以实验与分子动力学模拟方法相结合,研究了受阻酚/丁腈橡胶体系(AO-60/NBR、AO-70/NBR、AO-80/NBR)的氢键网络结构与阻尼性能之间的量化关系。红外光谱结果表明三体系中均存在分子间和分子内2种类型氢键;变温红外测试结果和高温下径向... 以实验与分子动力学模拟方法相结合,研究了受阻酚/丁腈橡胶体系(AO-60/NBR、AO-70/NBR、AO-80/NBR)的氢键网络结构与阻尼性能之间的量化关系。红外光谱结果表明三体系中均存在分子间和分子内2种类型氢键;变温红外测试结果和高温下径向分布函数的变化分析表明分子间氢键的强度大小顺序为:AO-80/NBR>AO-70/NBR>AO-60/NBR。分子模拟所得氢键网络结构与动态力学热分析数据表明,3种体系中分子间氢键数量对阻尼性能的影响趋势相似。分子间氢键数量的增加导致分子间相互作用增大,自由体积分数降低,分子链堆砌更紧密,因而断键与成键过程中耗能增大,阻尼性能得以提升。线性回归分析中,体系的结合能与阻尼性能线性相关(R2=0.95),证明分子间氢键是提高阻尼性能的主要因素。这一系列研究将对高阻尼橡胶复合材料的研制提供一定的参考数据和理论指导。 展开更多

目的研究在0℃条件下,不同气体环境对金红苹果贮藏生理及品质的影响。方法以金红苹果为实验材料,采用3个气调比例(O_2的体积分数均为2%~3%,CO_2的体积分数分别为2%~3%,5%~6%,8%~9%)分别处理金红苹果,定期对其相关生理指标进行测定。结... 目的研究在0℃条件下,不同气体环境对金红苹果贮藏生理及品质的影响。方法以金红苹果为实验材料,采用3个气调比例(O_2的体积分数均为2%~3%,CO_2的体积分数分别为2%~3%,5%~6%,8%~9%)分别处理金红苹果,定期对其相关生理指标进行测定。结果在贮藏期内,与对照组相比,气调贮藏有效推迟了果实呼吸高峰的出现,显著抑制了乙烯释放速率和呼吸强度的增大;增强了过氧化物酶(POD)活性,降低了多酚氧化酶(PPO)活性;果实硬度得到较好保持,减缓了可滴定酸(TA)含量和可溶性固形物(TSS)含量的下降。其中,O_2体积分数为2%~3%和CO_2体积分数为2%~3%的气体环境贮藏金红苹果的效果最好,果实肉质坚硬,酸甜适口,色泽红青鲜艳,具有较高的好果率,货架期间感官评分较高。结论低温贮藏金红苹果最适宜的气体环境为O_2体积分数2%~3%、CO_2体积分数2%~3%。 展开更多

The characteristics and formation mechanisms of the mixed siliciclastic-carbonate reservoirs of the Paleogene Shahejie Formation in the central Bohai Sea were examined based on polarized light microscopy and scanning electron microscopy observations, X-ray diffrac- tometry, carbon and oxygen stable isotope geochemistry, and integrated fluid inclusion analysis. High-quality reservoirs are mainly distributed in Type I and Type II mixed siliciclastic-carbonate sediments, and the dominant pore types include residual primary intergranular pores and intrafossil pores, feldspar dissolution pores mainly devel- oped in Type II sediments. Type I mixed sediments are characterized by precipitation of early pore-lining dolo- mite, relatively weak mechanical compaction during deep burial, and the occurrence of abundant oil inclusions in high-quality reservoirs. Microfacies played a critical role in the formation of the mixed reservoirs, and high-quality reservoirs are commonly found in high-energy environ- ments, such as fan delta underwater distributary channels, mouth bars, and submarine uplift beach bars. Abundant intrafossil pores were formed by bioclastic decay, and secondary pores due to feldspar dissolution further enhance reservoir porosity. Mechanical compaction was inhibited by the precipitation of pore-lining dolomite formed during early stage, and oil emplacement has further led to the preservation of good reservoir quality. 展开更多

