目的探讨miR-32-5p能否通过调节p53/SLC7A11信号通路介导的铁死亡影响甲状腺细胞增殖。方法将TPC-1细胞随机分为Control组、miR-NC组、miR-32-5p组、miR-32-5p+Fer-1组、miR-32-5p+si-p53组、miR-32-5p+Fer-1+si-p53组。RT-PCR检测细胞... 目的探讨miR-32-5p能否通过调节p53/SLC7A11信号通路介导的铁死亡影响甲状腺细胞增殖。方法将TPC-1细胞随机分为Control组、miR-NC组、miR-32-5p组、miR-32-5p+Fer-1组、miR-32-5p+si-p53组、miR-32-5p+Fer-1+si-p53组。RT-PCR检测细胞中miR-32-5p相对表达量;CCK-8法检测细胞增殖;检测细胞中Fe^(2+)、MDA、SOD、GSH及ROS的含量;JC-1法检测细胞中线粒体膜电位水平;蛋白质印迹检测细胞中p53、GPX4、SLC7A11蛋白表达。结果甲状腺癌细胞系BCPAP、TPC-1、SW1736细胞中miR-32-5p相对表达量均低于人正常甲状腺细胞系HT-ori3细胞(P<0.01)。和Control组相比,miR-32-5p组细胞中miR-32-5p相对表达量、细胞中MDA含量、Fe^(2+)含量和ROS荧光强度及细胞中p53蛋白表达明显增加,细胞存活率、细胞中GSH含量和SOD活性、线粒体膜电位水平及细胞中GPX4、SLC7A11蛋白表达明显降低(P<0.01);和miR-32-5p组相比,miR-32-5p+Fer-1组、miR-32-5p+si-p53组和miR-32-5p+Fer-1+si-p53组细胞中miR-32-5p相对表达量、细胞中MDA含量、Fe^(2+)含量和ROS荧光强度及细胞中p53蛋白表达均明显降低,细胞存活率、细胞中GSH含量和SOD活性、线粒体膜电位水平及细胞中GPX4、SLC7A11蛋白表达明显增加(P<0.01);且miR-32-5p+Fer-1+si-p53组细胞中miR-32-5p相对表达量、细胞中MDA含量、Fe^(2+)含量和ROS荧光强度及细胞中p53蛋白表达明显低于miR-32-5p+Fer-1组,细胞存活率、细胞中GSH含量和SOD活性、线粒体膜电位水平及细胞中GPX4、SLC7A11蛋白表达高于miR-32-5p+Fer-1组(P<0.05)。结论过表达miR-32-5p可通过促进铁死亡抑制甲状腺癌何丽琼细胞增殖,其可能和调控p53//SLC7A11信号通路有关。 展开更多

Psychological intervention based on social cognitive theory: Treating pain, anxiety, and depression in perioperative patients

BACKGROUND Surgery is an effective method for treating certain diseases.Factors such as disease,preoperative fear and tension,surgical stress,postoperative pain,and related complications directly affect the smooth progression and outcome of surgery.Patients may experience a series of psychological and physiological changes during the perioperative period,resulting in anxiety and depression,which may reduce the pain threshold and worsen their prognosis.METHODS We enrolled 200 patients who underwent surgical care at The First People’s Hospital of Lin’an District,Hangzhou between January and December 2023.They were categorized into a routine intervention group(n=103)and a psychological intervention group(n=97),based on the intervention strategies used.Various assessment tools,including the self-rating anxiety scale(SAS),the self-rating depression scale(SDS),and the Connor–Davidson Resilience scale,were used to measure patients’negative states and emotions.The pre-and post-intervention scores for these metrics in the two groups were then analyzed.RESULTS In the psychological intervention group,the SAS and SDS scores(31.56±5.18 and 31.46±4.57,respectively)were significantly reduced compared to the routine intervention group(P<0.05).The visual analog scale pain scores at 12 and 24 hours after intervention(6.85±1.21,4.24±0.72)were notably higher than those in the routine intervention group(P<0.05).The psychological intervention group also demonstrated superior scores in perseverance(36.08±3.29),self-reliance(22.63±2.91),optimism(11.42±1.98),and resilience(70.13±5.37),compared to the routine intervention group(P<0.05).Additionally,the psychological intervention group’s confrontation score(23.16±4.29)was higher(P<0.05).This group also reported lower scores in avoidance(9.28±1.94)and yielding(6.19±1.92)(P<0.05).Lastly,the Short Form 36 Health Survey scores were significantly higher in the psychological intervention group,indicating a better quality of life(P<0.05).CONCLUSION Psychological intervention measures based on SCT can effectively alleviate pain,anxiety,and depression in periop-erative patients. 展开更多

