Dehydroabietyl polyethylene glycol glycidyl ether-grafted hydroxyethyl cellulose(HEC)polymer surfactant(DA(EO)5GE-g-HEC)was prepared using ring-opening polymerization with biobased rosin and hydroxyethyl cellulose as ...Dehydroabietyl polyethylene glycol glycidyl ether-grafted hydroxyethyl cellulose(HEC)polymer surfactant(DA(EO)5GE-g-HEC)was prepared using ring-opening polymerization with biobased rosin and hydroxyethyl cellulose as feedstocks.Dehydroabietyl polyethylene glycol glycidyl ether(DA(EO)5GE)was formed by condensation of dehydroabietyl alcohol polyoxyethylene ether(Rosin derivative:DA(EO)5H)and epichlorohydrin.The grafting degree of DA(EO)5GE-g-HEC was manipulated by adjusting the mass ratio of HEC and DA(EO)5GE and confirmed by EA.According to the formula,when m(HEC)/m(DA(EO)2GE)was 1:1~1:5,the grafting rate of DA(EO)5GE in DA(EO)5GE-g-HEC varied from 34.43%to 38.33%.The surface activity and foam properties of DA(EO)5GE-g-HEC aqueous solution were studied.The results showed that with the increase in grafting rate,the critical micellar concentration(CMC)in aqueous solution changed from 1.28 to 0.96 g/L.The results of the thermogravimetric analysis showed that the temperature range of the main stage of mass loss of DA(EO)5GE-g-HEC was 310°C~410°C,and the thermal decomposition processes of the samples with five mass ratios were similar.An oil in water emulsion was prepared by choosing cyclohexane as the oil phase and DA(EO)5GE-g-HEC as the emulsifier.The effect of DA(EO)5GE-g-HEC mass fraction on emulsion particle size and stability was analyzed.The results suggested that when the oil-water ratio was 8:2 with 0.4%emulsifier,the emulsion droplets were the smallest in terms of particle size and were the most stable.The rheological test results showed that the apparent viscosity decreased with the increase in shear rate and showed a typical elastic gel phenomenon.展开更多
BACKGROUND Gastric cancer(GC)is a prevalent malignancy,leading to a high incidence of cancer-associated death.Cisplatin(DDP)-based chemotherapy is the principal therapy for clinical GC treatment,but DDP resistance is ...BACKGROUND Gastric cancer(GC)is a prevalent malignancy,leading to a high incidence of cancer-associated death.Cisplatin(DDP)-based chemotherapy is the principal therapy for clinical GC treatment,but DDP resistance is a severe clinical challenge and the mechanism remains poorly understood.Circular RNAs(circRNAs)have been identified to play crucial roles in modulating the chemoresistance of gastric cancer cells.AIM To explore the effect of circVAPA on chemotherapy resistance during GC progression.METHODS The effect of circVAPA on GC progression and chemotherapy resistance was analyzed by MTT assay,colony formation assay,Transwell assay,wound healing assay,and flow cytometry analysis in GC cells and DDP resistant GC cell lines,and tumorigenicity analysis in nude mice in vivo.The mechanism was investigated by luciferase reporter assay,quantitative real-time PCR,and Western blot analysis.RESULTS CircVAPA expression was up-regulated in clinical GC tissues compared with normal samples.CircVAPA depletion inhibited proliferation,migration,and invasion and increased apoptosis of GC cells.The expression of circVAPA,STAT3,and STAT3 downstream genes was elevated in DDP resistant SGC7901/DDP cell lines.CircVAPA knockdown attenuated the DDP resistance of GC cells.Mechanically,circVAPA was able to sponge miR-125b-5p,and miR-125b-5p could target STAT3 in the GC cells.MiR-125b-5p inhibitor reversed circVAPA depletion-enhanced inhibitory effect of DDP on GC cells,and STAT3 knockdown blocked circVAPA overexpression-induced proliferation of DDPtreated SGC7901/DDP cells.The depletion of STAT3 and miR-125b-5p inhibitor reversed circVAPA depletion-induced GC cell apoptosis.Functionally,circVAPA contributed to the tumor growth of SGC7901/DDP cells in vivo.CONCLUSION CircVAPA promotes chemotherapy resistance and malignant progression in GC by miR-125b-5p/STAT3 signaling.Our findings present novel insights into the mechanism by which circVAPA regulates chemotherapy resistance of GC cells.CircVAPA and miR-125b-5p may be considered as the potential targets for GC therapy.展开更多
Cotton (Gossypium hirsutum L.) is an importantfiber cash crop,but its root traits related to phosphorus (P) acquisition,including mycorrhizal root traits,are poorly understood.Eight cotton varieties bred in northweste...Cotton (Gossypium hirsutum L.) is an importantfiber cash crop,but its root traits related to phosphorus (P) acquisition,including mycorrhizal root traits,are poorly understood.Eight cotton varieties bred in northwestern China that were released between 1950 and 2013 were grown in pots with or without one arbuscular mycorrhizal fungal (AMF) species(Funneliformis mosseae) at three P supply levels (0,50 and 300 mg P as KH_(2)PO_(4)kg^(-1)).Eleven root traits were measured and calculated after 7 wk of growth.The more recent accessions had