BACKGROUND Liver fibrosis is a compensatory response during the tissue repair process in chronic liver injury,and finally leads to liver cirrhosis or even hepatocellular carcinoma.The pathogenesis of hepatic fibrosis ...BACKGROUND Liver fibrosis is a compensatory response during the tissue repair process in chronic liver injury,and finally leads to liver cirrhosis or even hepatocellular carcinoma.The pathogenesis of hepatic fibrosis is associated with the progressive accumulation of activated hepatic stellate cells(HSCs),which can transdiffer-entiate into myofibroblasts to produce an excess of the extracellular matrix(ECM).Myofibroblasts are the main source of the excessive ECM responsible for hepatic fibrosis.Therefore,activated hepatic stellate cells(aHSCs),the principal ECM producing cells in the injured liver,are a promising therapeutic target for the treatment of hepatic fibrosis.AIM To explore the effect of taurine on aHSC proliferation and the mechanisms involved.METHODS Human HSCs(LX-2)were randomly divided into five groups:Normal control group,platelet-derived growth factor-BB(PDGF-BB)(20 ng/mL)treated group,mmol/L,respectively)with PDGF-BB(20 ng/mL)treated group.Cell Counting Kit-8 method was performed to evaluate the effect of taurine on the viability of aHSCs.Enzyme-linked immunosorbent assay was used to estimate the effect of taurine on the levels of reactive oxygen species(ROS),malondialdehyde,glutathione,and iron concen-tration.Transmission electron microscopy was applied to observe the effect of taurine on the autophagosomes and ferroptosis features in aHSCs.Quantitative real-time polymerase chain reaction and Western blot analysis were performed to detect the effect of taurine on the expression ofα-SMA,Collagen I,Fibronectin 1,LC3B,ATG5,Beclin 1,PTGS2,SLC7A11,and p62.RESULTS Taurine promoted the death of aHSCs and reduced the deposition of the ECM.Treatment with taurine could alleviate autophagy in HSCs to inhibit their activation,by decreasing autophagosome formation,downregulating LC3B and Beclin 1 protein expression,and upregulating p62 protein expression.Meanwhile,treatment with taurine triggered ferroptosis and ferritinophagy to eliminate aHSCs characterized by iron overload,lipid ROS accumu-lation,glutathione depletion,and lipid peroxidation.Furthermore,bioinformatics analysis demonstrated that taurine had a direct targeting effect on nuclear receptor coactivator 4,exhibiting the best average binding affinity of-20.99 kcal/mol.CONCLUSION Taurine exerts therapeutic effects on liver fibrosis via mechanisms that involve inhibition of autophagy and trigger of ferroptosis and ferritinophagy in HSCs to eliminate aHSCs.展开更多
BACKGROUND Lung cancer is a common disease with high mortality,and psychological support is very important in the diagnosis and treatment of postoperative patients with cancer pain.AIM To explore the application effec...BACKGROUND Lung cancer is a common disease with high mortality,and psychological support is very important in the diagnosis and treatment of postoperative patients with cancer pain.AIM To explore the application effect of the narrative nursing method in postoperative lung cancer patients in the intensive care unit.METHODS A total of 120 patients diagnosed with lung cancer and experiencing cancer-related pain were randomly allocated into two groups:an observation group and a control group,each consisting of 60 cases.The control group was given routine analgesic and psychological care,while the research group applied the five-step narrative nursing method based on routine care,comparing the visual analogue scale scores,sleep status,anxiety and depression status,and quality of life of the two groups of patients before and after the intervention.RESULTS The pain scores,anxiety scores,and depression scores of the study group were lower than those of the control group after the intervention using the narrative nursing method,and the difference was statistically significant(P<0.05).CONCLUSION Using narrative nursing methods to intervene in patients with lung cancer combined with cancerous pain can help patients to correctly recognize their disease,adjust their mentality,establish confidence,alleviate patients'subjective pain feelings,and improve their adverse emotions.展开更多