AIM: To investigate and compare the effects of spinal D-(-)-2-amino-7-phosphonoheptanoic acid (AP-7) and 6-cyano-7-nitroquinoxaline-2,3-dione disodium (CNQX),two glutamate receptor antagonists, on the responses of dorsal horn neurons to colorectal distension (CRD) in adult rats exposed to neonatal colon irritation (CI).METHODS: Hypersensitive SD rats were generated by CI during postnatal days 8, 10 and 12. Experiments on adult rats were performed using extracellular single-unit recording. The effects of spinal application of AP-7 (0.001,0.01, 0.1, 1 mmoL) were tested on the CRD-evoked neuronal responses in 16 controls and 17 CI rats. The effects of CNQX (0.2, 2, 5, 10 μmoL) were also tested on the CRD-evoked responses of 17 controls and 18 CI neurons.RESULTS: (1) The average responses of lumbosacral neurons to all intensities of CRD in CI rats were significantly higher than those in control rats; (2) In control rats, AP-7 (0.01 mmoL) had no significant effect on the neuronal response to all intensities of CRD (20,40, 60, 80 mmHg); while AP-7 (0.1 mmoL) inhibited the neuronal response to 80-mmHg CRD. By contrast, in CI rats, AP-7 (0.01-1 mmoL) attenuated the CRD-evoked neuronal responses to all distention pressures in a dosedependent manner; (3) In control rats, CNQX (2 μmoL)had no significantly effect on the neuronal response to all intensities of CRD; however, CNQX (5 μmoL) significantly attenuated the responses to CRD in the 40-80 mmHg range. By contrast, CNQX (2-10 μmoL)significantly decreased the neuronal responses in CI rats to non-noxious and noxious CRD in a dose-dependent manner.CONCLUSION: Our results suggest that spinal N-methyl-D-aspartate (NMDA) and non-NMDA receptors may contribute to the processing of central sensitivity in a neonatal CI rat model, but they may play different roles in it. 展开更多

Flowering time is a key agronomic trait that directly affect the adaptation and yield of soybean.After whole genome duplications,about 75%of genes being represented by multiple copies in soybean.There are four TERMINAL FLOWER 1(TFL1)genes in soybean,and the TFL1b(Dt1)has been characterized as the determinant of stem growth habit.The function of other TFL1 homologs in soybean is still unclear.Here,we generated knockout mutants by CRISPR/Cas9 genome editing technology and found that the tfl1c/tfl1d double mutants flowered significantly earlier than wild-type plants.We investigated that TFL1c and TFL1d could physically interact with the b ZIP transcription factor FDc1 and bind to the promoter of APETALA1a(AP1a).RNA-seq and q RT-PCR analyses indicated that TFL1c and TFL1d repressed the expressions of the four AP1 homologs and delayed the flowering time in soybean.The two genes play important roles in the regulation of flowering time in soybean and mainly act as the flowering inhibitors under long-day conditions.Our results identify novel components in the flowering-time regulation network of soybean and will be invaluable for molecular breeding of improved soybean yield. 展开更多

To ensure a long-term quantum computational advantage,the quantum hardware should be upgraded to withstand the competition of continuously improved classical algorithms and hardwares.Here,we demonstrate a superconducting quantum computing systems Zuchongzhi 2.1,which has 66 qubits in a two-dimensional array in a tunable coupler architecture.The readout fidelity of Zuchongzhi 2.1 is considerably improved to an average of 97.74%.The more powerful quantum processor enables us to achieve larger-scale random quantum circuit sampling,with a system scale of up to 60 qubits and 24 cycles,and fidelity of FXEB=(3·66±0·345)×10^(-4).The achieved sampling task is about 6 orders of magnitude more difficult than that of Sycamore[Nature 574,505(2019)]in the classic simulation,and 3 orders of magnitude more difficult than the sampling task on Zuchongzhi 2.0[arXiv:2106.14734(2021)].The time consumption of classically simulating random circuit sampling experiment using state-of-the-art classical algorithm and supercomputer is extended to tens of thousands of years(about 4·8×104years),while Zuchongzhi 2.1 only takes about 4.2 h,thereby significantly enhancing the quantum computational advantage. 展开更多

目的 分析长链非编码RNA(lncRNA)SNHG6靶向调控miR-519d-3p/CCND1对三阴性乳腺癌(TNBC细胞增殖、迁移及凋亡的影响。方法 采用siRNA技术沉默TNBC细胞系MDA-MB-231中lncRNA SNHG6的表达,实验分为三组:正常人乳腺上皮细胞系MCF-10A(对照组... 目的 分析长链非编码RNA(lncRNA)SNHG6靶向调控miR-519d-3p/CCND1对三阴性乳腺癌(TNBC细胞增殖、迁移及凋亡的影响。方法 采用siRNA技术沉默TNBC细胞系MDA-MB-231中lncRNA SNHG6的表达,实验分为三组:正常人乳腺上皮细胞系MCF-10A(对照组)、MDA-MB-231和si-SNHG6(si组),采用实时荧光定量PCR(RT-qPCR)检测lncRNA SNHG6和miR-519d-3p表达,MTT法检测细胞增殖率,Transwell实验检测迁移能力,流式细胞术检测细胞凋亡率,Western blot实验检测CCND1蛋白表达。采用双荧光素酶报告基因检测SNHG6与miR-519d-3p及miR-519d-3p与CCND1的靶向关系。三组间计量资料采用单因素ANOVA分析和LSD-t法检验,P<0.05为差异有统计学意义。结果 连续培养48 h发现,与对照组比较,MDA-MB-231组lncRNA SNHG6与miR-519d-3p mRNA相对表达量、细胞增殖率、迁移率和CCND1蛋白水平均显著升高,细胞凋亡率显著下降(P<0.05);与MDA-MB-231组比较,si组lncRNASNHG6与miR-519d-3pmRNA相对表达量显著降低、细胞增殖率、迁移率和CCND1蛋白水平均显著下降(P<0.05),凋亡率上升;双荧光素酶报告基因检测显示,SNHG6能够靶向上调miR-519d-3p表达,miR-519d-3p能够靶向上调CCND1表达。结论 lncRNASNHG6能够通过靶向调控miR-519d-3p/CCND1功能进而影响TNBC细胞的增殖、迁移及凋亡能力,可能是TNBC发病的重要分子机制,也有望成为分子干预的靶点。 展开更多
