Methods:The Plasmid pcDNA3.1(+)-AP-2α was transfected into colorectal carcinoma SW620 cells line by liposome mediation for transient expression. After AP-2α transfected SW620 cells, the exogenous AP-2α mRNA and protein express were determined by the method of Real-time PCR and Western blot. In order to elucidate the effect of expression of exogenous AP-2α gene on the colorectal cancer cell SW620, the proliferation rates were analyzed by growth curves for cells including SW620, pcDNA3.1(+)/SW620 and pcDNA3.1(+)-AP-2α/SW620. At same time, the apoptotic rate of cells and the cell cycle analysis were also tested by flow cytometry. Results: The mRNA and protein expressions of AP-2α gene could be enhanced by transfecting of pcDNA3.1(+)-AP-2α recombinant plasmid into SW620 cell. The cell growth rates of SW620 cells transfected with pcDNA3.1(+)/AP-2α were slower than those transfected with pcDNA3.1(+) or untransfected. The apoptotic rates were increased compared with pcDNA3.1(+)/SW620 and SW620 (P<0.05), which indicated that over-expression of AP-2α gene could efficiently inhibit the growth of SW620 cells and induce apoptosis. FCM analyses indicated that the cells being arrested in G1 phase. Conclusion: AP-2α gene can be efficiently transfected into SW620 cells and over-expression AP-2α protein in the transfected cells. AP-2α induces G1 arrest and apoptosis, suppressive effect on cell growth.展开更多

Session-based recommendation aims to predict user preferences based on anonymous behavior sequences.Recent research on session-based recommendation systems has mainly focused on utilizing attention mechanisms on seque...Session-based recommendation aims to predict user preferences based on anonymous behavior sequences.Recent research on session-based recommendation systems has mainly focused on utilizing attention mechanisms on sequential patterns,which has achieved significant results.However,most existing studies only consider individual items in a session and do not extract information from continuous items,which can easily lead to the loss of information on item transition relationships.Therefore,this paper proposes a session-based recommendation algorithm(SGT)based on Gated Recurrent Unit(GRU)and Transformer,which captures user interests by learning continuous items in the current session and utilizes all item transitions on sessions in a more refined way.By combining short-term sessions and long-term behavior,user dynamic preferences are captured.Extensive experiments were conducted on three session-based recommendation datasets,and compared to the baseline methods,both the recall rate Recall@20 and the mean reciprocal rank MRR@20 of the SGT algorithm were improved,demonstrating the effectiveness of the SGT method.展开更多
A hallmark of systemic lupus erythematosus(SLE)is the breaking of B-cell tolerance with the generation of high-affinity autoantibodies;however,the antibody-independent features of the B-cell compartment in SLE are les...A hallmark of systemic lupus erythematosus(SLE)is the breaking of B-cell tolerance with the generation of high-affinity autoantibodies;however,the antibody-independent features of the B-cell compartment in SLE are less understood.In this study,we performed an extensive examination of B-cell subsets and their proinflammatory properties in a Chinese cohort of new-onset SLE patients.We observed that SLE patients exhibited an increased frequency of transitional B cells compared with healthy donors and rheumatoid arthritis patients.Plasma from SLE patients potently promoted the survival of transitional B cells in a type I IFN-dependent manner,which can be recapitulated by direct IFN-αtreatment.Furthermore,the effect of IFN-αon enhanced survival of transitional B cells was associated with NF-κB pathway activation and reduced expression of the pro-apoptotic molecule Bax.Transitional B cells from SLE patients harbored a higher capacity to produce proinflammatory cytokine IL-6,which was also linked to the overactivated type I IFN pathway.In addition,the frequency of IL-6-producing transitional B cells was positively correlated with disease activity in SLE patients,and these cells were significantly reduced after short-term standard therapies.Thus,the current study provides a direct link between type I IFN pathway overactivation and the abnormally high frequency and proinflammatory properties of transitional B cells in active SLE patients,which contributes to the understanding of the roles of type I IFNs and B cells in the pathogenesis of SLE.展开更多
Chinese herbal medicines(CHMs)play an increasingly important role in the field of medicine and affects public health in the world.Although more and more strict has been employed to ensure the quality and safety of CHM...Chinese herbal medicines(CHMs)play an increasingly important role in the field of medicine and affects public health in the world.Although more and more strict has been employed to ensure the quality and safety of CHMs,pesticide residues in CHMs remain a serious issue and are the bottleneck for the global development of CHMs.In this work,we applied molecularly imprinted membrane electrospray mass spectrometry(MIM-ESI MS)for rapid detecting 4 classes of pesticide residues in CHMs,including organophosphorus(OPP),carbamates,pyrethroids and neonicotinoids in CHMs.Compared with our previous ambient ionization method MESI,MIM-ESI is capable of achieving a~50-fold increase in the detection limit of conventional analytical methods owing to the specificity recognition and unique enrichment of MIM.The optimal experimental conditions were determined,and the method was further validated for its sensitivity and specificity.Our data showed that MIM-ESI MS is applicable for the direct quantitation of pesticide residues in CHMs.This detection technology may help to ensure the quality of CHMs in the future.展开更多