smaller root diameters,acquired less P and produced less biomass,indicating an (inadvertent) varietal selection for thinner roots that provided less cortical space for AMF,which then increased the need for a high P fertilizer level.At the two lower P levels,the mycorrhizal plants acquired more P and produced more biomass than non-mycorrhizal plants (3.2 vs.0.9 mg P per plant;1.8 vs.0.9 g biomass per plant at P_(0);14.5 vs.1.7 mg P per plant;and 4.7 vs.1.6 g biomass per plant at P_(50)).At the highest P level,the mycorrhizal plants acquired more P than non-mycorrhizal plants (18.8 vs.13.4 mg per P plant),but there was no difference in biomass (6.2 vs.6.3 g per plant).At the intermediate P level,root diameter was significantly positively correlated with shoot biomass,P concentration and the P content of mycorrhizal plants.The results of our study support the importance of the outsourcing model of P acquisition in the root economics space framework.Inadvertent varietal selection in the last decades,resulting in thinner roots and a lower benefit from AMF,has led to a lower productivity of cotton varieties at moderate P supply (i.e.,when mycorrhizal,the average biomass of older varieties 5.0 g per plant vs.biomass of newer varieties 4.4 g per plant),indicating the need to rethink cotton breeding efforts in order to achieve high yields without very high P input.One feasible way to solve the problem of inadvertent varietal selection for cotton is to be aware of the trade-offs between the root do-it-yourself strategy and the outsourcing towards AMF strategy,and to consider both morphological and mycorrhizal root traits when breeding cotton varieties.展开更多
Background Subarachnoid haemorrhage(SAH)can result in a highly unfavourable prognosis.In recent years,the study of SAH has focused on early brain injury(EBI),which is a crucial progress that contributes to adverse pro...Background Subarachnoid haemorrhage(SAH)can result in a highly unfavourable prognosis.In recent years,the study of SAH has focused on early brain injury(EBI),which is a crucial progress that contributes to adverse prognosis.SAH can lead to various complications,including mitochondrial dysfunction and DNA damage.Apurinic/apyrimidinic endonuclease 1(APE1)is an essential protein with multifaceted functionality integral to DNA repair and redox signalling.However,the role of APE1 in mitochondrial DNA damage repair after SAH is still unclear.Methods Our study involved an in vivo endovascular perforation model in rats and an in vitro neuron oxyhaemoglobin intervention.Then,the effects of APE1 on mitochondrial DNA damage repair were analysed by western blot,immunofluorescence,quantitative real-time PCR,mitochondrial bioenergetics measurement and neurobehavioural experiments.Results We found that the level of APE1 decreased while the mitochondria DNA damage and neuronal death increased in a rat model of SAH.Overexpression of APE1 improved short-term and long-term behavioural impairment in rats after SAH.In vitro,after primary neurons exposed to oxyhaemoglobin,APE1 expression significantly decreased along with increased mitochondrial DNA damage,a reduction in the subunit of respiratory chain complex levels and subsequent respiratory chain dysfunction.Overexpression of APE1 relieved energy metabolism disorders in the mitochondrial of neurons and reduced neuronal apoptosis.Conclusion In conclusion,APE1 is involved in EBI after SAH by affecting mitochondrial apoptosis via the mitochondrial respiratory chain.APE1 may potentially play a vital role in the EBI stage after SAH,making it a critical target for treatment.展开更多
The effects of NaCl salinity and NO^-3 on growth, root morphology, and nitrogen uptake of a halophyte Suaeda physophora were evaluated in a factorial experiment with four concentrations of NaCl (1, 150, 300, and 450 ...The effects of NaCl salinity and NO^-3 on growth, root morphology, and nitrogen uptake of a halophyte Suaeda physophora were evaluated in a factorial experiment with four concentrations of NaCl (1, 150, 300, and 450 mmol L^-1) and three NO^-3 levels (0.05, 5, and 10 mmol L^-1) in solution culture for 30 d. Addition of NO^-3 at 10 mmol L^-1 significantly improved the shoot (P 〈 0.001) and root (P 〈 0.001) growth and the promotive effect of NO^-3 was more pronounced on root dry weight despite the high NaCl concentration in the culture solution, leading to a significant increase in the root:shoot ratio (P 〈 0.01). Lateral root length, but not primary root length, considerably increased with increasing NaCl salinity and NO^-3 levels (P 〈 0.001), implying that Na^+ and NO3^- in the culture solution simultaneously stimulated lateral root growth. Concentrations of Na^+ in plant tissues were also significantly increased by higher NaCl treatments (P 〈 0.001). At 10 mmol L^-1 NO^-3, the concentrations of NO^-3 and total nitrogen and nitrate reductase activities in the roots were remarkably reduced by increasing salinity (P 〈 0.001), but were unaffected in the shoots. The results indicated that the fine lateral root development and effective nitrogen uptake of the shoots might contribute to high salt tolerance of S. physophora under adequate NO^-3 supply.展开更多

