Objective Gastrointestinal stromal tumors(GISTs)can rapidly proliferate through angiogenesis.Previous studies indicated the potential influence of microRNA on the progression of tumor immature angiogenesis.This study ...Objective Gastrointestinal stromal tumors(GISTs)can rapidly proliferate through angiogenesis.Previous studies indicated the potential influence of microRNA on the progression of tumor immature angiogenesis.This study aimed to explore the specific mechanism by which microRNA-409-5p(miR-409-5p)contributes to GIST.Methods To identify genes potentially involved in the development and progression of GIST,the differences of miR-409-5p between tumors and adjacent tissues were first analyzed.Following this analysis,target genes were predicted.To further investigate the function of miRNA in GIST cells,two GIST cell lines(GIST-T1 and GIST882)were transfected with lentiviruses that stably expressed miR-409-5p and scrambled miRNA(negative control).Later,the cells were subjected to Western blotting and ELSA to determine any differences in angiogenesis-related genes.Results In GISTs,there was a decrease in the expression levels of miR-409-5p compared to the adjacent tissues.It was observed that the upregulation of miR-409-5p in GIST cell lines effectively inhibited the proteins hypoxia-inducible transcription factor 1β(HIF1β)and vascular endothelial growth factor A(VEGF-A).Further investigations revealed that miR-409-5p acted as an inhibitor of angiogenesis by binding to the 3′-UTR of Lysine-specific demethylase 4D(KDM4D)mRNA.Moreover,the combination of miR-409-5p with imatinib enhanced its inhibitory effect on angiogenesis.Conclusion This study demonstrated that the miRNA-409-5p/KDM4D/HIF1β/VEGF-A signaling pathway could serve as a novel target for the development of therapeutic strategies for the treatment of imatinib-resistance in GIST patients.展开更多

Water management in proton exchange membrane fuel cells(PEMFC)is a topic of great importance for the optimization of these systems.Effective proton conductivity calls for moderate moisture content in the membrane,whil...Water management in proton exchange membrane fuel cells(PEMFC)is a topic of great importance for the optimization of these systems.Effective proton conductivity calls for moderate moisture content in the membrane,while uneven water distribution can lead to instability of the whole flow field,thereby decreasing the performance of the fuel cell.In the present study,a simplified two-tier hybrid structure is used to investigate the impact of the dynamic behavior of liquid water on the current density of the PEMFC.Simulation results show that water droplets attached to wall sides tend to increase current density.Visualization experiments confirm the existence of liquid droplets and the enhancement of current density,while indicating that the best performance and stability of fuel cell are attained for a cathode air flow rate of 300 ml/min.展开更多
Spondyloptosis in the clinic is rarely reported.We herein present a 47-year-old female,who suffered from a crush injury directly by a heavy cylindrical object from the lateral side.She was diagnosed to have traumatic ...Spondyloptosis in the clinic is rarely reported.We herein present a 47-year-old female,who suffered from a crush injury directly by a heavy cylindrical object from the lateral side.She was diagnosed to have traumatic L3 spondyloptosis with multiple traumas.Staged surgical procedures were conducted and a three-year follow-up was obtained.Eventually,normal spinal alignment was restored,and neurological deficits were gradually improved.At three years follow-up,the motor strength scores and function of the sphincters were incompletely improved.Previously published reports on traumatic lumbar spondyloptosis were reviewed and several critical points for management of this severe type of spinal injury were proposed.First,thoracolumbar and lumbosacral junction were mostly predilection sites.Second,numerous patients involving traumatic lumbar spondyloptosis were achieved to American Spinal Injury Association grade A.Third,lumbar spondyloptosis was commonly coupling withcauda equina injury.Finally,the outcomes were still with poorly prognosis and recovery of patients was correlation to spondyloptosis severity.Based on this case report and literatures review,we highlighted that the spinal alignment restoration relying on staged operations and following rehabilitation hereof are both important once facing with multiple traumas.Furthermore,we suggested to perform routine CT angiography during lumbar spondyloptosis to justify whether there are large vessel compression or injury.展开更多
Ferroptosis,a distinctive form of programmed cell death,is involved in numerous diseases with specific characteristics,including certain cell morphology,functions,biochemistry,and genetics,that differ from other forms...Ferroptosis,a distinctive form of programmed cell death,is involved in numerous diseases with specific characteristics,including certain cell morphology,functions,biochemistry,and genetics,that differ from other forms of programmed cell death,such as apoptosis.Many studies have explored ferroptosis and its associated mechanisms,drugs,and clinical applications in diseases such as kidney injury,stroke,ischemia-reperfusion injury,and prostate cancer.In this review,we summarize the regulatory mechanisms of some ferroptosis inducers,such as enzalutamide and erastin.These are current research focuses and have already been studied extensively.In summary,this review focuses on the use of ferroptosis induction as a therapeutic strategy for treating tumors of the urinary system.展开更多
