The impact of epigenetic modifications like DNA methylation on plant phenotypes has expanded the possibilities for crop development.DNA methylation plays a part in the regulation of both the chromatin structure and gene expression,and the enzyme involved,DNA methyltransferase,executes the methylation process within the plant genome.By regulating crucial biological pathways,epigenetic changes actively contribute to the creation of the phenotype.Therefore,epigenome editing may assist in overcoming some of the drawbacks of genome editing,which can have minor off-target consequences and merely facilitate the loss of a gene’s function.These drawbacks include gene knockout,which can have such off-target effects.This review provides examples of several molecular characteristics of DNA methylation,as well as some plant physiological processes that are impacted by these epigenetic changes in the plants.We also discuss how DNA alterations might be used to improve crops and meet the demands of sustainable and environmentally-friendly farming. 展开更多

Antibiotic resistance poses a significant global health threat, necessitating a thorough understanding of its prevalence in various ecological contexts. Medicinal plants, renowned for their therapeutic properties, host endophytic bacteria that produce bioactive compounds. Understanding antibiotic resistance dynamics in these bacteria is vital for human health and antibiotic efficacy preservation. In this study, we investigated antibiotic resistance profiles in endophytic bacteria from five medicinal plants: Thankuni, Neem, Aparajita, Joba, and Snake plant. We isolated and characterized 113 endophytic bacteria, with varying resistance patterns observed against multiple antibiotics. Notably, 53 strains were multidrug-resistant (MDR), with 14 exhibiting extensive drug resistance (XDR). Thankuni-associated bacteria displayed 44% MDR and 11% XDR, while Neem-associated bacteria showed higher resistance (60% MDR, 13% XDR). Aparajita-associated bacteria had lower resistance (22% MDR, 6% XDR), whereas Joba-associated bacteria exhibited substantial resistance (54% MDR, 14% XDR). Snake plant-associated bacteria showed 7% MDR and 4% XDR. Genus-specific distribution revealed Bacillus (47%), Staphylococcus (21%), and Klebsiella (11%) as major contributors to MDR. Our findings highlight diverse drug resistance patterns among plant-associated bacteria and underscore the complexity of antibiotic resistance dynamics in diverse plant environments. Identification of XDR strains emphasizes the severity of the antibiotic resistance problem, warranting further investigation into contributing factors. 展开更多

Human toxoplasmosis is caused by the intracellular protozoan parasite Toxoplasma gondii. Although T. gondii infection is generally asymptomatic for most of the immunocompetent adults, severe complications may occur particularly in pregnant women and immunocompromised individual. Host cell immunity plays a critical role in parasite differentiation and persistence in the host. Therefore, genetic polymorphism in the host immune genes, for instance interferon-γ gene could be linked with possibility of T. gondii infection. The objective of the study was to verify the link between the single nucleotide polymorphisms (SNPs) in the IFN-γ gene of pregnant women and T. gondii infection through correlating with anthropometric and sociodemographic parameters. In this study, ninety-two (N = 92) pregnant women (16 - 40 years) and healthy controls (N = 95) with similar age ranges were included. Among them, 25% (n = 23) pregnant women were seropositive for T. gondii IgG antibodies by Rapid Test Assay. Allelic and genotypic frequencies of IFN-γ +874T/A (rs2430561) SNPs were evaluated by using ARMS-PCR. The distribution of the A and T alleles in the specific position of the IFN-γ gene in the T. gondii-infected pregnant women and the control groups did not differ significantly, according to the data. However, we found a higher frequency (13.04%) of A/A genotype in T. gondii infected pregnant women as compared to non-infected individuals (8.70%), demonstrating that T. gondii infection susceptibility may be increased by homozygosity for the A allele. Further studies are to be needed to find out the link between host gene polymorphism and T. gondii infection in Bangladesh. 展开更多

Serologic Detection of <i>Toxoplasma gondii</i>in Cat Owners Residing at Dhaka Metropolitan Area of Bangladesh