The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to a...Objective: Mucin 1(MUC1/EMA) and sialyl Lewis X(sLe^(x)) indicate polarity reversal in invasive micropapillary carcinoma(IMPC). The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to assess their diagnostic and prognostic value in patients with IMPC.Methods: The expression of sLe^(x) and MUC1/EMA in 100 patients with IMPC and a control group of 89 patients with invasive ductal carcinoma not otherwise specified(IDC-NOS) were analyzed with IHC. Fresh tumor tissues were collected from patients with IMPC or IDC-NOS for primary culture and immunofluorescence analysis.Results: The rate of nodal metastasis was higher in patients with IMPC than those with IDC-NOS, and IMPC cells tended to express more sLe^(x) and MUC1/EMA in the cytomembranes(the stroma-facing surfaces of the micropapillary clusters) than IDC-NOS cells. In IMPC, high cytomembrane expression of sLe^(x), but not MUC1/EMA, indicated poor prognosis. In addition, among the 100 patients with IMPC, 10 patients had sLe^(x)+/EMA-expression patterns, and 8 patients had sLe^(x)-/EMA+ expression patterns. The primary IMPC cells were suspended, non-adherent tumor cell clusters, whereas the primary IDC cells were adherent tumor cells. Immunofluorescence analysis showed that MUC1/EMA and sLe^(x) were co-expressed on the cytomembranes in IMPC cell clusters and in the cytoplasm in IDC-NOS cells.Conclusions: sLe^(x) can be used as a prognostic indicator and can be combined with MUC1/EMA as a complementary diagnostic indicator to avoid missed IMPC diagnosis.展开更多
AIM: To compare the clinical efficacy of intravitreal injections of bevacizumab and ranibizumab for treating Chinese patients with neovascular age-related macular degeneration (AMD).METHODS: Among 60 Chinese patients ...AIM: To compare the clinical efficacy of intravitreal injections of bevacizumab and ranibizumab for treating Chinese patients with neovascular age-related macular degeneration (AMD).METHODS: Among 60 Chinese patients with exudative AMD (60 eyes), 28 received intravitreal bevacizumab injections (1.25mg) and 32 received intravitreal ranibizumab injections (0.5mg), once a month for 3 months and were followed for a total of 6 months. Monthly optical coherence tomography (OCT) was used to determine whether the patients received additional treatments during the follow-up. We compared the baseline and 6 -month follow-up values of mean best-corrected visual acuity (BCVA) and central retinal thickness (CRT) in both groups of patients. We also compared the occurrence of adverse events.RESULTS: At the 6-month follow-up, the mean BCVA (logMAR) of the bevacizumab and ranibizumab treatment groups improved from the baseline measurements of 0.72 ±0.23 and 0.73 ±0.22 to 0.47 ±0.14 and 0.45 ±0.20, respectively (P 【0.05 for both groups). However, the change was not significantly different between the two groups. As evaluated by OCT, CRT decreased from 366.71 ±34.72μm and 352 ±36.9μm at baseline to 250.86 ± 41.51μm and 243.22 ±41.38μm in the bevacizumab and ranibizumab groups, respectively (P 【0.05 for both groups). However, the change was not significantly different between the two groups. There were no severe local adverse reactions or systemic adverse events.CONCLUSION:Intravitreal bevacizumab and ranibizumab have equivalent effects on BCVA and CRT and appeare safe over the short-term.展开更多
Objective To evaluate the clinical and pathological features of Riedel's thyroiditis(RT),and current diagnostic and treatment methods for that disease.Methods Five RT cases identified by surgery and pathological e...Objective To evaluate the clinical and pathological features of Riedel's thyroiditis(RT),and current diagnostic and treatment methods for that disease.Methods Five RT cases identified by surgery and pathological examinations at Peking Union Medical College Hospital from 1985 to 2009 were analyzed and compared with the cases reported in the literature in terms of clinical and pathological features.Immunohistochemical staining of kappa and lambda light chains was carried out for RT tissues from all the five patients.Results All the five cases were females,aged 45-55 years.Elevation of serum thyroid autoantibodies was found in only one patient,who had longer disease duration than the others.Pathological examination revealed invasive fibrosclerosis of the thyroid follicles,thyroid capsule,and the surrounding tissues.In RT tissues,the number of cells containing lambda chains was a little higher than those containing kappa chains.Conclusions RT is a rare disease which might be more common in middle-aged females than in other populations.Pathological features include the destruction of thyroid follicle,extension into surrounding tissues by inflammatory cells and fibrous tissues.Immunohistochemical staining of kappa and lambda chains could help diagnose RT.展开更多