Background:Transzonal projections(TZPs)constitute a structural basis for the communication between the oocyte and its surrounding cumulus cells(CCs),which play critical roles in promoting the oocyte maturation.Previously we found that heat stress(HS)causes loss of TZPs in porcine cumulus-oocyte complexes(COCs)with decreased density of filamentous actin(F-actin).However,the time-course responses of F-actin and its monomeric actins(β-actin andγ-actin)during the in vitro maturation of oocytes remain unclear.Results:In this study,excised porcine ovaries were exposed to HS at 41.5°C for 1 h before COCs were isolated and matured in vitro for 44 h.HS significantly reduced oocyte quality,characterized by impaired cumulus expansion,delayed meiotic resumption and lower survival rate and polar body extrusion rate,as well as decreased expression of mitochondrial DNA-encoded genes and elevated mitochondrial reactive oxygen species concentration.Expression ofβ-actin andγ-actin in CCs increased gradually with oocytes maturation,which was significantly reduced in HS group,especially at 24 h and/or 44 h of in vitro maturation.By contrast,the number of TZPs and the fluorescence intensity of F-actin in zona pellucida decreased gradually during oocytes maturation,which were significantly reduced by HS at 24 h of in vitro maturation.Moreover,colocalization analyses revealed bothβ-actin andγ-actin contribute to the F-actin formation in porcine TZPs,and the colocalization of F-actin with GJ protein connexin 45 was significantly reduced in heat-exposed COCs.Conclusions:The results indicate that the suppression of actin expressions in CCs,which may lead to the F-actin unstabilization in TZPs,will subsequently contribute to the compromised quality of oocytes under HS. 展开更多

渤海古近系湖相沙河街组砂岩储层埋深较深,主要为超过3000米以下的中深层,表现为扇三角洲沉积体系。主要岩石类型为岩屑长石砂岩和长石岩屑砂岩,长石含量较高约占40.39%,石英含量较低,仅占20.39%。成岩作用过程中,压实作用总体体现为较... 渤海古近系湖相沙河街组砂岩储层埋深较深,主要为超过3000米以下的中深层,表现为扇三角洲沉积体系。主要岩石类型为岩屑长石砂岩和长石岩屑砂岩,长石含量较高约占40.39%,石英含量较低,仅占20.39%。成岩作用过程中,压实作用总体体现为较弱,而溶蚀作用占据主要位置。由于长石溶蚀的同时,自生矿物的形成尤其是粘土矿物和自生石英,再加上碳酸盐矿物的充填,导致孔隙度不断地减少。但总体来说,研究区砂岩物性较好,平均孔隙度20.8%,渗透率460.2 mD,能形成好的储层。另一方面,通过扫描电镜、电子显微镜、荧光分析等的观察,了解不同储层孔隙度和渗透率的特征,其目的是了解形成优质储层的原因。 展开更多

Establishment of Agrobacterium tumefaciens-mediated Genetic Transformation System for Aspergillus awamori

[ Objective ] This study aimed to establish Agrobacterium tumefaciens-mediated genetic transformation system for Aspergillus awamori and investigate the feasibility of expressing heterologous proteins in A. awamori. [ Method] Appropriate A. awamori host strains were determined according to the secretory protein profile. Selectable marker was selected for genetic transformation by drug sensitivity analysis. The established A. awamori genetic transformation system was used for transformation and expression analysis of Rhizomucor miehei lipase (RML). The feasibility of using A. awamori to express heterologous proteins was investigated by identification of transformants and property analysis. [ Result~ Based on the analysis of secretory protein profile, A. awamori strains CBS115.52 and CICC2257 were determined as the host strains for heterologous protein expression ; drug sensitivity analysis shows that hygromycin B resistance gene ( HygBr ) is an effective ge- netic seleetable marker; by using Agrobacterium tumefaciens-mediatcd transformation (ATMT) method, the plasmid pHGW-amdS containing HygBr was successfully transformed into A. awamori strain CBS115.52 to establish the genetic transformation system ofA. awamorl with HygBr as selectable marker. RML was transformed into A. awamori and its expression was validated by substrate hydrolysis test, SDS-PAGE and Western blot. [ Conclusion] This study demonstrates that the genetic transformation system of A. awamori mediated by Agrobacterium tumefaciens has potential feasibility for expression of heterologous proteins. 展开更多

Based on conventional meteorological observation data and Doppler radar data,the occurrence and development mechanism of mixed severe convective weather and evolution of convective storm in Guangxi on March 4,2018 were analyzed. The results showed that the dry line was the main trigger mechanism of this severe convective weather. Instable convection stratification of cold advection at middle layer and warm advection at low layer and abundant water vapor from low-level jet provided favorable stratification and water vapor conditions for the occurrence and development of severe convection. Cold trough at middle layer,low pressure and strong vertical wind shear at middle and lower layers may be main factors for the development and maintenance of strong storm system. Squall line developed along ground convergence line,and there was bow echo on reflectivity factor chart. Moving velocity of convective system was quick,and there was gale core and velocity ambiguity on velocity map. 展开更多

