We report the synthesis and characterization of a single-molecule magnet composed of triangular clusters of dysprosium ions.The structural study shows that the symmetry changes from one polar point group(mm2)at room t...We report the synthesis and characterization of a single-molecule magnet composed of triangular clusters of dysprosium ions.The structural study shows that the symmetry changes from one polar point group(mm2)at room temperature to another polar point group(m)at low temperature.Magnetic studies and theory calculations illustrate that the vortex distribution of magnetic dipoles in the triangular dysprosium clusters forms a toroidal magnetic moment.Interestingly,the analysis of AC magnetic susceptibility reveals the coexistence of three distinct magnetic relaxation processes,corresponding to the Raman,Orbach,and QTM relaxation pathways,respectively.The sum of three modified Debye functions is successfully used to describe the multiple relaxation behavior.展开更多

The anharmonicity of lattice vibration is mainly responsible for the coefficient of thermal expansion(CTE)of materials.External stimuli,such as magnetic and electric fields,thus cannot effectively change the CTE,much ...The anharmonicity of lattice vibration is mainly responsible for the coefficient of thermal expansion(CTE)of materials.External stimuli,such as magnetic and electric fields,thus cannot effectively change the CTE,much less the sign variation from positive to negative or vice versa.In this study,we report significant magnetic field effects on the CTE of zircon-and scheelite-type DyCrO_(4) prepared at ambient and high pressures,respectively.At zero field,the zircon-type DyCrO_(4) exhibits a negative CTE below the ferromagnetic-order temperature of 23 K.With increasing field up to≥1.0 T,however,the sign of the CTE changes from negative to positive.In the scheelite phase,magnetic field can change the initially positive CTE to be negative with a field up to 2.0 T,and then a reentrant positive CTE is induced by enhanced fields≥3.5 T.Both zircon and scheelite phases exhibit considerable magnetostrictive effects with the absolute values as high as~800 ppm at 2 K and 10 T.The strong spin–lattice coupling is discussed to understand the unprecedented sign changes of the CTE caused by applying magnetic fields.The current DyCrO_(4) provides the first example of field-induced sign change of thermal expansion,opening up a way to readily control the thermal expansion beyond the conventional chemical substitution.展开更多
Gibberellins(GAs)play crucial roles in a wide range of developmental processes and stress responses in plants.However,the roles of GA-responsive genes in tomato(Solanum lycopersicum)fruit development remain largely un...Gibberellins(GAs)play crucial roles in a wide range of developmental processes and stress responses in plants.However,the roles of GA-responsive genes in tomato(Solanum lycopersicum)fruit development remain largely unknown.Here,we identify 17 GASA(Gibberellic Acid-Stimulated Arabidopsis)family genes in tomato.These genes encode proteins with a cleavable signal peptide at their N terminus and a conserved GASA domain at their C terminus.The expression levels of all tomato GASA family genes were responsive to exogenous GA treatment,but adding ethylene eliminated this effect.Comprehensive expression profiling of SlGASA family genes showed that SlGASA1 follows a ripening-associated expression pattern,with low expression levels during fruit ripening,suggesting it plays a negative role in regulating ripening.Overexpressing SlGASA1 using a ripening-specific promoter delayed the onset of fruit ripening,whereas SlGASA1-knockdown fruits displayed accelerated ripening.Consistent with their delayed ripening,SlGASA1-overexpressing fruits showed significantly reduced ethylene production and carotenoid contents compared to the wild type.Moreover,ripening-related genes were downregulated in SlGASA1-overexpressing fruits but upregulated in SlGASA1-knockdown fruits compared to the wild type.Yeast two-hybrid,co-immunoprecipitation,transactivation,and DNA pull-down assays indicated that SlGASA1 interacts with the key ripening regulator FRUITFULL1 and represses its activation of the ethylene biosynthesis genes ACS2 and ACO1.Our findings shed new light on the role and mode of action of a GA-responsive gene in tomato fruit ripening.展开更多