[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performanc...[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performance of Sichuan white rabbits and reveal the regulatory role of miRNAs in their muscle growth and meat quality formation.[Methods]Three constructed skeletal muscle libraries of Sichuan white rabbits aged six months were sequenced by the solexa technology to identify known miRNAs,predict new miRNAs and construct an expression profile of muscle miRNAs.[Results]A total of 511 known miRNAs and 42 miRNAs were detected in 34089472 pure sequences,and the proportion of miRNAs with a length of 22 nt was the highest.The number of known miRNA sequences accounted for 71.38%of pure sequences,which was much higher than the proportion of other types of RNAs.The proportion of sequences from exons was 0.38%,indicating a low degree of mRNA degradation in the samples.Base U had the highest proportion at the first position,and the bases with the highest proportions at positions 8 and 10 were U and A,respectively.Muscle-specific miRNAs(miR-1,miR-133,and miR-206)ranked in the top 10 in terms of expression level.The number and expression levels of new miRNAs were lower than those of known miRNAs.The length distribution,base bias at different positions and expression profile characteristics of miRNAs might be related to the biological function of miRNAs in regulating muscle proliferation and differentiation and the action mechanisms with target genes.[Conclusions]The identification and expression of miRNAs in muscle tissues of Sichuan white rabbits will help to understand the complex molecular mechanisms of meat production performance and provide a theoretical basis for the functional research of miRNAs in meat rabbits.展开更多

Hence, status of epigenetics is dramatically important in maintaining of cell identity. Histone modification is another factor of epigenetic alterations during human oncogenesis. Histones, one of main components of chromatin, can be modified post-translationally. Histone tail modifications are regulated by corresponding modification enzymes. The interaction between laminin-γ2(LAMC2) and integrin-β1(ITGB1) plays a role in premetastatic niche signaling, which may induce epithelial mesenchymal transformation(EMT) and lead to metastasis.AIM To investigate the effects of alcohol on CRC metastasis from the molecular mechanism of the premetastatic niche.METHODS The interaction between LAMC2 and ITGB1 was measured by Duolink assay, and the expression levels of LAMC2, ITGB1 and focal adhesion kinase(FAK), snail, fibronectin, N-cadherin and special AT-rich sequence binding protein 1(SATB1) were measured by quantitative real-time polymerase chain reaction, immunohistochemistry and western blotting. Interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) and IL-6 levels were measured via enzyme-linked immunosorbent assay, histopathological assessment via hematoxylin eosin staining, and determination of aberrant crypt foci via methylene blue.RESULTS The lymph node metastasis rate was higher in the alcohol group than non-alcohol group. There was a significant increase in interaction signals between LAMC2 and ITGB1, and an increase in phosphorylate-FAK/FAK, snail, fibronectin, N-cadherin and SATB1, whereas E-cadherin was reduced in the alcohol group compared to the non-alcohol group in both animal and clinical samples. Serum IL-1β, TNF-α and IL-6 were higher in alcohol group than in non-alcohol group. Alcohol may promote CRC metastasis by influencing the molecular mechanism of the premetastatic niche.CONCLUSION Our study suggests that alcohol promotes EMT-mediated premetastatic niche formation of CRC by activating the early interaction between LAMC2 and ITGB1 and lead to CRC metastasis.展开更多
