In this paper, the dynamic text representation method XLNet is used to generate word vectors with context-dependent dependencies to distribute the text information of food safety network public opinion. Then, the word vector is input into the CNN-BiLSTM network for local semantic feature and context semantic extraction. The attention mechanism is introduced to give different weights according to the importance of features, and the emotional tendency analysis is carried out. Based on sentiment analysis, sentiment value time series data is obtained, and a time series model is constructed to predict sentiment trends. The sentiment analysis model proposed in this paper can well classify the sentiment of food safety network public opinion, and the time series model has a good effect on the prediction of food safety network public opinion sentiment trend. .展开更多

BACKGROUND China's most frequent malignancy is gastric cancer(GC),which has a very poor survival rate,and the survival rate for patients with advanced GC is dismal.Pyroptosis has been connected to the genesis and ...BACKGROUND China's most frequent malignancy is gastric cancer(GC),which has a very poor survival rate,and the survival rate for patients with advanced GC is dismal.Pyroptosis has been connected to the genesis and development of cancer.The function of pyroptosis-related long non-coding RNAs(PRLs)in GC,on the other hand,remains uncertain.AIM To explore the construction and comprehensive analysis of the prognostic characteristics of long non-coding RNA(lncRNA)related to pyroptosis in GC patients.METHODS The TCGA database provided us with 352 stomach adenocarcinoma samples,and we obtained 28 pyroptotic genes from the Reactome database.We examined the correlation between lncRNAs and pyroptosis using the Pearson correlation coefficient.Prognosis-related PRLs were identified through univariate Cox analysis.A predictive signature was constructed using stepwise Cox regression analysis,and its reliability and independence were assessed.To facilitate clinical application,a nomogram was created based on this signature.we analyzed differences in immune cell infiltration,immune function,and checkpoints between the high-risk group(HRG)and low-risk group(LRG).RESULTS Five hundred and twenty-three PRLs were screened from all lncRNAs(absolute correlation coefficient>0.4,P<0.05).Nine PRLs were included in the risk prediction signature that was created through stepwise Cox regression analysis.We determined the risk score for GC patients and employed the median value as the dividing line between HRG and LRG.The ability of the risk signature to predict the overall survival(OS)of GC is demonstrated by the Kaplan-Meier analysis,risk curve,receiver operating characteristic curve,and decision curve analysis curve.The risk signature was shown to be an independent prognostic factor for OS in both univariate and multivariate Cox regression analyses.HRG showed a more efficient local immune response or modulation compared to LRG,as indicated by the predicted signal pathway analysis and examination of immune cell infiltration,function,and checkpoints(P<0.05).CONCLUSION In general,we have created a brand-new prognostic signature using PRLs,which may provide ideas for immunotherapy in patients with GC.展开更多

METHODS: Recombinant plasmid psil-HBV was constructed and transfected into HepG2.2.15 cells. The transfected cells were cultured in lamivudine-containing medium (0.05 μmol/L) and harvested at 48, 72 and 96 h. The concentration of HBeAg and HBsAg was determined using ELISA. HBV DNA replication was examined by real- time PCR and the level of HBV mRNA was measured by RT-PCR. RESULTS: In HepG2.2.15 cells treated with combination of siRNA and lamivudine, the secretion of HBeAg and HBsAg into the supernatant was found to be inhibited by 91.80% and 82.40% (2.89 ± 0.48 vs 11.73 ± 0.38, P < 0.05; 4.59 ± 0.57 vs 16.25 ± 0.48, P < 0.05) at 96 h, respectively; the number of HBV DNA copies within culture medium was also significantly decreased at 96 h (1.04 ± 0.26 vs 8.35 ± 0.33, P < 0.05). Moreover, mRNA concentration in HepG2.2.15 cells treated with combination of siRNA and lamivudine was obviously lower compared to those treated either with siRNA or lamivudine (19.44 ± 0.17 vs 33.27 ± 0.21 or 79.9 ± 0.13, P < 0.05). CONCLUSION: Combination of siRNA and lamivudine is more effective in inhibiting HBV replication as compared to the single use of siRNA or lamivudine in HepG2.2.15 cells.展开更多