Methods: To examine the expressed level of...Objective: To investigate the relationship between P-glycoprotein (P-gp) and adhesion molecule CD44 expression as well as their clinical significance in esophageal carcinoma. Methods: To examine the expressed level of P-gp and CD44 by flow cytometry (FCM) in the operated samples of 70 cases with esophageal carcinoma and their normal mucosa of esophageal incision, and to evaluate their relationship with clinicopathological factors. Results: Among the 70 cases with esophageal car- cinoma, the expression of P-gp in the 27 cases (38.6%) was negative (positive cells <25%); 11 cases (15.7%) were 25%–40% expression of P-gp positive cells; 14 cases (20%) were 41%–60% expression of P-gp positive cells; 18 cases (25.7%) were the high expression (positive cells >60%) of P-gp. Of the cases with the tumor sizes being more than 4 cm, the expression of CD44 showed a significant difference (P<0.05) in 25 cases with P-gp positive, compared with 19 cases with P-gp negative. Of the cases with high-mild differentiated esophageal carcinoma, the expression of CD44 showed a significant difference (P<0.05) in 22 cases with P-gp positive, compared with 17 cases with P-gp negative. Of the cases with clinical III--IV stage, the expression of CD44 showed a significant difference (P<0.05) in 26 cases with P-gp positive, compared with 10 cases with P-gp negative. Of the cases with lymph node metastasis, the CD44 expression showed a significant difference (P=0.050) in 27 cases with P-gp positive, compared with 11 cases with P-gp negative. Of the cases of the patients’ age being more than 56 years, the expression of CD44 showed a significant difference (P<0.01) in 27 cases with P-gp positive, compared with 12 cases with P-gp negative. When the P-gp and CD44 expression were positive, the clinical II stage and III–IV stage in esophageal carcinoma was showed a significant difference (P<0.05). Conclusion: When the CD44 and P-gp both have the positive high expression, it will be signifi- cantly associated with the esophageal carcinoma progression and metastasis, so both were a positive expression in esophageal carcinoma, it might suggest a poor and unfavorable prognosis result.展开更多

Male Tssk6 knockout mice are infertile owing to sperrnatogenic impairment,including sperm count reduction,a decrease in motile sperm number and motility rates,and an increase in the number of sperms with abnormal morphology. We investigated the possible association between variations oftbe TSSK6 gene and spermatogenic impairment in humans. Mutation screening of TSSK6 was carried out in 519 patients with azoospermia (n = 273) or severe oligozoospermia (n = 246) and in 359 controls with normozoospermia by denaturing high-performance liquid chromatography and DNA sequencing. The frequencies of alleles and genotypes of gene polymorphism were compared between patients and controls. A novel triallelic polymorphism in TSSK6,c.822+126T〉G/C,was identified. The frequencies of genotype TT and allele T were increased dramatically in infertile patients compared with controls,whereas genotype TG,allele G and allele C frequencies were significantly higher in controls than in patients. Further study revealed that the allele C frequency of controls was remarkably higher than that of patients with oligospermia. Our findings,for the first time,suggested an association of c.822+I26T〉G/C in TSSK6 with spermatogenic impairment in humans in which allele T may be a risk factor for male infertility,while alleles C and G may decrease susceptibility to male infertility.展开更多

This study aimed to invest...Objective: Chronic inflammation plays a fatal role in tumor metastasis. Pterostilbene(PTE) is a natural dimethylated analogue of resveratrol with anticancer and anti-inflammatory activities. This study aimed to investigate the inhibitory effect of PTE on inflammation-associated metastasis and explore the underlying mechanisms.Methods: Lipopolysaccharide(LPS)-induced lung inflammation and melanoma metastasis models were established in mice. After PTE treatment for four weeks, the organ index, histological changes, proinflammatory cytokines, and the expression and activity of neutrophil elastase(NE), a biomarker of neutrophil influx in the lungs, were analysed. Additionally, direct effects of PTE on NE-induced B16 cell migration were explored in wound healing and Transwell assays, and the expression of thrombospondin-1(TSP-1) and epithelial-mesenchymal transition(EMT) markers were also detected.Results: PTE obviously attenuated the LPS-induced metastasis of circulatory B16 cells to lungs by reducing the number of metastatic nodules on the lung surfaces and the lung weight/body weight ratio. PTE treatment also significantly reduced LPS-activated increase levels of tumor necrosis factor(TNF)-a and interleukin(IL)-6 in the lungs of tumor-bearing mice. In addition, increased expression and enzyme activity of NE and decreased expression of TSP-1 were observed, and these were blocked by PTE. In vitro, PTE at concentrations without cytotoxicity also markedly suppressed NE-triggered B16 cell migration, prevented NE-induced TSP-1 proteolysis and reversed the expression of vimentin, N-cadherin and Ecadherin.Conclusion: PTE could block inflammation-enhanced tumor metastasis, and the underlying mechanism might be associated with the inhibition of NE-mediated TSP-1 degradation.展开更多