Infection with the para... <i>Toxoplasma gondii</i> is a zoonotic protozoan that can infect any warm-blooded mammal. <i>T. gondii</i> infects about one-third of the human population on the planet. Infection with the parasite in human causes toxoplasmosis that may pose a high risk in immunocompromised individuals under certain clinical conditions. Cats are the ultimate hosts of <i>T. gondii</i> where oocysts are formed through mating of male and female gametes. Infected cats can expel <i>T. gondii</i> oocysts in their feces, and thereby capable of pass on a disease to humans and other animals through consumption of foods, vegetables and water that are polluted with cat feces. The study was conducted to detect the presence of anti-<i>T. gondii</i> IgM and IgG antibodies in the blood of individuals with or without cat contact to determine if there is any relationship between cat contacts and <i>T. gondii</i> infection in humans. To address this, we enrolled subjects who contacted with the cat as target group and individuals with no cat contact as control group. Following register of different demographic data (including age, sex, education, foods habit, income status, etc.), whole blood from each enrolled subject of both the target group and control group was collected for serum preparation. <i>T. gondii</i> infected subjects were detected by Toxo Rapid test kit through identifying anti-<i>T. gondii</i> IgM and IgG antibodies in their serum. We found that only three out of twenty subjects who were in contact with cat showed positive IgG response while IgM antibody response was absent for all subjects. When compared with the data from control group, we did not find any significant association (p = 0.33) of cat contact with the transmission <i>of T. gondii</i> into human. However, with this small number of study subjects, we cannot conclusively say that there is no impact of cat contact on the transmission of <i>T. gondii</i> into human. Whether any association exists or not can be ascertained with a large number of subjects from different areas of Bangladesh in a future study in the population. 展开更多

Toxoplasma gondii (T. gondii) an intracellular protozoan parasite, infects mammals including human population world-wide. Upon primary infection, the parasite contributes to mild flu like symptoms in immune competent host, but life threatening complication is seen in immune compromised patients and in pregnant women. Understanding the host-parasite interaction is critical for understanding the pathogenesis and biology parasite reactivation in the host. In this study, we used proteotrasncriptomics analyses by integrating the transcriptomics and proteomics data of T. gondii infected mouse liver to uncover the effector molecules responsible for disease pathogenesis that can be used as candidate markers for diagnosis and drug target. With this aim, we systematically integrated transcriptomicand proteomic data, representing the parasite infected mouse liver. Out of 2758 differentially expressed genes (DEGs) and 301 differentially expressed proteins (DEPs), 159 overlapping genes were identified. Among them, 86 genes were upregulated and 72 were downregulated in their respective mRNA and protein levels in the infected condition. Gene Ontology (GO) analysis revealed that the upregulated genes were mostly associated with immune system processes whereas the downregulated genes were involved in oxidation-reduction process and metabolism of lipid, and fatty acids. Protein-protein interaction (PPI) network analysis uncovered an interaction-hub including, Psmb8, Psmb9 and Tap1 for upregulated proteins and Cyp1A2, Cyp4A10 and Cyp3A11 for down-regulated proteins. Further studies are needed to validating these effector molecules. These molecules are likely to play a vital role in disease pathogenesis, as well as can be used as potential diagnostic marker and drug target candidates. 展开更多

Furthermore, among low-income countries including Bangladesh, individuals have a high risk for developing breast cancer. This study aimed to identify candidate mitochondrial DNA (mtDNA) biomarkers for breast cancer diagnosis in Bangladeshi women to be used as a preventive approach. We screened the blood samples from 24 breast cancer patients and 20 healthy controls to detect polymorphisms in the D-loop and the ND3-and ND4-coding regions of mtDNA by direct sequencing. Among 14 distinct mutations, 10 polymorphisms were found in the D-loop, 3 were found in the ND3-coding region, and 1 was found in the ND4-coding region. The frequency of two novel polymorphisms in the D-loop, one at position 16290 (Tins) and the other at position 16293 (A-del), was higher in breast cancer patients than in control subjects (position 16290: odds ratio = 6.011, 95% confidence interval = 1.2482 to 28.8411, P = 0.002; position 16293: odds ratio = 5.6028, 95% confidence interval = 1.4357 to 21.8925, P = 0.010). We also observed one novel mutation in the ND3-coding region at position 10316 (A > G) in 69% of breast cancer patients but not in control subjects. The study suggests that two novel polymorphisms in the D-loop may be candidate biomarkers for breast 展开更多

