Suppression of roots and/or their symbiotic microorganisms,such as mycorrhizal fungi and rhizobia,is an effective way for alien plants to outcompete native plants.However,little is known about how invasive and native ...Suppression of roots and/or their symbiotic microorganisms,such as mycorrhizal fungi and rhizobia,is an effective way for alien plants to outcompete native plants.However,little is known about how invasive and native plants interact with the quantity and activity of nutrient-acquisition agents.Here a pot experiment was conducted with monoculture and mixed plantings of an invasive plant,Xanthium strumarium,and a common native legume,Glycine max.We measured traits related to root and nodule quantity and activity and mycorrhizal colonization.Compared to the monoculture,fine root quantity(biomass,surface area)and activity(root nitrogen(N)concentration,acid phosphatase activity)of G.max decreased in mixed plantings;nodule quantity(biomass)decreased by 45%,while nodule activity in Nfixing via rhizobium increased by 106%;mycorrhizal colonization was unaffected.Contribution of N fixation to leaf N content in G.max increased in the mixed plantings,and this increase was attributed to a decrease in the rhizosphere soil N of G.max in the mixed plantings.Increased root quantity and activity,along with a higher mycorrhizal association was observed in X.strumarium in the mixed compared to monoculture.Together,the invasive plant did not directly scavenge N from nodule-fixed N,but rather depleted the rhizosphere soil N of the legume,thereby stimulating the activity of N-fixation and increasing the dependence of the native legume on this N source.The quantity-activity framework holds promise for future studies on how native legumes respond to alien plant invasions.展开更多

The strawberry species Fragaria nilgerrensis Schlechtendal ex J.Gay,renowned for its distinctive white,fragrant peach-like fruits and strong disease resistance,is an exceptional research material.In a previous study,a...The strawberry species Fragaria nilgerrensis Schlechtendal ex J.Gay,renowned for its distinctive white,fragrant peach-like fruits and strong disease resistance,is an exceptional research material.In a previous study,an ethyl methane sulfonate(EMS)mutant library was established for this species,resulting in various yellow leaf mutants.Leaf yellowing materials are not only the ideal materials for basic studies on photosynthesis mechanism,chloroplast development,and molecular regulation of various pigments,but also have important utilization value in ornamental plants breeding.The present study focused on four distinct yellow leaf mutants:mottled yellow leaf(MO),yellow green leaf(YG),light green leaf(LG),and buddha light leaf(BU).The results revealed that the flavonoid content and carotenoid-to-chlorophyll ratio exhibited a significant increase among these mutants,while experiencing a significant decrease in chlorophyll and carotenoid contents compared to the wild type(WT).To clarify the regulatory mechanisms and network relationships underlying these mutants,the RNA-seq and weighted gene coexpression network(WGCNA)analyses were employed.The results showed flavonoid metabolism pathway was enriched both in MO and YG mutants,while the chlorophyll biosynthesis pathway and carotenoid degradation pathway were only enriched in MO and YG mutants,respectively.Subsequently,key structural genes and transcription factors were identified on metabolic pathways of three pigments through correlation analyses and quantitative experiments.Furthermore,a R2R3-MYB transcription factor,FnMYB4,was confirmed to be positively correlated with flavonoid synthesis through transient overexpression,virus-induced gene silencing(VIGS),and RNA interference(RNAi),accompanying by reoccurrence and attenuation of mutant phenotype.Finally,dual-luciferase(LUC)and yeast one-hybrid assays confirmed the binding of FnMYB4 to the FnFLS and FnF3H promoters,indicating that FnMYB4 positively regulates flavonoid synthesis.In addition,correlation analyses suggested that FnMYB4 also might be involved in chlorophyll and carotenoid metabolisms.These findings demonstrated the pivotal regulatory role of FnMYB4 in strawberry leaf coloration.展开更多
This study investigated the effects of a xylitol-casein non-covalent complex(XC)on parameters related to type 2 diabetes mellitus(T2DM),in addition to related changes in gut microbiome composition and functions.High-f...This study investigated the effects of a xylitol-casein non-covalent complex(XC)on parameters related to type 2 diabetes mellitus(T2DM),in addition to related changes in gut microbiome composition and functions.High-fat-diet(HFD)+streptozotocin(STZ)-induced T2DM mice were treated with xylitol(XY),casein(CN),and XC,after which fecal samples were collected for gut microbiota composition and diversity analyses based on 16S rRNA high-throughput sequencing and multivariate statistics.XC decreased body weight and improved glucose tolerance,insulin sensitivity,pancreas impairment,blood lipid levels,and liver function in T2DM mice compared to XY-and CN-treated mice.Furthermore,XC modulated theα-diversity,β-diversity and gut microbiota composition.Based on Spearman’s correlation analysis,the relative abundances of Alistipes,Bacteroides,and Faecalibaculum were positively correlated and those of Akkermansia,Lactobacillus,Bifidobacterium,and Turicibacter were negatively correlated with the phenotypes related to the improvement of T2DM.In conclusion,we found that XC alleviated insulin resistance by restoring the gut microbiota of T2DM mice.Our results provide strong evidence for the beneficial effects of XC on T2DM and motivation for further investigation in animal models and,eventually,human trials.展开更多