The species richness and propagule number of arbuscular mycorrhizal fungi(AMF)are high in intensively-managed agricultural soils.Past research has shown that AMF improve crop phosphorus(P)uptake under low soil P condi...The species richness and propagule number of arbuscular mycorrhizal fungi(AMF)are high in intensively-managed agricultural soils.Past research has shown that AMF improve crop phosphorus(P)uptake under low soil P conditions,however it is unclear if AMF play a role in high Olsen-P soils.In this study,we investigated whether native fungal benefits exist under high P input field conditions in-situ and contribute to P utilization.We installed in-grow tubes which were sealed with different membrane pore sizes(30 or 0.45μm)to allow or prevent AMF hyphae access to the hyphal compartment and prevent cotton roots from penetrating the chamber.We used the depletion of soil available P(Olsen-P)in the hyphae accessed compartment to indicate P uptake by the native AMF community.Our results showed that the native AMF mediated P depletion and microbial biomass P(MBP)turnover and caused the largest Olsen-P depletion ratio and MBP turnover ratio in the high P treatments(Olsen-P:78.29 mg kg^(-1)).The cotton roots in each fertilization regime were colonized by a unique AMF community and Glomus and Paraglomus were the dominant genera,implying the longterm fertilization regimes domesticated the AMF community.We conclude that native AMF caused the P depletion and P turnover even under high soil Olsen-P conditions.展开更多

METHODS: Ophthalmologic examinations were performed on the sporadic RP patient, 144 genes associated with retinal diseases were scanned with capture next generation sequencing (CNGS) approach. Two heterozygous mutations in PDE6B were confirmed in the pedigree by Sanger sequencing subsequently. The carrier frequency of PDE6B mutations of reported PDE5B mutations based on the available two public exome databases (1000 Genomes Project and ESP6500 Genomes Project) and one in-house exome database was investigated. RESULTS: We identified compound heterozygosity of two novel nonsense mutations c.1133G>A (p.W378X) and c.2395C>T (p.R799X) in PDE6B, one reported causative gene for RP. Neither of the two mutations in our study was presented in three exome databases. Two mutations (p.R74C and p.T6041) in PDE5B have relatively high frequencies in the ESP6500 and in-house databases, respectively, while no common dominant mutation in each of the database or across all databases. CONCLUSION: We demonstrates that compound heterozygosity of two novel nonsense mutations in PDE6B could lead to RP. These results collectively point to enormous potential of next-generation sequencing in determining the genetic etiology of RP and how various mutations in PDE9B contribute to the genetic heterogeneity of RP.展开更多

In this work, patterned macropores with a diameter larger than 100 μm were introduced to pristine three-dimensional (3D) nanofibrous bacterial cellulose (BC) scaffolds by using the infrared laser micromachining techn...In this work, patterned macropores with a diameter larger than 100 μm were introduced to pristine three-dimensional (3D) nanofibrous bacterial cellulose (BC) scaffolds by using the infrared laser micromachining technique in an attempt to create an in vitro model for the culture of breast cancer cells. The morphology, pore structure, and mechanical performance of the obtained patterned macroporous BC (PM-BC) scaffolds were characterized by scanning electron microscopy (SEM), mercury intrusion porosimeter, and mechanical testing. A human breast cancer cell (MDA-MB-231) line was cultured onto the PM-BC scaffolds to investigate the role of macropores in the control of cancer cell behavior. MTT assay, SEM, and hematoxylin and eosin (H&E) staining were employed to determine cell adhesion, growth, proliferation, and infiltration. The PM-BC scaffolds were found to be able to promote cellular adhesion and proliferation on the scaffolds, and further to allow for cell infiltration into the PM-BC scaffolds. The results demonstrated that BC scaffolds with laser-patterned macropores were promising for the in vitro 3D culture of breast cancer cells.展开更多