A novel program based on bootstrap for DOA estimation is performed to compared with different number of snapshots in this paper. We have resampled the received signals for 200-1000 times to create new data, therefore the arrival angle is estimated by the music algorithm in the conditions of confidence interval. The demo results show that higher estimation probability and smaller mean square error can be achieved in the situation of fewer snapshots received by passive radar system than that of traditional algorithm.展开更多

Respiratory syncytial virus(RSV)is a nonsegmented,negative strand RNA virus that has caused severe lower respiratory tract infections of high mortality rates in infants and the elderly,yet no effective vaccine or anti...Respiratory syncytial virus(RSV)is a nonsegmented,negative strand RNA virus that has caused severe lower respiratory tract infections of high mortality rates in infants and the elderly,yet no effective vaccine or antiviral therapy is available.The RSV genome encodes the nucleoprotein(N)that forms helical assembly to encapsulate and protect the RNA genome from degradation,and to serve as a template for transcription and replication.Previous crystal structure revealed a decameric ring architecture of N in complex with the cellular RNA(N-RNA)of 70 nucleotides(70-nt),whereas cryo-ET reconstruction revealed a low-resolution left-handed filament,in which the crystal monomer structure was docked with the helical symmetry applied to simulate a nucleocapsid-like assembly of RSV.However,the molecular details of RSV nucleocapsid assembly remain unknown,which continue to limit our complete understanding of the critical interactions involved in the nucleocapsid and antiviral development that may target this essential process during the viral life cycle.Here we resolve the near-atomic cryo-EM structure of RSV N-RNA that represents roughly one turn of the helical assembly that unveils critical interaction interfaces of RSV nucleocapsid and may facilitate development of RSV antiviral therapy.展开更多

METHODS: Transwell chamber invasive assays were used to investiga...OBJECTIVE: To study the anticancer mechanism of polyphyllin I (PPI), a Traditional Chinese Medicine, on the ovarian cancer cell line HO-8910PM invitro. METHODS: Transwell chamber invasive assays were used to investigate the inhibitory capacity of PPI on HO-8910PM metastasis. Gene expression profiling chips was used to screen differentially expressed genes between experiment group and control group. Reverse transcription PCR and Western blotting were used to determine mRNA and protein levels. RESULTS: With increasing PPI concentration, the metastatic capacity of cells decreased, with significance differences between the experimental and control groups (P<0.01) as well as between two concentration groups. Gene expression profiling identified 123 differentially expressed genes, of which 70 were downregulated and 53 were upregulated. The genes were involved in multiple signal transduction pathways, including apoptosis, proliferation and metastasis. Real-time PCR (RT-PCR) showed that differential genes PIK3C2B, Caspase 9and Wnt5A were downregulated with increasing PPI, showing an evident dose-effect relationship. The c-Jun was an exception. As the PPI dosage increased and the exposure time was extended, c-Jun relative expression showed an upward trend. There were significant differences between the experiment and control (P<0.05). Western blot analyses showed that PPI treatment decreased levels of Caspase 9, Wnt5A and PIK3C2B and increased activatedCaspase9,c-Junandp-c-Junexpressionlevels. CONCLUSION: PPI has strong antitumor and anti transfer activity. It can activate c-Jun expression and the JNK signaling pathway, elicit cell apoptosis via the mitochondrial-mediated Caspase activation pathway, and finally inhibit tumor growth and mi- gration in vitro. The downregulation of PIK3C2B and Wnt5A jointly inhibit the proliferation and metastasis of HO-8910PM. PPI may be a novel treatment for ovarian cancer.展开更多
Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences be...Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.展开更多

