除风益损颗粒质量控制方法研究

目的:采用薄层色谱法和高效液相色谱法建立除风益损颗粒的质量控制方法。方法:采用薄层色谱法定性鉴别除风益损颗粒中赤芍、牡丹皮、当归、川芎、大黄、防风、女贞子;高效液相色谱法(HPLC)对黄芩中黄芩苷进行含量测定,采用Waters Atlantis d C_(18)(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.4%磷酸溶液(47∶53)为流动相,检测波长280 nm,流速1.0 mL·min^(-1),柱温25℃,进样量10μL。结果:采用薄层色谱法建立了除风益损颗粒中赤芍、牡丹皮、当归、川芎、大黄、防风、女贞子的定性鉴别方法,斑点清晰,阴性无干扰;采用高效液相色谱法建立了除风益损颗粒中黄芩苷含量测定的方法,专属性强,黄芩苷在0.101~1.01μg(r=0.9997)范围内线性关系良好,平均回收率为100.22%,RSD为0.84%。结论:建立的薄层色谱法鉴别及含量测定方法专属性强、重复性好,可用于除风益损颗粒的质量控制。

P-HPLC法同时测定不同产地不同采收期墨旱莲中6种成分的含量

目的建立同时测定不同产地、不同采收期墨旱莲药材中咖啡酸、木犀草苷、异绿原酸B、异绿原酸C、木犀草素、蟛蜞菊内酯6种化学成分的HPLC含量测定方法。方法采用高效液相色谱法,色谱柱为Agilent C18(250 mm×4.6 mm,5μm);流动相为乙腈-0.3%甲酸水溶液,梯度洗脱;检测波长340 nm;流速0.9 mL·min^(-1);柱温30℃。结果咖啡酸线性范围为0.02008~0.20080μg(r=0.9998),平均回收率(n=6)为99.07%;木犀草苷线性范围为0.04084~0.40840μg(r=0.9999),平均回收率(n=6)为99.13%;异绿原酸B线性范围为0.08186~0.81860μg(r=1.0000),平均回收率(n=6)为98.46%;异绿原酸C线性范围为0.07966~0.79660μg(r=0.9995),平均回收率(n=6)为97.84%;木犀草素线性范围为0.08104~0.81040μg(r=1.0000),平均回收率(n=6)为97.89%;蟛蜞菊内酯线性范围为0.40280~4.02800μg(r=0.9992),平均回收率(n=6)为98.18%。不同产地的墨旱莲药材6种成分含量差异较大,江苏宿迁含量最高。结论该方法稳定可靠、准确、重复性好。适合于墨旱莲药材中多种成分的含量测定研究,为墨旱莲药材寻找更佳产地及采收期提供了科学依据。

人参益元颗粒质量控制方法研究

目的:采用薄层色谱法和高效液相色谱法建立人参益元颗粒的质量控制方法。方法:采用薄层色谱法定性鉴别人参益元颗粒中人参、黄芪、炙甘草、白术;高效液相色谱法(HPLC)对人参益元颗粒中芍药苷进行含量测定。结果:采用薄层色谱法建立了益元颗粒中人参、黄芪、炙甘草、白术的定性鉴别方法,斑点清晰,阴性无干扰。采用高效液相色谱法建立了益元颗粒中芍药苷含量测定的方法,具体为采用Alphasil VC-C 18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.2%磷酸溶液(17∶83)为流动相,检测波长230 nm,流速1.0 mL·min^(-1),柱温25℃,进样量10μL。该方法专属性强,芍药苷在23.35~116.74μg·mL^(-1)(r=0.9998)范围内线性关系良好,平均回收率为99.56%,RSD为1.40%。结论:本研究所建立的薄层色谱法鉴别及HPLC含量测定方法专属性强、重现性好,可用于人参益元颗粒的质量控制。

藤黄健骨丸HPLC指纹图谱的建立及7种成分的含量测定

目的建立藤黄健骨丸HPLC指纹图谱,并同时测定淫羊藿苷、朝藿定A、朝藿定B、朝藿定C、柚皮苷、松果菊苷、毛蕊花糖苷的含量。方法采用高效液相色谱法,Agilent ZORBAX Extend C18(4.6 mm×250 mm,5μm)色谱柱,流动相:乙腈-0.1%磷酸溶液,梯度洗脱;流速1.0 ml/min;柱温为25℃;检测波长280 nm;进样量为10μl。结合《中药色谱指纹图谱相似度评价系统》(2012年版)建立11批藤黄健骨丸的指纹图谱并进行相似度评价。结果从11批藤黄健骨丸指纹图谱中共标定20个共有峰,相似度均在0.98以上。7种成分在各自范围内线性关系良好(r≥0.9997),平均加标回收率97.70%~99.25%,RSD 0.53%~1.57%。结论所建指纹图谱重复性好,准确,可用于藤黄健骨丸的质量控制。

高效液相色谱一测多评法同时检测藤黄健骨丸中7种成分含量

目的 建立高效液相色谱一测多评法同步检测藤黄健骨丸中松果菊苷、麦角甾苷、柚皮苷、淫羊藿定A、淫羊藿定B、淫羊藿定C、淫羊藿苷含量的方法。方法 采用高效液相色谱法,Agilent ZORBAX Extend C(4.6mm×250mm,5μm)色谱柱,流动相:乙腈-0.1%磷酸溶液,梯度洗脱;流速1.0 ml/min;检测波长280 nm;柱温为25℃;进样量为10μl。以淫羊藿苷为内参物,建立其他6种成分的相对校正因子,并计算各成分含量。采用外标法测定藤黄健骨丸中7种成分含量与一测多评法计算结果的准确性进行比较。结果 测定了11批藤黄健骨丸中上述成分的含量,7种成分在各自范围内线性关系良好(r≥0.999 7),平均加标回收率为97.70%~99.25%,RSD为0.53%~1.57%(n=6)。一测多评法所测结果与外标法无明显差异。结论 该方法可用于藤黄健骨丸中7种成分含量的同步测定,达到多指标质量控制的目的。