Effects of liraglutide on soluble tumor necrosis factor receptor in patients with diabetic nephropathy

Objective:To observe the effects of Liraglutide combined with sequential dialysis on soluble tumor necrosis factor receptor in patients with diabetic nephropathy.Methods: A total of 110 patients with early diabetic nephropathy who had been seeking treatment in the hospital between December 2016 and December 2017 were selected and according to the random number table method, these patients were randomly divided into two groups, with 55 cases in each group. Patients in the control group were given conventional symptomatic treatment such as hypoglycemic therapy, whereas the observation group was treated with Liraglutide based on the conventional symptomatic treatment given to the control group. The renal function, blood glucose metabolism level, serum indexes, vascular endothelial function and adverse drug reactions were compared between the two groups.Results: After treatment, the levels of 24 hUpor, UAER and Scr in the two groups were both shown to be lower than those before treatment, and the levels in the observation group were shown to be lower than those in the control group, with the differences being statistically significant. After treatment, the levels of PBG, FPG, HOMA-IR , HbA1c and FIns in the two groups were both significantly lower than before treatment, and the levels of PBG, FPG, HOMA-IR , HbA1c and FIns in the observation group were shown to be lower than those in the control group, where the differences were statistically significant. After treatment, the levels of TNF-α, sTNFR1 and hs-CRP in the two groups were significantly reduced than before treatment, and the levels in the observation group were shown to be significantly lower than the control group, with statistically significant differences. After treatment, the NO levels in both groups were significantly increased and ET-1 level significantly reduced than before treatment, and the NO level in the observation group was shown to be higher and the ET-1 level lower than the control group, where the difference was statistically significant.Conclusion: Liraglutide is with higher safety for patients with diabetic nephropathy, which can effectively improve their renal function and blood glucose, enhance insulin sensitivity, reduce the levels of inflammatory factors, and improve the endothelial function. 展开更多

Characteristics of Bi-2223 HTS Tapes Under Short-Circuit Current Impacts in Hybrid Energy Transmission Pipelines

A hybrid energy transmission pipeline is proposed with the aim of long-distance cooperative transmission of electricity and chemical fuels, which is composed of an inner high-temperature superconducting (HTS) power cable and outer liquefied natural gas (LNG) pipeline. The flowing LNG could maintain the operating temperature of the inner HTS power cable within the range of 85 K-90 K, thus the Bi-2223 superconductors in the HTS power cable produce little Joule loss with the transmission current below the critical current. Owing to the advantages of high power density, low transmission losses and economical manufacturing costs, the hybrid energy transmission pipeline is expected to be widely utilized in the near future. In order to ensure the safety of the HTS power cable and explosive LNG in case of short-circuit faults, this paper tests and analyzes the characteristics of Bi-2223 HTS tapes of the Type HT-CA, Type HT-SS and Type H models under short-circuit current impacts at the LNG cooling temperature (85 K-90 K). An experimental platform is designed and established for the ampacity tests of HTS tapes above LN2 cooling temperature (77 K). The AC over-current impact tests at 85 K-90 K are carried out on each sample of Bi-2223 tapes respectively, and the experimental results are analyzed and compared to evaluate their performances under different operating conditions. The results indicate that the Type HT-CA tape can withstand 50 Hz short-circuit current impact with the amplitude of 1108 A (10 times of critical current Ic ) for 100 ms at 90 K, and its resistance is the smallest of the three tested samples under similar current impacts. Therefore, the Type HT-CA Bi-2223 tape is the optimal superconductor of the HTS power cable in the hybrid energy transmission pipeline. 展开更多