文摘Gibberellins(GAs)play crucial roles in a wide range of developmental processes and stress responses in plants.However,the roles of GA-responsive genes in tomato(Solanum lycopersicum)fruit development remain largely unknown.Here,we identify 17 GASA(Gibberellic Acid-Stimulated Arabidopsis)family genes in tomato.These genes encode proteins with a cleavable signal peptide at their N terminus and a conserved GASA domain at their C terminus.The expression levels of all tomato GASA family genes were responsive to exogenous GA treatment,but adding ethylene eliminated this effect.Comprehensive expression profiling of SlGASA family genes showed that SlGASA1 follows a ripening-associated expression pattern,with low expression levels during fruit ripening,suggesting it plays a negative role in regulating ripening.Overexpressing SlGASA1 using a ripening-specific promoter delayed the onset of fruit ripening,whereas SlGASA1-knockdown fruits displayed accelerated ripening.Consistent with their delayed ripening,SlGASA1-overexpressing fruits showed significantly reduced ethylene production and carotenoid contents compared to the wild type.Moreover,ripening-related genes were downregulated in SlGASA1-overexpressing fruits but upregulated in SlGASA1-knockdown fruits compared to the wild type.Yeast two-hybrid,co-immunoprecipitation,transactivation,and DNA pull-down assays indicated that SlGASA1 interacts with the key ripening regulator FRUITFULL1 and represses its activation of the ethylene biosynthesis genes ACS2 and ACO1.Our findings shed new light on the role and mode of action of a GA-responsive gene in tomato fruit ripening.

文摘Objective: To investigate the relationship between P-glycoprotein (P-gp) and adhesion molecule CD44 expression as well as their clinical significance in esophageal carcinoma. Methods: To examine the expressed level of P-gp and CD44 by flow cytometry (FCM) in the operated samples of 70 cases with esophageal carcinoma and their normal mucosa of esophageal incision, and to evaluate their relationship with clinicopathological factors. Results: Among the 70 cases with esophageal car- cinoma, the expression of P-gp in the 27 cases (38.6%) was negative (positive cells <25%); 11 cases (15.7%) were 25%–40% expression of P-gp positive cells; 14 cases (20%) were 41%–60% expression of P-gp positive cells; 18 cases (25.7%) were the high expression (positive cells >60%) of P-gp. Of the cases with the tumor sizes being more than 4 cm, the expression of CD44 showed a significant difference (P<0.05) in 25 cases with P-gp positive, compared with 19 cases with P-gp negative. Of the cases with high-mild differentiated esophageal carcinoma, the expression of CD44 showed a significant difference (P<0.05) in 22 cases with P-gp positive, compared with 17 cases with P-gp negative. Of the cases with clinical III--IV stage, the expression of CD44 showed a significant difference (P<0.05) in 26 cases with P-gp positive, compared with 10 cases with P-gp negative. Of the cases with lymph node metastasis, the CD44 expression showed a significant difference (P=0.050) in 27 cases with P-gp positive, compared with 11 cases with P-gp negative. Of the cases of the patients’ age being more than 56 years, the expression of CD44 showed a significant difference (P<0.01) in 27 cases with P-gp positive, compared with 12 cases with P-gp negative. When the P-gp and CD44 expression were positive, the clinical II stage and III–IV stage in esophageal carcinoma was showed a significant difference (P<0.05). Conclusion: When the CD44 and P-gp both have the positive high expression, it will be signifi- cantly associated with the esophageal carcinoma progression and metastasis, so both were a positive expression in esophageal carcinoma, it might suggest a poor and unfavorable prognosis result.

文摘TSSK6 is a member of the testis-specific serine/threonine kinase family. Male Tssk6 knockout mice are infertile owing to sperrnatogenic impairment,including sperm count reduction,a decrease in motile sperm number and motility rates,and an increase in the number of sperms with abnormal morphology. We investigated the possible association between variations oftbe TSSK6 gene and spermatogenic impairment in humans. Mutation screening of TSSK6 was carried out in 519 patients with azoospermia (n = 273) or severe oligozoospermia (n = 246) and in 359 controls with normozoospermia by denaturing high-performance liquid chromatography and DNA sequencing. The frequencies of alleles and genotypes of gene polymorphism were compared between patients and controls. A novel triallelic polymorphism in TSSK6,c.822+126T〉G/C,was identified. The frequencies of genotype TT and allele T were increased dramatically in infertile patients compared with controls,whereas genotype TG,allele G and allele C frequencies were significantly higher in controls than in patients. Further study revealed that the allele C frequency of controls was remarkably higher than that of patients with oligospermia. Our findings,for the first time,suggested an association of c.822+I26T〉G/C in TSSK6 with spermatogenic impairment in humans in which allele T may be a risk factor for male infertility,while alleles C and G may decrease susceptibility to male infertility.