Salidroside is the active ingredient extracted from Rhodiola rosea,and has been reported to show protective effects in cerebral ischemia,but the exact mechanisms of neuronal protective effects are still unrevealed.In ...Salidroside is the active ingredient extracted from Rhodiola rosea,and has been reported to show protective effects in cerebral ischemia,but the exact mechanisms of neuronal protective effects are still unrevealed.In this study,the protective effects of salidroside(1 jimol/L)in ameliorating neuronal injuries induced by oxygen-glucose deprivation(OGD),which is a classical model of cerebral ischemia,were clarified.The results showed that after 8 h of OGD,the mouse hippocampal neuronal cell line HT22 cells showed increased cell death,accompanied with mitochondrial fragmentation and augmented mitophagy.However,the cell viability of HT22 cells showed significant restoration after salidroside treatment.Mitochondrial morphology and mitochondrial function were effectively preserved by salidroside treatment.The protective effects of salidroside were further related to the prevention of mitochondrial over-fission.The results showed that mTOR could be recruited to the mitochondria after salidroside treatment,which might be responsible for inhibiting excessive mitophagy caused by OGD.Thus,salidroside was shown to play a protective role in reducing neuronal death under OGD by safeguarding mitochondrial function,which may provide evidence for further translational studies of salidroside in ischemic diseases.展开更多
BACKGROUND Extracellular matrix(ECM)remodeling and stiffening,which are correlated with tumor malignancy,drives tumor development.However,the relationship between ECM remodeling and rat experimental model of 1,2-dimet...BACKGROUND Extracellular matrix(ECM)remodeling and stiffening,which are correlated with tumor malignancy,drives tumor development.However,the relationship between ECM remodeling and rat experimental model of 1,2-dimethylhyrazine(DMH)-induced colorectal cancer(CRC)imposed by cold and capsaicin exposure remains unclear.AIM To explore the effects of cold exposure and capsaicin on ECM remodeling and ECM enzymes in DMH-induced CRC.METHODS For histopathological analysis,the sections of colon tissues were stained with hematoxylin and eosin,Masson’s trichrome,Picrosirius red,and Weigert’s Resorcin-Fuchsin to observe the remodeling of collagen and elastin.Additionally,the protein expression level of type I collagen(COL I),type 3 collagen(COL III0,elastin,matrix metalloproteinase(MMP)1,MMP2,MMP9,and tissue-specific matrix metalloproteinase 1(TIMP1)was assessed by immunohistochemistry.The messenger RNA(mRNA)levels of COL I,COL III,elastin,and lysyl oxidase-like-2(LOXL2)in the colon tissues of rats was measured by reverse-transcriptase quantitative polymerase chain reaction.RESULTS Although no differences were observed in the proportion of adenomas,a trend towards the increase of invasive tumors was observed in the cold and capsaicin group.The cold exposure group had a metastasis rate compared with the other groups.Additionally,abnormal accumulation of both collagen and elastin was observed in the cold exposure and capsaicin group.Specifically,collagen quantitative analysis showed increased length,width,angle,and straightness compared with the DMH group.Collagen deposition and straightness were significantly increased in the cold exposure group compared with the capsaicin group.Cold exposure and capsaicin significantly increased the protein levels of COL I,elastin,and LOXL2 along with increases in their mRNA levels in the colon tissues compared with the DMH group,while COL III did not show a significant difference.Furthermore,in immunohistochemical evaluations,MMP1,MMP2,MMP9,and TIMP1 staining increased in the cold exposure and capsaicin group compared with the DMH group.CONCLUSION These results suggest that chronic cold and capsaicin exposure further increased the deposition of collagen and elastin in the colonic tissue.Increased COL I and elastin mRNA and protein levels expression may account for the enhanced ECM remodel and stiffness variations of colon tissue.The upregulated expression of the LOXL2 and physiological imbalance between MMP/TIMP activation and deactivation could contribute to the progression of the CRC resulting from cold and capsaicin exposure.展开更多

A new positioning a...Limited by the sampling capacity of the mobile devices, many real-time indoor location systems have such problems as low accuracy, large variance, and non-smooth movement of the estimated position. A new positioning algorithm and a new processing method for sampled data are proposed. Firstly, a positioning algorithm is designed based on the cluster-based nearest neighbour or probability. Secondly, a weighted average method with sliding window is used to process the sampled data as to overcome the mobile devices’ weak capability of signal sampling. Experimental results show that, for the general mobile devices, the accuracy of indoor position estimation increases from 56.5% to 76.6% for a 2-meter precision, and from 77.4% to 90.9% for a 3-meter precision. Therefore, the proposed methods can significantly and stably improve the positioning accuracy.展开更多

