Tumor necrosis factor-stimulated gene-6 ameliorates early brain injury after subarachnoid hemorrhage by suppressing NLRC4 inflammasome-mediated astrocyte pyroptosis 被引量:2

Subarachnoid hemorrhage is associated with high morbidity and mortality and lacks effective treatment.Pyroptosis is a crucial mechanism underlying early brain injury after subarachnoid hemorrhage.Previous studies have confirmed that tumor necrosis factor-stimulated gene-6(TSG-6)can exert a neuroprotective effect by suppressing oxidative stress and apoptosis.However,no study to date has explored whether TSG-6 can alleviate pyroptosis in early brain injury after subarachnoid hemorrhage.In this study,a C57BL/6J mouse model of subarachnoid hemorrhage was established using the endovascular perforation method.Our results indicated that TSG-6 expression was predominantly detected in astrocytes,along with NLRC4 and gasdermin-D(GSDMD).The expression of NLRC4,GSDMD and its N-terminal domain(GSDMD-N),and cleaved caspase-1 was significantly enhanced after subarachnoid hemorrhage and accompanied by brain edema and neurological impairment.To explore how TSG-6 affects pyroptosis during early brain injury after subarachnoid hemorrhage,recombinant human TSG-6 or a siRNA targeting TSG-6 was injected into the cerebral ventricles.Exogenous TSG-6 administration downregulated the expression of NLRC4 and pyroptosis-associated proteins and alleviated brain edema and neurological deficits.Moreover,TSG-6 knockdown further increased the expression of NLRC4,which was accompanied by more severe astrocyte pyroptosis.In summary,our study revealed that TSG-6 provides neuroprotection against early brain injury after subarachnoid hemorrhage by suppressing NLRC4 inflammasome activation-induced astrocyte pyroptosis. 展开更多

A series of mixed alkali-zinc borosilicate glasses with various r values(r=molar ratio of[ZnO]/([R^(2)O]+[ZnO]))from 0.00 to 1.00 were fabricated to probe the mixed alkali-zinc effects on thermo-mechanical properties.The nonlinear evolution of glass transition temperature(T_(g))with the addition of ZnO is ascribed to the competition of two converse factors,i e,the T_(g)depression as one of the colligative properties for a solution,on the one hand,and the enhancement of T_(g)due to the higher field strength of zinc cations compared to that of alkali ions.However,the nonlinear evolution of elastic moduli and coefficients of thermal expansion with r is attributed to the variance of intermediate-range clusters,which is confirmed by infrared and Raman scattering spectra.These findings are very helpful in tailoring the performance of borosilicate glasses. 展开更多

BACKGROUND Development of distant metastasis(DM)is a major concern during treatment of nasopharyngeal carcinoma(NPC).However,studies have demonstrated im-proved distant control and survival in patients with advanced NPC with the addition of chemotherapy to concomitant chemoradiotherapy.Therefore,precise prediction of metastasis in patients with NPC is crucial.AIM To develop a predictive model for metastasis in NPC using detailed magnetic resonance imaging(MRI)reports.METHODS This retrospective study included 792 patients with non-distant metastatic NPC.A total of 469 imaging variables were obtained from detailed MRI reports.Data were stratified and randomly split into training(50%)and testing sets.Gradient boosting tree(GBT)models were built and used to select variables for predicting DM.A full model comprising all variables and a reduced model with the top-five variables were built.Model performance was assessed by area under the curve(AUC).RESULTS Among the 792 patients,94 developed DM during follow-up.The number of metastatic cervical nodes(30.9%),tumor invasion in the posterior half of the nasal cavity(9.7%),two sides of the pharyngeal recess(6.2%),tubal torus(3.3%),and single side of the parapharyngeal space(2.7%)were the top-five contributors for predicting DM,based on their relative importance in GBT models.The testing AUC of the full model was 0.75(95%confidence interval[CI]:0.69-0.82).The testing AUC of the reduced model was 0.75(95%CI:0.68-0.82).For the whole dataset,the full(AUC=0.76,95%CI:0.72-0.82)and reduced models(AUC=0.76,95%CI:0.71-0.81)outperformed the tumor node-staging system(AUC=0.67,95%CI:0.61-0.73).CONCLUSION The GBT model outperformed the tumor node-staging system in predicting metastasis in NPC.The number of metastatic cervical nodes was identified as the principal contributing variable. 展开更多

