METHODS: HCT116 cells were...AIM:To study the effect of caffeic add phenethyl ester (CAPE) on proliferation, cell cycle, apoptosis and expression of β-catenin in cultured human colorectal cancer (CRC) cell line HCT116. METHODS: HCT116 cells were treated with CAPE at serial concentrations of 80,40,20,10,5,2.5 mg/L. The proliferative status of HCT116 cells was measured by using methaben-zthiazuron (MTT) assay. Cell cycle was analyzed by using flow cytometry (FCM) with propidium iodide (PI) labeling method. The rate of apoptosis was detected by using FCM with annexin V-FITC and PI double labeling method, β-catenin levels were determined by Western blotting, β-catenin localization in HCT116 was determined by indirect immunofluorescence. RESULTS: After HCT116 cells were exposed to CAPE (80, 40, 20, 10, 5, and 2.5 mg/L) for 24, 48, 72, 96 h, CAPE displayed a strong growth inhibitory effect in a dose- and time-dependent manner against HCT116 cells. FCM analysis showed that the ratio of G0/G1 phase cells increased, S phase ratio decreased and apoptosis rate increased after HCT116 cells were exposed to CAPE (10, 5, and 2.5 mg/L) for 24 h. CAPE treatment was associated with decreased cytoplasmic β-catenin, nuclear p-catenin and a concurrent increase in β-catenin protein expression at cell-cell junctions. CONCLUSION: CAPE could inhibit HCT116 cell proliferation and induce cell cycle arrest and apoptosis. Decreased β-catenin protein expression may mediate the anti-proliferative effects of CAPE.展开更多
The Bayanhot Basin is a superimposed basin that experienced multiple-staged tectonic movements;it is in the eastern Alxa Block,adjacent to the North China Craton(NCC)and the North Qilian Orogenic Belt(NQOB).There are ...The Bayanhot Basin is a superimposed basin that experienced multiple-staged tectonic movements;it is in the eastern Alxa Block,adjacent to the North China Craton(NCC)and the North Qilian Orogenic Belt(NQOB).There are well-developed Paleozoic-Cenozoic strata in this basin,and these provide a crucial window to a greater understanding of the amalgamation process and source-to-sink relationships between the Alxa Block and surrounding tectonic units.However,due to intensive post-depositional modification,and lack of subsurface data,several fundamental issues-including the distribution and evolution of the depositional systems,provenance supplies and source-to-sink relationships during the Carboniferous-Permian remain unclear and thus hinder hydrocarbon exploration and limit the geological understanding of this basin.Employing integrated outcrop surveys,new drilling data,and detrital zircon dating,this study examines the paleogeographic distribution and evolution,and provenance characteristics of the Carboniferous-Permian strata in the Bayanhot Basin.Our results show that the Bayanhot Basin experienced a long-term depositional evolution process from transgression to retrogression during the Carboniferous-late Permian.The transgression extent could reach the central basin in the early Carboniferous.The maximum regional transgression occurred in the early Permian and might connect the Qilian and North China seas with each other.Subsequently,a gradual regression followed until the end of the Permian.The northwestern NCC appeared as a paleo-uplift area and served as a sediments provenance area for the Alxa Block at that time.The NCC,Bayanwula Mountain,and NQOB jointly served as major provenances during the Carboniferous-Permian.There was no ocean separation,nor was there an orogenic belt between the Alxa Block and the NCC that provided sediments for both sides during the Carboniferous-Permian.The accretion of the Alxa and North China blocks should have been completed before the Carboniferous period.展开更多