INTRODUCTION In the Earth’s history,the Late Ediacaran to Early Cambrian period witnessed a series of profound changes in both Earth’s interior and surficial systems which may have made this planet more habitable fo...INTRODUCTION In the Earth’s history,the Late Ediacaran to Early Cambrian period witnessed a series of profound changes in both Earth’s interior and surficial systems which may have made this planet more habitable for the first radiation of early animals(known as the global occurrence of Ediacaran biota;Xiao and Narbonne,2020).Among those changes,the Late Ediacaran glaciation,soon followed and accompanied by the early complex life of Ediacaran biota,attracted much attention of scientists and may have provided a good opportunity to investigate the interactions between the Earth’s surficial environment and the biosphere(e.g.,Xu et al.,2024;Zhang et al.,2023;Hoffman et al.,2017).To achieve this goal,the nature of the glaciation itself needs to be understood first.Until now,32 or more Ediacaran glacial deposits have been documented on almost all the existing continents(Niu et al.,2024;Wang et al.,2023a,b,c),showing a similar distribution pattern to the two Cryogenian(Sturtian and Marinoan)Snowball Earth glaciations if plotted in one single paleogeographic map(Figure 1;Niu et al.,2024;Li et al.,2013).But further compared to the Cryogenian Snowball Earth glaciations,the Late Ediacaran glaciation has some fundamentally different characteristics:(1)as shown in Figure 1,most glacial deposits are not stratigraphically overlain by the typical cap carbonate while it usually happened in the aftermath of a Snowball Earth glaciation and represented an abrupt climatic switch from extreme icehouse(global freezing)to greenhouse(global warming)condition(Hoffman et al.,2017).展开更多
Background:The ongoing coronavirus disease 2019(COVID-19)pandemic has put radiologists at a higher risk of infection during the computer tomography(CT)examination for the patients.To help settling these problems,we ad...Background:The ongoing coronavirus disease 2019(COVID-19)pandemic has put radiologists at a higher risk of infection during the computer tomography(CT)examination for the patients.To help settling these problems,we adopted a remote-enabled and automated contactless imaging workflow for CT examination by the combination of intelligent guided robot and automatic positioning technology to reduce the potential exposure of radiologists to 2019 novel coronavirus(2019-nCoV)infection and to increase the examination efficiency,patient scanning accuracy and better image quality in chest CT imaging.Methods:From February 10 to April 12,2020,adult COVID-19 patients underwent chest CT examinations on a CT scanner using the same scan protocol except with the conventional imaging workflow(CW group)or an automatic contactless imaging workflow(AW group)in Wuhan Leishenshan Hospital(China)were retrospectively and prospectively enrolled in this study.The total examination time in two groups was recorded and compared.The patient compliance of breath holding,positioning accuracy,image noise and signal-to-noise ratio(SNR)were assessed by three experienced radiologists and compared between the two groups.Results:Compared with the CW group,the total positioning time of the AW group was reduced((118.0±20.0)s vs.(129.0±29.0)s,P=0.001),the proportion of scanning accuracy was higher(98%vs.93%),and the lung length had a significant difference((0.90±1.24)cm vs.(1.16±1.49)cm,P=0.009).For the lesions located in the pulmonary centrilobular and subpleural regions,the image noise in the AW group was significantly lower than that in the CW group(centrilobular region:(140.4±78.6)HU vs.(153.8±72.7)HU,P=0.028;subpleural region:(140.6±80.8)HU vs.(159.4±82.7)HU,P=0.010).For the lesions located in the peripheral,centrilobular and subpleural regions,SNR was significantly higher in the AW group than in the CW group(centrilobular region:6.6±4.3 vs.4.9±3.7,P=0.006;subpleural region:6.4±4.4 vs.4.8±4.0,P<0.001).Conclusions:The automatic contactless imaging workflow using intelligent guided robot and automatic posi-tioning technology allows for reducing the examination time and improving the patient’s compliance of breath holding,positioning accuracy and image quality in chest CT imaging.展开更多