Outer leaves are photosynthetic organs that usually senesce at the late heading stage in Chinese cabbage, and premature leaf senescence often reduces leafy head yield and quality. In this study, 11 premature leaf senescence mutants were screened from an ethyl methanesulfonate-mutagenized population of the double haploid line ‘FT' in Chinese cabbage. At the early heading stage, the mutants exhibited edge yellowing within its outer leaves, and at the mature stage, its leafy head weight decreased significantly. Genetic analysis revealed that the mutated trait of all 11 mutants corresponds to single gene recessive inheritance. Semi-diallel cross tests showed that 5 of the 11 were allelic mutants. MutMap and Kompetitive Allele Specific PCR genotyping revealed that BraA01g001400.3C was the candidate gene, which is orthologous of Arabidopsis SUPPRESSOR OF rps4-RLD 1, encoding an immune regulator, so we named it as BrSRFR1. All the BrSRFR1 in the five allelic mutants exhibited single nucleotide polymorphisms at different positions on their exons and led to premature translation termination, which confirmed that defect in BrSRFR1 led to premature leaf senescence. These results verify the role of Br SRFR1 on leaf senescence and provide a new insight into the mechanisms of leaf senescence in Chinese cabbage, which reveals a novel function of SRFR1 in plant development.展开更多
The accumulation processes of loess in northeastern(NE)China record the varying characteristics of the East Asian Monsoons(EAM)and the evolution of the local environment.In this study,grain size end-member analysis of...The accumulation processes of loess in northeastern(NE)China record the varying characteristics of the East Asian Monsoons(EAM)and the evolution of the local environment.In this study,grain size end-member analysis of the Dajiugang(DJG)section deposited since the Last Glacial in Chifeng City,NE China was used to reveal the dynamic depositional characteristics of loess and environmental evolution of NE China.Results showed that the Chifeng loess comprises three grain size end-members(EM),EM1(7.38μm),EM2(49.4μm)and EM3(90.00μm),indicating the three transport dynamics of dust delivered to the region.The EM1 represents atmospheric dust as the background.The EM2,mainly derived from relatively distant-sources deposition,and EM3,material transported over a short distance,correspond to the intensity of southwesterly winds and the East Asian Winter Monsoon(EAWM),respectively.Results of climate reconstruction,combined with other records,showed that the climate was generally cold and dry with a strong EAWM and a weak East Asian summer monsoon(EASM)during the stadial-period[Marine Isotope Stage(MIS)4 and MIS 2 stage].The climate was predominantly warm and humid,with weak EAWM and surface winds and strong EASM in interstadial-period(MIS 3 and MIS 1 stage).Comparisons between indicators,e.g.,EM3,the LR04 benthicδ18O stack and summer insolation at 65°N suggest that the strong EAWM is induced by increasing the Northern Hemisphere ice volume and reduced summer solar radiation.展开更多
Straw mulching is a widespread practice for reducing the soil carbon loss caused by erosion.However,the effects of straw mulching on dissolved organic matter(DOM)runoff loss from black soil are not well studied.How st...Straw mulching is a widespread practice for reducing the soil carbon loss caused by erosion.However,the effects of straw mulching on dissolved organic matter(DOM)runoff loss from black soil are not well studied.How straw mulching affects the composition and loss of runoff DOM by changing soil aggregates remains largely unclear.Here,a straw mulching treatment was compared to a no mulching treatment(as a control)on sloping farmland with black soil erosion in Northeast China.We divided the soil into large macroaggregates(>2 mm),small macroaggregates(0.25-2 mm),and microaggregates(<0.25 mm).After five rain events,the effects of straw mulching on the concentration(characterized by dissolved organic carbon(DoC)and composition(analyzed by fluorescence spectroscopy)of runoff and soil aggregate DOM were studied.The results showed that straw mulching reduced the runoff amount by 54.7%.Therefore,although straw mulching increased the average DOc concentration in runoff,it reduced the total runoff DOM loss by 48.3%.The composition of runoff DOM is similar to that of soil,as both contain humic-like acid and protein-like components.With straw mulching treatment,the protein-like components in small macroaggregates accumulated and the protein-like components in runoff declined with rain events.Fluorescence spectroscopy technology may help in understanding the hydrological paths of rain events by capturing the dynamic changes of runoff and soil DOM characteristics.A variation partitioning analysis(VPA)indicated that the DOM concentration and composition of microaggregates explained 68.2%of the change in runoff DOM from no mulching plots,while the change in runoff DOM from straw mulching plots was dominated by small macroaggregates at a rate of 55.1%.Taken together,our results demonstrated that straw mulching reduces the fragmentation of small macroaggregates and the loss of microaggregates,thus effecting DOM compositions in soil and reducing the DOM loss in runoff.These results provide a theoretical basis for reducing carbon loss in sloping farmland.展开更多
Commercial sterility does not guarantee the sustained stability of ultrahigh temperature(UHT)milk over 6 months shelf life.We explore the microbiota presented in normal(SZ)and quality deteriorated UHT milk(QY and WY)p...Commercial sterility does not guarantee the sustained stability of ultrahigh temperature(UHT)milk over 6 months shelf life.We explore the microbiota presented in normal(SZ)and quality deteriorated UHT milk(QY and WY)products from the same brand.Based on high-throughput sequencing research results,11 phyla and 54 genera were identified as dominant microbiota.Pseudomonas,Streptococcus,and Acinetobacter as core functional microbiota significantly influenced the UHT milk quality properties.Moreover,principal component analysis(PCA)and multivariate analyses were used to examine the quality characteristics,including 11 physicochemical parameters,10 fatty acids,and 2 enzyme activities,in normal and quality deteriorated UHT milk.We found that the abundance of Pseudomonas increased in quality deteriorated milk(WY)and showed a significant positive correlation with heat-resistant protease content.Acinetobacter in quality deteriorated milk(QY)also considerably contributed to the content of heat-resistant lipase,which resulted in spoilage deterioration of UHT milk.展开更多

