永生化人甲状旁腺细胞系的建立及功能鉴定

目的 建立永生化的人甲状旁腺细胞系并进行功能鉴定。方法 采用慢病毒感染的方式将SV40(simian vacuolating virus 40,SV40)大T抗原导入细胞,转染后用嘌呤霉素筛选转染成功的细胞。通过细胞形态学、实时荧光定量聚合酶链式反应(real tim... 目的 建立永生化的人甲状旁腺细胞系并进行功能鉴定。方法 采用慢病毒感染的方式将SV40(simian vacuolating virus 40,SV40)大T抗原导入细胞,转染后用嘌呤霉素筛选转染成功的细胞。通过细胞形态学、实时荧光定量聚合酶链式反应(real time quantitative polymerase chain reaction,RT-qPCR)及免疫荧光检测转染后传代甲状旁腺细胞表达甲状旁腺激素(parathyroid hormone,PTH)、钙敏感受体(calcium sensitive receptor,CaSR)和神经胶质细胞缺失因子2(glial cells missing homolog 2,GCM2)mRNA及蛋白的表达情况。通过细胞增殖实验检测转染的细胞对Ca~(2+)的反应性。结果 转染成功的甲状旁腺细胞传代后大部分呈梭形,细胞核椭圆形,胞浆均质透明;RT-qPCR结果显示甲状旁腺细胞表达CaSR、GCM2以及PTH mRNA;免疫荧光显示永生化的甲状旁腺细胞系表达甲状旁腺相关蛋白CaSR、GCM2以及PTH。细胞增殖实验显示低钙能够促进甲状旁腺细胞系增殖。结论 本研究成功建立了永生化的甲状旁腺细胞系,为进一步探讨继发性甲状旁腺功能亢进的发病机制及治疗提供了实验基础。 展开更多

目的:探讨血液透析(HD)甲状旁腺切除术(PTx)患者COMM域蛋白5(COMMD5)与血压下降之间的关系。方法:用ELISA方法检测HD患者PTx前后COMMD5水平,并在体外使用COMMD5重组蛋白刺激大鼠血管内皮细胞,观察心房利钠肽(ANP)和内皮型一氧化氮合酶(e... 目的:探讨血液透析(HD)甲状旁腺切除术(PTx)患者COMM域蛋白5(COMMD5)与血压下降之间的关系。方法:用ELISA方法检测HD患者PTx前后COMMD5水平,并在体外使用COMMD5重组蛋白刺激大鼠血管内皮细胞,观察心房利钠肽(ANP)和内皮型一氧化氮合酶(eNOS)等血管活性物质的表达情况。结果:(1)HD患者PTx术后血压低于术前(收缩压140.58±20.55 mmHg vs 148.81±18.14 mmHg,P<0.05;舒张压75.48±12.83 mmHg vs 88.74±10.92 mmHg,P<0.000 1);(2) PTx术后血液COMMD5较术前升高[0.97(0.64~1.79)ng/mL vs 0.65(0.43~1.07) ng/mL,P=0.02]。(3)PTx术前后收缩压下降与COMMD5相关,r=-0.57;P=0.000 7;舒张压下降与COMMD5相关,r=-0.36;P=0.043 6;(4)体外COMMD5重组蛋白刺激血管内皮细胞24h,ANP蛋白的表达量是空白对照组的2.39倍(P=0.017 9);eNOS蛋白的表达量是空白对照组的2.70倍(P=0.000 5)。结论:HD患者PTx后血清COMMD5水平升高,可能通过COMMD5上调ANP、eNOS表达,参与PTx后血压下降。 展开更多

