白芍通过调控M2巨噬细胞极化有效缓解何首乌引发的特异质肝损伤

特异质药物性肝损伤(IDILI)是一种因药物引起的严重不良反应,通常发生在少数易感人群中。何首乌(PM)虽是一个常用的补益药,但许多临床数据已证明PM会引发IDILI。大多数IDILI的发生与机体免疫应激相关,然而对于PM导致IDILI的内在机制仍... 特异质药物性肝损伤(IDILI)是一种因药物引起的严重不良反应,通常发生在少数易感人群中。何首乌(PM)虽是一个常用的补益药,但许多临床数据已证明PM会引发IDILI。大多数IDILI的发生与机体免疫应激相关,然而对于PM导致IDILI的内在机制仍尚未被阐明。白芍(PRA)在临床上常与其他中药配伍,具有增效减毒缓解炎症等功效。但PRA是否能够缓解PM引发的肝损伤及内在机制仍未可知。在本研究中,通过网络药理学分析预测了PRA配伍PM在天然免疫中的相关靶点可能与巨噬细胞极化相关。同时采用非肝毒性剂量的LPS复制免疫应激模型,通过肝功能指标及病理等结果显示,当PM与PRA联用后,肝损伤的程度有所改善,相关促炎因子TNF-α和IL-6下降,M2巨噬细胞相关因子也有所上升。在体外,PM抑制了小鼠骨髓来源巨噬细胞(BMDMs)中IL-4诱导的Arg1及M2巨噬细胞标记物的表达,PRA则能够促进M2巨噬细胞极化,当二者配伍时,PRA可以改善PM对M2巨噬细胞极化的抑制作用。总之,这些研究结初步表明PRA能够通过促进M2巨噬细胞极化,降低炎症因子的表达,从而缓解PM引发的肝损伤,为解决何首乌引发的IDILI提供了新的思路和方案。 展开更多

近年来,新型冠状病毒感染疫情给全球医疗、政治、经济领域带来了巨大的影响。自新型冠状病毒感染疫情流行开始以来,我们对新型冠状病毒感染影响的了解有了几何数级的增长,近期国内新型冠状病毒感染疫情变化迅速,对于肺部发现肿瘤性病变... 近年来,新型冠状病毒感染疫情给全球医疗、政治、经济领域带来了巨大的影响。自新型冠状病毒感染疫情流行开始以来,我们对新型冠状病毒感染影响的了解有了几何数级的增长,近期国内新型冠状病毒感染疫情变化迅速,对于肺部发现肿瘤性病变患者的外科手术如何开展也因近期疫情的变化而出现了争议。一些研究表明,接受肺癌手术的患者感染新型冠状病毒后较普通患者更易发生呼吸衰竭和围术期死亡事件,然而,癌症治疗的推迟也与患者死亡率的上升有关。特别是新型冠状病毒奥密克戎变异株具有更高的传播性,并有可能逃避通过以前的新型冠状病毒感染、疫苗接种获得的免疫力,为了将手术患者发生新型冠状病毒感染的风险降至最低,有必要制定新的治疗指南、专家共识和预防措施,但目前疫情的快速变化导致我们没有充足的时间和证据来制定指南和共识。胸外科医生急需在术前评估发生严重并发症风险较高的特定患者人群的标准,权衡外科治疗的获益与新型冠状病毒感染的风险孰轻孰重。因此,我们尝试结合在新型冠状病毒感染流行情况下不同地区肿瘤学及胸外科学组织对于肺癌手术的指南及共识,给出一些目前国内疫情期间肺部结节及肺癌患者新型冠状病毒感染后肺部手术时机的初步建议,并希望能引发更深入的讨论和建议。 展开更多