H10 subtype avian influenza viruses(AIV)have been circulating in China for 40 years.H10 AIVs in China have expanded their host range from wild birds to domestic poultry and mammals,even human.Most of the H10 subtype A...H10 subtype avian influenza viruses(AIV)have been circulating in China for 40 years.H10 AIVs in China have expanded their host range from wild birds to domestic poultry and mammals,even human.Most of the H10 subtype AIVs reported in China were isolate from the southeast part.We isolated an H10N3 AIV,A/Chicken/Liaoning/SY1080/2021(SY1080),from live poultry market(LPM)in Liaoning Province of the Northeast China.SY1080replicated efficiently in mice lungs and nasal turbinates without prior adaptation.We systematically compared SY1080 with other H10 subtype isolates in China.Phylogenetic analysis showed that SY1080 and most of the H10strains belonged to the Eurasian lineage.H10 AIVs in China have formed 63 genotypes.SY1080 as well as the H10N3 strains from human infections belonged to G60 genotype.H10Nx AIV acquired multiple mammalian adaptive and virulence related mutations during circulation and the recent reassortants derived internal genes from chicken H9N2 AIVs.The H10Nx subtypes AIVs posed potential threat to public health.These results suggested we should strengthen the surveillance and evaluation of H10 subtype strains.展开更多
Blast disease,caused by the hemibiotrophic ascomycete fungus,Magnaporthe oryzae,is a significant threat to sustainable rice production worldwide.Studies have shown that the blast fungus secretes vast arrays of functio...Blast disease,caused by the hemibiotrophic ascomycete fungus,Magnaporthe oryzae,is a significant threat to sustainable rice production worldwide.Studies have shown that the blast fungus secretes vast arrays of functionally diverse proteins into the host cell for a successful disease progression.However,the final destinations of these effector proteins inside the host cell and their role in advancing fungal pathogenesis remain a mystery.Here,we reported that a putative mitochondrial targeting non-classically secreted protein(MoMtp)positively regulates conidiogenesis and appressorium maturation in M.oryzae.Moreover,MoM TP gene deletion mutant strains triggered a hypersensitive response when inoculated on rice leaves displaying that MoMtp is essential for the virulence of M.oryzae.In addition,cell wall and oxidative stress results indicated that MoMtp is likely involved in the maintenance of the structural integrity of the fungus cell.Our study also demonstrates an upregulation in the expression pattern of the MoMTP gene at all stages of infection,indicating its possible regulatory role in host invasion and the infectious development of M.oryzae.Furthermore,Agrobacterium infiltration and sheath inoculation confirmed that MoMtpGFP protein is predominantly localized in the host mitochondria of tobacco leaf and rice cells.Taken together,we conclude that MoMtp protein likely promotes the normal conidiation and pathogenesis of M.oryzae and might have a role in disturbing the proper functioning of the host mitochondria during pathogen invasion.展开更多

Soybean mosaic virus(SMV),an RNA virus,is the most common and destructive pathogenic virus in soybean fields.The newly developed CRISPR/Cas immune system has provided a novel strategy for improving plant resistance to...Soybean mosaic virus(SMV),an RNA virus,is the most common and destructive pathogenic virus in soybean fields.The newly developed CRISPR/Cas immune system has provided a novel strategy for improving plant resistance to viruses;hence,this study aimed to engineer SMV resistance in soybean using this system.Specifically,multiple sgRNAs were designed to target positive-and/or negative-sense strands of the SMV HC-Pro gene.Subsequently,the corresponding CRISPR/CasRx vectors were constructed and transformed into soybeans.After inoculation with SMV,39.02%,35.77%,and 18.70%of T_(1)plants were confirmed to be highly resistant(HR),resistant(R),and mildly resistant(MR)to SMV,respectively,whereas only 6.50%were identified as susceptible(S).Additionally,qRT-PCR and DAS-ELISA showed that,both at 15 and 30 d post-inoculation(dpi),SMV accumulation significantly decreased or was even undetectable in HR and R plants,followed by MR and S plants.Additionally,the expression level of the CasRx gene varied in almost all T_(1)plants with different resistance level,both at 15 and 30 dpi.Furthermore,when SMV resistance was evaluated in the T_(2)generation,the results were similar to those recorded for the T_(1)generation.These findings provide new insights into the application of the CRISPR/CasRx system for soybean improvement and offer a promising alternative strategy for breeding for resistance to biotic stress that will contribute to the development of SMV-immune soybean germplasm to accelerate progress towards greater soybean crop productivity.展开更多
Pinus sylvestris var.mongolica(P.sylvestris)plantations are extensively established in the boreal zone.Increasing stand biomass of these plantations can effectively enhance carbon stock,which is crucial for mitigating...Pinus sylvestris var.mongolica(P.sylvestris)plantations are extensively established in the boreal zone.Increasing stand biomass of these plantations can effectively enhance carbon stock,which is crucial for mitigating climate change.However,the current understanding of optimizing plantation strategies to maximize stand biomass is primarily derived from experiments in tropical and subtropical zones,which is difficult to extend to the boreal due to substantial climatic differences.Based on a comprehensive dataset from 1,076 sample plots of P.sylvestris plantations in the boreal zone of China,we evaluated the effects of tree species richness and stand density on tree height,diameter at breast height(DBH),and stand biomass to investigate the optimal plantation strategy.Furthermore,we examined how these effects changed with stand age and investigated their relative importance.We found that monocultures at a high stand density of 2,000–2,500ha^(−1) were the optimal plantation strategy to maximize stand biomass(107.5Mg·ha^(−1)),and this held true at almost all stand ages.Unfortunately,this strategy resulted in low species richness and small individual trees(10.6m height and 9.8cm DBH),thus presenting a trade-off.In addition,as stand age increased,the effect of tree species richness on stand biomass shifted from positive to negative,but the effect of stand density was always positive.Overall,stand age had the greatest effect on stand biomass,followed by stand density and then tree species richness.Our findings reveal a distinct plantation strategy for optimizing stand biomass of P.sylvestris plantations in the boreal zone.More importantly,this study highlights that(1)maximizing stand biomass in the boreal zone may compromise tree species richness;(2)net effects of tree species richness on stand biomass are not always positive,as negative selection effects offset positive complementary effects.展开更多