Mitophagy is regulated by the post-translational modification of mitophagy receptors. Fun14 domain-containing protein 1 (FUNDC1) was reported to be a new receptor for hypoxia-induced mitophagy in mammalian cells and interact with micro-tubule-associated protein light chain 3 beta (LC3B) through its LC3 interaction region (LIR). Moreover, the phosphorylation modification of FUNDC1 affects its binding affinity for LC3B and regulates selective mitophagy. However, the structural basis of this regulation mechanism remains unclear. Here, we present the crystal structure of LC3B in complex with a FUNDCI LIR peptide phosphorylated at Ser17 (pS17), demonstrating the key residues of LC3B for the specific recognition of the phosphorylated or dephosphorylated FUNDC1. Intriguingly, the side chain of LC3B Lys49 shifts remarkably and forms a hydrogen bond and electrostatic interaction with the phosphate group of FUNDC1 pS17. Alternatively, phosphorylated Tyr18 (PY18) and Ser13 (PS13) in FUNDC1 significantly obstruct their interaction with the hydrophobic pocket and Arg10 of LC3B, respectively. Structural observations are further validated by mutation and isothermal titration calorimetry (ITC) assays. Therefore, our structural and biochemical results reveal a working model for thespecific recognition of FUNDCI by LC3B and imply that the reversible phosphorylation modification of mitophagy receptors may be a switch for selective mitophagy.展开更多

文摘[Objectives]miRNAs play an important role in the proliferation and differentiation of different myoblasts.This study was conducted to elucidate the complex genetic mechanisms that affect the meat production performance of Sichuan white rabbits and reveal the regulatory role of miRNAs in their muscle growth and meat quality formation.[Methods]Three constructed skeletal muscle libraries of Sichuan white rabbits aged six months were sequenced by the solexa technology to identify known miRNAs,predict new miRNAs and construct an expression profile of muscle miRNAs.[Results]A total of 511 known miRNAs and 42 miRNAs were detected in 34089472 pure sequences,and the proportion of miRNAs with a length of 22 nt was the highest.The number of known miRNA sequences accounted for 71.38%of pure sequences,which was much higher than the proportion of other types of RNAs.The proportion of sequences from exons was 0.38%,indicating a low degree of mRNA degradation in the samples.Base U had the highest proportion at the first position,and the bases with the highest proportions at positions 8 and 10 were U and A,respectively.Muscle-specific miRNAs(miR-1,miR-133,and miR-206)ranked in the top 10 in terms of expression level.The number and expression levels of new miRNAs were lower than those of known miRNAs.The length distribution,base bias at different positions and expression profile characteristics of miRNAs might be related to the biological function of miRNAs in regulating muscle proliferation and differentiation and the action mechanisms with target genes.[Conclusions]The identification and expression of miRNAs in muscle tissues of Sichuan white rabbits will help to understand the complex molecular mechanisms of meat production performance and provide a theoretical basis for the functional research of miRNAs in meat rabbits.

文摘Formation of malignant tumor originating from normal healthy cell is a multistep process including genetic and epigenetic lesions. Previous studies of cell line model systems displayed that early important epigenetic events happened in stepwise fashion prior to cell immortalization. Once these epigenetic alterations are integrated into chromatin, they will perform vertical propagation through cell subculture. Hence, status of epigenetics is dramatically important in maintaining of cell identity. Histone modification is another factor of epigenetic alterations during human oncogenesis. Histones, one of main components of chromatin, can be modified post-translationally. Histone tail modifications are regulated by corresponding modification enzymes. This review focuses on the description of relationship between the main sites of histone modification and oncogenesis.