Restructuring tilth layers(RTL)is a tillage method that exchanges the 0–20 and 20–40 cm soil layers that can be applied during cotton cultivation to increase cotton yield,eliminate weeds and alleviate severe disease,including Verticillium wilt.However,the mechanism by which RTL inhibits Verticillium wilt is unclear.Therefore,we investigated the distribution of microbial communities after rotary tillage(CK)and RTL treatments to identify the reasons for the reduction of Verticillium wilt in cotton fields subjected to RTL.Illumina high-throughput sequencing was used to sequence the bacterial and fungal genes.The disease incidence and severity of Verticillium wilt decreased by 28.57%and 42.64%,respectively,after RTL.Moreover,RTL significantly enhanced bacterial richness and evenness at 20–40 cm and-reduced the differences in fungal evenness and richness between soil depths of 0–20 and 20–40 cm.The number of Verticillium dahliae decreased,while the relative abundance of biocontrol bacteria such as Bacillus and Pseudoxanthomonas increased significantly following RTL.Overall RTL improved bacterial diversity,decreased the number of Verticillium dahliae and increased the relative abundance of biocontrol bacteria,which may have suppressed the occurrence of Verticillium wilt in cotton fields. 展开更多

背景与目的静脉血栓栓塞症(venous thromboembolism, VTE)是一种公认的在肺癌患者中有较高发病率和死亡率的并发症。本研究目的是明确我们中心肺癌患者入院时下肢深静脉血栓(lower extremity venous thrombosis, LEDVT)的发生率,并揭示... 背景与目的静脉血栓栓塞症(venous thromboembolism, VTE)是一种公认的在肺癌患者中有较高发病率和死亡率的并发症。本研究目的是明确我们中心肺癌患者入院时下肢深静脉血栓(lower extremity venous thrombosis, LEDVT)的发生率,并揭示其入院时LEDVT发生的危险因素。方法选择天津医科大学总医院肺部肿瘤外科在2017年7月-2017年12月收治的231例肺癌患者,入院时即行双下肢静脉彩超检查,以分析肺癌患者LEDVT的发生率;同时,对肺良性疾病患者入院时LEDVT的发生率进行比较。在肺癌患者中,进一步分析LEDVT发生与其临床特征的关系,寻找LEDVT发生的可能危险因素;同时,亦分析这些患者的血浆D-二聚体(D-Dimmer)、纤维蛋白原(fibrinogen, FIB)、凝血酶时间(thrombin time, TT)、活化部分凝血酶时间(activated partial thrombin time,APTT)、凝血酶原时间(prothrombin time, PT)及血小板(platelet, PLT)之间的差异。结果在231例肺癌患者中,入院时发生LEDVT者12例,其发生率为5.2%(12/231);而77例肺良性疾病患者入院时均未查见LEDVT的发生,提示肺癌患者入院时LEDVT的发生率明显高于肺良性疾病(P<0.05)。在肺癌患者中进一步分析发现,伴有远处转移(包括N3淋巴结转移)的肺癌患者较不伴有转移者更易发生LEDVT(11.29%, 7/62 vs 2.96%, 5/169)(P<0.05)。肺癌LEDVT组患者入院时的D-Dimer的中位值为1,534mg/L(369 mg/L-10,000 mg/L),明显高于非LEDVT组患者(539 mg/L, 126 mg/L-1,000 mg/L)(P<0.05);而FIB、TT、APTT、PT和PLT在两组患者之间无明显统计学差异(P>0.05)。结论我们中心肺癌患者入院时LEDVT总体发生率约为5%,明显高于肺良性疾病患者。入院时伴有远处转移(包括N3淋巴结转移)的肺癌患者更易发生LEDVT,其中,D-Dimer值较高的患者应考虑到VTE事件发生的可能。 展开更多

Application Effect of Chinese Medicine Rehabilitation in the Treatment of Adolescent Ankle Sports Injuries

