OBJECTIVE To investigate the expression of the RECK gene in human breast (cancer) cell lines, and to determine the relationship between RECK gene expression and the invasive capacity of the breast cancer cell lines....OBJECTIVE To investigate the expression of the RECK gene in human breast (cancer) cell lines, and to determine the relationship between RECK gene expression and the invasive capacity of the breast cancer cell lines.METHODS The invasive capacity of breast (cancer) cell lines including HBL-100, MCF-7 and MDA-MB-435S were determined by the Tran- swell method. The protein expression levels of RECK, MMP-2 and MMP- 9 genes in these three cell lines were measured by immunocytochemical methods. The expressions of the RECK gene and protein level were measured hv RT-PCR and Westrn hints in the cell lines respectively.RESULTS The order of the nvasive capacity of the breast (cancer) cell lines was MDA-MB-435S, being the highest, and HBL-100, being the lowest. The invasive capacity difference between any two groups among the three groups was significant (P〈0.01). The protein expression level of the RECK gene in the HBL-100 cell line was highest, and no expression was detected in MDA-MB-435S cells. Moreover, the expression of the RECK gene was negatively correlated with the expression of the MMP-2 and MMP-9 genes. The mRNA level of the RECK gene in HBL-100 cells was the highest, but no expression was found in the MDA-MB-435S cells (P〈0.001).CONCLUSION There was a significant negative correlation between the expression level of the RECK gene and invasive capacity in vitro, and the RECK gene expression showed an inverse proportion to that of the MMP-2, MMP-9 genes.展开更多
Vascularization and bone regeneration are closely related in the process of bone remodeling,and designing a bioactive scaffold with pro-angiogenic and osteogenic properties may accelerate the repair of bone defects.In...Vascularization and bone regeneration are closely related in the process of bone remodeling,and designing a bioactive scaffold with pro-angiogenic and osteogenic properties may accelerate the repair of bone defects.In this work,an iron-based metal-organic framework(MIL-88)was developed as a carrier for loading a pro-angiogenic small molecular drug(dimethyloxallyl glycine,DMOG),and then embedded into the PLGA nanofibrous scaffolds to repair cranial defects in rats.Imaging and histological evaluation indicated that PLGA/MIL@D scaffold markedly enhanced vascularization and bone regeneration in vivo.Moreover,in vitro assay showed that co-delivery system significantly promoted angiogenesis by stimulating endothelial cell migration,tube formation,and enhanced osteogenesis by promoting expression of osteoblast related proteins.In addition,PLGA/MIL@D scaffold promotes angiogenesis by activating the hypoxia-inducible factor-1(HIF-1)/vascular endothelial growth factor(VEGF)signaling pathway.Altogether,this bioactive PLGA/MIL@D scaffold can combine angiogenesis with osteogenesis,and will be a bright strategy for the repair of bone defects.展开更多

Innovative scaffold was here constructed by immobilizing exosomes derived from human bone mesenchymal stem cells(h BMSCs) onto porous polymer meshes which developed by PLGA and Cu-based MOF(PLGA/Cu BDC@Exo). The synthesized exosome-laden scaffold capable of providing a dual cooperative controllable release of bioactive copper ions and exosomes that promote osteogenesis and angiogenesis, thereby achieving cell-free bone regeneration. In vitro assay revealed the composite stent not only substantially upregulated the expression of osteogenic-related proteins(ALP, Runx2, Ocn)and VEGF in h BMSCs, but promoted the migration and tube formation of the human umbilical vein endothelial cells(HUVECs). In vivo evaluation further confirmed this scaffold dramatically stimulated bone regeneration and angiogenesis in critical-sized defects in rats. Altogether, this composite scaffold carrying therapeutic exosomes had an osteogenic-angiogenic coupling effect and offered a new idea for cell-free bone tissue engineering.展开更多

Although metal-organic framework (MOF) scaffolds are promisingstrategies for bone tissue regeneration, their potential use for exosome loading remains unexplored. In this study,motivated by the potential advantages of hADSCs-Exos and Mg-GA MOF, we designed and synthesized anexosome-functionalized cell-free PLGA/Mg-GA MOF (PLGA/Exo-Mg-GA MOF) scaffold, taking using of thebenefits of hADSCs-Exos, Mg2+, and gallic acid (GA) to construct unique nanostructural interfaces to enhanceosteogenic, angiogenic and anti-inflammatory capabilities simultaneously. Our in vitro work demonstrated thebeneficial effects of PLGA/Exo-Mg-GA MOF composite scaffolds on the osteogenic effects in human bonemarrow-derived mesenchymal stem cells (hBMSCs) and angiogenic effects in human umbilical endothelial cells(HUVECs). Slowly released hADSCs-Exos from composite scaffolds were phagocytosed by co-cultured cells,stabilized the bone graft environment, ensured blood supply, promoted osteogenic differentiation, and acceleratedbone reconstruction. 文摘In this paper, we introduce stochasticity into an SIR epidemic model with vaccina- tion. The stochasticity in the model is a standard technique in stochastic population modeling. When the perturbations are small, by the method of stochastic Lyapunov functions, we carry out a detailed analysis on the dynamical behavior of the stochastic model regarding of the basic reproduction number R0. If R0 ≤ 1, the solution of the model is oscillating around a steady state, which is the disease-free equilibrium of the corresponding deterministic model. If R0 〉 1, there is a stationary distribution and the solution has the ergodic property, which means that the disease will prevail.

文摘In this paper,we analyze a higher-order stochastically perturbed multigroup staged-progression model for the transmission of HlV with saturated incidence rate.We obtainsufficient conditions for the existence and uniqueness of an ergodic stationary distribu-tion of positive solutions to the system by establishing a suitable stochastic Lyapunovfunction.In addition,we make up adequate conditions for complete eradication and wip-ing out the infectious disease.In a biological interpretation,the existence of a stationarydistribution implies that the disease will prevail and persist in the long term.Finally,examples and numerical simulations are introduced to validate our theoretical results.

文摘In this paper,we study the dynamical behavior of a stochastic two-compartment model of B-cell chronic lymphocytic leukemia,which is perturbed by white noise.Firstly,by constructing suitable Lyapunov functions,we establish sufficient conditions for the existence of a unique ergodic stationary distribution.Then,conditions for extinction of the disease are derived.Furthermore,numerical simulations are presented for supporting the theoretical results.Our results show that large noise intensity may contribute to extinction of the disease.

文摘This paper discusses the dynamics of a Gilpin-Ayala competition model of two interacting species perturbed by white noise.We obtain the existence of a unique global positive solution of the system and the solution is bounded in pth moment.Then,we establish sufficient and necessary conditions for persistence and the existence of an ergodic stationary distribution of the model.We also establish sufficient conditions for extinction of the model.Moreover,numerical simulations are carried out for further support of present research.