文摘BACKGROUND Colorectal cancer(CRC) is a common malignant tumor. Alcohol consumption is positively correlated with CRC malignant metastasis;however, the mechanism is unclear. The interaction between laminin-γ2(LAMC2) and integrin-β1(ITGB1) plays a role in premetastatic niche signaling, which may induce epithelial mesenchymal transformation(EMT) and lead to metastasis.AIM To investigate the effects of alcohol on CRC metastasis from the molecular mechanism of the premetastatic niche.METHODS The interaction between LAMC2 and ITGB1 was measured by Duolink assay, and the expression levels of LAMC2, ITGB1 and focal adhesion kinase(FAK), snail, fibronectin, N-cadherin and special AT-rich sequence binding protein 1(SATB1) were measured by quantitative real-time polymerase chain reaction, immunohistochemistry and western blotting. Interleukin-1β(IL-1β), tumor necrosis factor-α(TNF-α) and IL-6 levels were measured via enzyme-linked immunosorbent assay, histopathological assessment via hematoxylin eosin staining, and determination of aberrant crypt foci via methylene blue.RESULTS The lymph node metastasis rate was higher in the alcohol group than non-alcohol group. There was a significant increase in interaction signals between LAMC2 and ITGB1, and an increase in phosphorylate-FAK/FAK, snail, fibronectin, N-cadherin and SATB1, whereas E-cadherin was reduced in the alcohol group compared to the non-alcohol group in both animal and clinical samples. Serum IL-1β, TNF-α and IL-6 were higher in alcohol group than in non-alcohol group. Alcohol may promote CRC metastasis by influencing the molecular mechanism of the premetastatic niche.CONCLUSION Our study suggests that alcohol promotes EMT-mediated premetastatic niche formation of CRC by activating the early interaction between LAMC2 and ITGB1 and lead to CRC metastasis.

文摘Salidroside is the active ingredient extracted from Rhodiola rosea,and has been reported to show protective effects in cerebral ischemia,but the exact mechanisms of neuronal protective effects are still unrevealed.In this study,the protective effects of salidroside(1 jimol/L)in ameliorating neuronal injuries induced by oxygen-glucose deprivation(OGD),which is a classical model of cerebral ischemia,were clarified.The results showed that after 8 h of OGD,the mouse hippocampal neuronal cell line HT22 cells showed increased cell death,accompanied with mitochondrial fragmentation and augmented mitophagy.However,the cell viability of HT22 cells showed significant restoration after salidroside treatment.Mitochondrial morphology and mitochondrial function were effectively preserved by salidroside treatment.The protective effects of salidroside were further related to the prevention of mitochondrial over-fission.The results showed that mTOR could be recruited to the mitochondria after salidroside treatment,which might be responsible for inhibiting excessive mitophagy caused by OGD.Thus,salidroside was shown to play a protective role in reducing neuronal death under OGD by safeguarding mitochondrial function,which may provide evidence for further translational studies of salidroside in ischemic diseases.

文摘BACKGROUND Extracellular matrix(ECM)remodeling and stiffening,which are correlated with tumor malignancy,drives tumor development.However,the relationship between ECM remodeling and rat experimental model of 1,2-dimethylhyrazine(DMH)-induced colorectal cancer(CRC)imposed by cold and capsaicin exposure remains unclear.AIM To explore the effects of cold exposure and capsaicin on ECM remodeling and ECM enzymes in DMH-induced CRC.METHODS For histopathological analysis,the sections of colon tissues were stained with hematoxylin and eosin,Masson’s trichrome,Picrosirius red,and Weigert’s Resorcin-Fuchsin to observe the remodeling of collagen and elastin.Additionally,the protein expression level of type I collagen(COL I),type 3 collagen(COL III0,elastin,matrix metalloproteinase(MMP)1,MMP2,MMP9,and tissue-specific matrix metalloproteinase 1(TIMP1)was assessed by immunohistochemistry.The messenger RNA(mRNA)levels of COL I,COL III,elastin,and lysyl oxidase-like-2(LOXL2)in the colon tissues of rats was measured by reverse-transcriptase quantitative polymerase chain reaction.RESULTS Although no differences were observed in the proportion of adenomas,a trend towards the increase of invasive tumors was observed in the cold and capsaicin group.The cold exposure group had a metastasis rate compared with the other groups.Additionally,abnormal accumulation of both collagen and elastin was observed in the cold exposure and capsaicin group.Specifically,collagen quantitative analysis showed increased length,width,angle,and straightness compared with the DMH group.Collagen deposition and straightness were significantly increased in the cold exposure group compared with the capsaicin group.Cold exposure and capsaicin significantly increased the protein levels of COL I,elastin,and LOXL2 along with increases in their mRNA levels in the colon tissues compared with the DMH group,while COL III did not show a significant difference.Furthermore,in immunohistochemical evaluations,MMP1,MMP2,MMP9,and TIMP1 staining increased in the cold exposure and capsaicin group compared with the DMH group.CONCLUSION These results suggest that chronic cold and capsaicin exposure further increased the deposition of collagen and elastin in the colonic tissue.Increased COL I and elastin mRNA and protein levels expression may account for the enhanced ECM remodel and stiffness variations of colon tissue.The upregulated expression of the LOXL2 and physiological imbalance between MMP/TIMP activation and deactivation could contribute to the progression of the CRC resulting from cold and capsaicin exposure.