In this paper,we utilized the deep convolutional neural network D-LinkNet,a model for semantic segmentation,to analyze the Himawari-8 satellite data captured from 16 channels at a spatial resolution of 0.5 km,with a f...In this paper,we utilized the deep convolutional neural network D-LinkNet,a model for semantic segmentation,to analyze the Himawari-8 satellite data captured from 16 channels at a spatial resolution of 0.5 km,with a focus on the area over the Yellow Sea and the Bohai Sea(32°-42°N,117°-127°E).The objective was to develop an algorithm for fusing and segmenting multi-channel images from geostationary meteorological satellites,specifically for monitoring sea fog in this region.Firstly,the extreme gradient boosting algorithm was adopted to evaluate the data from the 16 channels of the Himawari-8 satellite for sea fog detection,and we found that the top three channels in order of importance were channels 3,4,and 14,which were fused into false color daytime images,while channels 7,13,and 15 were fused into false color nighttime images.Secondly,the simple linear iterative super-pixel clustering algorithm was used for the pixel-level segmentation of false color images,and based on super-pixel blocks,manual sea-fog annotation was performed to obtain fine-grained annotation labels.The deep convolutional neural network D-LinkNet was built on the ResNet backbone and the dilated convolutional layers with direct connections were added in the central part to form a string-and-combine structure with five branches having different depths and receptive fields.Results show that the accuracy rate of fog area(proportion of detected real fog to detected fog)was 66.5%,the recognition rate of fog zone(proportion of detected real fog to real fog or cloud cover)was 51.9%,and the detection accuracy rate(proportion of samples detected correctly to total samples)was 93.2%.展开更多
Phosphorus-solubilizing microbes play key roles in improving phosphorus availability and in alleviating phosphorus nutrient limitation in soils. However, we did not have a comprehensive understanding of the overall re...Phosphorus-solubilizing microbes play key roles in improving phosphorus availability and in alleviating phosphorus nutrient limitation in soils. However, we did not have a comprehensive understanding of the overall research progress and development trend of phosphorus solubilizing microorganisms. In this study, we obtain documents from the Web of Science (WOS) core collection between 2002 and 2022, and a comprehensive review of the progress of global research on soil phosphate solubilizing microorganisms was conducted by using the VOSviewer bibliometric analysis tool. The results showed an increasing trend in the number of published articles from 2002 to 2022. India, accounting for 28% of the total number of published articles, became the most productive country. However, Canada was the country with the highest average citation frequency of articles. Chinese Academy of Sciences (CAS) was the greatest contributor with the most publications. Among the published journals, Frontiers in Microbiology, Applied Soil Ecology and Plant and Soil were the top three core journals in this field. Based on the keyword analysis, the assessment of the mechanisms between phosphorus solubilizing microbes and the soil carbon cycles with the different management practices became the new research trend among the scientific communities. These findings would provide an important reference value for future in-depth research on soil phosphate solubilizing microorganisms.展开更多

文摘Agriculture is the biggest water user in China,and the development of agricultural water-saving has great significance to the national economy and social development. In this paper,the present situation of water used in agriculture irrigation and water-saving potential were analyzed,and the " bottleneck" and main problems existing in water-saving irrigation in China were discussed. From the aspects of engineering investment channels,agricultural water-saving policies and management system,reform of agricultural water price and water right transfer,improvement of farmers' water-saving consciousness,and promotion of rural land transfer,suggestions were proposed for the development of China's agricultural water-saving in future,which will provide a technical support for the sustainable use of agricultural water resources in China.

文摘Constipation is a common intestinal disease.Kiwi berries can effectively prevent constipation.However,studies have yet to be done to determine how kiwi berries prevent constipation.For two weeks,mice in this study were continually orally gavaged with kiwi berry,loperamide,or a combination of the 2.This study found that the kiwi group's feces had more water than the constipated mice.In addition,kiwi berries can speed up gastrointestinal transit(GI),shorten the time it takes to pass the first dark stool,and dramatically enhance body weight gain.In the interstitial cells of Caj al(ICC)cells and colon tissues,alterations in the protein expression of vasoactive intestinal peptide(VIP),cyclic adenosine monophosphate(cAMP),protein kinase A(PKA),and aquapcrin-3(AQP3)were found.At 3,6,and 12 h of ICC cells and mouse colon,the kiwi group's VIP,cAMP,PKA,and AQP3 protein expression levels were lower than those of the constipated mice.The kiwi berry can decrease the Firmicutes to Bacteroidetes ratio and boost the diversity and quantity of gut microbiota.By influencing the gut microbiota and VIP-cAMP-PKA-AQP3 signaling pathway,kiwi berries prevent constipation.

