We characterized the genome sequences of defective-interfering(DI) particle DNA of porcine circovirus type 2(PCV2) by sequencing and bioinformatics analyses. DI particles were both generated by serial passage of PCV2 in PK15 cells and obtained from sera of pigs with postweaning multisystemic wasting syndrome(PMWS). These subviral isolates ranged from 358 nt to 1 125 nt genomes. Investigating the complexity and diversity of PCV2 DI in vivo and in vitro can help elucidating the evolutionary history of PCV2. 展开更多

Recently, a novel porcine circovirus-like virus P1 with a circular DNA genome of 0.648 kb was identified. P1 antigen was detected both in vitro and in vivo by synthetic peptide-derived polyclonal antibody-based immunochemistry. The designed peptides were synthesized by solid-phase technique, purified by high performance liquid chromatography, coupled to Keyhole limpet hemocyanin, and injected into rabbits to prepare polyclonal antibody. The emergence of positive cells revealed that synthetic peptide could elicit antibodies against P1 and viral protein could be synthesized. The polyclonal peptide antibodies described here was successfully applied to immunochemical staining and proved helpful in diagnosing P1. 展开更多

PDGF signaling from pharyngeal pouches promotes arch artery morphogenesis

The great vessels of the heart originate from the pharyngeal arch arteries(PAAs).Anomalies of the PAAs often occur together with pharyngeal pouch malfo rmations,but the reasons for this phenomenon are not fully understood.In the current study,we show that platelet-derived growth factor(PDGF)signaling derived from the pharyngeal pouches plays an important function in PAA vasculogenesis,During PAA development in zebrafish embryos,pdgfaa and pdgfab are expressed in the developing pharyngeal pouches.Results from loss-of-function experiments revealed a critical role of these genes in PAA formation.We found that nitroreductase(NTR)-mediated pouch ablation distinctly decreased PDGF receptor tyrosine phosphorylation,yielding a severe loss of PAAs.Importantly,pouch-specific overexpression of pdgfaa in pdgfaa-/-;pdgfab-/-mutants significantly relieved the PAA defects,which indicated a primary role of pharyngeal pouch-expressed PDGF ligands in signal activation and PAA morphogenesis.Our findings further showed that PDGF signaling was indispensable for the proliferation of PAA angioblasts.Together,these results established a role for PDGFaa-and PDGFab-mediated tissuetissue interaction during PAA development. 展开更多

Background:Physiological signal-based research has been a hot topic in affective computing.Previous works mainly focus on some strong,short-lived emotions(e.g.,joy,anger),while the attention,which is a weak and long-lasting emotion,receives less attraction.In this paper,we present a study of attention recognition based on electrocardiogram(ECG)signals,which contain a wealth of information related to emotions.Methods:The ECG dataset is derived from 10 subjects and specialized for attention detection.To relieve the impact of noise of baseline wondering and power-line interference,we apply wavelet threshold denoising as preprocessing and extract rich features by pan-tompkins and wavelet decomposition algorithms.To improve the generalized ability,we tested the performance of a variety of combinations of different feature selection algorithms and classifiers.Results:Experiments show that the combination of generic algorithm and random forest achieve the highest correct classification rate(CCR)of 86.3%.Conclusion:This study indicates the feasibility and bright future of ECG-based attention research. 展开更多