We developed a GPU based single-pulse search pipeline(GSP)with a candidate-archiving database.Largely based upon the infrastructure of the open source PulsaR Exploration and Search Toolkit(PRESTO),GSP implements GPU a...We developed a GPU based single-pulse search pipeline(GSP)with a candidate-archiving database.Largely based upon the infrastructure of the open source PulsaR Exploration and Search Toolkit(PRESTO),GSP implements GPU acceleration of the de-dispersion and integrates a candidate-archiving database.We applied GSP to the data streams from the Commensal Radio Astronomy FAST Survey(CRAFTS),which resulted in quasi-real-time processing.The integrated candidate database facilitates synergistic usage of multiple machine-learning tools and thus improves efficient identification of radio pulsars such as rotating radio transients(RRATs)and fast radio bursts(FRBs).We first tested GSP on pilot CRAFTS observations with the FAST Ultra-Wide Band(UWB)receiver.GSP detected all pulsars known from the the Parkes multibeam pulsar survey in the corresponding sky area covered by the FAST-UWB.GSP also discovered 13 new pulsars.We measured the computational efficiency of GSP to be~120 times faster than the original PRESTO and~60 times faster than an MPI-parallelized version of PRESTO.展开更多
We report here Atacama Large Millimeter/submillimeter Array(ALMA)N2H+(1-0)images of the Orion Molecular Cloud 2 and 3(OMC-2/3)with high angular resolution(3"or 1200 au)and high spatial dynamic range.Combining a d...We report here Atacama Large Millimeter/submillimeter Array(ALMA)N2H+(1-0)images of the Orion Molecular Cloud 2 and 3(OMC-2/3)with high angular resolution(3"or 1200 au)and high spatial dynamic range.Combining a dataset from the ALMA main array,Atacama Compact Array(ACA),Nobeyama 45-m Telescope and Very Large Array(VLA)(providing temperature measurement on matching scales),we find that most of the dense gas in OMC-2/3 is subsonic(σQNT/cs=0.62)with a mean line width(△v)of 0.39 kms-1 full width at half maximum(FWHM).This is markedly different from the majority of previous observations of massive star-forming regions.In contrast,line widths from the Nobeyama Telescope are transonic at 0.69 km s-1(σNT/cs=1.08).We demonstrated that the larger line widths obtained by the single-dish telescope arose from unresolved sub-structures within their respective beams.The dispersions from larger scalesσls(as traced by the Nobeyama Telescope)can be decomposed into three components such thatσls2=σss2+σbm2+σrd2,where small-scaleσss is the line dispersion of each ALMA beam,bulk motionσbm is dispersion between peak velocity of each ALMA beam andσrd is the residual dispersion.Such decomposition,though purely empirical,appears to be robust throughout our data cubes.Apparent supersonic line widths,commonly found in massive molecular clouds,are thus likely due to the effect of poor spatial resolution.The observed non-thermal line dispersion(sometimes referred to as’turbulence’)transits from supersonic to subsonic at~0.05 pc scales in the OMC-2/3 region.Such transition could be commonly found with sufficient spatial(not just angular)resolution,even in regions with massive young clusters,such as the Orion molecular clouds studied here.展开更多

Such missing information can be studied by the new generation of Chinese radio telescopes through a blind search of 21-cm absorption systems. We forecast the capabilities of surveys of 21-cm absorption systems by two representative radio telescopes in China - the Five-hundred-meter Aperture Spherical radio Telescope (FAST) and Tianlai 21-cm cosmology experiment (Tianlai). Facilitated by either the high sensitivity (FAST) or wide field of view (Tianlai) of these telescopes, more than a thousand 21-cm absorption systems can be discovered in a few years, representing orders of magnitude improvement over the cumulative discoveries in the past half a century.展开更多
The increasing radio frequency interference(RFI)is a well-recognized problem in radio astronomy research.Pulsars and Fast Radio Bursts(FRBs)are high-priority science targets of the ongoing Commercial Radio Astronomy F...The increasing radio frequency interference(RFI)is a well-recognized problem in radio astronomy research.Pulsars and Fast Radio Bursts(FRBs)are high-priority science targets of the ongoing Commercial Radio Astronomy FAST Survey(CRAFTS).To improve the quality of RFI removal in searches of pulsars and FRBs based on CRAFTS multi-beam data,we here propose an intuitive but powerful RFI mitigation pipeline(CCF-ST).The“CCF-ST”is a spatial filter constructed by signal cross-correlation function(CCF)and Sum-Threshold(ST)algorithm.The RFI marking result is saved in a“mask”file,a binary format for RFI masks in PRESTO.Three known pulsars,PSR B0525-21,PSR B0621-04,and PSR J0943+2252 from CRAFTS L-band 19 beams data are used for evaluation of the performance of CCF-ST in comparison with other methods,such as PRESTO’s“rfifind”,ArPLS-ST and ArPLS-SF.The result shows that CCF-ST can reduce effective data loss rate and improves the detected signal-to-noise ratio of the pulsations by~26%and~18%respectively compared with PRESTO’s“rfifind”and ArPLS-ST.The CCF-ST also has the advantage of low computational cost,e.g.,reducing the time consumption by~40%and memory consumption by~90%compared with ArPLS-SF.We expect that the new RFI mitigation and analysis toolkit(CCF-ST)demonstrated in this paper can be applied to CRAFTS and other multi-beam telescope observations to improve the data quality and efficiency of pulsar and FRB searches.展开更多
We report the discovery of PSR J1909+0122 by the Five-hundred-meter Aperture Spherical Radio Telescope(FAST)as part of the Commensal Radio Astronomy FAST Survey.PSR J1909+0122 has a spin period of 1.257 s and a disper...We report the discovery of PSR J1909+0122 by the Five-hundred-meter Aperture Spherical Radio Telescope(FAST)as part of the Commensal Radio Astronomy FAST Survey.PSR J1909+0122 has a spin period of 1.257 s and a dispersion measure of 186.2 pc cm^(-3).The averaged pulse profile shows two distinct components.We performed a single-pulse study based on a one-hour observation at 1.25 GHz on 2021 August 23.We used a threshold of 5σ_(ep) to measure the nulling fraction(NF)as 63%±1.5%.The longitude-resolved fluctuation spectra and fast Fourier transform spectra of the binary sequences revealed the quasi-periodicity of nulling with a period of 30 rotation periods.We examined the reliability of the periodicity by comparing it to random noise injection.The NF,E,and modulation periodicity P_(M) of PSR J1909+0122 were compared with other periodic nulling pulsars,showing that the source of J1909+0122 has the second largest NF in the population.Long-term timing observations over six months were used to derive the phase-connected ephemeris of this pulsar.The measured P and P values disfavor dipolar geometry for polar gap models,and the prediction for a space-charge-limited flow model in the case of inverse Compton scattering is only just above the death line.In this work,PSR J1909+0122 has revealed possible correlations between nulling behavior and pulsar properties,which will help to shed light on the pulsar emission mechanism and its temporal evolution in future observations.展开更多
We investigate the single-pulse emission variations of two pulsars,PSRs J0211+4235 and J0553+4111,observed with the Five-hundred-meter Aperture Spherical radio Telescope at the 1.25 GHz central frequency.The observati...We investigate the single-pulse emission variations of two pulsars,PSRs J0211+4235 and J0553+4111,observed with the Five-hundred-meter Aperture Spherical radio Telescope at the 1.25 GHz central frequency.The observation sessions span from 2020 December to 2021 July,with 21 and 22 observations for them respectively.The integrated pulse profile of PSR J0211+4235 shows that there is a weak pulse component following the main component,and PSR J0553+4111 displays a bimodal profile with a bridge component in the middle.PSR J0211+4235 presents significant nulling phenomenon with nulling duration lasting from 2 to 115 pulses and burst duration lasting from 2 to 113 pulses.The NF of each observation is determined to be 45%-55%.No emission greater than threeσis found in the mean integrated profile of all nulling pulses.In most cases,the pulse energy changes abruptly during the transition from null to burst,while in the transition from burst to null there are two trends:abrupt and gradual.We find that the nulling phenomenon of PSR J0211+4235 is periodic by the Fourier transform of the null and burst state.In addition,the single-pulse modulation characteristics of these two pulsars are investigated,and the distributions of modulation index,LRFS and 2DFS are analyzed with PSRSALSA.The left peak of PSR J0553+4111 has intensity modulation.Finally,the polarization properties of these two pulsars are obtained through polarization calibration,and their characteristics are analyzed.The possible physical mechanisms of these phenomena are discussed.展开更多