文摘Suppression of roots and/or their symbiotic microorganisms,such as mycorrhizal fungi and rhizobia,is an effective way for alien plants to outcompete native plants.However,little is known about how invasive and native plants interact with the quantity and activity of nutrient-acquisition agents.Here a pot experiment was conducted with monoculture and mixed plantings of an invasive plant,Xanthium strumarium,and a common native legume,Glycine max.We measured traits related to root and nodule quantity and activity and mycorrhizal colonization.Compared to the monoculture,fine root quantity(biomass,surface area)and activity(root nitrogen(N)concentration,acid phosphatase activity)of G.max decreased in mixed plantings;nodule quantity(biomass)decreased by 45%,while nodule activity in Nfixing via rhizobium increased by 106%;mycorrhizal colonization was unaffected.Contribution of N fixation to leaf N content in G.max increased in the mixed plantings,and this increase was attributed to a decrease in the rhizosphere soil N of G.max in the mixed plantings.Increased root quantity and activity,along with a higher mycorrhizal association was observed in X.strumarium in the mixed compared to monoculture.Together,the invasive plant did not directly scavenge N from nodule-fixed N,but rather depleted the rhizosphere soil N of the legume,thereby stimulating the activity of N-fixation and increasing the dependence of the native legume on this N source.The quantity-activity framework holds promise for future studies on how native legumes respond to alien plant invasions.

文摘The addition of ectomycorrhizal fungi(ECMF),beneficial rhizosphere microorganisms,to the soil can pro-mote plant growth and resistance.Here,Populus davidi-ana×Populus bolleana tissue culture seedlings were grown for 3 months in soils inoculated with one of the species,then seedlings were assessed for mycorrhizal colonization rate and growth,physiological and root traits.Suillus luteus and Populus involutus each formed ectomycorrhizal associa-tions with the seedlings.Seedling height,ground diameter,biomass,and leaf area were significantly greater after treat-ment with ECMF than in the non-inoculated controls.Treat-ment improved all physiological and root variables assessed(chlorophylls and carotenoids,cellulose,and soluble sugars and proteins;root length,surface area,projected area,mean diameter,volume,number of root tips).Seedlings inocu-lated with S.luteus outperformed those inoculated with P.involutus.

文摘The strawberry species Fragaria nilgerrensis Schlechtendal ex J.Gay,renowned for its distinctive white,fragrant peach-like fruits and strong disease resistance,is an exceptional research material.In a previous study,an ethyl methane sulfonate(EMS)mutant library was established for this species,resulting in various yellow leaf mutants.Leaf yellowing materials are not only the ideal materials for basic studies on photosynthesis mechanism,chloroplast development,and molecular regulation of various pigments,but also have important utilization value in ornamental plants breeding.The present study focused on four distinct yellow leaf mutants:mottled yellow leaf(MO),yellow green leaf(YG),light green leaf(LG),and buddha light leaf(BU).The results revealed that the flavonoid content and carotenoid-to-chlorophyll ratio exhibited a significant increase among these mutants,while experiencing a significant decrease in chlorophyll and carotenoid contents compared to the wild type(WT).To clarify the regulatory mechanisms and network relationships underlying these mutants,the RNA-seq and weighted gene coexpression network(WGCNA)analyses were employed.The results showed flavonoid metabolism pathway was enriched both in MO and YG mutants,while the chlorophyll biosynthesis pathway and carotenoid degradation pathway were only enriched in MO and YG mutants,respectively.Subsequently,key structural genes and transcription factors were identified on metabolic pathways of three pigments through correlation analyses and quantitative experiments.Furthermore,a R2R3-MYB transcription factor,FnMYB4,was confirmed to be positively correlated with flavonoid synthesis through transient overexpression,virus-induced gene silencing(VIGS),and RNA interference(RNAi),accompanying by reoccurrence and attenuation of mutant phenotype.Finally,dual-luciferase(LUC)and yeast one-hybrid assays confirmed the binding of FnMYB4 to the FnFLS and FnF3H promoters,indicating that FnMYB4 positively regulates flavonoid synthesis.In addition,correlation analyses suggested that FnMYB4 also might be involved in chlorophyll and carotenoid metabolisms.These findings demonstrated the pivotal regulatory role of FnMYB4 in strawberry leaf coloration.

文摘This study investigated the effects of a xylitol-casein non-covalent complex(XC)on parameters related to type 2 diabetes mellitus(T2DM),in addition to related changes in gut microbiome composition and functions.High-fat-diet(HFD)+streptozotocin(STZ)-induced T2DM mice were treated with xylitol(XY),casein(CN),and XC,after which fecal samples were collected for gut microbiota composition and diversity analyses based on 16S rRNA high-throughput sequencing and multivariate statistics.XC decreased body weight and improved glucose tolerance,insulin sensitivity,pancreas impairment,blood lipid levels,and liver function in T2DM mice compared to XY-and CN-treated mice.Furthermore,XC modulated theα-diversity,β-diversity and gut microbiota composition.Based on Spearman’s correlation analysis,the relative abundances of Alistipes,Bacteroides,and Faecalibaculum were positively correlated and those of Akkermansia,Lactobacillus,Bifidobacterium,and Turicibacter were negatively correlated with the phenotypes related to the improvement of T2DM.In conclusion,we found that XC alleviated insulin resistance by restoring the gut microbiota of T2DM mice.Our results provide strong evidence for the beneficial effects of XC on T2DM and motivation for further investigation in animal models and,eventually,human trials.

