Methods: The indirect immun-ofluorescence method with anti-GCNF antiserum was used to investigate the GCNF expression in mice at day 8, 10, 14, 17, 20, 28, 35, 70, and 420 after birth and in sperm before and after capacitation. Results: With the proceeding of spermatogenesis, GCNF was first detected in the nuclei of spermatogonia and a few early stage primary sperma-tocytes at day 8, which was increased gradually at day 10 to 14 inclusive. From day 17 to day 20, the GCNF was concentrated in round spermatids, while both spermatogonia and early stage primary spermatocytes became GCNF negative. From day 28 until day 420, strong GCNF expression was shown in round spermatids and pachytene spermatocytes, while spermatogonia, early primary spermatocytes and elongating spermatids were all GCNF negative. In addition, it was also found that GCNF was localized on the acrosomal cap region of spermatozoa and there was a big change in GCNF expression during capacitation, from 98 % GCNF positive before capacitation to about 20 % positive following capacitation. The localization of GCNF in caput and cauda spermatozoa was similar. Conclusion: GCNF may play important roles in spermatogenesis, capacitation and fertilization.展开更多

Inter simple sequence repeats (ISSRs) and random amplified polymorphic DNAs (RAPDs) technologies were applied to detect the genetic variation of 3 bulking samples and individually sampled populations in nowadays Xiamen sea areas (Xiekou, Nanxian and Huangcuo) where the amphioxuses are alive. For the bulking sampled populations, 5 ISSR and 10 RAPD primers generated 357 bands, of which 181 (50.7%) were polymorphic. Nei index and UPGMA statistical analysis indicated that amphioxuses in these 3 areas could be divided into 2 groups. The genetic distance between animals in Nanxian and Huangcuo areas was 0.07 and classified into 1 group, while the population in Xiekou belonged to another group because its genetic distances in Nanxian and Huangcuo were 0.12 and 0.14, respectively. The result was in accordance with the morphological comparison among animals from those areas. For individually sampled population, Shannon's index of genetic diversity was used to partition the diversity of the animals among these 3 sea areas, and the results showed that the indices in populations of Xiekou, Nanxian and Huangcuo, were 0.583, 0.482 and 0.374, respectively. The linear regression equation analysis for amphioxus' genetic diversity versus the environment factors revealed that granularity/sorting coefficient and water depth were the most important factors that affect amphioxus genetic diversity. On the basis of the results, the suggestions for amphioxus resource conservation in Xiamen sea areas are put forward.展开更多

Methods: The epididymal sections from different postnatal days were stained for GCNF by the indirect immunofluorescence technique and digital photographs were taken by a Carl Zeiss confocal microscope. Results: GCNF was first detected on day 12 in mouse epididymis and day 14 in rat epididymis. The highest expression of GCNF was observed on day 35 in both mouse and rat epididymis. In adults, GCNF exhibited a region-specific expression pattern, i.e., it was expressed predominantly in the initial segment, caput and proximal corpus of rat epididymis and was abundant in the proximal corpus of mouse epididymis. GCNF could be found in the nuclei of the principal, apical, narrow, clear and halo cells. Conclusion: GCNF may play an important role in epididymal differentiation and development and in sperm maturation.展开更多

In this paper, a robust subspace-based DOA estimation and arrayauto-calibration algorithm is proposed for uniformly linear array (ULA), when the arraymutual coupling is present. Based on a banded symmetric Toeplitz matrix model for themutual coupling of ULA, the algorithm provides an accurate and high-resolution DOAestimate without any knowledge of the array mutual couplings. Moreover, a favorableestimate of mutual coupling matrix can also be achieved simultaneously for arrayauto-calibration. The algorithm is realized just via one-dimensional search or polynomialrooting, with no multidimensional nonlinear search or convergence burden involved. Theproblem of parameter ambiguity, statistically consistence and efficiency of the newestimator are also analyzed. Monte-Carlo simulation results are also provided todemonstrate the effectiveness and behavior of the proposed algorithm.展开更多
Salt-tolerant gene, CSRG1, which was isolated from a kind of salt-tolerant mangroves, Avicennia marina, constructed the transgenic plasmid, pGAM189/CSRG1. CSRG1, GUS, Kmr and Hyg^r could be transferred into tobacco ge...Salt-tolerant gene, CSRG1, which was isolated from a kind of salt-tolerant mangroves, Avicennia marina, constructed the transgenic plasmid, pGAM189/CSRG1. CSRG1, GUS, Kmr and Hyg^r could be transferred into tobacco genome by the ameliorated leaf discs method of agro-bacterium-mediate transformation. Thirteen stable resistant lines were obtained when fifty transgenic explants were selected through 50 mg/L hygromycin and 150 mg/L kanamycin. Assessments of PCR amplification, Southern blot analysis and GUS histochemical staining showed that CSRG1 has been integrated into the genome of the eleven transgenic lines (frequency of transformation was 22%). Northern bolt analysis revealed that CSRG1 had expressed in transgenic lines. The assessments of salt-tolerant ability and photosyn-thetic rates indicated that the survival rate of the transgenic lines is 80%—90% and the transgenic lines could increase by 30%—40% in plant height, even when they were cultivated in MS medium containing 2% NaCl and the total seawater (salinity 24). It is supposed that the special physiologic metabolic pathway formed by the products of CSRG1 can really endow the tobacco plants with the high salt-tolerant ability, not only to Na^+ stress, but also to the comprehensive stress of various ions.展开更多
