conclude that the produced bioforumlated agent was more efficient as fungicide when compared with the other chemical synthesized fungicides,safe for human and the environment and economy.

文摘Gibberellins(GAs)are an important hormone in regulating plant growth and development,and DELLA protein is an essential negative regulator of GA signal transduction.The aim of the study was to clone a GAinhibiting protein DELLA from trifoliate orange(Poncirus trifoliata L.Raf.)and to analyze the bioinformations and expression patterns of the protein gene in tissues and in response to drought stress.A DELLA protein was isolated from trifoliate orange and named as PtGai(Genebank number:MZ170959).The PtGai protein had 1731 bp open reading frames,along with 576 amino acid codes,and also grouped with sweet orange(XM_006430552.4).The PtGai protein sequence was 65%homology with the sequences of DELLA proteins in other plant families.PtGai protein existed in the nucleus based on the prediction of subcellular localization.PtGai protein could be expressed in roots,stems,and leaves,along with the highest expression in stems.PtGai was upregulated by drought stress in leaves and roots,along with the decrease of root total GA concentration and the inhibition of shoot and root biomass production.It indicated the characteristics of PtGai protein and the roles of PtGai in GA synthesis and plant growth.
文摘Many analytic strategies have emerged to estimate plant responses to Fusarium wilt. The demand for fast and reliable method (diagnosis, prediction) to determine isolate strength accurately is not established yet. Early determination of pathogen strength helps in plant medication. The aim of this study was to develop a faster strategy and method for early determination of fungal isolates strength in correlation to plant response. Till now, the scientists have no consensus on the most correlated parameters that could express wilt precisely. In this study, 30 isolates of Fusarium oxysporum isolated from Lupinus termis L. were used to provide an explicit image about the real strength of Fusarium isolates and its impact on the plant. Wilting percentage ranged from 26.67% to 93.33% of the infected plants depending on isolate virulence. Some of cellular, morphological and physical measurements were conducted on 8 out of 30 isolates, including root (length, fresh weight (FW) and dry weight (DW)), nodules (water content (WC), FW, DW), stem (height, WC, FW, DW), total leaves/plant (WC, FW, DW) and the fourth leaf (WC, FW, DW, leaf area, epidermal cell area, epidermal cell number, succulence). Hierarchical clustering was used to determine the variance between the isolates. Detrended correspondence analysis (DCA) and canonical correspondence analysis (CCA) were used to determine the most important growth parameters that could express wilting accurately. The CCA results showed that most of the measured parameters on the fourth leaf, except for leaf epidermal cell number, were highly and positively correlated to wilt. That makes these specific parameters valuable and sensitive for any changes in isolates strength. Accordingly, a mathematical model was created to be helpful in the quick determination of isolate strength and precise medication.
文摘The direct impact of seed-borne fungi on seed is considerable. Many fungi are serious parasites of seed primordial and maturing seeds and reduce yield of seed both quantitatively and qualitatively. Other fungi, including saprophytes and very weak parasites, may lower the quality of seeds by causing discoloration which may seriously depreciate the commercial value of seeds, particularly of grain when graded for consumption. Studies by using scanning electron microscopy (SEM) confirmed the importance of the seed coat, and seed cells as infection sites as well as location of the mycelium of the investigated fungus. Macrophominaphaseolina The present investigation is undertaken to study the colonization, infection and fungal establishment on different sesame seed parts by (SEM). A successful colonization of M. phaseolina to seed tissues was also detected. Different forms of pycnidial shapes were also observed.

文摘Taxonomic names are key links between various databases that store information on different organisms.Several global fungal nomenclural and taxonomic databases(notably Index Fungorum,Species Fungorum and MycoBank)can be sourced to find taxonomic details about fungi,while DNA sequence data can be sourced from NCBI,EBI and UNITE databases.Although the sequence data may be linked to a name,the quality of the metadata is variable and generally there is no corresponding link to images,descriptions or herbarium material.There is generally no way to establish the accuracy of the names in these genomic databases,other than whether the submission is from a reputable source.To tackle this problem,a new database(FacesofFungi),accessible at www.facesoffungi.org(FoF)has been established.This fungal database allows deposition of taxonomic data,phenotypic details and other useful data,which will enhance our current taxonomic understanding and ultimately enable mycologists to gain better and updated insights into the current fungal classification system.In addition,the database will also allow access to comprehensive metadata including descriptions of voucher and type specimens.This database is user-friendly,providing links and easy access between taxonomic ranks,with the classification system based primarily on molecular data(from the literature and via updated web-based phylogenetic trees),and to a lesser extent on morphological data when molecular data are unavailable.In FoF species are not only linked to the closest phylogenetic representatives,but also relevant data is provided,wherever available,on various applied aspects,such as ecological,industrial,quarantine and chemical uses.The data include the three main fungal groups(Ascomycota,Basidiomycota,Basal fungi)and fungus-like organisms.The FoF webpage is an output funded by the Mushroom Research Foundation which is an NGO with seven directors with mycological expertise.The webpage has 76 curators,and with the help of these specialists,FoF will provide an updated natural classification of the fungi,with illustrated accounts of species linked to molecular data.The present paper introduces the FoF database to the scientific community and briefly reviews some of the problems associated with classification and identification of the main fungal groups.The structure and use of the database is then explained.We would like to invite all mycologists to contribute to these web pages.