BACKGROUND Circular RNAs(circRNAs)have attracted extensive attention as therapeutic targets in gastric cancer(GC).Circ_0003356 is known to be downregulated in GC tissues,but its cellular function and mechanisms remain undefined.AIM To investigate the role of circ_0003356 in GC at the molecular and cellular level.METHODS Circ_0003356,miR-668-3p,and SOCS3 expression were assessed via quantitative real time-polymerase chain reaction(qRT-PCR).Wound healing,EdU,CCK-8,flow cytometry and transwell assays were used to analyze the migration,proliferation,viability,apoptosis and invasion of GC cells.The subcellular localization of circ_0003356 was monitored using fluorescence in situ hybridization.The interaction of circ_0003356 with miR-668-3p was confirmed using RIP-qRT-PCR,RNA pull-down,and dual luciferase reporter assays.We observed protein levels of genes via western blot.We injected AGS cells into the upper back of mice and performed immunohistochemistry staining for examining E-cadherin,N-cadherin,Ki67,and SOCS3 expressions.TUNEL staining was performed for the assessment of apoptosis in mouse tumor tissues.RESULTS Circ_0003356 and SOCS3 expression was downregulated in GC cells,whilst miR-668-3p was upregulated.Exogenous circ_0003356 expression and miR-668-3p silencing suppressed the migration,viability,proliferation,epithelial to mesenchy-mal transition(EMT)and invasion of GC cells and enhanced apoptosis.Circ_0003356 overexpression impaired tumor growth in xenograft mice.Targeting of miR-668-3p by circ_0003356 was confirmed through binding assays and SOCS3 was identified as a downstream target of miR-668-3p.The impacts of circ_0003356 on cell proliferation,apoptosis,migration,invasion and EMT were reversed by miR-668-3p up-regulation or SOCS3 downregulation in GC cells.CONCLUSION Circ_0003356 impaired GC development through its interaction with the miR-668-3p/SOCS3 axis. 展开更多

We investigated the mixed alkali effect on the thermal properties and elastic response to temperature in the borosilicate glasses system with the composition of 70.65Si O_(2)·21.09B_(2)O_(3)·1.88Al_(2)O_(3)·(6.38-x)Li_(2)O·x Na_(2)O glasses,where x=0.00,1.595,3.19,4.785,and 6.38.Except for the expected positive and negative deviations from linearity for the coefficients of thermal expansion,room temperature E and G,we observed a new mixed alkali efiect on the response of elastic moduli to temperature.Fourier transform infrared spectra were obtained to elucidate the possible structural origin of the mixed alkali efiects.This work provides a valuable insight into the structural and mechanical properties of mixed-alkali borosilicate glasses. 展开更多

AIM:To investigate the therapeutic effects of four strains of probiotics(E.feacalis,L.acidophilus,C.butyricum and B.adolescentis) on dextran sulphate sodium(DSS)-induced experimental colitis in Balb/c mice.METHODS:Eighty Balb/c mice were randomly divided into 8 groups.Weight-loss,fecal character,fecal occult blood and hematochezia were recorded daily.Disease activity index(DAI) scores were also evaluated everyday.Length of colon was measured and histological scores were evaluated on the 13th day.Myeloperoxidase(MPO) activity was detected.Interleukin-1(IL-1) and IL-4 expression was detected by ELISA and RT-PCR.RESULTS:The four strains of probiotics relieved the inflammatory condition of DSS-induced experimental colitis in mice.Weight loss was slowed down in all probiotics-treated mice.Even weight gain was observed by the end of probiotics treatment.The DAI and histological scores of probiotics-treated mice were lower than those of mice in the control group(1.9 ± 0.2 vs 8.6 ± 0.4,P < 0.05 for E.faecalis).The length of colon of probiotics-treated mice was longer than that of mice in the control group(10.3 ± 0.34 vs 8.65 ± 0.77,P < 0.05 for E.faecalis).The four strains of probiotics decreased the MP activity and the IL-1 expression,but increased the IL-4 expression.E.faecalis had a better effect on DSS-induced experimental colitis in mice than the other three strains.CONCLUSION:The four strains of probiotics have beneficial effects on experimental colitis in mice.E.faecalis has a better effect on DSS-induced experimental colitis in mice than the other three strains.Supplement of probiotics provides a new therapy for UC. 展开更多