The phenomena of accelerated urbanisation is the main culprit, wherein besides bringing higher standard of living, it has also brought problems, as growth of dense and unplanned residential areas, environmental pollution, non-availability of services and amenities, solid waste etc. Remote sensing satellite data is suitable for urban land use mapping to get detail and up- to-date information for environmental management. Where as GIS helps in developing database system for urban information, which supports decision making process. Development of digital database on all aspects of land use and urban planning is the next crucial task for the future in which remote sensing based informa-tion is going to play a major role. In Delhi, rise in population and growth in economic activity has led to en-vironmental degradation. With this view an attempt has been made to study the quality of urban environment in the East district of Delhi, which is experiencing very high urban growth with 98.75% urban population in 2001. For this study Landsat ASTER (MSS) data of year 2001 (15 m Ground resolution), Guide map of the year 1982 and demographic and environmental data has been used. Eight parameters were selected, which affect the urban environmental quality, namely built-up area, open spaces, household density, occupancy ra-tio, population density, accessibility to roads, noise and smell affected area. The study shows that the quality of environment has been degraded when we compare 1982 and 2003 data. Most of the East district was in a better state of environment in 1982, but in 2003 things have been changed and now 50% area is in very good, fair and desirable condition. The public participation and involvement should be encouraged planning and decisions making for the improvement in better urban environmental quality. 展开更多

Objective: To evaluate the ameliorating effects of Raphanus sativus leaves(RSL)against sodium arsenite(Sa)-induced adverse effects through mice experiments.Methods: Swiss albino mice were divided into four equal groups: control, Sa, RSL,RSL + Sa. Sa(10 mg/kg body weight/day), and powder form of RSL(50 mg/kg body weight/day) were provided as food supplement orallty. Blood indices were measured using commercially available kits through colorimetric methods.Results: It was observed that lactate dehydrogenase, alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase activities were significantly(P < 0.05)higher in Sa-treated mice than those in the control group. RSL significantly reduced Sainduced elevation of the activities of these enzymes in serum significantly(P < 0.05).Serum butyrylcholinesterase activity and high density lipoproteins cholesterol levels in Satreated mice were significantly(P < 0.05) lower than the control group, and the food supplementation of RSL could significantly(P < 0.05) prevent the reduction of Sa-mediated serum butyryl cholinesterase activity and high density lipoproteins cholesterol levels. RSL could also reduce the Sa-induced elevation of serum urea level significantly(P < 0.05).Conclusions: Results of this study suggest the protective or ameliorating effects of RSL on Sa-induced perturbation of blood indices are related to the hepatic, cardiovascular and kidney dysfunction. Therefore, RSL may be useful to reduce arsenic toxicity in human in the future. 展开更多

