双硫死亡相关基因对结直肠癌预后和药物敏感性的预测价值

目的通过生物信息学方法建立基于双硫死亡相关基因表达量预测结直肠癌(colorectal cancer,CRC)预后和药物敏感性的评分模型,并结合CRC患者来源类器官(CRC patient-derived organoids,CRC-PDOs)验证。方法利用NMF(non-negative Matrix Fa... 目的通过生物信息学方法建立基于双硫死亡相关基因表达量预测结直肠癌(colorectal cancer,CRC)预后和药物敏感性的评分模型,并结合CRC患者来源类器官(CRC patient-derived organoids,CRC-PDOs)验证。方法利用NMF(non-negative Matrix Factorization)算法、Cox和LASSO回归分析明确对CRC预后有预测价值的双硫死亡相关基因,构建双硫死亡相关风险评分公式,结合GO(Gene Ontology)和KEGG(Kyoto Encyclopedia of Genes and Genomes)分析不同集群之间的差异基因及富集通路。通过GDSC数据库预测不同分组CRC患者对化疗药物的敏感性,并利用CRC-PDOs进行验证。结果NMF算法结果显示基于双硫死亡相关基因可建立CRC患者分群,COX回归分析显示LRPPRC和SLC7A11是能够预测CRC患者预后的潜在基因(P=0.047、0.033),CRC患者肿瘤中的SLC7A11低表达或LRPPRC高表达与总生存率(overall survival,OS)显著相关(P=0.004、0.003)。基于LASSO回归分析,双硫死亡风险评分公式为:Risk score=LRPPRC×(-0.6705)+SLC7A11×0.3112,且能将GSE161158数据集分群为高风险和低风险组。双硫死亡高低危群集间有125个差异表达基因(differentially expressed genes,DEGs),结合GO和KEGG结果,高危群集患者的上调基因主要富集于免疫调节,如白细胞趋化性、粒细胞迁移和Toll样受体结合。低风险集群的特征是与硫化物代谢和运输相关的途径,如硫化物跨膜转运蛋白活性。基于GDSC数据库,双硫死亡相关的双基因表达水平可预测化疗药物敏感性,化疗药物(伊立替康)对于CRC-PDOs的生长抑制结果提示患者的结直肠癌组织中SLC7A11和LRPPRC的相对表达量与其类器官映射疗效呈线性相关(P=0.007、0.040)。结论根据生物信息学分析和CRC-PDOs实验结果,双硫死亡相关评分能够预测结直肠癌患者预后和药物敏感性,具有潜在的临床应用前景。 展开更多

45岁以下青年发生急性冠脉综合征的影响因素

目的:探讨血尿酸(UA)水平对45岁以下急性冠状动脉综合征(ACS)患者的临床意义。方法:选择2020年1月—2022年1月福建医科大学附属泉州第一医院收治的因“胸闷、胸痛”入院的可疑ACS患者224例为研究对象,根据诊断分为ACS组与非ACS组,比较两... 目的:探讨血尿酸(UA)水平对45岁以下急性冠状动脉综合征(ACS)患者的临床意义。方法:选择2020年1月—2022年1月福建医科大学附属泉州第一医院收治的因“胸闷、胸痛”入院的可疑ACS患者224例为研究对象,根据诊断分为ACS组与非ACS组,比较两组UA及其他指标的差异,运用Logistic回归法筛选ACS的主要危险因素;经KruslalWallis秩和检验分析冠状动脉病变支数与尿酸水平相关性。结果:ACS组吸烟、饮酒、高血压、高脂血症、UA、总胆固醇、低密度脂蛋白胆固醇及Gensini积分明显高于非ACS组,差异有统计学意义(P<0.05);而两组血小板、血小板分布宽度、甘油三酯、高密度脂蛋白胆固醇、尿素氮、肌酐比较,差异无统计学意义(P>0.05)。Logistic回归分析结果显示高尿酸是发生ACS的重要危险因素;秩和检验显示,冠状动脉病变支数越多,尿酸水平越高,差异有统计学意义(Z=12.872,P<0.001)。结论:高尿酸血症是青年ACS重要危险因素,冠状动脉病变支数与尿酸的呈中等强度相关,可为临床治疗提供参考。 展开更多

为了研究绿豆蔻精油的化学成分以及抑菌活性,本文采用水蒸气盐析辅助蒸馏法提取绿豆蔻精油,通过气相色谱-质谱联用法(GC-MS)对绿豆蔻精油成分进行分析;并以枯草芽孢杆菌、金黄色葡萄球菌、大肠杆菌、沙门氏菌为供菌种,测定绿豆蔻精油的... 为了研究绿豆蔻精油的化学成分以及抑菌活性,本文采用水蒸气盐析辅助蒸馏法提取绿豆蔻精油,通过气相色谱-质谱联用法(GC-MS)对绿豆蔻精油成分进行分析;并以枯草芽孢杆菌、金黄色葡萄球菌、大肠杆菌、沙门氏菌为供菌种,测定绿豆蔻精油的抑菌活性。结果表明,绿豆蔻的最佳提取工艺条件为:Na Cl浓度为5%,液料比为12∶1(m L∶g),提取时间为3.0 h,在此条件下,精油的提取率最高可达5.6%;对绿豆蔻精油进行GC-MS分析,检测出20种化合物,其中萜类化合物18种;进一步的抑菌活性研究显示,绿豆蔻精油对金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌、沙门氏菌均有良好的抑菌作用,其中对大肠杆菌的抑菌效果最好。 展开更多