文摘Leaf senescence is the final stage of leaf development, where the nutrients and energy of senescent leaves are redistributed to developing tissues or organs for plant growth, reproduction, and defense. Outer leaves are photosynthetic organs that usually senesce at the late heading stage in Chinese cabbage, and premature leaf senescence often reduces leafy head yield and quality. In this study, 11 premature leaf senescence mutants were screened from an ethyl methanesulfonate-mutagenized population of the double haploid line ‘FT' in Chinese cabbage. At the early heading stage, the mutants exhibited edge yellowing within its outer leaves, and at the mature stage, its leafy head weight decreased significantly. Genetic analysis revealed that the mutated trait of all 11 mutants corresponds to single gene recessive inheritance. Semi-diallel cross tests showed that 5 of the 11 were allelic mutants. MutMap and Kompetitive Allele Specific PCR genotyping revealed that BraA01g001400.3C was the candidate gene, which is orthologous of Arabidopsis SUPPRESSOR OF rps4-RLD 1, encoding an immune regulator, so we named it as BrSRFR1. All the BrSRFR1 in the five allelic mutants exhibited single nucleotide polymorphisms at different positions on their exons and led to premature translation termination, which confirmed that defect in BrSRFR1 led to premature leaf senescence. These results verify the role of Br SRFR1 on leaf senescence and provide a new insight into the mechanisms of leaf senescence in Chinese cabbage, which reveals a novel function of SRFR1 in plant development.

文摘The accumulation processes of loess in northeastern(NE)China record the varying characteristics of the East Asian Monsoons(EAM)and the evolution of the local environment.In this study,grain size end-member analysis of the Dajiugang(DJG)section deposited since the Last Glacial in Chifeng City,NE China was used to reveal the dynamic depositional characteristics of loess and environmental evolution of NE China.Results showed that the Chifeng loess comprises three grain size end-members(EM),EM1(7.38μm),EM2(49.4μm)and EM3(90.00μm),indicating the three transport dynamics of dust delivered to the region.The EM1 represents atmospheric dust as the background.The EM2,mainly derived from relatively distant-sources deposition,and EM3,material transported over a short distance,correspond to the intensity of southwesterly winds and the East Asian Winter Monsoon(EAWM),respectively.Results of climate reconstruction,combined with other records,showed that the climate was generally cold and dry with a strong EAWM and a weak East Asian summer monsoon(EASM)during the stadial-period[Marine Isotope Stage(MIS)4 and MIS 2 stage].The climate was predominantly warm and humid,with weak EAWM and surface winds and strong EASM in interstadial-period(MIS 3 and MIS 1 stage).Comparisons between indicators,e.g.,EM3,the LR04 benthicδ18O stack and summer insolation at 65°N suggest that the strong EAWM is induced by increasing the Northern Hemisphere ice volume and reduced summer solar radiation.

文摘Straw mulching is a widespread practice for reducing the soil carbon loss caused by erosion.However,the effects of straw mulching on dissolved organic matter(DOM)runoff loss from black soil are not well studied.How straw mulching affects the composition and loss of runoff DOM by changing soil aggregates remains largely unclear.Here,a straw mulching treatment was compared to a no mulching treatment(as a control)on sloping farmland with black soil erosion in Northeast China.We divided the soil into large macroaggregates(>2 mm),small macroaggregates(0.25-2 mm),and microaggregates(<0.25 mm).After five rain events,the effects of straw mulching on the concentration(characterized by dissolved organic carbon(DoC)and composition(analyzed by fluorescence spectroscopy)of runoff and soil aggregate DOM were studied.The results showed that straw mulching reduced the runoff amount by 54.7%.Therefore,although straw mulching increased the average DOc concentration in runoff,it reduced the total runoff DOM loss by 48.3%.The composition of runoff DOM is similar to that of soil,as both contain humic-like acid and protein-like components.With straw mulching treatment,the protein-like components in small macroaggregates accumulated and the protein-like components in runoff declined with rain events.Fluorescence spectroscopy technology may help in understanding the hydrological paths of rain events by capturing the dynamic changes of runoff and soil DOM characteristics.A variation partitioning analysis(VPA)indicated that the DOM concentration and composition of microaggregates explained 68.2%of the change in runoff DOM from no mulching plots,while the change in runoff DOM from straw mulching plots was dominated by small macroaggregates at a rate of 55.1%.Taken together,our results demonstrated that straw mulching reduces the fragmentation of small macroaggregates and the loss of microaggregates,thus effecting DOM compositions in soil and reducing the DOM loss in runoff.These results provide a theoretical basis for reducing carbon loss in sloping farmland.