OBJECTIVE To isolate and characterize bioactive compound from traditionally important medicinal plant Mikania cordata,and to investigate muti-faced pharmacological activities of the isolated compound.METHODS A triterpeenoid,16-hydroxy betulinic acid(16 HBA) was isolated from Mikania cordata leaf and the structure of the compound was determined by NMR spectroscopic means.Antimicrobial activity of 16 HBA was tested by disc diffusion method and minimum inhibitory concentration(MIC) against the tested microorganisms was determined.The analgesic property of 16 HBA was tested using acetic acid-induced writhes in mice and hot plate thermal stimulation in rats.The anti-inflammatory activity was studied using carrageenin-induced paw edema method.The antipyretic potential of 16 HBA was evaluated by using yeast-induced hyperthermia in rats.RESULTS The triterpenoid 16 HBA showed potent antibacterial activity with inhibition zone of diameter ranging from 12.0~17.5 mm and antifungal activity with mycelial growth inhibition ranges from 37.6%~54.5%.The MIC values for antibacterial and antifungal activities ranged from 31.5~125 and 250~1000 μg·mL-1 respectively.The compound(50 and100 mg·kg^(-1) body weight) showed potent peripheral and central analgesic activity having 55.19% and41% of writhing inhibition at 90 min after administration of the compound and the highest 55.98%,79.18% elongation of reaction time,respectively.In anti-inflammatory activity screening,the compound(100 mg·kg^(-1)) revealed the highest 77.08% edema inhibition at 4 h after administration of carrageenan.In antipyretic assay,16 HBA exhibited a strong antipyretic effect in yeast-induced rats.CONCLUSION The present study confirmed that 16-hydroxy betulinic acid isolated from Mikania cordata has potent antimicrobial,analgesic,anti-inflammatory and antipyretic properties.Our results can be seen as scientific support for the traditional and folklore usage of Mikania cordata in Bangladesh for the treatment of different ailments and provide opportunities to explore this plant as a source bioactive compounds for biochemical and pharmaceutical industries. 展开更多

It can potentially infect almost all mammalian and avian hosts including one-third of the human population world-wide. The major target group of the parasite includes immunocompromised patients (e.g. AIDS, cancer, organ transplantation) and fetus bearing pregnant women where it develops toxoplasmic encephalitis, myocarditis, chorioretinitis and abnormal fetal brain development or stillbirths respectively. In this review, we have presented the current status of T. gondii infection in livestock animals and human population in Bangladesh to assess the country-wide relative risk. Although exact prevalence is difficult to predict due to the scarcity of data, nevertheless existing literature suggests that 16% - 39% humans and 8% - 70% domestic animals are infected with T. gondii, which implies Bangladeshi population is at high risk of toxoplasmosis. Furthermore, we have proposed a potential area of research to decipher the genetic diversity and transmission routes of T. gondii infection into Bangladeshi population. 展开更多

An understanding of the amount and type of aerosols present in the atmosphere is required for the atmospheric correction of satellite imagery. A sensitivity analysis of the atmospheric inputs to the MOD09 software has shown that uncertainty in the estimation of Aerosol Optical Depth (AOD) has the greatest impact on the accuracy of atmospheric correction of MODIS data. The MOD09 software used for the generation of surface reflectance products estimates the AOD of the atmosphere at the time of image acquisition. AOD measurements retrieved from MODIS were compared with near-simultaneous AErosol RObotic NETwork (AERONET) data over three sites in Australia, using time-series of MODIS surface reflectance products. The results of the study provide an important independent validation of ACRES MODIS Surface Reflectance Products. This procedure may be applied to long time series MODIS data for estimating the accuracy of MOD09 retrieved AOD. 展开更多

The crisis of coronavirus disease(COVID-19)is growing and has a potential impact on low-and-middle-income countries(LMICs),particularly in Asian regions,which,have reported less so far.The main purpose of this article is to discuss the challenges of COVID-19 management in Asian LMICs and propose some future guidelines to control the transmission and spread.Due to a highly dense population,low-resourced health system,lack of testing kit,and tracing,the ultimate impact could be lasting.It is highly likely that Asian LMICs were not in the center of receiving high priority by the international community in order to take effective prevention strategies.This might upsurge caseloads.Like the symptoms,mode of transmission varies largely due to a number of unknown clinical features of the virus,defining a risk group only based on some limited criteria such as age and disease with comorbidity could be risky.Recently some of the Asian LMICs launched contact tracing app.However,the study has identified several healthcare challenges in Asian LMICs such as shortage of supply of PPE,inadequate long-term care services,the socio-cultural differences which require utmost consideration.In addition,to control infection and enhance prevention,some pertinent problems need to be addressed.Based on that,some multifactorial approaches can be taken in line with international guidelines.Finally,this paper recommends developing a strong and resilient healthcare monitoring system. 展开更多