Background:As a surrogate marker of systemic inflammation,the lymphocyte-to-monocyte ratio(LMR) is an independent prognostic factor for various malignancies.This study investigated the prognostic significance of the pre-chemotherapy LMR in patients with previously untreated metastatic colorectal cancer(mCRC) receiving chemotherapy.Methods:The present study included newly diagnosed mCRC patients treated between January 2005 and December 2013 with FOLFOX chemotherapy,specifically oxaliplatin 180 mg/m^2 on day 1,with leucovorin 400 mg/m^2administered as a 2-hour infusion before the administration of 5-fluorouracil 400 mg/m^2 as an intravenous bolus injection,and 5-fluorouracil 2400 mg/m^2 as a 46-h infusion immediately after 5-fluorouracil bolus injection.The LMR was calculated as the absolute count of lymphocytes divided by the absolute count of monocytes.COX proportional hazards analysis was performed to evaluate the association of LMR with survival outcomes.Results:A total of 488 patients were included.Patients with high pre-chemotherapy LMR experienced significant improvements in progression-free survival(PFS,9.2 vs.7.6 months,P < 0.001) and overall survival(OS,19.4 vs.16.6 months,P < 0.001) compared with patients with low pre-chemotherapy LMR.Subsequent COX multivariate analysis showed that high pre-chemotherapy LMR(>3.11) was an independent favorable prognostic factor for PFS and OS.Additionally,patients whose LMR remained high(high-high subgroup),increased(low-high subgroup),or decreased(high-low subgroup) following chemotherapy showed better results in terms of PFS and OS than patients whose LMR remained low(low-low subgroup) after chemotherapy.Conclusions:For patients with previously untreated mCRC receiving FOLFOX chemotherapy,an elevated pre-chemotherapy LMR is an independent favorable prognostic factor for PFS and OS,and changes in the LMR before and after chemotherapy seem to predict the benefit of chemotherapy. 展开更多

In a prospective study, 42 048 adults residing in Zhongshan City, Guangdong, China, were followed for 16 years, and 171 of them developed nasopharyngeal carcinoma (NPC). Although Epstein-Barr virus (EBV) antibody levels of the cohort fluctuated, the antibody levels of 93% of the patients with NPC were raised and maintained at high levels for up to 10 years prior to diagnosis. This suggests that the serologic window affords an opportunity to monitor tumor progression during the preclinical stage of NPC development, facilitating early NPC detection. We reviewed the clinical records of the 171 patients with NPC in the prospective study to assess the efficacy of early NPC detection by serologic screening and clinical examination. Of the 171 patients, 51 had Stage I tumor (44 were among the 73 patients detected by clinical examination and 7 were among the 98 patients presented to outpatient department). Initial serologic screening predicted 58 (95.1%) of the 61 patients detected within 2 years. The risk of the screened population (58/3093) raised 13 times relative to cohort (61/42 048) during this period. Clinical examination detected all the 58 predicted cases, and 35 (60.3%) of which were diagnosed with Stage I tumor. The serologic prediction rate fell to 33.6% (37/110) 2 to 16 years after screening. The proportion of cases detected by clinical examination fell to 40.5% (15/37). The proportion of Stage I tumors among the cases detected by clinical examination during both periods remained at about 60%. We concluded that early detection of NPC can be accomplished by repeated serologic screening to maintain high prediction rates and by promptly examining screened subjects to detect tumors before the symptoms develop. 展开更多