Objective:To analyze the clinical effect of traditional Chinese medicine rehabilitation in treating adolescent ankle sports injuries.Methods:From December 2020 to December 2022,adolescent patients with ankle joint sports injuries admitted to our hospital were selected as observation objects,and 67 patients were divided into a control group(n=33,routine rehabilitation treatment)and an experimental group(n=34,traditional Chinese medicine rehabilitation treatment)according to the computer grouping method.Compare the treatment results.Results:(i)The treatment received in the experimental group had an efficacy of 94.11%,which was higher than that of the control group(75.75%),and statistical significance was established(P<0.05).(ii)Before treatment,there was no difference in ankle joint pain,ankle joint activity,and ankle joint function scores between the control group and the experimental group(P>0.05);after treatment,the ankle joint pain,ankle joint activity,and ankle joint function scores in the experimental group were significantly higher than those in the control group(P<0.05).(iii)The experimental group had higher scores on physiological function,physiological function,physical pain,general health status,energy,social function,emotional function,and mental health compared to the control group(P<0.05).Conclusion:Traditional Chinese medicine rehabilitation is effective in treating ankle injuries caused by sports in adolescents and improve their quality of life.Therefore,it should be popularized. 展开更多

Colorectal distension was used to establish a rat model of chronic visceral hypersensitivity. Immunohistochemistry was used to detect P2X2 and P2X3 receptor expression in dorsal root ganglia from rats with chronic visceral hypersensitivity. Results demonstrated that abdominal withdrawal reflex scores obviously increased following establishment of the model, indicating visceral hypersensitivity. Simultaneously, P2X2 and P2X3 receptor expression increased in dorsal root ganglia. After bilateral electroacupuncture at Shangjuxu and Tianshu, abdominal withdrawal reflex scores and P2X2 and P2X3 receptor expression decreased in rats with visceral hypersensitivity. These results indicated that electroacupuncture treatment improved visceral hypersensitivity in rats with irritable bowel syndrome by reducing P2X2 and P2X3 receptor expression in dorsal root ganglia. 展开更多

The convective heat transfer of supercritical-pressure RP-3(Rocket Propellant 3)aviation kerosene in a horizontal circular tube has been numerically studied,focusing mainly on the non-uniform heat transfer deterioration along the circumferential direction.The governing equations of mass,momentum and energy have been solved using the pressure-based segregated solver based on the finite volume method.The re-normalization group(RNG)k-εturbulence model with an enhanced wall treatment was selected.Considering the heat conduction in the solid wall,the mechanism of heat transfer deterioration and the buoyancy effect on deteriorated heat transfer were discussed.The evolution of secondary flow was analyzed.Effects of the outer-wall heat flux,mass flux,pressure and tube thermal conductivity on heat transfer were investigated.Moreover,the buoyancy criterion and the heat transfer correlation were obtained.Results indicate that the poor flow performance of near-wall fluid causes the pseudo-film boiling,further leads to the heat transfer deterioration.The strong buoyancy has an effect of enhancing the heat transfer at the bottom of tube,and weakening the heat transfer at the top of tube,which results in the non-uniform inner-wall temperature and heat flux distributions.Decreasing the ratio of outer-wall heat flux and mass flux,increasing the pressure could weaken the heat transfer difference along the circumferential direction,while the effect of thermal conductivity of tube on the circumferential parameters distributions is more complicated.When the buoyancy criterion of(Grq/Grth)max≤0.8 is satisfied,the effect of buoyancy could be ignored.The new correlations work well for non-uniform heat transfer predictions. 展开更多

为研究碳系材料干电极性能,采用化学气相沉积技术,制备掺硼金刚石(BDD)及垂直生长多孔石墨烯薄膜,以SEM、TEM、XPS及Raman等手段进行形貌与结构的表征,并对比这两种碳系干电极在采集脑电信号时的响应特征。在眨眼、咬牙、睁闭眼范式下... 为研究碳系材料干电极性能,采用化学气相沉积技术,制备掺硼金刚石(BDD)及垂直生长多孔石墨烯薄膜,以SEM、TEM、XPS及Raman等手段进行形貌与结构的表征,并对比这两种碳系干电极在采集脑电信号时的响应特征。在眨眼、咬牙、睁闭眼范式下采集脑电信号,石墨烯和BDD电极与商用Ag/AgCl湿电极的相关度均不低于95%。研究表明,该BDD电极是由大量金刚石晶体紧密堆垛而成,其(100)和(111)晶面硼掺杂水平的差异造成(111)晶面具有更高的导电效果,构成不同取向“5~10μm微电极”集合,即使不依赖液体导电层,也能与头皮紧密接触而传递电子;直立石墨烯阵列起到电极与头皮之间固态导电介质的作用,其三维多级孔隙结构构筑长程导电网,而开放的多级孔表面则构筑短程离子传输通道,可有效降低电极与头皮之间的界面阻抗。实验验证了二者作为干式脑电传感电极使用的可行性。 展开更多