文摘Commercial sterility does not guarantee the sustained stability of ultrahigh temperature(UHT)milk over 6 months shelf life.We explore the microbiota presented in normal(SZ)and quality deteriorated UHT milk(QY and WY)products from the same brand.Based on high-throughput sequencing research results,11 phyla and 54 genera were identified as dominant microbiota.Pseudomonas,Streptococcus,and Acinetobacter as core functional microbiota significantly influenced the UHT milk quality properties.Moreover,principal component analysis(PCA)and multivariate analyses were used to examine the quality characteristics,including 11 physicochemical parameters,10 fatty acids,and 2 enzyme activities,in normal and quality deteriorated UHT milk.We found that the abundance of Pseudomonas increased in quality deteriorated milk(WY)and showed a significant positive correlation with heat-resistant protease content.Acinetobacter in quality deteriorated milk(QY)also considerably contributed to the content of heat-resistant lipase,which resulted in spoilage deterioration of UHT milk.

文摘The black chokeberry is rich in polyphenols,including flavonoids with anthocyanins,flavanols and lavonols as the main components and a variety of phenolic acids represented by chlorogenic acid.Because of these polyphenols,black chokeberry has the effect of preventing and adjuvant therapy diseases.This study summarized the current research results on the types and contents of functional components in black chokeberry,and analyzed their digestion,absorption and metabolism in human body.On this basis,the disease control functions that have been proved effective in clinical research were reviewed and analyzed.These studies showed that black chokeberry have good prevention and adjuvant therapy effects on hyperlipidemia,hypertension,diabetes and inflammation.Because there are different functional components in black chokeberry,its prevention and treatment of the same disease can come from multiple pathways,which provides a more reliable effectiveness for the disease control of different populations.

文摘H10 subtype avian influenza viruses(AIV)have been circulating in China for 40 years.H10 AIVs in China have expanded their host range from wild birds to domestic poultry and mammals,even human.Most of the H10 subtype AIVs reported in China were isolate from the southeast part.We isolated an H10N3 AIV,A/Chicken/Liaoning/SY1080/2021(SY1080),from live poultry market(LPM)in Liaoning Province of the Northeast China.SY1080replicated efficiently in mice lungs and nasal turbinates without prior adaptation.We systematically compared SY1080 with other H10 subtype isolates in China.Phylogenetic analysis showed that SY1080 and most of the H10strains belonged to the Eurasian lineage.H10 AIVs in China have formed 63 genotypes.SY1080 as well as the H10N3 strains from human infections belonged to G60 genotype.H10Nx AIV acquired multiple mammalian adaptive and virulence related mutations during circulation and the recent reassortants derived internal genes from chicken H9N2 AIVs.The H10Nx subtypes AIVs posed potential threat to public health.These results suggested we should strengthen the surveillance and evaluation of H10 subtype strains.

文摘Blast disease,caused by the hemibiotrophic ascomycete fungus,Magnaporthe oryzae,is a significant threat to sustainable rice production worldwide.Studies have shown that the blast fungus secretes vast arrays of functionally diverse proteins into the host cell for a successful disease progression.However,the final destinations of these effector proteins inside the host cell and their role in advancing fungal pathogenesis remain a mystery.Here,we reported that a putative mitochondrial targeting non-classically secreted protein(MoMtp)positively regulates conidiogenesis and appressorium maturation in M.oryzae.Moreover,MoM TP gene deletion mutant strains triggered a hypersensitive response when inoculated on rice leaves displaying that MoMtp is essential for the virulence of M.oryzae.In addition,cell wall and oxidative stress results indicated that MoMtp is likely involved in the maintenance of the structural integrity of the fungus cell.Our study also demonstrates an upregulation in the expression pattern of the MoMTP gene at all stages of infection,indicating its possible regulatory role in host invasion and the infectious development of M.oryzae.Furthermore,Agrobacterium infiltration and sheath inoculation confirmed that MoMtpGFP protein is predominantly localized in the host mitochondria of tobacco leaf and rice cells.Taken together,we conclude that MoMtp protein likely promotes the normal conidiation and pathogenesis of M.oryzae and might have a role in disturbing the proper functioning of the host mitochondria during pathogen invasion.

文摘Soybean mosaic virus(SMV),an RNA virus,is the most common and destructive pathogenic virus in soybean fields.The newly developed CRISPR/Cas immune system has provided a novel strategy for improving plant resistance to viruses;hence,this study aimed to engineer SMV resistance in soybean using this system.Specifically,multiple sgRNAs were designed to target positive-and/or negative-sense strands of the SMV HC-Pro gene.Subsequently,the corresponding CRISPR/CasRx vectors were constructed and transformed into soybeans.After inoculation with SMV,39.02%,35.77%,and 18.70%of T_(1)plants were confirmed to be highly resistant(HR),resistant(R),and mildly resistant(MR)to SMV,respectively,whereas only 6.50%were identified as susceptible(S).Additionally,qRT-PCR and DAS-ELISA showed that,both at 15 and 30 d post-inoculation(dpi),SMV accumulation significantly decreased or was even undetectable in HR and R plants,followed by MR and S plants.Additionally,the expression level of the CasRx gene varied in almost all T_(1)plants with different resistance level,both at 15 and 30 dpi.Furthermore,when SMV resistance was evaluated in the T_(2)generation,the results were similar to those recorded for the T_(1)generation.These findings provide new insights into the application of the CRISPR/CasRx system for soybean improvement and offer a promising alternative strategy for breeding for resistance to biotic stress that will contribute to the development of SMV-immune soybean germplasm to accelerate progress towards greater soybean crop productivity.

