Since the demonstration of direct reprogramming of differentiated cells such as fibroblasts, to a malleable, pluripotent state by defined transcription factors great effort has been given to isolate induced pluripotent stem cells (iPSCs) with the same developmental potential as embryonic stem cells (ESCs) derived from blastocysts. Various selection and morphological criteria have led to the isolation of iPSCs with differential pluripotent capacity, but without the addition of small molecules very few lines have been able to undergo the most stringent pluripotent test - generate viable "all iPS cell mice" by tetraploid complementation. 展开更多