The current collection, taken from the observation during the FRB active phase from April to June 2021, is the largest burst sample detected for any FRB so far. The standard PSRFITs format is adopted, including dynamic spectra of the burst, and the time information of the dynamic spectra, in addition, mask files help readers to identify the pulse positions are also provided. Correlation power analysis(CPA)combined with genetic algorithms(GA)now achieves greater attack efficiency and can recover all subkeys simultaneously.However,two issues in GA-based CPA still need to be addressed:key de...Correlation power analysis(CPA)combined with genetic algorithms(GA)now achieves greater attack efficiency and can recover all subkeys simultaneously.However,two issues in GA-based CPA still need to be addressed:key degeneration and slow evolution within populations.These challenges significantly hinder key recovery efforts.This paper proposes a screening correlation power analysis framework combined with a genetic algorithm,named SFGA-CPA,to address these issues.SFGA-CPA introduces three operations designed to exploit CPA characteris-tics:propagative operation,constrained crossover,and constrained mutation.Firstly,the propagative operation accelerates population evolution by maximizing the number of correct bytes in each individual.Secondly,the constrained crossover and mutation operations effectively address key degeneration by preventing the compromise of correct bytes.Finally,an intelligent search method is proposed to identify optimal parameters,further improving attack efficiency.Experiments were conducted on both simulated environments and real power traces collected from the SAKURA-G platform.In the case of simulation,SFGA-CPA reduces the number of traces by 27.3%and 60%compared to CPA based on multiple screening methods(MS-CPA)and CPA based on simple GA method(SGA-CPA)when the success rate reaches 90%.Moreover,real experimental results on the SAKURA-G platform demonstrate that our approach outperforms other methods.展开更多
BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cance...BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cancer lacks a comprehensive analysis,and its role and mechanism in hepatocellular carcinoma(HCC)are still unclear.The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Profiling Interactive Analysis,and Tumor Immune Estimation Resource databases were used to analyze B56εexpression,prognostic mutations,somatic copy number alterations,and tumor immune characteristics in 33 tumors.The relationships between B56εexpression levels and drug sensitivity,immuno-therapy,immune checkpoints,and human leukocyte antigen(HLA)-related genes were further analyzed.Gene Set Enrichment Analysis(GSEA)was performed to reveal the role of B56εin HCC.The Cell Counting Kit-8,plate cloning,wound healing,and transwell assays were conducted to assess the effects of B56εinterference on the malignant behavior of HCC cells.RESULTS In most tumors,B56εexpression was upregulated,and high B56εexpression was a risk factor for adrenocortical cancer,HCC,pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma(all P<0.05).B56εexpression levels were correlated with a variety of immune cells,such as T helper 17 cells,B cells,and macro-phages.There was a positive correlation between B56εexpression levels with immune checkpoint genes and HLA-related genes(all P<0.05).The expression of B56εwas negatively correlated with the sensitivity of most chemotherapy drugs,but a small number showed a positive correlation(all P<0.05).GSEA analysis showed that B56εexpression was related to the cancer pathway,p53 downstream pathway,and interleukin-mediated signaling in HCC.Knockdown of B56εexpression in HCC cells inhibited the proliferation,migration,and invasion capacity of tumor cells.Core Tip:The expression of protein phosphatase 2A(PP2A)subunit B56εis up-regulated in most tumors,and its high expression is a risk factor for adrenocortical cancer,hepatocellular carcinoma(HCC),pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma.B56εexpression levels correlate with immune cells,immune checkpoint genes,human leukocyte antigen-related genes,and the sensitivity of chemotherapy drugs.In HCC,B56εexpression is related to the cancer pathway.Knockdown of B56εexpression in HCC cells can inhibit the proliferation,migration and invasion capacity of tumor cells.Our study supports PP2A subunit B56εas a prognostic marker and potential therapeutic target for HCC.展开更多
Recent submillimeter dust thermal emission observations have unveiled a significant number of inter-arm massive molecular clouds in M31.However,the effectiveness of this technique is limited to its sensitivity,making ...Recent submillimeter dust thermal emission observations have unveiled a significant number of inter-arm massive molecular clouds in M31.However,the effectiveness of this technique is limited to its sensitivity,making it challenging to study more distant galaxies.This study introduces an alternative approach,utilizing optical extinctions derived from space-based telescopes,with a focus on the forthcoming China Space Station Telescope(CSST).We first demonstrate the capability of this method by constructing dust extinction maps for 17 inter-arm massive molecular clouds in M31 using the Panchromatic Hubble Andromeda Treasury data.Our analysis reveals that inter-arm massive molecular clouds with an optical extinction(A_(V)) greater than 1.6 mag exhibit a notable A_(V) excess,facilitating their identification.The majority of these inter-arm massive molecular clouds show an A_(V) around 1 mag,aligning with measurements from our JCMT data.Further validation using a mock CSST RGB star catalog confirms the method's effectiveness.We show that the derived A_(V)values using CSST z and y photometries align more closely with the input values.Molecular clouds with A_(V)> 1.6 mag can also be identified using the CSST mock data.We thus claim that future CSST observation clouds provide an effective way for the detection of inter-arm massive molecular clouds with significant optical extinction in nearby galaxies.展开更多