This improvement has led to the discovery of new molecular markers for cancer diagnosis and prognosis as well as new molecular targets for cancer treatment and intervention. Continuous emergence of some new developing area in molecular profi ling, new therapeutic agents, tissue microenvironment and systems biology have made signifi cant progress in clinical oncology. Clinical research and investi-gation that focus on these new developments have begun to show exciting results that indicate future promises in improving patient management and survival.
文摘With the rapid development of global transportation track equipment, the running speed of modern rail transit equipment keeps refreshing records. High-speed EMUs, inter-city EMUs and urban rail vehicles have become the main types of domestic passenger transport vehicles, especially the lightweight production and manufacture of high-speed EMUs and urban rail vehicles, which are the main topics of railway transport modernization research and discussion. Track manufacturing is a successful example of China's industrial transformation and strategic restructuring through systematic innovation since the reform and opening up. In this paper, the application of some advanced manufacturing technologies in the track equipment manufacturing industry and the opportunities and challenges faced by the whole track equipment manufacturing industry were mainly studied, hoping to provide some help for the development of the track equipment manufacturing industry in China.

文摘Objective: Chronic inflammation plays a fatal role in tumor metastasis. Pterostilbene(PTE) is a natural dimethylated analogue of resveratrol with anticancer and anti-inflammatory activities. This study aimed to investigate the inhibitory effect of PTE on inflammation-associated metastasis and explore the underlying mechanisms.Methods: Lipopolysaccharide(LPS)-induced lung inflammation and melanoma metastasis models were established in mice. After PTE treatment for four weeks, the organ index, histological changes, proinflammatory cytokines, and the expression and activity of neutrophil elastase(NE), a biomarker of neutrophil influx in the lungs, were analysed. Additionally, direct effects of PTE on NE-induced B16 cell migration were explored in wound healing and Transwell assays, and the expression of thrombospondin-1(TSP-1) and epithelial-mesenchymal transition(EMT) markers were also detected.Results: PTE obviously attenuated the LPS-induced metastasis of circulatory B16 cells to lungs by reducing the number of metastatic nodules on the lung surfaces and the lung weight/body weight ratio. PTE treatment also significantly reduced LPS-activated increase levels of tumor necrosis factor(TNF)-a and interleukin(IL)-6 in the lungs of tumor-bearing mice. In addition, increased expression and enzyme activity of NE and decreased expression of TSP-1 were observed, and these were blocked by PTE. In vitro, PTE at concentrations without cytotoxicity also markedly suppressed NE-triggered B16 cell migration, prevented NE-induced TSP-1 proteolysis and reversed the expression of vimentin, N-cadherin and Ecadherin.Conclusion: PTE could block inflammation-enhanced tumor metastasis, and the underlying mechanism might be associated with the inhibition of NE-mediated TSP-1 degradation.

文摘The bootstrap technique is a powerful method for assessing the accuracy of parameters estimator, that have been widely applied on statistical and signal processing problems. A novel program based on bootstrap for DOA estimation is performed to compared with different number of snapshots in this paper. We have resampled the received signals for 200-1000 times to create new data, therefore the arrival angle is estimated by the music algorithm in the conditions of confidence interval. The demo results show that higher estimation probability and smaller mean square error can be achieved in the situation of fewer snapshots received by passive radar system than that of traditional algorithm.

文摘Respiratory syncytial virus(RSV)is a nonsegmented,negative strand RNA virus that has caused severe lower respiratory tract infections of high mortality rates in infants and the elderly,yet no effective vaccine or antiviral therapy is available.The RSV genome encodes the nucleoprotein(N)that forms helical assembly to encapsulate and protect the RNA genome from degradation,and to serve as a template for transcription and replication.Previous crystal structure revealed a decameric ring architecture of N in complex with the cellular RNA(N-RNA)of 70 nucleotides(70-nt),whereas cryo-ET reconstruction revealed a low-resolution left-handed filament,in which the crystal monomer structure was docked with the helical symmetry applied to simulate a nucleocapsid-like assembly of RSV.However,the molecular details of RSV nucleocapsid assembly remain unknown,which continue to limit our complete understanding of the critical interactions involved in the nucleocapsid and antiviral development that may target this essential process during the viral life cycle.Here we resolve the near-atomic cryo-EM structure of RSV N-RNA that represents roughly one turn of the helical assembly that unveils critical interaction interfaces of RSV nucleocapsid and may facilitate development of RSV antiviral therapy.