文摘Pinus sylvestris var.mongolica(P.sylvestris)plantations are extensively established in the boreal zone.Increasing stand biomass of these plantations can effectively enhance carbon stock,which is crucial for mitigating climate change.However,the current understanding of optimizing plantation strategies to maximize stand biomass is primarily derived from experiments in tropical and subtropical zones,which is difficult to extend to the boreal due to substantial climatic differences.Based on a comprehensive dataset from 1,076 sample plots of P.sylvestris plantations in the boreal zone of China,we evaluated the effects of tree species richness and stand density on tree height,diameter at breast height(DBH),and stand biomass to investigate the optimal plantation strategy.Furthermore,we examined how these effects changed with stand age and investigated their relative importance.We found that monocultures at a high stand density of 2,000–2,500ha^(−1) were the optimal plantation strategy to maximize stand biomass(107.5Mg·ha^(−1)),and this held true at almost all stand ages.Unfortunately,this strategy resulted in low species richness and small individual trees(10.6m height and 9.8cm DBH),thus presenting a trade-off.In addition,as stand age increased,the effect of tree species richness on stand biomass shifted from positive to negative,but the effect of stand density was always positive.Overall,stand age had the greatest effect on stand biomass,followed by stand density and then tree species richness.Our findings reveal a distinct plantation strategy for optimizing stand biomass of P.sylvestris plantations in the boreal zone.More importantly,this study highlights that(1)maximizing stand biomass in the boreal zone may compromise tree species richness;(2)net effects of tree species richness on stand biomass are not always positive,as negative selection effects offset positive complementary effects.

文摘Blueberry,kiwifruit,Rosa roxburghii,and raspberry are the characteristic fruits planted in Guizhou Province.However,in recent years,harmful factors such as plant diseases and insect pests,pesticides and heavy metal residues have affected the quality and safety of blueberry,kiwifruit,R.roxburghii,raspberry and other berries.These problems mainly include the frequent occurrence of plant diseases and insect pests,pesticide residues and heavy metal pollution,which not only seriously affect the quality and safety of berries,but also restrict the healthy development of berry industry.Therefore,it is very important to study the detection and monitoring of key hazard factors affecting the quality and safety of blueberry,kiwifruit,R.roxburghii and raspberry,as well as the standardized production technology.Using literature analysis,field investigation,questionnaire survey,comprehensive analysis,SWOT analysis,laboratory testing and other methods,this paper made a comprehensive and in-depth study of the berry industry in Guizhou Province.Through the analysis of the current situation of the berry industry in Guizhou Province,the problems and shortcomings in the planting,management,sales and other aspects of the industry were revealed.In order to solve these problems,a series of practical measures were put forward,including strengthening pest control,optimizing pesticide application technology,and strictly controlling heavy metal pollution,so as to ensure the healthy and stable development of berry industry.The implementation of these measures will help to improve the overall quality level of the berry industry in Guizhou Province.

文摘In this paper,we utilized the deep convolutional neural network D-LinkNet,a model for semantic segmentation,to analyze the Himawari-8 satellite data captured from 16 channels at a spatial resolution of 0.5 km,with a focus on the area over the Yellow Sea and the Bohai Sea(32°-42°N,117°-127°E).The objective was to develop an algorithm for fusing and segmenting multi-channel images from geostationary meteorological satellites,specifically for monitoring sea fog in this region.Firstly,the extreme gradient boosting algorithm was adopted to evaluate the data from the 16 channels of the Himawari-8 satellite for sea fog detection,and we found that the top three channels in order of importance were channels 3,4,and 14,which were fused into false color daytime images,while channels 7,13,and 15 were fused into false color nighttime images.Secondly,the simple linear iterative super-pixel clustering algorithm was used for the pixel-level segmentation of false color images,and based on super-pixel blocks,manual sea-fog annotation was performed to obtain fine-grained annotation labels.The deep convolutional neural network D-LinkNet was built on the ResNet backbone and the dilated convolutional layers with direct connections were added in the central part to form a string-and-combine structure with five branches having different depths and receptive fields.Results show that the accuracy rate of fog area(proportion of detected real fog to detected fog)was 66.5%,the recognition rate of fog zone(proportion of detected real fog to real fog or cloud cover)was 51.9%,and the detection accuracy rate(proportion of samples detected correctly to total samples)was 93.2%.

文摘Phosphorus-solubilizing microbes play key roles in improving phosphorus availability and in alleviating phosphorus nutrient limitation in soils. However, we did not have a comprehensive understanding of the overall research progress and development trend of phosphorus solubilizing microorganisms. In this study, we obtain documents from the Web of Science (WOS) core collection between 2002 and 2022, and a comprehensive review of the progress of global research on soil phosphate solubilizing microorganisms was conducted by using the VOSviewer bibliometric analysis tool. The results showed an increasing trend in the number of published articles from 2002 to 2022. India, accounting for 28% of the total number of published articles, became the most productive country. However, Canada was the country with the highest average citation frequency of articles. Chinese Academy of Sciences (CAS) was the greatest contributor with the most publications. Among the published journals, Frontiers in Microbiology, Applied Soil Ecology and Plant and Soil were the top three core journals in this field. Based on the keyword analysis, the assessment of the mechanisms between phosphorus solubilizing microbes and the soil carbon cycles with the different management practices became the new research trend among the scientific communities. These findings would provide an important reference value for future in-depth research on soil phosphate solubilizing microorganisms.