Objective: To investigate patients' perception of service quality at hospitals in nine Chinese cities and propose some measures for improvement. Methods: The ServQ ual scale method was used in a survey involving patients at out-patient and in-patient facilities in Shanghai, Chongqing, Chengdu, Nanning, Guilin and Laibin of Guangxi, Honghezhou of Yunnan, Wulumuqi of Xinjiang and Zhongshan of Guangdong. The data collected were entered and analyzed using SPSS 20.0. Statistical analyses included descriptive statistics, factor analyses, reliability analyses, product-moment correlations, independent-sample t-tests, One-way ANOVA and regression analyses. Results: The Kaiser-Meyer-Olkin value for the factor analysis of the scale was 0.979. The Cronbach's α for the reliability analysis was 0.978. All the Pearson correlation coei cients were positive and statistically signii cant. Visitors to out-patient facilities reported more positive perception tacilities on tangibles(t = 4.168, P(t = 1.979, P <han visitors to in-patient f 0.05). Patients of 60 years of age and above reported mor< 0.001) and reliability e positive perception th<an those between 40 and 49 on reliability(F = 3.311, P = 0.010), assurances(F = 2.751, P 0.05) and empathy(F = 4.009, P = 0.003). For the i ve dimensions of the scale, patients in Laibin, Guangxi reported the most positive perceived service quality, followed by patients in Shanghai. On the other hand, patients in Chongqing and Nanning and Guilin of Guangxi reported relatively poor perceptions of service quality. Standardized regression coei cients showed statistically significant(P < 0.001) positive values for all Serv Qual dimensions. Empathy(β = 0.267) and reliability(β uality. = 0.239) most strongly predicted perception of service qConclusions: Chinese patients perceived service quality as satisfactory. Achieving high-quality perovskite films with uniform morphology and homogeneous crystallinity is challenging owing to the coffee ring effect(CRE) in the spray-coating technologies. In this study, an evaporation/spray-coating two-step deposition method is used to fabricate Cs_(0.19)FA_(0.81)PbI_(2.5)Br_(0.5)light harvesters for perovskite solar cells(PSCs). Considering the solid–liquid reaction, we establish a reaction-dependent regulating strategy that inhibits CRE successfully and prepare a high-quality perovskite layer, wherein the solvent for the FAI/Br solution during the spraying process is changed from isopropanol to n-butyl alcohol(NBA). The retarded-drying-enhanced spreading of the NBA solution inhibits contact line pinning to suppress the capillary flows and increases the reaction between metal halides(CsI/PbI_(2)) and organic salts(FAI/Br), which result in a reduction in the accumulation of solutes in the periphery effectively inhibiting CRE. Consequently, we obtain a high performance Cs_(0.19)FA_(0.81)PbI_(2.5)Br_(0.5) PSC with a power conversion efficiency(PCE) of 19.17%. An enlarged perovskite film(10 × 10 cm^(2)) containing 40 sub-cells is prepared. The average PCE of these devices is 18.33 ± 0.56%, proving the reliability of the "coffee ring" regulating strategy. This study provides an effective approach for CRE controlment in spraying technology to achieve high repeatability devices with good performance. 展开更多

Pipelines are one of the most important modern energy transportation methods,used especially for the transportation of certain dangerous energy media materials such as crude oil,natural gas,and chemical raw materials.New requirements have been put forward for the health monitoring and early security warning of pipelines because of the large-scale and complicated development trend of the pipe network system.To achieve an accurate assessment of the health conditions of pipeline infrastructure,obtaining as many precise operating parameters as possible,particularly at some critical parts of the pipeline,is necessary.Therefore,a novel type of fiber grating strain sensor array is proposed herein to monitor the pipeline hoop strain.The sensor utilizes fiber grating characteristics such as light weight,corrosion resistance,remote transmission,and strong environmental adaptability.The fiber containing the grating measurement points is implanted into the composite material to complete the sensitization encapsulation and protection of the bare fiber grating.The design of the sensor array fulfills the requirements for monitoring pipeline mass data,making it easy to form a pipeline health monitoring sensor network.The sensor sensitivity is researched by using a combination of theoretical and experimental analysis.A sensitivity test,as well as linearity and stability tests,are performed on the sensor.The experimental results show that the average sensitivity of the sensor is 14.86 pm/με,and the error from the theoretical calculation analysis value is 8.75%.Due to its high reliability,good linear response and long-term stability,and the ability to reflect the exact strain change of the outer wall of the pipeline,the designed sensor can support longterm online pipeline monitoring.The fiber grating sensor array network has successfully realized the monitoring of the pipeline’s internal operation by using external strain changes.In addition to the performance benefits,there are other merits associated with the applicability of the sensor namely simple structure,compact size,manufacturing ease,and exterior installation ease. 展开更多