METHODS: Recombinant plasmid psiI-HBV was constructed and transfected into HepG2.2.15 cells. At 48 h, 72 h and 96 h after transfection, culture media were collected and cells were harvested for HBV replication assay. HBsAg and HBeAg in the cell culture medium were detected by enzyme-linked immunoadsorbent assay (ELISA). Intracellular viral DNA and covalently closed circular DNA (cccDNA) were quantified by real-time polymerase chain reaction (PCR). HBV viral mRNA was reverse transcribed and quantified by reverse-transcript PCR (RT-PCR). RESULTS: siRNAs showed marked anti-HBV effects. siRNAs could specifically inhibit the expression of HBsAg and the replication of HBV DNA in a dosedependent manner. Furthermore, combination of siRNAs, compared with individual use of each siRNA, exerted a stronger inhibition on antigen expression and viral replication. More importantlycombination of siRNAs significantly suppressed HBV cccDNA amplification. CONCLUSION: Combination of siRNAs mediates a stronger inhibition on viral replication and antigenexpression in HepG2.2.15 cells, especially on cccDNA amplification.展开更多

These factors' sensitivity analysis is to adopt the Taguchi experimental design technology and point out the dimension of the quadrature domain with the largest influence on the computational accuracy of the present MLPGM for shells and give out the optimum combination of these factors. A few examples are given to verify the reliability and good convergence of MLPGM for shell problems compared to the theoretical or the finite element results.展开更多
Fast Radio Bursts (FRBs) are intense radio flashes from the sky that are characterized by mil- lisecond durations and Jansky-level flux densities. We carried out a statistical analysis on FRBs that have been discove...Fast Radio Bursts (FRBs) are intense radio flashes from the sky that are characterized by mil- lisecond durations and Jansky-level flux densities. We carried out a statistical analysis on FRBs that have been discovered. Their mean dispersion measure, after subtracting the contribution from the interstellar medium of our Galaxy, is found to be ~ 660 pc cm-3, supporting their being from a cosmological origin. Their energy released in the radio band spans about two orders of magnitude, with a mean value of ~ 10-39 erg. More interestingly, although the study of FRBs is still in a very early phase, the published collection of FRBs enables us to derive a useful intensity distribution function. For the 16 non-repeating FRBs detected by the Parkes telescope and the Green Bank Telescope, the intensity distribution can be described as dN/dFobs = (4.1± 1.3) × 103 F-obs1.1±0.2 sky-1 d-l, where Fobs is the observed radio obs fluence in units of Jy ms. Here the power-law index is significantly flatter than the expected value of 2.5 for standard candles distributed homogeneously in a flat Euclidean space. Based on this intensity distribution function, the Five-hundred-meter Aperture Spherical radio Telescope (FAST) is predicted to be able to detect about five FRBs for every 1000 h of observation time.展开更多
文摘Emotion has a nearly decisive role in behavior, which will directly affect netizens’ views on food safety public opinion events, thereby affecting the development direction of public opinion on the event, and it is of great significance for food safety network public opinion to predict emotional trends to do a good job in food safety network public opinion guidance. In this paper, the dynamic text representation method XLNet is used to generate word vectors with context-dependent dependencies to distribute the text information of food safety network public opinion. Then, the word vector is input into the CNN-BiLSTM network for local semantic feature and context semantic extraction. The attention mechanism is introduced to give different weights according to the importance of features, and the emotional tendency analysis is carried out. Based on sentiment analysis, sentiment value time series data is obtained, and a time series model is constructed to predict sentiment trends. The sentiment analysis model proposed in this paper can well classify the sentiment of food safety network public opinion, and the time series model has a good effect on the prediction of food safety network public opinion sentiment trend. .
文摘The Submillimeter Wave Astronomy Satellite(SWAS)was the first space telescope capable of high spectral resolution observations of terahertz spectral lines.We have investigated the integration ability of its two receivers and spectrometer during five and a half years of on-orbit operation.The CI,O_(2),H_(2)O,and^(13)CO spectra taken toward all observed Galactic sources were analyzed.The present results are based on spectra with a total integration time of up to 2.72×10~4hr(■10~8s).The noise in the spectra is generally consistent with that expected from the radiometer equation,without any sign of approaching a noise floor.This noise performance reflects the extremely stable performance of the passively cooled front end as well as other relevant components in the SWAS instrument throughout its mission lifetime.