文摘A counterbalance between immune cells and tumour cells is key to fighting tumours,and immune escape is an important mechanism for the survival of tumour cells in the body.Tumor cells and their cytokines impair the activity of T cells,NK cells,macrophages and other immune cells through various ways,and change the expression of their own surface antigens so as to avoid the clearance of the immune system.Changes in major histocompatibility complex molecules,high expression of programmed death-ligand 1,and the presence of immunosuppressive cells in the tumor microenvironment(TME)are main means by which tumors impair the function of immune cells.During the development of tumours of the digestive system,different mechanisms acting on tumour cells,the TME,and immune cells lead to immune escape and promote tumour progression.In this paper,the mechanisms of immune escape in tumour cells of the digestive system are reviewed to provide a theoretical basis for the immunotherapy of gastrointestinal tumours.
文摘Limited by the sampling capacity of the mobile devices, many real-time indoor location systems have such problems as low accuracy, large variance, and non-smooth movement of the estimated position. A new positioning algorithm and a new processing method for sampled data are proposed. Firstly, a positioning algorithm is designed based on the cluster-based nearest neighbour or probability. Secondly, a weighted average method with sliding window is used to process the sampled data as to overcome the mobile devices’ weak capability of signal sampling. Experimental results show that, for the general mobile devices, the accuracy of indoor position estimation increases from 56.5% to 76.6% for a 2-meter precision, and from 77.4% to 90.9% for a 3-meter precision. Therefore, the proposed methods can significantly and stably improve the positioning accuracy.
文摘Naturally, like the web, integrated software systems in Internet will have to be distributed and heterogeneous. To im-prove the interoperability of services for SAAS, it is crucial to build requirements semantics that will cross the entire lifecycle of services especially on requirements stage. In this paper, a requirements semantics interoperability extend-ing approach called Connecting Ontologies (CO) that will act as semantics information carrier designing to facilitate the requirements identification and services composition is proposed. Semantic measurement of Chinese scenario is explored. By adopting the approach, a series of tools support for transport domain are developed and applied based on CO and DPO (Domain Problem Ontology) to enforce requirements engineering of networked software efficiently.

文摘INTRODUCTION In the Earth’s history,the Late Ediacaran to Early Cambrian period witnessed a series of profound changes in both Earth’s interior and surficial systems which may have made this planet more habitable for the first radiation of early animals(known as the global occurrence of Ediacaran biota;Xiao and Narbonne,2020).Among those changes,the Late Ediacaran glaciation,soon followed and accompanied by the early complex life of Ediacaran biota,attracted much attention of scientists and may have provided a good opportunity to investigate the interactions between the Earth’s surficial environment and the biosphere(e.g.,Xu et al.,2024;Zhang et al.,2023;Hoffman et al.,2017).To achieve this goal,the nature of the glaciation itself needs to be understood first.Until now,32 or more Ediacaran glacial deposits have been documented on almost all the existing continents(Niu et al.,2024;Wang et al.,2023a,b,c),showing a similar distribution pattern to the two Cryogenian(Sturtian and Marinoan)Snowball Earth glaciations if plotted in one single paleogeographic map(Figure 1;Niu et al.,2024;Li et al.,2013).But further compared to the Cryogenian Snowball Earth glaciations,the Late Ediacaran glaciation has some fundamentally different characteristics:(1)as shown in Figure 1,most glacial deposits are not stratigraphically overlain by the typical cap carbonate while it usually happened in the aftermath of a Snowball Earth glaciation and represented an abrupt climatic switch from extreme icehouse(global freezing)to greenhouse(global warming)condition(Hoffman et al.,2017).