文摘Dear Editor,Since its appearance in May 2022,monkeypox has spread to>100 countries and afflicted tens of thousands of people.Individuals infected with monkeypox present a fever,an extensive characteristic rash,and usually swollen lymph nodes(Chatterjee et al.,2022).The number of confirmed cases worldwide continues to grow at a rapid rate,but the treatment to this highly infectious viral disease is still very limited.Identification of new targeted-therapies will be crucial to control of this emerging public health threat.

文摘Colorectal cancer(CRC)is the third most common cancer and the second leading cause of cancer death worldwide.Clinical research results have provided more treatment opportunities for CRC patients,showing that an optimal combination of existing drugs and new drugs is needed to mitigate the burden of this disease.In this review,we have summarized recent advances in drug clinical research for CRC in 2022,including chemotherapy,targeted therapy,and immunotherapy,to find opportunities for substantial improvements in drug discovery and clinical development methods.

文摘OBJECTIVE: To study the anticancer mechanism of polyphyllin I (PPI), a Traditional Chinese Medicine, on the ovarian cancer cell line HO-8910PM invitro. METHODS: Transwell chamber invasive assays were used to investigate the inhibitory capacity of PPI on HO-8910PM metastasis. Gene expression profiling chips was used to screen differentially expressed genes between experiment group and control group. Reverse transcription PCR and Western blotting were used to determine mRNA and protein levels. RESULTS: With increasing PPI concentration, the metastatic capacity of cells decreased, with significance differences between the experimental and control groups (P<0.01) as well as between two concentration groups. Gene expression profiling identified 123 differentially expressed genes, of which 70 were downregulated and 53 were upregulated. The genes were involved in multiple signal transduction pathways, including apoptosis, proliferation and metastasis. Real-time PCR (RT-PCR) showed that differential genes PIK3C2B, Caspase 9and Wnt5A were downregulated with increasing PPI, showing an evident dose-effect relationship. The c-Jun was an exception. As the PPI dosage increased and the exposure time was extended, c-Jun relative expression showed an upward trend. There were significant differences between the experiment and control (P<0.05). Western blot analyses showed that PPI treatment decreased levels of Caspase 9, Wnt5A and PIK3C2B and increased activatedCaspase9,c-Junandp-c-Junexpressionlevels. CONCLUSION: PPI has strong antitumor and anti transfer activity. It can activate c-Jun expression and the JNK signaling pathway, elicit cell apoptosis via the mitochondrial-mediated Caspase activation pathway, and finally inhibit tumor growth and mi- gration in vitro. The downregulation of PIK3C2B and Wnt5A jointly inhibit the proliferation and metastasis of HO-8910PM. PPI may be a novel treatment for ovarian cancer.

文摘Tomato(Solanum lycopersicum)is a major horticultural crop worldwide and has emerged as a preeminent model for metabolic research.Although many research efforts have focused on the analysis of metabolite differences between varieties and species,the dynamics of metabolic changes during the tomato growth cycle and the regulatory networks that underlie these changes are poorly understood.In this study,we integrated high-resolution spatio-temporal metabolome and transcriptome data to systematically explore the metabolic landscape across 20 major tomato tissues and growth stages.In the resulting MicroTom Metabolic Network,the 540 detected metabolites and their co-expressed genes could be divided into 10 distinct clusters based on their biological functions.Using this dataset,we constructed a global map of the major metabolic changes that occur throughout the tomato growth cycle and dissected the underlying regulatory network.In addition to verifying previously well-established regulatory networks for important metabolites,we identified novel transcription factors that regulate the biosynthesis of important secondary metabolites such as steroidal glycoalkaloids and flavonoids.Our findings provide insights into spatiotemporal changes in tomato metabolism and generate a valuable resource for the study of metabolic regulatory processes in model plants.