文摘BACKGROUND China's most frequent malignancy is gastric cancer(GC),which has a very poor survival rate,and the survival rate for patients with advanced GC is dismal.Pyroptosis has been connected to the genesis and development of cancer.The function of pyroptosis-related long non-coding RNAs(PRLs)in GC,on the other hand,remains uncertain.AIM To explore the construction and comprehensive analysis of the prognostic characteristics of long non-coding RNA(lncRNA)related to pyroptosis in GC patients.METHODS The TCGA database provided us with 352 stomach adenocarcinoma samples,and we obtained 28 pyroptotic genes from the Reactome database.We examined the correlation between lncRNAs and pyroptosis using the Pearson correlation coefficient.Prognosis-related PRLs were identified through univariate Cox analysis.A predictive signature was constructed using stepwise Cox regression analysis,and its reliability and independence were assessed.To facilitate clinical application,a nomogram was created based on this signature.we analyzed differences in immune cell infiltration,immune function,and checkpoints between the high-risk group(HRG)and low-risk group(LRG).RESULTS Five hundred and twenty-three PRLs were screened from all lncRNAs(absolute correlation coefficient>0.4,P<0.05).Nine PRLs were included in the risk prediction signature that was created through stepwise Cox regression analysis.We determined the risk score for GC patients and employed the median value as the dividing line between HRG and LRG.The ability of the risk signature to predict the overall survival(OS)of GC is demonstrated by the Kaplan-Meier analysis,risk curve,receiver operating characteristic curve,and decision curve analysis curve.The risk signature was shown to be an independent prognostic factor for OS in both univariate and multivariate Cox regression analyses.HRG showed a more efficient local immune response or modulation compared to LRG,as indicated by the predicted signal pathway analysis and examination of immune cell infiltration,function,and checkpoints(P<0.05).CONCLUSION In general,we have created a brand-new prognostic signature using PRLs,which may provide ideas for immunotherapy in patients with GC.

文摘AIM: To observe the inhibition of hepatitis B virus (HBV) replication and expression by combination of siRNA and lamivudine in HepG2.2.15 cells. METHODS: Recombinant plasmid psil-HBV was constructed and transfected into HepG2.2.15 cells. The transfected cells were cultured in lamivudine-containing medium (0.05 μmol/L) and harvested at 48, 72 and 96 h. The concentration of HBeAg and HBsAg was determined using ELISA. HBV DNA replication was examined by real- time PCR and the level of HBV mRNA was measured by RT-PCR. RESULTS: In HepG2.2.15 cells treated with combination of siRNA and lamivudine, the secretion of HBeAg and HBsAg into the supernatant was found to be inhibited by 91.80% and 82.40% (2.89 ± 0.48 vs 11.73 ± 0.38, P < 0.05; 4.59 ± 0.57 vs 16.25 ± 0.48, P < 0.05) at 96 h, respectively; the number of HBV DNA copies within culture medium was also significantly decreased at 96 h (1.04 ± 0.26 vs 8.35 ± 0.33, P < 0.05). Moreover, mRNA concentration in HepG2.2.15 cells treated with combination of siRNA and lamivudine was obviously lower compared to those treated either with siRNA or lamivudine (19.44 ± 0.17 vs 33.27 ± 0.21 or 79.9 ± 0.13, P < 0.05). CONCLUSION: Combination of siRNA and lamivudine is more effective in inhibiting HBV replication as compared to the single use of siRNA or lamivudine in HepG2.2.15 cells.

文摘The Five-hundred-meter Aperture Spherical radio Telescope(FAST) is by far the largest telescope of any kind ever built. FAST produced its first light in September 2016 and it is now under commissioning, with normal operation to commence in late 2019. During testing and early science operation, FAST has started making astronomical discoveries, particularly pulsars of various kinds, including millisecond pulsars, binaries, gamma-ray pulsars, etc. The papers in this mini-volume propose ambitious observational projects to advance our knowledge of astronomy, astrophysics and fundamental physics in many ways.Although it may take FAST many years to achieve all the goals explained in these papers, taken together they define a powerful strategic vision for the next decade.