Potassium-ion capacitors(KICs)are promising for sustainable and eco-friendly energy storage technologies,yet their slow reaction kinetics and poor cyclability induced by large K-ion size are a major obstacle toward pr...Potassium-ion capacitors(KICs)are promising for sustainable and eco-friendly energy storage technologies,yet their slow reaction kinetics and poor cyclability induced by large K-ion size are a major obstacle toward practical applications.Herein,by employing black phosphorus nanosheets(BPNSs)as a typical high-capacity anode material,we report that BPNS anodes armored with an ultrathin oriented-grown metal–organic-framework(MOF)interphase layer(BPNS@MOF)exhibit regulated potassium storage behavior for highperformance KICs.The MOF interphase layers as protective layer with ordered pores and high chemical/mechanical stability facilitate K ion diffusion and accommodate the volume change of electrode,beneficial for improved reaction kinetics and enhanced cyclability,as evidenced by substantial characterizations,kinetics analysis and DFT calculations.Consequently,the BPNS@MOF electrode as KIC anodes exhibits outstanding cycle performance outperforming most of the reported state-of-art KICs so far.展开更多
BACKGROUND Waldenström macroglobulinemia(WM)is a distinct clinicopathologic entity characterized by the infiltration of the bone marrow by clonal lymphoplasmacytic cells that produce monoclonal immunoglobulin M(I...BACKGROUND Waldenström macroglobulinemia(WM)is a distinct clinicopathologic entity characterized by the infiltration of the bone marrow by clonal lymphoplasmacytic cells that produce monoclonal immunoglobulin M(IgM)in the blood,and patients may present with symptoms related to the infiltration of the hematopoietic tissues or the effects of monoclonal IgM in the blood.Funduscopic abnormalities were noted in some of the patients due to hyperviscosity or other retinal lesions.Optical coherence tomography angiography(OCTA)as a noninvasive imaging tool can give qualitative and quantitative information about the status of retinal and choroidal vessels,which might be useful for diagnosing patients with WM-associated retinopathy.CASE SUMMARY The patient was a 67-year-old man who presented with sudden visual disturbance in both eyes.Ophthalmic tests showed that best corrected visual acuity(BCVA)for this patient was 20/100 in the right eye and 20/1000 in the left eye.Fundus examination,optical coherence tomography(OCT),and OCTA revealed substantial bilateral optic disc edema,dilated and tortuous retinal veins,and diffuse intraretinal blot hemorrhages and edema which were consistent with bilateral central retinal vein occlusion(CRVO).Meanwhile,remarkable bilateral serous macular detachments(SMD)were noticed on OCT.Systemic examinations showed that the patient had anemia and extremely high level of monoclonal IgM and infiltration of clonal lymphoplasmacytic cells in bone marrow.The diagnosis of WM with hyperviscosity and retinopathy was made based on the clinical manifestation and laboratory findings.He was subsequently treated with intravitreal ranibizumab injection,plasmapheresis,and bortezomib plus rituximab with dexamethasone.Six months after treatments,the central macular volume decreased by 16.1%in the right eye and 28.6%in the left eye on OCT,and the patient’s BCVA was improved to 20/60 in the right eye and 20/400 in the left eye.Very good partial response was obtained after systemic treatment.CONCLUSION WM may affect visual function and present as bilateral CRVO.OCTA can show characteristic changes in both retina and choroid vasculatures,which might be of great value for diagnosing or following patients with WM retinopathy.Intravitreal anti-vascular endothelial growth factor treatment combined with systemic therapy might be beneficial for WM patients with retinopathy(SMD and CRVO).展开更多

文摘Dehydroabietyl polyethylene glycol glycidyl ether-grafted hydroxyethyl cellulose(HEC)polymer surfactant(DA(EO)5GE-g-HEC)was prepared using ring-opening polymerization with biobased rosin and hydroxyethyl cellulose as feedstocks.Dehydroabietyl polyethylene glycol glycidyl ether(DA(EO)5GE)was formed by condensation of dehydroabietyl alcohol polyoxyethylene ether(Rosin derivative:DA(EO)5H)and epichlorohydrin.The grafting degree of DA(EO)5GE-g-HEC was manipulated by adjusting the mass ratio of HEC and DA(EO)5GE and confirmed by EA.According to the formula,when m(HEC)/m(DA(EO)2GE)was 1:1~1:5,the grafting rate of DA(EO)5GE in DA(EO)5GE-g-HEC varied from 34.43%to 38.33%.The surface activity and foam properties of DA(EO)5GE-g-HEC aqueous solution were studied.The results showed that with the increase in grafting rate,the critical micellar concentration(CMC)in aqueous solution changed from 1.28 to 0.96 g/L.The results of the thermogravimetric analysis showed that the temperature range of the main stage of mass loss of DA(EO)5GE-g-HEC was 310°C~410°C,and the thermal decomposition processes of the samples with five mass ratios were similar.An oil in water emulsion was prepared by choosing cyclohexane as the oil phase and DA(EO)5GE-g-HEC as the emulsifier.The effect of DA(EO)5GE-g-HEC mass fraction on emulsion particle size and stability was analyzed.The results suggested that when the oil-water ratio was 8:2 with 0.4%emulsifier,the emulsion droplets were the smallest in terms of particle size and were the most stable.The rheological test results showed that the apparent viscosity decreased with the increase in shear rate and showed a typical elastic gel phenomenon.