A novel holmium complex [Ho(HIA)_2(H_2O)_4(NO_3)](NO_3)_2(1, HIA = isonicotinic acid) has been synthesized through hydrothermal reactions and characterized by single-crystal X-ray diffraction. Complex 1 crystallizes in the C2/c space group of monoclinic system: a = 14.4797(7), b = 12.4768(2), c = 13.3471(5) ?, β = 118.690(4)°, V = 2115.26(13) ?~3, C_(12)H_(16)HoN_5O_(17), Mr = 667.23, Z = 4, Dc = 2.095 g/cm^3, μ(Mo Kα) = 3.838 mm^(–1) and F(000) = 1304. The crystal structure of 1 is characterized by an isolated structure. Solid-state photoluminescence experiment uncovers that it shows yellow light emission. The emission bands are originated from the characteristic emission of the 4 f electrons intrashell transition of the ~5S_2 → ~5I_8 and ~5F_5 → ~5I_8 of the Ho^(3+) ions. Energy transfer mechanism is explained by the energy level diagram of the Ho3+ ion and the isonicotinic acid ligand. It has remarkable CIE chromaticity coordinates of(0.4929, 0.4632), so it may be a promising color converter for lighting and displays. 展开更多

Complex 1 crystallizes in the C2/c space group of monoclinic system with a = 24.2347(5), b = 20.8342(6), c = 15.3206(3) ?, β = 128.257(2)°, V = 6074.3(2) ?3, C36H41Cl13Hg4N6O20Tb2, Mr = 2458.80, Z = 4, Dc = 2.689 g/cm3, μ(Mo Kα) = 13.014 mm–1 and F(000) = 4520. The crystal structure of 1 is characterized by a one-dimensional(1-D) chain-like structure. Solid-state UV/Vis diffuse reflectance spectrum reveals the existence of a wide optical band gap of 3.36 eV. Solid-state photoluminescence experiment uncovers that it shows reddish brown upconversion emission. The emission bands are originated from the characteristic emission of the 4 f electrons intrashell transition of the 5D4 → 7 FJ(J = 6, 5, 4) of the Tb3+ ions. Energy transfer mechanism is explained by the energy level diagram of the Tb3+ ion and the isonicotinic acid ligand. It shows a remarkable CIE chromaticity coordinates(0.4158, 0.4005). 展开更多

目的了解泉州市乙型病毒性肝炎的流行病学特征,为制定乙肝防控策略提供科学依据。方法对泉州市2004~2019年乙肝流行病学资料进行描述性统计分析。结果泉州市2004~2019年累计报告158 743例乙肝病例,年均发病率为122.73/10万,死亡32例,年... 目的了解泉州市乙型病毒性肝炎的流行病学特征,为制定乙肝防控策略提供科学依据。方法对泉州市2004~2019年乙肝流行病学资料进行描述性统计分析。结果泉州市2004~2019年累计报告158 743例乙肝病例,年均发病率为122.73/10万,死亡32例,年均死亡率为0.025/10万,15岁以下乙肝报告年均发病率逐渐下降。年均发病率前3位的地区为泉港区、丰泽区、惠安县,年均发病率分别为210.30/10万、141.47/10万、137.52/10万,沿海发病高于山区(P<0.01);男性发病高于女性(P<0.01),男女发病之比为2.54∶1;病例主要集中在20~44岁(62.41%),以农民、工人、家政和待业人员为主(65.80%)。结论泉州市2004~2019年乙肝发病率总体呈下降趋势。 展开更多

An unprecedented 4f-5d material(La6Hg5Br26)[4(HgBr2)](2Br)(1)was syn-thesized by hydrothermal reactions and structurally characterized by single-crystal X-ray diffraction method.Complex 1 crystallizes in the Pbam space group of orthorhombic system with a=13.0980(10),b=13.6650(9),c=28.010(2)?,V=5013.4(6)?^3,Br36Hg9La6,Mr=5515.53,Z=2,Dc=3.613 g/cm^3,μ(MoKα)=29.457 mm^–1 and F(000)=4598.Compound 1 is characteristic of a two-dimensional(2D)layered structure.The photoluminescent measurements with solid-state samples reveal that compound 1 has a strong emission in the green region of light spectrum.It has remarkable CIE chromaticity coordinates of(0.2499,0.3589).A wide optical band gap of 3.41 eV is discovered by the solid-state UV/vis diffuse reflectance spectrum.The variable-temperature magnetic susceptibility obeys the Curie-Weiss law(cm=c/(T–q))with C=2.48 K and a negative Weiss constantθ=–169.07 K as revealed by the magnetic measurements,indicating the existence of an antiferromagnetic interaction in compound 1. 展开更多