文摘We developed a GPU based single-pulse search pipeline(GSP)with a candidate-archiving database.Largely based upon the infrastructure of the open source PulsaR Exploration and Search Toolkit(PRESTO),GSP implements GPU acceleration of the de-dispersion and integrates a candidate-archiving database.We applied GSP to the data streams from the Commensal Radio Astronomy FAST Survey(CRAFTS),which resulted in quasi-real-time processing.The integrated candidate database facilitates synergistic usage of multiple machine-learning tools and thus improves efficient identification of radio pulsars such as rotating radio transients(RRATs)and fast radio bursts(FRBs).We first tested GSP on pilot CRAFTS observations with the FAST Ultra-Wide Band(UWB)receiver.GSP detected all pulsars known from the the Parkes multibeam pulsar survey in the corresponding sky area covered by the FAST-UWB.GSP also discovered 13 new pulsars.We measured the computational efficiency of GSP to be~120 times faster than the original PRESTO and~60 times faster than an MPI-parallelized version of PRESTO.

文摘We report here Atacama Large Millimeter/submillimeter Array(ALMA)N2H+(1-0)images of the Orion Molecular Cloud 2 and 3(OMC-2/3)with high angular resolution(3"or 1200 au)and high spatial dynamic range.Combining a dataset from the ALMA main array,Atacama Compact Array(ACA),Nobeyama 45-m Telescope and Very Large Array(VLA)(providing temperature measurement on matching scales),we find that most of the dense gas in OMC-2/3 is subsonic(σQNT/cs=0.62)with a mean line width(△v)of 0.39 kms-1 full width at half maximum(FWHM).This is markedly different from the majority of previous observations of massive star-forming regions.In contrast,line widths from the Nobeyama Telescope are transonic at 0.69 km s-1(σNT/cs=1.08).We demonstrated that the larger line widths obtained by the single-dish telescope arose from unresolved sub-structures within their respective beams.The dispersions from larger scalesσls(as traced by the Nobeyama Telescope)can be decomposed into three components such thatσls2=σss2+σbm2+σrd2,where small-scaleσss is the line dispersion of each ALMA beam,bulk motionσbm is dispersion between peak velocity of each ALMA beam andσrd is the residual dispersion.Such decomposition,though purely empirical,appears to be robust throughout our data cubes.Apparent supersonic line widths,commonly found in massive molecular clouds,are thus likely due to the effect of poor spatial resolution.The observed non-thermal line dispersion(sometimes referred to as’turbulence’)transits from supersonic to subsonic at~0.05 pc scales in the OMC-2/3 region.Such transition could be commonly found with sufficient spatial(not just angular)resolution,even in regions with massive young clusters,such as the Orion molecular clouds studied here.

文摘Neutral hydrogen clouds are known to exist in the Universe, however their spatial distributions and physical properties are poorly understood. Such missing information can be studied by the new generation of Chinese radio telescopes through a blind search of 21-cm absorption systems. We forecast the capabilities of surveys of 21-cm absorption systems by two representative radio telescopes in China - the Five-hundred-meter Aperture Spherical radio Telescope (FAST) and Tianlai 21-cm cosmology experiment (Tianlai). Facilitated by either the high sensitivity (FAST) or wide field of view (Tianlai) of these telescopes, more than a thousand 21-cm absorption systems can be discovered in a few years, representing orders of magnitude improvement over the cumulative discoveries in the past half a century.

文摘The increasing radio frequency interference(RFI)is a well-recognized problem in radio astronomy research.Pulsars and Fast Radio Bursts(FRBs)are high-priority science targets of the ongoing Commercial Radio Astronomy FAST Survey(CRAFTS).To improve the quality of RFI removal in searches of pulsars and FRBs based on CRAFTS multi-beam data,we here propose an intuitive but powerful RFI mitigation pipeline(CCF-ST).The“CCF-ST”is a spatial filter constructed by signal cross-correlation function(CCF)and Sum-Threshold(ST)algorithm.The RFI marking result is saved in a“mask”file,a binary format for RFI masks in PRESTO.Three known pulsars,PSR B0525-21,PSR B0621-04,and PSR J0943+2252 from CRAFTS L-band 19 beams data are used for evaluation of the performance of CCF-ST in comparison with other methods,such as PRESTO’s“rfifind”,ArPLS-ST and ArPLS-SF.The result shows that CCF-ST can reduce effective data loss rate and improves the detected signal-to-noise ratio of the pulsations by~26%and~18%respectively compared with PRESTO’s“rfifind”and ArPLS-ST.The CCF-ST also has the advantage of low computational cost,e.g.,reducing the time consumption by~40%and memory consumption by~90%compared with ArPLS-SF.We expect that the new RFI mitigation and analysis toolkit(CCF-ST)demonstrated in this paper can be applied to CRAFTS and other multi-beam telescope observations to improve the data quality and efficiency of pulsar and FRB searches.