文摘BACKGROUND Gastric cancer(GC)is a prevalent malignancy,leading to a high incidence of cancer-associated death.Cisplatin(DDP)-based chemotherapy is the principal therapy for clinical GC treatment,but DDP resistance is a severe clinical challenge and the mechanism remains poorly understood.Circular RNAs(circRNAs)have been identified to play crucial roles in modulating the chemoresistance of gastric cancer cells.AIM To explore the effect of circVAPA on chemotherapy resistance during GC progression.METHODS The effect of circVAPA on GC progression and chemotherapy resistance was analyzed by MTT assay,colony formation assay,Transwell assay,wound healing assay,and flow cytometry analysis in GC cells and DDP resistant GC cell lines,and tumorigenicity analysis in nude mice in vivo.The mechanism was investigated by luciferase reporter assay,quantitative real-time PCR,and Western blot analysis.RESULTS CircVAPA expression was up-regulated in clinical GC tissues compared with normal samples.CircVAPA depletion inhibited proliferation,migration,and invasion and increased apoptosis of GC cells.The expression of circVAPA,STAT3,and STAT3 downstream genes was elevated in DDP resistant SGC7901/DDP cell lines.CircVAPA knockdown attenuated the DDP resistance of GC cells.Mechanically,circVAPA was able to sponge miR-125b-5p,and miR-125b-5p could target STAT3 in the GC cells.MiR-125b-5p inhibitor reversed circVAPA depletion-enhanced inhibitory effect of DDP on GC cells,and STAT3 knockdown blocked circVAPA overexpression-induced proliferation of DDPtreated SGC7901/DDP cells.The depletion of STAT3 and miR-125b-5p inhibitor reversed circVAPA depletion-induced GC cell apoptosis.Functionally,circVAPA contributed to the tumor growth of SGC7901/DDP cells in vivo.CONCLUSION CircVAPA promotes chemotherapy resistance and malignant progression in GC by miR-125b-5p/STAT3 signaling.Our findings present novel insights into the mechanism by which circVAPA regulates chemotherapy resistance of GC cells.CircVAPA and miR-125b-5p may be considered as the potential targets for GC therapy.

文摘Cotton (Gossypium hirsutum L.) is an importantfiber cash crop,but its root traits related to phosphorus (P) acquisition,including mycorrhizal root traits,are poorly understood.Eight cotton varieties bred in northwestern China that were released between 1950 and 2013 were grown in pots with or without one arbuscular mycorrhizal fungal (AMF) species(Funneliformis mosseae) at three P supply levels (0,50 and 300 mg P as KH_(2)PO_(4)kg^(-1)).Eleven root traits were measured and calculated after 7 wk of growth.The more recent accessions had smaller root diameters,acquired less P and produced less biomass,indicating an (inadvertent) varietal selection for thinner roots that provided less cortical space for AMF,which then increased the need for a high P fertilizer level.At the two lower P levels,the mycorrhizal plants acquired more P and produced more biomass than non-mycorrhizal plants (3.2 vs.0.9 mg P per plant;1.8 vs.0.9 g biomass per plant at P_(0);14.5 vs.1.7 mg P per plant;and 4.7 vs.1.6 g biomass per plant at P_(50)).At the highest P level,the mycorrhizal plants acquired more P than non-mycorrhizal plants (18.8 vs.13.4 mg per P plant),but there was no difference in biomass (6.2 vs.6.3 g per plant).At the intermediate P level,root diameter was significantly positively correlated with shoot biomass,P concentration and the P content of mycorrhizal plants.The results of our study support the importance of the outsourcing model of P acquisition in the root economics space framework.Inadvertent varietal selection in the last decades,resulting in thinner roots and a lower benefit from AMF,has led to a lower productivity of cotton varieties at moderate P supply (i.e.,when mycorrhizal,the average biomass of older varieties 5.0 g per plant vs.biomass of newer varieties 4.4 g per plant),indicating the need to rethink cotton breeding efforts in order to achieve high yields without very high P input.One feasible way to solve the problem of inadvertent varietal selection for cotton is to be aware of the trade-offs between the root do-it-yourself strategy and the outsourcing towards AMF strategy,and to consider both morphological and mycorrhizal root traits when breeding cotton varieties.

文摘Background Subarachnoid haemorrhage(SAH)can result in a highly unfavourable prognosis.In recent years,the study of SAH has focused on early brain injury(EBI),which is a crucial progress that contributes to adverse prognosis.SAH can lead to various complications,including mitochondrial dysfunction and DNA damage.Apurinic/apyrimidinic endonuclease 1(APE1)is an essential protein with multifaceted functionality integral to DNA repair and redox signalling.However,the role of APE1 in mitochondrial DNA damage repair after SAH is still unclear.Methods Our study involved an in vivo endovascular perforation model in rats and an in vitro neuron oxyhaemoglobin intervention.Then,the effects of APE1 on mitochondrial DNA damage repair were analysed by western blot,immunofluorescence,quantitative real-time PCR,mitochondrial bioenergetics measurement and neurobehavioural experiments.Results We found that the level of APE1 decreased while the mitochondria DNA damage and neuronal death increased in a rat model of SAH.Overexpression of APE1 improved short-term and long-term behavioural impairment in rats after SAH.In vitro,after primary neurons exposed to oxyhaemoglobin,APE1 expression significantly decreased along with increased mitochondrial DNA damage,a reduction in the subunit of respiratory chain complex levels and subsequent respiratory chain dysfunction.Overexpression of APE1 relieved energy metabolism disorders in the mitochondrial of neurons and reduced neuronal apoptosis.Conclusion In conclusion,APE1 is involved in EBI after SAH by affecting mitochondrial apoptosis via the mitochondrial respiratory chain.APE1 may potentially play a vital role in the EBI stage after SAH,making it a critical target for treatment.