Methods: Twenty-four rabbits were firstly divided into void group (n=...Objective: To explore the effect of tumor necrosis factor-α(TNF-α)on the occurrence of steroid-induced avascular necrosis of femoral head(SANFH). Methods: Twenty-four rabbits were firstly divided into void group (n=12) and model group (n=12) at random. Prednisone Acetate was injected to the model group(0.32 mg·kg^(-1)·d^(-1)). In the 6 th and 8 th week, two rabbits of the two groups were killed respectively to observe whether the model was successful. The level of TNF-α in serum of the residual rabbits of the two groups was examined in Radioimmunoassay method. Results: The level of TNF-α in model group is significantly higher than that in void group(P<0.001) under the premise of the model of SANFH success by histological observation. Conclusion: The rise of level of TNF-α may be one of the most important factors in the occurrence of SANFH.展开更多

The measured power dependence of the fluorescence signals provided direct evidence for TPE. Ltd. were analyzed. Considering steelmaking processconditions, the optimum power-input diagrams were investigated, which can save energy, lowerconsumption, and raise productivity. This has been verified by production contrast tests.Application of the optimum curve in 50 t UHP-EAF indicates that research method is correct, the 50 tUHP-EAF runs smoothly with lowering electrical energy by 29.35 kW-h/t and shortening power on timeby 3 min. However, there were some researchers ...Based on three major theories, the Chinese local officials' promotion incentive could be influenced by economic performances, political relationships, and financial performances. However, there were some researchers focusing on these promotion mechanisms and then they got different outcomes. This paper achieves the goal for exploring the influence factors of officials' promotion by using OLS empirical test. From collecting the data of Chinese local officials and cities' economic conditions, we find that economic performances and political relationships can have a significant effect on officials' promotion, but financial performances cannot have this effect. And the experiences and lesson of this research have some reference for exploration of Chinese local officials' promotion incentive.展开更多

Methods: Mice were pretreated with LPS of different doses at 48 and 24 h before femoral medial lon- gitudinal incision was made and infected with different bacteria. Results: It is showed that 0.5 mg/kg/time ofLPS pretreatment can significantly alleviate the inflammation in mouse model infected with methicillin-resistances Staphylococcus aureus, methicillin-sensitive S. aureus, Pseudomonas aeruginosa, or Escherichia coil compared with doses of 0.25 mg/kg/time, 1 mg/ kg/time, and 1.5 mg/kg/time. Conclusions: LPS pretreatment can alleviate the inflammation in mouse model and the optimal dose is 0.5 mg/kg/time, and meanwhile it does not damage organs' function.展开更多
Implicit-explicit (IMEX) linear multistep methods are popular techniques for solving partial differential equations (PDEs) with terms of different types. While fixed timestep versions of such schemes have been dev...Implicit-explicit (IMEX) linear multistep methods are popular techniques for solving partial differential equations (PDEs) with terms of different types. While fixed timestep versions of such schemes have been developed and studied, implicit-explicit schemes also naturally arise in general situations where the temporal smoothness of the solution changes. In this paper we consider easily implementable variable step-size implicit-explicit (VSIMEX) linear multistep methods for time-dependent PDEs. Families of order-p, pstep VSIMEX schemes are constructed and analyzed, where p ranges from 1 to 4. The corresponding schemes are simple to implement and have the property that they reduce to the classical IMEX schemes whenever constant time step-sizes are imposed. The methods are validated on the Burgers' equation. These results demonstrate that by varying the time step-size, VSIMEX methods can outperform their fixed time step counterparts while still maintaining good numerical behavior.展开更多

In this paper, we study the feas...The echo-enabled harmonic generation (EEHG) scheme holds promising prospects for efficiently generat- ing intense coherent radiation at very high harmonics of the initial seed laser. In this paper, we study the feasibility of operating the Shanghai X-ray free-electron laser test facility with a single stage EEHG setup. With a combina- tion of existing numerical codes, start-to-end simulations considering various three-dimensional effects have been carried out to show the possible performance of the EEHG FEL. Simulation results demonstrate that a single stage EEHG can generate high power soft X-ray radiation pulses with narrow bandwidth directly from UV seed lasers. With the help of the harmonic lasing technique or upgraded linac, we also show the possibility of generating coherent soft X-ray radiation in the "water window" with a single stage EEHG.展开更多