文摘We report the discovery of PSR J1909+0122 by the Five-hundred-meter Aperture Spherical Radio Telescope(FAST)as part of the Commensal Radio Astronomy FAST Survey.PSR J1909+0122 has a spin period of 1.257 s and a dispersion measure of 186.2 pc cm^(-3).The averaged pulse profile shows two distinct components.We performed a single-pulse study based on a one-hour observation at 1.25 GHz on 2021 August 23.We used a threshold of 5σ_(ep) to measure the nulling fraction(NF)as 63%±1.5%.The longitude-resolved fluctuation spectra and fast Fourier transform spectra of the binary sequences revealed the quasi-periodicity of nulling with a period of 30 rotation periods.We examined the reliability of the periodicity by comparing it to random noise injection.The NF,E,and modulation periodicity P_(M) of PSR J1909+0122 were compared with other periodic nulling pulsars,showing that the source of J1909+0122 has the second largest NF in the population.Long-term timing observations over six months were used to derive the phase-connected ephemeris of this pulsar.The measured P and P values disfavor dipolar geometry for polar gap models,and the prediction for a space-charge-limited flow model in the case of inverse Compton scattering is only just above the death line.In this work,PSR J1909+0122 has revealed possible correlations between nulling behavior and pulsar properties,which will help to shed light on the pulsar emission mechanism and its temporal evolution in future observations.

文摘We investigate the single-pulse emission variations of two pulsars,PSRs J0211+4235 and J0553+4111,observed with the Five-hundred-meter Aperture Spherical radio Telescope at the 1.25 GHz central frequency.The observation sessions span from 2020 December to 2021 July,with 21 and 22 observations for them respectively.The integrated pulse profile of PSR J0211+4235 shows that there is a weak pulse component following the main component,and PSR J0553+4111 displays a bimodal profile with a bridge component in the middle.PSR J0211+4235 presents significant nulling phenomenon with nulling duration lasting from 2 to 115 pulses and burst duration lasting from 2 to 113 pulses.The NF of each observation is determined to be 45%-55%.No emission greater than threeσis found in the mean integrated profile of all nulling pulses.In most cases,the pulse energy changes abruptly during the transition from null to burst,while in the transition from burst to null there are two trends:abrupt and gradual.We find that the nulling phenomenon of PSR J0211+4235 is periodic by the Fourier transform of the null and burst state.In addition,the single-pulse modulation characteristics of these two pulsars are investigated,and the distributions of modulation index,LRFS and 2DFS are analyzed with PSRSALSA.The left peak of PSR J0553+4111 has intensity modulation.Finally,the polarization properties of these two pulsars are obtained through polarization calibration,and their characteristics are analyzed.The possible physical mechanisms of these phenomena are discussed.

文摘Fast radio bursts(FRBs) are highly dispersed millisecond-duration radio bursts,[1,2]of which the physical origin is still not fully understood. FRB 20201124A is one of the most actively repeating FRBs. In this paper, we present the collection of 1863 burst dynamic spectra of FRB 20201124A measured with the Five-hundred-meter Aperture Spherical radio Telescope(FAST). The current collection, taken from the observation during the FRB active phase from April to June 2021, is the largest burst sample detected for any FRB so far. The standard PSRFITs format is adopted, including dynamic spectra of the burst, and the time information of the dynamic spectra, in addition, mask files help readers to identify the pulse positions are also provided. The dataset is available in Science Data Bank, with the link https://www.doi.org/10.57760/sciencedb.j00113.00076.

文摘Correlation power analysis(CPA)combined with genetic algorithms(GA)now achieves greater attack efficiency and can recover all subkeys simultaneously.However,two issues in GA-based CPA still need to be addressed:key degeneration and slow evolution within populations.These challenges significantly hinder key recovery efforts.This paper proposes a screening correlation power analysis framework combined with a genetic algorithm,named SFGA-CPA,to address these issues.SFGA-CPA introduces three operations designed to exploit CPA characteris-tics:propagative operation,constrained crossover,and constrained mutation.Firstly,the propagative operation accelerates population evolution by maximizing the number of correct bytes in each individual.Secondly,the constrained crossover and mutation operations effectively address key degeneration by preventing the compromise of correct bytes.Finally,an intelligent search method is proposed to identify optimal parameters,further improving attack efficiency.Experiments were conducted on both simulated environments and real power traces collected from the SAKURA-G platform.In the case of simulation,SFGA-CPA reduces the number of traces by 27.3%and 60%compared to CPA based on multiple screening methods(MS-CPA)and CPA based on simple GA method(SGA-CPA)when the success rate reaches 90%.Moreover,real experimental results on the SAKURA-G platform demonstrate that our approach outperforms other methods.

文摘BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cancer lacks a comprehensive analysis,and its role and mechanism in hepatocellular carcinoma(HCC)are still unclear.The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Profiling Interactive Analysis,and Tumor Immune Estimation Resource databases were used to analyze B56εexpression,prognostic mutations,somatic copy number alterations,and tumor immune characteristics in 33 tumors.The relationships between B56εexpression levels and drug sensitivity,immuno-therapy,immune checkpoints,and human leukocyte antigen(HLA)-related genes were further analyzed.Gene Set Enrichment Analysis(GSEA)was performed to reveal the role of B56εin HCC.The Cell Counting Kit-8,plate cloning,wound healing,and transwell assays were conducted to assess the effects of B56εinterference on the malignant behavior of HCC cells.RESULTS In most tumors,B56εexpression was upregulated,and high B56εexpression was a risk factor for adrenocortical cancer,HCC,pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma(all P<0.05).B56εexpression levels were correlated with a variety of immune cells,such as T helper 17 cells,B cells,and macro-phages.There was a positive correlation between B56εexpression levels with immune checkpoint genes and HLA-related genes(all P<0.05).The expression of B56εwas negatively correlated with the sensitivity of most chemotherapy drugs,but a small number showed a positive correlation(all P<0.05).GSEA analysis showed that B56εexpression was related to the cancer pathway,p53 downstream pathway,and interleukin-mediated signaling in HCC.Knockdown of B56εexpression in HCC cells inhibited the proliferation,migration,and invasion capacity of tumor cells.Core Tip:The expression of protein phosphatase 2A(PP2A)subunit B56εis up-regulated in most tumors,and its high expression is a risk factor for adrenocortical cancer,hepatocellular carcinoma(HCC),pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma.B56εexpression levels correlate with immune cells,immune checkpoint genes,human leukocyte antigen-related genes,and the sensitivity of chemotherapy drugs.In HCC,B56εexpression is related to the cancer pathway.Knockdown of B56εexpression in HCC cells can inhibit the proliferation,migration and invasion capacity of tumor cells.Our study supports PP2A subunit B56εas a prognostic marker and potential therapeutic target for HCC.