文摘The effects of NaCl salinity and NO^-3 on growth, root morphology, and nitrogen uptake of a halophyte Suaeda physophora were evaluated in a factorial experiment with four concentrations of NaCl (1, 150, 300, and 450 mmol L^-1) and three NO^-3 levels (0.05, 5, and 10 mmol L^-1) in solution culture for 30 d. Addition of NO^-3 at 10 mmol L^-1 significantly improved the shoot (P 〈 0.001) and root (P 〈 0.001) growth and the promotive effect of NO^-3 was more pronounced on root dry weight despite the high NaCl concentration in the culture solution, leading to a significant increase in the root:shoot ratio (P 〈 0.01). Lateral root length, but not primary root length, considerably increased with increasing NaCl salinity and NO^-3 levels (P 〈 0.001), implying that Na^+ and NO3^- in the culture solution simultaneously stimulated lateral root growth. Concentrations of Na^+ in plant tissues were also significantly increased by higher NaCl treatments (P 〈 0.001). At 10 mmol L^-1 NO^-3, the concentrations of NO^-3 and total nitrogen and nitrate reductase activities in the roots were remarkably reduced by increasing salinity (P 〈 0.001), but were unaffected in the shoots. The results indicated that the fine lateral root development and effective nitrogen uptake of the shoots might contribute to high salt tolerance of S. physophora under adequate NO^-3 supply.

文摘Objective: Mucin 1(MUC1/EMA) and sialyl Lewis X(sLe^(x)) indicate polarity reversal in invasive micropapillary carcinoma(IMPC). The purpose of this study was to evaluate the expression of MUC1/EMA and sLe^(x) and to assess their diagnostic and prognostic value in patients with IMPC.Methods: The expression of sLe^(x) and MUC1/EMA in 100 patients with IMPC and a control group of 89 patients with invasive ductal carcinoma not otherwise specified(IDC-NOS) were analyzed with IHC. Fresh tumor tissues were collected from patients with IMPC or IDC-NOS for primary culture and immunofluorescence analysis.Results: The rate of nodal metastasis was higher in patients with IMPC than those with IDC-NOS, and IMPC cells tended to express more sLe^(x) and MUC1/EMA in the cytomembranes(the stroma-facing surfaces of the micropapillary clusters) than IDC-NOS cells. In IMPC, high cytomembrane expression of sLe^(x), but not MUC1/EMA, indicated poor prognosis. In addition, among the 100 patients with IMPC, 10 patients had sLe^(x)+/EMA-expression patterns, and 8 patients had sLe^(x)-/EMA+ expression patterns. The primary IMPC cells were suspended, non-adherent tumor cell clusters, whereas the primary IDC cells were adherent tumor cells. Immunofluorescence analysis showed that MUC1/EMA and sLe^(x) were co-expressed on the cytomembranes in IMPC cell clusters and in the cytoplasm in IDC-NOS cells.Conclusions: sLe^(x) can be used as a prognostic indicator and can be combined with MUC1/EMA as a complementary diagnostic indicator to avoid missed IMPC diagnosis.
文摘AIM: To compare the clinical efficacy of intravitreal injections of bevacizumab and ranibizumab for treating Chinese patients with neovascular age-related macular degeneration (AMD).METHODS: Among 60 Chinese patients with exudative AMD (60 eyes), 28 received intravitreal bevacizumab injections (1.25mg) and 32 received intravitreal ranibizumab injections (0.5mg), once a month for 3 months and were followed for a total of 6 months. Monthly optical coherence tomography (OCT) was used to determine whether the patients received additional treatments during the follow-up. We compared the baseline and 6 -month follow-up values of mean best-corrected visual acuity (BCVA) and central retinal thickness (CRT) in both groups of patients. We also compared the occurrence of adverse events.RESULTS: At the 6-month follow-up, the mean BCVA (logMAR) of the bevacizumab and ranibizumab treatment groups improved from the baseline measurements of 0.72 ±0.23 and 0.73 ±0.22 to 0.47 ±0.14 and 0.45 ±0.20, respectively (P 【0.05 for both groups). However, the change was not significantly different between the two groups. As evaluated by OCT, CRT decreased from 366.71 ±34.72μm and 352 ±36.9μm at baseline to 250.86 ± 41.51μm and 243.22 ±41.38μm in the bevacizumab and ranibizumab groups, respectively (P 【0.05 for both groups). However, the change was not significantly different between the two groups. There were no severe local adverse reactions or systemic adverse events.CONCLUSION:Intravitreal bevacizumab and ranibizumab have equivalent effects on BCVA and CRT and appeare safe over the short-term.
