In order to make use of ocean clay reasonably and excavate its potential applications the properties of ocean clay in Chinese investigating area of the East Pacific have been tested with the chemical analysis, XRFS, I...In order to make use of ocean clay reasonably and excavate its potential applications the properties of ocean clay in Chinese investigating area of the East Pacific have been tested with the chemical analysis, XRFS, ICP/MAS, granulometry and SSA, etc. The results show that the clay sediments of this area are rich in clay minerals, fine granularity, bigger SSA, bigger water absorbability and lower ion changing ability etc. To improve the adaptability of ocean clay and macromolecule materials, using the dry milling method with silane, aluminate, stearic acid coupling agents etc the properties of ocean clay are improved. The effect has been evaluated by testing the water absorbing of the modified ocean clay and their dispersing in paraffin liquid. The result is that KH550 is better than any other active reagents, and the best activation technics is that 1.5% (volume) of KH550 quantity, 20min of milling time. The DTA, XRD, SEM and IR analyses and SSA testing show that the organic activation can not change the mineral structure but can densify the structure of clay, and activation mechanism is chemical covered mainly and chemical combined secondly.展开更多
Genome-wide physical protein±protein interaction(PPI)mapping remains a major challenge for current technologies.Here,we reported a high-efficiency BiFC-seq method,yeastenhanced green fluorescent protein-based bim...Genome-wide physical protein±protein interaction(PPI)mapping remains a major challenge for current technologies.Here,we reported a high-efficiency BiFC-seq method,yeastenhanced green fluorescent protein-based bimolecular fluorescence complementation(y EGFPBiFC)coupled with next-generation DNA sequencing,for interactome mapping.We first applied y EGFP-BiFC method to systematically investigate an intraviral network of the Ebola virus.Two-thirds(9/14)of known interactions of EBOV were recaptured,and five novel interactions were discovered.Next,we used the BiFC-seq method to map the interactome of the tumor protein p53.We identified 97 interactors of p53,more than three-quarters of which were novel.Furthermore,in a more complex background,we screened potential interactors by pooling two BiFC libraries together and revealed a network of 229 interactions among 205 proteins.These results show that BiFC-seq is a highly sensitive,rapid,and economical method for genome-wide interactome mapping.展开更多
Algal blooms have become a worldwide environmental concern due to water eutrophication.Dianchi Lake in Yunnan Province,China is suffering from severe eutrophication and is listed in the Three Important Lakes Restorati...Algal blooms have become a worldwide environmental concern due to water eutrophication.Dianchi Lake in Yunnan Province,China is suffering from severe eutrophication and is listed in the Three Important Lakes Restoration Act of China.Hydrothermal liquefaction allows a promising and direct conversion of algal biomass into biocrude oil.In this study,algal samples were collected from Dianchi Lake after a separation procedure including dissolved air flotation with polyaluminum chloride and centrifugation during four months,April,June,August and October.The algal biochemical components varied over the period;lipids from 0.7%to 2.1%ash-free dry weight(afdw),protein from 20.9%to 33.4%afdw and ash from 36.6%to 45.2%dry weight.The algae in June had the highest lipid and protein concentrations,leading to a maximum biocrude oil yield of 24.3%afdw.Biodiversity analysis using pyrosequencing revealed different distributions of microbial communities,specifically Microcystis in April(89.0%),June(63.7%)and August(84.0%),and Synechococcus in April(2.2%),June(12.0%)and August(1.0%).This study demonstrated remarkable temporal changes in the biochemical composition and biodiversity of algae harvested from Dianchi Lake and changes in biocrude oil production potential.展开更多