文摘Recent submillimeter dust thermal emission observations have unveiled a significant number of inter-arm massive molecular clouds in M31.However,the effectiveness of this technique is limited to its sensitivity,making it challenging to study more distant galaxies.This study introduces an alternative approach,utilizing optical extinctions derived from space-based telescopes,with a focus on the forthcoming China Space Station Telescope(CSST).We first demonstrate the capability of this method by constructing dust extinction maps for 17 inter-arm massive molecular clouds in M31 using the Panchromatic Hubble Andromeda Treasury data.Our analysis reveals that inter-arm massive molecular clouds with an optical extinction(A_(V)) greater than 1.6 mag exhibit a notable A_(V) excess,facilitating their identification.The majority of these inter-arm massive molecular clouds show an A_(V) around 1 mag,aligning with measurements from our JCMT data.Further validation using a mock CSST RGB star catalog confirms the method's effectiveness.We show that the derived A_(V)values using CSST z and y photometries align more closely with the input values.Molecular clouds with A_(V)> 1.6 mag can also be identified using the CSST mock data.We thus claim that future CSST observation clouds provide an effective way for the detection of inter-arm massive molecular clouds with significant optical extinction in nearby galaxies.

文摘AIM: To observe the inhibition of hepatitis B virus (HBV) replication and expression in HepG2.2.15 cells by combination of small interfering RNAs (siRNAs). METHODS: Recombinant plasmid psiI-HBV was constructed and transfected into HepG2.2.15 cells. At 48 h, 72 h and 96 h after transfection, culture media were collected and cells were harvested for HBV replication assay. HBsAg and HBeAg in the cell culture medium were detected by enzyme-linked immunoadsorbent assay (ELISA). Intracellular viral DNA and covalently closed circular DNA (cccDNA) were quantified by real-time polymerase chain reaction (PCR). HBV viral mRNA was reverse transcribed and quantified by reverse-transcript PCR (RT-PCR). RESULTS: siRNAs showed marked anti-HBV effects. siRNAs could specifically inhibit the expression of HBsAg and the replication of HBV DNA in a dosedependent manner. Furthermore, combination of siRNAs, compared with individual use of each siRNA, exerted a stronger inhibition on antigen expression and viral replication. More importantlycombination of siRNAs significantly suppressed HBV cccDNA amplification. CONCLUSION: Combination of siRNAs mediates a stronger inhibition on viral replication and antigenexpression in HepG2.2.15 cells, especially on cccDNA amplification.

文摘The objectives of this study are to employ the meshless local Petrov-Galerkin method (MLPGM) to solve three-dimensional shell problems. The computational accuracy of MLPGM for shell problems is affected by many factors, including the dimension of compact support domain, the dimension of quadrture domain, the number of integral cells and the number of Gauss points. These factors' sensitivity analysis is to adopt the Taguchi experimental design technology and point out the dimension of the quadrature domain with the largest influence on the computational accuracy of the present MLPGM for shells and give out the optimum combination of these factors. A few examples are given to verify the reliability and good convergence of MLPGM for shell problems compared to the theoretical or the finite element results.

文摘Fast Radio Bursts (FRBs) are intense radio flashes from the sky that are characterized by mil- lisecond durations and Jansky-level flux densities. We carried out a statistical analysis on FRBs that have been discovered. Their mean dispersion measure, after subtracting the contribution from the interstellar medium of our Galaxy, is found to be ~ 660 pc cm-3, supporting their being from a cosmological origin. Their energy released in the radio band spans about two orders of magnitude, with a mean value of ~ 10-39 erg. More interestingly, although the study of FRBs is still in a very early phase, the published collection of FRBs enables us to derive a useful intensity distribution function. For the 16 non-repeating FRBs detected by the Parkes telescope and the Green Bank Telescope, the intensity distribution can be described as dN/dFobs = (4.1± 1.3) × 103 F-obs1.1±0.2 sky-1 d-l, where Fobs is the observed radio obs fluence in units of Jy ms. Here the power-law index is significantly flatter than the expected value of 2.5 for standard candles distributed homogeneously in a flat Euclidean space. Based on this intensity distribution function, the Five-hundred-meter Aperture Spherical radio Telescope (FAST) is predicted to be able to detect about five FRBs for every 1000 h of observation time.