目的探讨miR-429在膀胱癌中的表达及作用机制。方法选取广东省内三家医院2014年1月~2016年12月期间收集的膀胱癌标本120例,同时选取距离肿瘤边缘大于2 cm的癌旁组织作为对照。SYBR green实时荧光定量PCR法检测miR-429在膀胱癌、癌旁组... 目的探讨miR-429在膀胱癌中的表达及作用机制。方法选取广东省内三家医院2014年1月~2016年12月期间收集的膀胱癌标本120例,同时选取距离肿瘤边缘大于2 cm的癌旁组织作为对照。SYBR green实时荧光定量PCR法检测miR-429在膀胱癌、癌旁组织中的相对表达量,并分析miR-429与临床相关参数的关系,同时观察miR-429过表达或者敲除时T24细胞(膀胱癌细胞系)增殖和凋亡情况。结果膀胱癌组织miR-429的相对表达水平高于正常膀胱组织(P<0.05);miR-429的表达水平与膀胱癌的肿瘤大小、有无淋巴结转移,及TNM分期有关(P<0.05);肿瘤直径大于5 cm组miR-429的表达水平高于肿瘤直径小于5 cm组;有淋巴结转移的膀胱癌组miR-429的表达水平高于无淋巴结转移的膀胱癌组;Ⅲ+Ⅳ期膀胱癌组miR-429的表达水平高于Ⅰ+Ⅱ期膀胱癌(P<0.05)。miR-429的表达水平与膀胱癌患者的年龄、性别无关(P>0.05)。miR-429过表达时,膀胱癌细胞(T24细胞系)增殖增加,凋亡减少;而miR-429敲除时,膀胱癌细胞(T24细胞系)增殖减少,凋亡增加。结论 miR-429在膀胱癌的发生发展中起着重要作用,其表达水平对膀胱癌的严重程度及预后情况具有一定的参考价值。 展开更多

The relationship between serum trimethylamine N-Oxide level and carotid intima media thickness in untreated essential hypertension patients

Background Circulating trimethylamine N-Oxide (TMAO) level has been linked to adverse cardiovascular outcome and mortality in the general population, and atherosclerosis is a window of the cardiovascular disease. Therefore, the present study was designed to investigate the relationship between TMAO level and atherosclerosis in untreated essential hypertension patients. Methods We measured serum TMAO level in atherosclerosis, subclinical atherosclerosis and controls matched by age and sex. The link between serum TMAO level and CIMT was subsequently assessed. Results The level of serum TMAO was significantly higher in atherosclerosis pa- tients than in controls. Serum TMAO level was positively correlated with carotid intima media thickness (r = 0.783, P 〈 0.001), and logistic regression indicated that TMAO was a risk factor of atherosclerosis (OR, 1.904; 95% CI, 1.197- 2.733, P 〈 0.001). Conclusions Serum TMAO concentration positively correlates to carotid intima media thickness, and should be a good predicted biomarker for atherosclerosis. 展开更多

Central aortic systolic blood pressure can better predict prolonged QRS duration than brachial artery systolic blood pressure in rural community residents

Background Central aortic systolic blood pressure(CASP) has been shown to be a stronger predictor of target-organ damage and cardiovascular events than brachial systolic blood pressure(BSBP), but there was no data about whether CASP can predict prolonged QRS duration more than BSBP. We examined the association of CASP and BSBP with QRS duration in rural community residents. Methods We retrospectively analyzed 490 rural community residents. Standard resting 12-lead ECG and central aortic blood pressure(CABP) were measured noninvasively in all subjects at baseline. The QRS duration was equal to or more than 120 ms being defined as prolonged QRS duration. Results The prolonged QRS duration group showed higher CASP(139.38 ± 11.67 vs. 135.36 ± 16.22, P = 0.031) and BSBP(136.03 ± 6.74 vs. 124.44 ± 13.01, P 〈 0.001) as compared with controls. Multivariate linear regression analysis showed that CASP, BSBP and heart rate were independently affecting QRS duration. Logistic regression analyses showed that CASP(OR 1.057, 95%CI: 1.027, 1.088, P 〈 0.001)and BSBP(OR 1.056, 95%CI: 1.027, 1.086, P = 0.032) were independent predictors of prolonged QRS duration after adjustment for age, sex, body mass index, heart rate. CASP had a better predictive value for prolonged QRS duration than(AUC: 0.793 vs. 0.601, P 〈 0.001) BSBP. Conclusions Our findings demonstrate that both CASP and BSBP are risks for prolonged QRS duration, but CASP can predict prolonged QRS duration better than BSBP. 展开更多
