Anterior cruciate ligament(ACL)injuries of the knee are one of the most common and serious athletic injuries.The widely used cortical suspension fixation buttons for ligament reconstruction are permanent implants,particularly those made from conventional steel or titanium alloys.In this study,a biodegradable Zn-0.45Mn-0.2Mg(ZMM42)alloy with the yield strength of 300.4 MPa and tensile strength of 329.8 MPa was prepared through hot extrusion.The use of zinc alloys in the preparation of cortical suspension fixation buttons was proposed for the first time.After 35 d of immersion in simulated body fluids,the ZMM42 alloy fixation buttons were degraded at a rate of 44μm/a,and the fixation strength was retained(379.55 N)in the traction loops.Simultaneously,the ZMM42 alloy fixation buttons exhibited an increase in MC3T3-E1 cell viability and high antibacterial activity against Escherichia coli and Staphylococcus aureus.These results reveal the potential of biodegradable zinc alloys for use as ligament reconstruction materials and for developing diverse zinc alloy cortical suspension fixation devices. 展开更多

小鼠是最广泛使用的模式生物,构建基因改造的小鼠模型是研究基因功能和疾病发生机制的重要手段。从20世纪80年代发展至今,小鼠基因改造技术更新迭代,主要包括胚胎干细胞介导的同源重组策略、配子介导的转基因策略、以及最新基于CRISPR/C... 小鼠是最广泛使用的模式生物,构建基因改造的小鼠模型是研究基因功能和疾病发生机制的重要手段。从20世纪80年代发展至今,小鼠基因改造技术更新迭代,主要包括胚胎干细胞介导的同源重组策略、配子介导的转基因策略、以及最新基于CRISPR/Cas9技术的遗传改造策略等。由我国科学家自主研发的全新小鼠基因改造技术——“人造精子细胞”(或“类精子干细胞”)介导的半克隆技术与以往策略相比,具有小鼠构建周期短、效率高、成本低、应用场景兼容性强等特点。为了摆脱我国小鼠实验动物“卡脖子”风险,填补标签蛋白质小鼠资源库的国际空白,中国科学院分子细胞科学卓越创新中心进行了战略资源布局,于2017年启动了基因组标签计划(genome tagging project,GTP),并成立了GTP研发中心致力于建立全新的标签细胞和小鼠战略资源库。GTP研发中心通过对“人造精子细胞”基因组上编码蛋白质的基因原位系统性地添加标签序列,建立了可用液氮保存的标签细胞资源库,并作为生产标签小鼠的种子贮备。当需要标签小鼠时,利用半克隆技术将标签细胞注射到小鼠卵子中,一步法获得标签小鼠。标签小鼠可通过标准化的标签抗体监测特定蛋白质从而实现蛋白质的在体、实时、动态研究。目前,GTP研发中心已构建1532个标签细胞和277个标签小鼠,初步形成了具有特色标签产品战略资源库;已有242个标签小鼠品系被分发到3个囯家15个地区的32个研究机构,供66个研究团队使用。同时,GTP研发中心已建立完备的标签产品信息,定期在GTP官网上发布更新,供科研人员查询、订购和定制。后续标签小鼠的配繁规律、标签蛋白质组织图谱描绘、发表文献等也会陆续更新到GTP官网。GTP研发中心将对标行业尖端,打造全球独特性战略资源,为蛋白质功能研究提供标准化平台,推动生命科学的发展和临床转化。 展开更多

No. 1 document of central government proposed innovating agricultural production and operation system and establishing new agricultural business entities. Seizing these customers becomes a great challenge for pesticide enterprises in the new trend. Therefore,pesticide enterprises need to find out key factors driving customer equity,so as to carry out pertinent marketing and grab the maximum market share. Based on the first-hand survey data,this paper analyzed the influence of value equity,brand equity and relation equity on customer equity by factor analysis and structural equation analysis. It found that the relation equity has the highest driving effect,especially training,community building and visiting experience. Finally,it came up with some recommendations to make pertinent marketing. 展开更多

Oxygen-deficient titania as alternative support for Pt catalysts for the oxygen reduction reaction

Insufficient electrochemical stability is a major challenge for carbon materials in oxygen reduction reaction (ORR) due to carbon corrosion and insufficient metal-support interactions. In this work, titania is explored as an alternative support for Pt catalysts. Oxygen deficient titania samples including TiO2-x and TiO2_xNy were obtained by thermal treatment of anatase TiO2 under flowing H2 and NH3, respectively. Pt nanoparticles were deposited on the titania by a modified ethylene glycol method. The samples were characterized by N2-physisorption, X-ray diffraction and X-ray photoelectron spectroscopy. The ORR activity and long-term stability of supported Pt catalysts were evaluated using linear sweep voltammetry and chronoamperometry in 0.1 mol/L HC104. Pt/TiO2_x and Pt/TiO2_xNy showed higher ORR activities than Pt/TiO2 as indicated by higher onset potentials. Oxygen deficiency in TiO2-x and TiO2-xNy contributed to the high ORR activity due to enhanced charge transfer, as disclosed by electrochemical impedance spectroscopy studies. Electrochemical stability studies revealed that Pt/TiOE_x exhibited a higher stability with a lower current decay rate than commercial Pt/C, which can be attributed to the stable oxide support and strong interaction between Pt nanoparticles and the oxygen-deficient TiO2-x support. 展开更多

Embryonic stem cells possess fascinating capacity of self-renewal and developmental potential,leading to significant progress in understanding the molecular basis of pluripotency,disease modeling,and reprogramming technology.Recently,2-cell-like embryonic stem cells(ESCs)and expanded potential stem cells or extended pluripotent stem cells(EPSCs)generated from early-cleavage embryos display some features of totipotent embryos.These cell lines provide valuable in vitro models to study underlying principles of totipotency,cell plasticity,and lineage segregation.In this review,we summarize the current progress in this filed and highlight the application potentials of these cells in the future. 展开更多