Antiferroelectric PbZrO_(3)(AFE PZO)films have great potential to be used as the energy storage dielectrics due to the unique electric field(E)-induced phase transition character.However,the phase transition process always accompanies a polarization(P)hysteresis effect that induces the large energy loss(Wloss)and lowers the breakdown strength(EBDS),leading to the inferior energy storage density(Wrec)as well as low efficiency.In this work,the synergistic strategies by doping smaller ions of Li^(+)–Al^(3+)to substitute Pb2+and lowering the annealing temperature(T)from 700 to 550℃are proposed to change the microstructures and tune the polarization characters of PZO films,except to dramatically improve the energy storage performances.The prepared Pb_((1−x))(Li_(0.5)Al_(0.5))_(x)ZrO_(3)(P_((1−x))(L_(0.5)A_(0.5))_(x)ZO)films exhibit ferroelectric(FE)-like rather than AFE character once the doping content of Li^(+)–Al^(3+)ions reaches 6 mol%,accompanying a significant improvement of W_(rec) of 49.09 J/cm^(3),but the energy storage efficiency(η)is only 47.94%due to the long-correlation of FE domains.Accordingly,the low-temperature annealing is carried out to reduce the crystalline degree and the P loss.P_(0.94)(L_(0.5)A_(0.5))_(0.06)ZO films annealed at 550℃deliver a linear-like polarization behavior rather than FE-like behavior annealed at 700℃,and the lowered remanent polarization(P_(r))as well as improved E_(BDS)(4814 kV/cm)results in the superior Wrec of 58.7 J/cm^(3) and efficiency of 79.16%,simultaneously possessing excellent frequency and temperature stability and good electric fatigue tolerance. 展开更多

In recent years,PbZrO_(3)(PZO)films have become favorable electric storage materials due to the unique electric field-induced phase transition behavior,but the severe hysteresis effect leads to low energy storage density and efficiency.In this work,inserting Al_(2)O_(3)(AO)insulation nanolayers is proposed to tune the polarization behavior of flexible PZO films,anticipating optimization of energy storage performance.The results show that the thickness of the AO nanolayers has a deep influence on the polarization behavior of the PZO films,and PZO/AO/PZO(PAP)sandwiched films with 8 nm AO interlayer deliver relaxor ferroelectric-like polarization instead of antiferroelectric counterpart.To further utilize the AO nanolayers as top/bottom layers,the linear-like polarization and the highest breakdown strength are achieved in the AO/PZO/AO/PZO/AO(APAPA8)multilayer films,leading to both high discharged energy storage density of 35.2 J/cm^(3) and efficiency of 92.9%,as well as excellent fatigue and bending endurance,good temperatures,and frequency stability.The tunable polarization induced by growing the AO nanolayers makes antiferroelectric PZO films have great potential to be used as energy storage dielectrics. 展开更多

磷石膏填充薄壁方钢管短柱轴压性能试验研究

提出了一种新型的冷弯薄壁型钢墙架柱——磷石膏填充薄壁方钢管短柱,它是由磷石膏填充到薄壁方钢管中形成的组合构件,满足冷弯薄壁型钢结构“轻质”的特点。在构件中,核心磷石膏的支撑作用可延缓薄壁方钢管的局部屈曲,钢管的约束作用可... 提出了一种新型的冷弯薄壁型钢墙架柱——磷石膏填充薄壁方钢管短柱,它是由磷石膏填充到薄壁方钢管中形成的组合构件,满足冷弯薄壁型钢结构“轻质”的特点。在构件中,核心磷石膏的支撑作用可延缓薄壁方钢管的局部屈曲,钢管的约束作用可提升核心磷石膏的强度,从而充分发挥两种材料的力学性能,提升构件的轴压承载力。为了探究磷石膏填充薄壁方钢管短柱的轴压性能,以薄壁方钢管内是否填充磷石膏、磷石膏水灰比、空心率、薄壁方钢管壁厚为试验参数,设计并制作了3个空钢管试件与24个磷石膏填充薄壁方钢管短柱试件,对其开展轴压试验,研究了磷石膏填充薄壁方钢管短柱的力学性能,分析了各试件的破坏模式,荷载-变形曲线、荷载-应变曲线及轴压承载力。研究结果表明:空钢管试件呈局部屈曲破坏模式,填充试件呈腰鼓型破坏模式,核心磷石膏的支撑作用不仅可延缓薄壁钢管的局部屈曲,而且可改变试件的破坏模式,与空钢管试件相比,填充试件的轴压承载力提升了65.52%;试件空心率的变化对试件承载力的影响较为显著,这是由于含水率对磷石膏自身强度影响较大,改变试件空心率的大小会导致磷石膏含水率出现差异进而影响试件的承载力;填充磷石膏水灰比降低时,试件的轴压承载力得到一定程度提升,当磷石膏的水灰比由0.9下降到0.8、0.7时,试件的承载力分别提升了6.36%、12.42%;增加薄壁钢管壁厚,可显著提升试件的轴压性能,当钢管壁厚由0.85 mm增加到1.48,1.93 mm时,试件的轴压承载力分别提升了115.36%,176.55%,提升效果明显。基于叠加理论推导了空钢管试件与填充试件的轴压承载力计算公式,结果表明:所推导的计算公式有较好的计算精度,可较为准确地预测试件的轴压承载力。基于上述研究结果,磷石膏填充薄壁方钢管短柱具有良好的轴压力学性能,其应用前景广阔,该类构件的研发也可为磷石膏的综合应用提供一条切实可行的路径。 展开更多

