The underlying causes of biodiversity disparities among geographic regions have long been a fundamental theme in ecology and evolution.However,the patterns of phylogenetic diversity(PD) and phylogenetic beta diversity...The underlying causes of biodiversity disparities among geographic regions have long been a fundamental theme in ecology and evolution.However,the patterns of phylogenetic diversity(PD) and phylogenetic beta diversity(PBD) of congeners that are disjunctly distributed between eastern Asia-eastern North America(EA-ENA disjuncts) and their associated factors remain unknown.Here we investigated the standardized effect size of PD(SES-PD),PBD,and potentially associated factors in 11natural mixed forest sites(five in EA and six in ENA) where abundant EA-ENA disjuncts occur.We found that the disjuncts in ENA possessed higher SES-PD than those in EA at the continental scale(1.96vs-1.12),even though the number of disjunct species in ENA is much lower than in EA(128 vs 263).SESPD of the EA-ENA disjuncts tended to decrease with increasing latitude in 11 sites.The latitudinal diversity gradient of SES-PD was stronger in EA sites than in ENA sites.Based on the unweighted unique fraction metric(UniFrac) distance and the phylogenetic community dissimilarity,PBD showed that the two northern sites in EA were more similar to the six-site ENA group than to the remaining southern EA sites.Based on the standardized effect size of mean pairwise distances(SES-MPD),nine of eleven studied sites showed a neutral community structure(-1.96 ≤SES-MPD ≤1.96).Both Pearson’s r and structural equation modeling suggested that SES-PD of the EA-ENA disjuncts was mostly associated with mean divergence time.Moreover,SES-PD of the EA-ENA disjuncts was positively correlated with temperaturerelated climatic factors,although negatively correlated with mean diversification rate and community structure.By applying approaches from phylogenetics and community ecology,our work sheds light on historical patterns of the EA-ENA disjunction and paves the way for further research.展开更多

Chromosome spreads were prepared by conventional squashing. The meiotic index and the average configuration were higher, whereas the frequency of aberrance (chromosomal bridges, fragments, or micronuclei) was lower, in all 10 taxa compared with other gynmosperms. The meiotic index, average configuration, and frequency of irregularity were found to be uniform among the species. It was shown that the genomes of the Pinus species investigated were highly stable, confirming results of previous mitotic analyses in this genus. However, slight differentiation of homologous chromosomes among genomes was revealed by analysis of meiotic configurations in Pinus nigra var. poiretiana. Quadrivalents were observed in 9.31% of PMCs in this species. This is the first time that quadrivalents have been observed in gymnosperms.展开更多

It is revealed that there were slightly lower chiasma frequency, lower ring bivalent frequency, lower meiotic index and distinctly higher frequency of aberrance (chromosomal bridges, fragments or micronuclei) in pollen mother cells (PMCs) of the hybrid (F1) than those of the parental species, which showed a certain degree of differentiation between homologous chromosomes of the two parents. However, relatively higher frequency of ring bivalents and higher meiotic index in all the three entities indicate the great stability of genomes of parental species, and the differentiation of genomes between the two parents must have been slight. Total nineteen signal loci of 18S rDNA were observed in nine bivalents of the hybrid (F1), among which one bivalent bears two loci, while the others have only one. It is suggested that distinct differentiation at genetic level existed in homologous chromosomes of the two parental species, whereas only slight differentiation at karyotypic and genomic levels take place between the parent species.展开更多
Scrophularia ningpoensis has long been used in the Chinese Materia Medica for inflammation.Like other herbal medicines,S.ningpoensis collected from different localities may considerably differ in their therapeutic eff...Scrophularia ningpoensis has long been used in the Chinese Materia Medica for inflammation.Like other herbal medicines,S.ningpoensis collected from different localities may considerably differ in their therapeutic efficacy,and the one grown in Zhejiang Province is recognized as geo-authentic.However,it is difficult to confirm the geo-graphical authenticity by similar morphological characteristics.In the present study,inter-simple sequence repeat(ISSR) markers were conducted to detect S.ningpoensis from different origins.A 1 259-bp fragment amplified by primer UBC874 was found only in geo-authentic ones.By cloning and sequencing that specific band,sequence characterized amplified region(SCAR) markers were designed to distinguish geo-authentic S.ningpoensis from others.This is a rapid and easy method that can be used to identify the geographical authenticity of S.ningpoensis.展开更多