文摘Background:The ongoing coronavirus disease 2019(COVID-19)pandemic has put radiologists at a higher risk of infection during the computer tomography(CT)examination for the patients.To help settling these problems,we adopted a remote-enabled and automated contactless imaging workflow for CT examination by the combination of intelligent guided robot and automatic positioning technology to reduce the potential exposure of radiologists to 2019 novel coronavirus(2019-nCoV)infection and to increase the examination efficiency,patient scanning accuracy and better image quality in chest CT imaging.Methods:From February 10 to April 12,2020,adult COVID-19 patients underwent chest CT examinations on a CT scanner using the same scan protocol except with the conventional imaging workflow(CW group)or an automatic contactless imaging workflow(AW group)in Wuhan Leishenshan Hospital(China)were retrospectively and prospectively enrolled in this study.The total examination time in two groups was recorded and compared.The patient compliance of breath holding,positioning accuracy,image noise and signal-to-noise ratio(SNR)were assessed by three experienced radiologists and compared between the two groups.Results:Compared with the CW group,the total positioning time of the AW group was reduced((118.0±20.0)s vs.(129.0±29.0)s,P=0.001),the proportion of scanning accuracy was higher(98%vs.93%),and the lung length had a significant difference((0.90±1.24)cm vs.(1.16±1.49)cm,P=0.009).For the lesions located in the pulmonary centrilobular and subpleural regions,the image noise in the AW group was significantly lower than that in the CW group(centrilobular region:(140.4±78.6)HU vs.(153.8±72.7)HU,P=0.028;subpleural region:(140.6±80.8)HU vs.(159.4±82.7)HU,P=0.010).For the lesions located in the peripheral,centrilobular and subpleural regions,SNR was significantly higher in the AW group than in the CW group(centrilobular region:6.6±4.3 vs.4.9±3.7,P=0.006;subpleural region:6.4±4.4 vs.4.8±4.0,P<0.001).Conclusions:The automatic contactless imaging workflow using intelligent guided robot and automatic posi-tioning technology allows for reducing the examination time and improving the patient’s compliance of breath holding,positioning accuracy and image quality in chest CT imaging.

文摘Mitophagy is an essential intracellular process that eliminates dysfunctional mitochondria and maintains cellular homeostasis. Mitophagy is regulated by the post-translational modification of mitophagy receptors. Fun14 domain-containing protein 1 (FUNDC1) was reported to be a new receptor for hypoxia-induced mitophagy in mammalian cells and interact with micro-tubule-associated protein light chain 3 beta (LC3B) through its LC3 interaction region (LIR). Moreover, the phosphorylation modification of FUNDC1 affects its binding affinity for LC3B and regulates selective mitophagy. However, the structural basis of this regulation mechanism remains unclear. Here, we present the crystal structure of LC3B in complex with a FUNDCI LIR peptide phosphorylated at Ser17 (pS17), demonstrating the key residues of LC3B for the specific recognition of the phosphorylated or dephosphorylated FUNDC1. Intriguingly, the side chain of LC3B Lys49 shifts remarkably and forms a hydrogen bond and electrostatic interaction with the phosphate group of FUNDC1 pS17. Alternatively, phosphorylated Tyr18 (PY18) and Ser13 (PS13) in FUNDC1 significantly obstruct their interaction with the hydrophobic pocket and Arg10 of LC3B, respectively. Structural observations are further validated by mutation and isothermal titration calorimetry (ITC) assays. Therefore, our structural and biochemical results reveal a working model for thespecific recognition of FUNDCI by LC3B and imply that the reversible phosphorylation modification of mitophagy receptors may be a switch for selective mitophagy.