The phenomena associated with the entrainment of free-stream turbulence (FST) into boundary-layer flows are relevant for a number of subjects. It has been be- lieved that the continuous spectra of the Orr-Sommerfeld (O-S)/Squire equations describe the entrainment process, and thus they are used to specify the inlet condition in simulation of bypass transition. However, Dong and Wu (Dong, M. and Wu, X. On continuous spectra of the Orr-Sommerfeld/Squire equations and entrainment of free-stream vortical disturbances. Journal of Fluid Mechanics, 732, 616-659 (2013)) pointed out that continuous spectra exhibit several non-physical features due to neglecting the non-parallelism. They further proposed a large-Reynolds-number asymptotic approach, and showed that the non-parallelism is a leading-order effect even for the short-wavelength disturbance, for which the response concentrates in the edge layer. In this paper, the asymptotic solution is verified numerically by studying its evolution in incompressible boundary layers. It is found that the numerical results can be accurately predicted by the asymptotic solution, implying that the latter is adequate for moderate Reynolds numbers. By introducing a series of such solutions as the inflow perturbations, the bypass transition is investigated via the direct numerical simulation (DNS). The transition processes, including the evolution of streaks, the amplification of secondary-instability modes, and the emergence of turbulent spots, agree with the experimental observations. 展开更多

Forward-/backward-facing steps in boundary-layer flows are often seen in engineering applications, and they have potential impacts on laminar-turbulent transition through scattering of the oncoming instability modes(e.g., Tollmien-Schlichting(T-S) waves). This issue is studied in the present paper by applying a local scattering framework, which is a rather generic mathematical framework on describing the mode scattering process. In this framework, a high-Reynolds-number triple-deck formalism is employed, and a transmission coefficient, defined as the ratio of the asymptotic amplitude of the instability mode downstream of the step to that upstream, is introduced. Through the systematical study, it has been found that both the forward-and backward-facing steps have a destabilizing effect on the oncoming T-S waves in subsonic boundary layers, this effect increases with the height of the step and/or the frequency of the T-S wave, and a backward-facing step(BFS) always has a greater impact than a forward-facing step(FFS). These facts agree with most of the previous investigations.However, one numerical study(WORNER, A., RIST, U., and WAGNER, S. Humps/steps influence on stability characteristics of two-dimensional laminar boundary layer. AIAA Journal, 41, 192–197(2003)), which was based on an ad-hoc configuration, showed an opposite impact of an FFS. Through the investigation on the specific configuration, it is revealed that the wrong conclusion was drawn by misinterpreting the numerical results. 展开更多

Circular RNA(circRNA)is a newly discovered non-coding RNA with special structure,which is widely expressed in eukaryotic organisms and mainly located in the cytoplasm.circRNAs participate in gene regulation by working as miRNA sponges that block the inhibitory effect of miRNA on its target genes.In addition,circRNAs can bind to RNA binding proteins to regulate gene expression.Based on characteristics of stability,expression specificity and participation in gene regulation,circRNAs are expected to be biomarkers for early diagnosis of cancer or potential targets for cancer therapy.With the help of bioinformatics analysis,circRNA microarray analysis and high-throughput sequencing technology,more circRNAs were discovered to participate in the progression of gastric cancer(GC)over the past three years.This article gives an overview of these recent research focusing on the roles of circRNAs in GC and highlights the advances. 展开更多