In general,the material properties,loads,resistance of the prestressed concrete continuous rigid frame bridge in different construction stages are time-varying.So,it is essential to monitor the internal force state when the bridge is in construction.Among them,how to assess the safety is one of the challenges.As the continuous monitoring over a long-term period can increase the reliability of the assessment,so,based on a large number of monitored strain data collected from the structural health monitoring system(SHMS)during construction,a calculation method of the punctiform time-varying reliability is proposed in this paper to evaluate the stress state of this type bridge in cantilever construction stage by using the basic reliability theory.At the same time,the optimal stress distribution function in the bridge mid-span base plate is determined when the bridge is closed.This method can provide basis and direction for the internal force control of this type bridge in construction process.So,it can reduce the bridge safety and quality accidents in construction stages. 展开更多

Background:Macrovascular invasion(MaVI)occurs in nearly half of hepatocellular carcinoma(HCC)patients at diagnosis or during follow-up,which causes severe disease deterioration,and limits the possibility of surgical approaches.This study aimed to investigate whether computed tomography(CT)-based radiomics analysis could help predict development of MaVI in HCC.Methods:A cohort of 226 patients diagnosed with HCC was enrolled from 5 hospitals with complete MaVI and prognosis follow-ups.CT-based radiomics signature was built via multi-strategy machine learning methods.Afterwards,MaVI-related clinical factors and radiomics signature were integrated to construct the final prediction model(CRIM,clinical-radiomics integrated model)via random forest modeling.Cox-regression analysis was used to select independent risk factors to predict the time of MaVI development.Kaplan-Meier analysis was conducted to stratify patients according to the time of MaVI development,progression-free survival(PFS),and overall survival(OS)based on the selected risk factors.Results:The radiomics signature showed significant improvement for MaVI prediction compared with conventional clinical/radiological predictors(P<0.001).CRIM could predict MaVI with satisfactory areas under the curve(AUC)of 0.986 and 0.979 in the training(n=154)and external validation(n=72)datasets,respectively.CRIM presented with excellent generalization with AUC of 0.956,1.000,and 1.000 in each external cohort that accepted disparate CT scanning protocol/manufactory.Peel9_fos_InterquartileRange[hazard ratio(HR)=1.98;P<0.001]was selected as the independent risk factor.The cox-regression model successfully stratified patients into the high-risk and low-risk groups regarding the time of MaVI development(P<0.001),PFS(P<0.001)and OS(P=0.002).Conclusions:The CT-based quantitative radiomics analysis could enable high accuracy prediction of subsequent MaVI development in HCC with prognostic implications. 展开更多

