Early detection and intervention are key strategies to reduce mortality,increase longterm survival,and improve the therapeutic effects of hepatocellular carcinoma(HCC)patients.Herein,the isobaric tag for relative and absolute quantitation(iTRAQ)-based quantitative proteomic strategy was used to study the secretomes in conditioned media from HCC cancerous tissues,surrounding noncancerous tissues,and distal noncancerous tissues to identify diagnostic and prognostic biomarkers for HCC.In total,22 and 49 dysregulated secretory proteins were identified in the cancerous and surrounding noncancerous tissues,respectively,compared with the distal noncancerous tissues.Among these proteins,carbonic anhydrase II(CA2)was identified to be significantly upregulated in the secretome of cancerous tissues;correspondingly,the serum concentrations of CA2 were remarkably increased in HCC patients compared with that in normal populations.Interestingly,a significant increase of serum CA2 in recurrent HCC patients after radical resection was also confirmed compared with HCC patients without recurrence,and the serum level of CA2 could act as an independent prognostic factor for time to recurrence and overall survival.Regarding the mechanism,the secreted CA2 enhances the migration and invasion of HCC cells by activating the epithelial mesenchymal transition pathway.Taken together,this study identified a novel biomarker for HCC diagnosis and prognosis,and provided a valuable resource of HCC secretome for investigating serological biomarkers. 展开更多

吡咯啉-5-羧酸(P5C)、哌啶-6-羧酸(P6C)是一类同代谢反应和疾病诊疗密切相关的环形亚胺化合物,在机体内氨基酸代谢中发挥重要作用。本研究利用Thermomicrobium roseum肌氨酸氧化酶对-C-N-键的催化能力建立了一种酶法制备工艺并进行了条... 吡咯啉-5-羧酸(P5C)、哌啶-6-羧酸(P6C)是一类同代谢反应和疾病诊疗密切相关的环形亚胺化合物,在机体内氨基酸代谢中发挥重要作用。本研究利用Thermomicrobium roseum肌氨酸氧化酶对-C-N-键的催化能力建立了一种酶法制备工艺并进行了条件优化。通过薄层层析(TLC)、液质联用(LC-MS)和~1H-NMR核磁共振鉴别出化合物结构,并建立了含量测定方法。单因素实验分析反应温度、pH和底物浓度对产量的影响,并通过响应面得到P5C最佳反应条件:温度为79.33℃、pH为8.54、底物浓度为112.82 mmol/L,P6C最佳反应条件:温度80.19℃、pH为8.31、底物浓度83.58 mmol/L。在此条件下反应30 min,P5C和P6C的产量分别为340.15±9.12, 450.87±8.54 mg/L。反应48 h后,转化率分别达到81.94%和99.99%。研究结果为酶法制备P5C和P6C奠定了理论基础,为吡咯啉、哌啶类化合物的合成提供了新的思路。 展开更多

The Jin-Neelin model for the El Nio–Southern Oscillation(ENSO for short) is considered for which the authors establish existence and uniqueness of global solutions in time over an unbounded channel domain. The result is proved for initial data and forcing that are sufficiently small. The smallness conditions involve in particular key physical parameters of the model such as those that control the travel time of the equatorial waves and the strength of feedback due to vertical-shear currents and upwelling; central mechanisms in ENSO dynamics.From the mathematical view point, the system appears as the coupling of a linear shallow water system and a nonlinear heat equation. Because of the very different nature of the two components of the system, the authors find it convenient to prove the existence of solution by semi-discretization in time and utilization of a fractional step scheme. The main idea consists of handling the coupling between the oceanic and temperature components by dividing the time interval into small sub-intervals of length k and on each sub-interval to solve successively the oceanic component, using the temperature T calculated on the previous sub-interval, to then solve the sea-surface temperature(SST for short) equation on the current sub-interval. The passage to the limit as k tends to zero is ensured via a priori estimates derived under the aforementioned smallness conditions. 展开更多