Background:Renal cell carcinoma (RCC) is frequently associated with paraneoplastic inflammatory syndrome (PIS).This study aimed at exploring the connections between the survival rate and specific gene alterations and the potential mechanism.Methods:We retrospectively studied 69 surgical RCC cases from August 2014 to February 2016,including 18 cases of clear cell RCC (ccRCC) demonstrating elevated pretreatment serum C-reactive protein (CRP,Group A).Twelve of the 18 cases were symptomized with febrile episode.We also selected 49 cases ofccRCC with normal pretreatment CRP (Group B).Using 22 microsatellite markers,we compared the incidence of loss of heterozygosity (LOH) between Group A and Group B.All statistical tests are two-sided.Results:The 3p LOH was common in both Group A (89%) and Group B (92%).The frequency of 14q LOH in Group A (16 of 18) was higher than Group B (4 of 49,χ^2 =40.97 P 〈 0.0001).The 3p and 14q LOH were the characteristics of ccRCC with elevated acute phase reactants,including PIS,regardless of the presence of metastasis.On the contrary,14q LOH was a rare genomic alternation in advanced-staged ccRCC without PIS.The overall survival of patients with elevated CRP (33.3%) was lower than its counterparts (6.1%,hazard ratio=1.852,P 〈 0.0001) in Kaplan-Meier curve.Conclusions:The results imply that the disruption ofa 14q gene(s) might result in not only the inflammatory manifestations in the tumor host but also the poor survival rate as well.The isolation of the gene(s) on 14q might be a vital goal in the treatment of PIS-associated RCC. 展开更多

In this contribution,we present the results of recent neutron irradiation campaign performed in the material test reactor BR2(Belgium)on pure tungsten.We have applied various irradiation conditions and sample geometry to assess the effect of neu-tron irradiation on hardness,bending,tensile and fracture mechanical properties.The investigated material is a commercially pure tungsten plate fabricated according to the international thermonuclear experimental reactor(ITER)specification for the application in the divertor plasma-facing components.The neutron irradiation covers a large span of temperatures and damage doses,ranging from 600 to 1200°C and 0.1-1 dpa.The obtained mechanical properties were analyzed to deduce the shift of the ductile to brittle transition temperature(DBTT)applying bending,tensile and fracture toughness-testing procedures.Then,a correlation of the fracture toughness with the change of the hardness was established.The obtained results are compared with the already published results on another ITER specification grade produced in the form of a rod.The presented and discussed results show that the performance of the compared grades in terms of the irradiation-induced embrittlement is similar,and that the irradiation in the high-temperature region(600-800°C)causes a considerable DBTT shift already at 0.2-0.5 dpa. 展开更多

A Water-soluble Conjugated Polymer for Thiol Detection Based on "Tu rn-off" Effect

We investigated a novel water-soluble conjugated polymer (WSCP) for thiol detection based on "turn-off" ef- fect. This WSCP was modified with poly(ethylene glycol) (PEG) by disulfide linkages to achieve good solubility in aqueous solution (34 mg/mL) and high quantum yield (0.47). The separation of water-soluble PEG chains from the conjugated backbone induced by the cleavage of the disulfide linkages would lead to a significant decrease of the water solubility and a dramatical fluorescence quenching of the probe. The combined intuitive images and fluores- cence spectrophotometer further confirmed that decreased solubility produced an aggregation of the hydrophobic conjugated backbone. The fluorescence intensity of the probe showed a good linear relationship with glutathione (GSH) (1-200 nmol·L^-1), and the detection limit was 16 nmol·L^-1. This WSCP probe was confirmed to be a good sensing material with high selectivity to thiols by testing various biological molecules. And this WSCP probe ex- hibited good detection effect to intracellular thiols by testing Hela cells. Considering the good sensitivity and selec- tivity, the probe could be further used in vivo. In conclusion, this conjugated polymer probe made up for the draw- backs of the micromolecue probes and contributed to the development of new probes based on conjugated poly- mers. 展开更多
