目的探讨紫檀芪(pterostilbene,PTE)对慢性不可预知军事应激(chronic unpredictable military stress,CUMS)暴露小鼠记忆功能的影响及其作用机制。方法将36只健康7~8周龄雄性C57BL/6J小鼠随机分为对照组(CON)、慢性不可预知军事应激暴露...目的探讨紫檀芪(pterostilbene,PTE)对慢性不可预知军事应激(chronic unpredictable military stress,CUMS)暴露小鼠记忆功能的影响及其作用机制。方法将36只健康7~8周龄雄性C57BL/6J小鼠随机分为对照组(CON)、慢性不可预知军事应激暴露组(CUMS)和紫檀芪干预组(PTE)。CUMS组及PTE组小鼠连续30d暴露于不可预知军事应激条件下,PTE组小鼠每日给予紫檀芪(100mg/kg.BW)灌胃干预。暴露期间,记录小鼠体质量及进食量变化;暴露结束后,检测各组小鼠血清皮质酮水平,水迷宫记忆测试实验检测小鼠空间记忆功能,透射电镜观察海马神经元超微结构,SeahorseXFHSMini分析仪检测离体小鼠海马线粒体呼吸功能,RT-qPCR检测海马mtDNA相对拷贝数及Pgc-1α、Tfam基因表达,以及Western Blot检测海马PGC-1α蛋白表达。结果与CON组相比,CUMS组小鼠体质量及进食量显著下降(P<0.05),血清皮质酮含量升高(P<0.0001),PTE组小鼠较CUMS组小鼠体重及进食量下降减少(P<0.05),血清皮质酮升高程度减低(P<0.001);水迷宫探查实验发现,与CON组相比,CUMS组小鼠对平台的记忆准确性降低,而PTE组小鼠表现优于CUMS组。透射电镜显示CUMS暴露后,小鼠海马神经元脂褐素增多、线粒体肿胀和嵴消失,而PTE组脂褐素较CUMS组少,线粒体肿胀较轻。CUMS组离体海马线粒体基础呼吸(P<0.05)、最大呼吸(P<0.001)、储备呼吸(P<0.01)较CON组显著下降;而PTE组最大呼吸(P<0.05)、ATP生成能力(P<0.05)及呼吸控制率(P<0.05)较CUMS组明显改善。同时,发现CUMS组海马mtDNA拷贝数降低(P<0.01),Pgc-1α(P<0.0001)、Tfam(P<0.01)基因相对表达量及PGC-1α蛋白(P<0.01)含量降低,PTE组较CUMS组海马mtDNA拷贝数升高(P<0.05),Pgc-1α、Tfam基因相对表达量及PGC-1α蛋白含量升高(P<0.05)。结论紫檀芪可改善CUMS暴露小鼠记忆功能,可能与其有效遏制CUMS暴露所致PGC-1α表达下降,进而减轻海马线粒体结构及功能损伤有关。展开更多

This study aim...Background Recent studies have revealed that the reason for the low surgical resection rate of pancreatic carcinoma partly lies in its biological behavior, which is characterized by neural infiltration. This study aimed to investigate the clinical significance of radical pancreatoduodenectomy combined with retroperitoneal nerve, lymph, and soft-tissue dissection for carcinoma of the pancreatic head.Methods Forty-six patients with pancreatic head cancer were treated in our hospital from 1995 to 2005. The patients were divided into two groups: radical pancreatoduodenectomy combined with retroperitoneal nerve, lymph and soft-tissue dissection (group A, n=25) and routine Whipple's operation (group B, n=-21). There were no significant differences between the two groups in relation to age, gender and preoperative risk factors, and perioperative conditions, patholoclical data and survival rates were studied.Results There were no significant differences in tumor size, surgical procedure time, postoperative complications, and time of hospitalization. However, the number and positive rate of resected lymph nodes in group A were significantly higher than those in group B (P〈0.05). The 1- and 3-year survival rate in group A were 80% and 53%, respectively, which was higher than those in group B (P〈0.05). There were significant differences in the survival rates between patients with and without nerve infiltration in group A (P〈0.05).Conclusions Radical pancreatoduodenectomy combined with retroperitoneal nerve, lymph and soft-tissue dissection, can effectively remove the lymph and nerve tissues that were infiltrated by tumor. Meanwhile, this method can reduce the local recurrence rate so as to improve the long-term survival of patients.展开更多