Methods: Patients with cor pulmonale complicated by respiratory failure who were treated in Dongfeng People's Hospital between May 2014 and February 2017 were selected and randomly divided into the sequential group who received sequential mechanical ventilation combined with conventional therapy and the control group who received invasive positive pressure ventilation combined with conventional therapy. The serum levels of cardiac function-related neurohumoral indicators, endothelial injury indicators and oxidative stress response indicators were detected before treatment as well as 3 d and 7 d after treatment. Results: 3 d and 7 d after treatment, serum NT-proBNP, Copeptin, Ang-II, ALD, ET-1, vWF, sST2 levels of both groups of patients were significantly lower than those before treatment while NO, SOD, GSH-Px and T-AOC levels were significantly higher than those before treatment;serum NT-proBNP, Copeptin, Ang-II, ALD, ET-1, vWF, sST2, NO, 8-iso-PGF2a, MDA, SOD, GSH-Px and T-AOC levels of sequential group 3 d after treatment were not significantly different from those of control group;serum NT-proBNP, Copeptin, Ang-II, ALDET-1, vWF, sST2, 8-iso-PGF2a and MDA levels of sequential group 7 d after treatment were significantly lower than those of control group while NO, SOD, GSH-Px and T-AOC levels were significantly higher than those of control group. Conclusion: Sequential mechanical ventilation for cor pulmonale can improve the cardiac function and reduce the degree of endothelial injury and oxidative stress response. 展开更多

Objective:To compare the expression of Epstein-Barr virus encoded LMP1 and E-cadherin/β-catenin in primary and metastatic nasopharyngeal carcinoma(NPC)for the purpose of understanding their relationship.Methods:Twenty-two pairs of biopsies taken from the nasopharynx and cervical lymph node(s)of the same patient with nasopharyngeal carcinoma were collected.The expression of LMP1,E-cadherin andβ-catenin was observed on immunostained slides using LSAB method.Results:The expression rate of LMP1 in the 22 metastatic tumors(86.36%,19/22)was significantly higher than that in the 22 primary growths(68.18%,15/22),P<0.05.The mean expression percentages of E-cadherin andβ-catenin in metastatic tumors(50.11%±22.53%and 66.36±21.05%,respectively)were significantly lower than those in primary growths(71.52±24.34%and 79.40%±15.05%,respectively),P<0.05.There was a positive correlation between the expressions of E-cadherin andβ-catenin either in primary growths or metastatic tumors.Conclusion:The LMP1 is more likely to be expressed in metastatic neoplastic cells of NPC than in primary carcinoma cells,and on the contrary the expression of E-cadherin/β-catenin in metastatic cells was decreased.Accordingly,the LMP1 might have the ability to downregulate the expression of E-cadherin/β-catenin,resulting in enhancement of the invasive capacity of metastatic NPC cells. 展开更多

Objective To investigate the changes of hipbone biomechanics after the resection of ischiopubic tumors and their relationships with the complications in the convalescent stage, and directing the postoperative pelvic reconstruction. Methods DICOM data were used to create an intact hipbone finite element model and postoperative model. The biomechanical indices on the same region in the two models under the same boundary condition were compared. The differences of displacement, stress, and strain of the two models were analyzed with statistical methods. Results The distribution areas of the hipbone nodes' displacement, stress, and strain were similar before and after the simulated operation. The sacroiliac joint nodes' displacement (P=0.040) and strain (P=0.000), and the acetabular roof nodes' stress (P=0.000) and strain (P=0.005) of two models had significant differences, respectively. But the sacroiliac joint nodes' stress (P=0.076) and the greater sciatic notch nodes' stress (P=0.825) and strain (P=0.506) did not have significant differences. Conclusions The resection of ischiopubic tumors mainly affect the biomechanical states of the homolateral sacroiliac joint and acetabular roof. The complications in the convalescent stage are due to the biomechanical changes of the sacroiliac joint and the acetabular roof and disappearances of the stabilization and connection functions of the pubic symphysis and superior ramus of pubis. 展开更多

Since 1980's, innovations in management-have seen the application of several accounting techniques such as activity-based costing, balanced scorecard and target costing. However, subsequent surveys have shown that the application of these innovative management accounting techniques is not widespread. Indeed, the application of many techniques has been decidedly ephemeral. The authors suggest two important and interrelated reasons for the apparent failure to adopt such techniques. First, the absence of a conceptual framework needed to envision the collection and integration of multi-dimensional data needed in the modern organization. The second reason is the failure to exploit the data handling potentialities offered by modem computer systems. Reconstruction of contemporary management accounting system using insights from principles of events based accounting is offered as possible solution. 展开更多