文摘Objective To evaluate the clinical and pathological features of Riedel's thyroiditis(RT),and current diagnostic and treatment methods for that disease.Methods Five RT cases identified by surgery and pathological examinations at Peking Union Medical College Hospital from 1985 to 2009 were analyzed and compared with the cases reported in the literature in terms of clinical and pathological features.Immunohistochemical staining of kappa and lambda light chains was carried out for RT tissues from all the five patients.Results All the five cases were females,aged 45-55 years.Elevation of serum thyroid autoantibodies was found in only one patient,who had longer disease duration than the others.Pathological examination revealed invasive fibrosclerosis of the thyroid follicles,thyroid capsule,and the surrounding tissues.In RT tissues,the number of cells containing lambda chains was a little higher than those containing kappa chains.Conclusions RT is a rare disease which might be more common in middle-aged females than in other populations.Pathological features include the destruction of thyroid follicle,extension into surrounding tissues by inflammatory cells and fibrous tissues.Immunohistochemical staining of kappa and lambda chains could help diagnose RT.

文摘The species richness and propagule number of arbuscular mycorrhizal fungi(AMF)are high in intensively-managed agricultural soils.Past research has shown that AMF improve crop phosphorus(P)uptake under low soil P conditions,however it is unclear if AMF play a role in high Olsen-P soils.In this study,we investigated whether native fungal benefits exist under high P input field conditions in-situ and contribute to P utilization.We installed in-grow tubes which were sealed with different membrane pore sizes(30 or 0.45μm)to allow or prevent AMF hyphae access to the hyphal compartment and prevent cotton roots from penetrating the chamber.We used the depletion of soil available P(Olsen-P)in the hyphae accessed compartment to indicate P uptake by the native AMF community.Our results showed that the native AMF mediated P depletion and microbial biomass P(MBP)turnover and caused the largest Olsen-P depletion ratio and MBP turnover ratio in the high P treatments(Olsen-P:78.29 mg kg^(-1)).The cotton roots in each fertilization regime were colonized by a unique AMF community and Glomus and Paraglomus were the dominant genera,implying the longterm fertilization regimes domesticated the AMF community.We conclude that native AMF caused the P depletion and P turnover even under high soil Olsen-P conditions.
文摘Well-organized mesoporous titania particles and thin films were successfully synthesized by using tetrabutyl titanate as the inorganic precursor and triblock copolymer (Pluronic P123) as the template via evaporationinduced self-assembly process. The resulting materials were characterized by high resolution transmission electron microscopy (HRTEM), X-ray diffraction (XRD). Fourier-transform infrared spectroscopy (FT-IR),Brunauer-Emmett-Teller (BET) and atomic force microscopy (AFM). Macro shape of mesoporous titania would greatly influence the mesostructure of materials, and the probable reasons were also discussed.

文摘AIM: To identify the genetic defects of a Chinese patient with sporadic retinitis pigmentosa (RP). METHODS: Ophthalmologic examinations were performed on the sporadic RP patient, 144 genes associated with retinal diseases were scanned with capture next generation sequencing (CNGS) approach. Two heterozygous mutations in PDE6B were confirmed in the pedigree by Sanger sequencing subsequently. The carrier frequency of PDE6B mutations of reported PDE5B mutations based on the available two public exome databases (1000 Genomes Project and ESP6500 Genomes Project) and one in-house exome database was investigated. RESULTS: We identified compound heterozygosity of two novel nonsense mutations c.1133G>A (p.W378X) and c.2395C>T (p.R799X) in PDE6B, one reported causative gene for RP. Neither of the two mutations in our study was presented in three exome databases. Two mutations (p.R74C and p.T6041) in PDE5B have relatively high frequencies in the ESP6500 and in-house databases, respectively, while no common dominant mutation in each of the database or across all databases. CONCLUSION: We demonstrates that compound heterozygosity of two novel nonsense mutations in PDE6B could lead to RP. These results collectively point to enormous potential of next-generation sequencing in determining the genetic etiology of RP and how various mutations in PDE9B contribute to the genetic heterogeneity of RP.

Mutation were screened in recruited family members by polymerase chain reaction(PCR) of the two reported genes(CRYAA and GJA8) which were linked to human total cataracts and direct sequencing of the PCR product. The molecular consequences of the identified mutation was dissected. The plasmids carrying wild-type and mutant mouse ORF of Gja8, coding for connexin 50(Cx50), were generated and ectopic expressed in 293 cells. Recombinant protein expression and cellular localization of recombinated Cx50 were assessed by confocal microscopy.RESULTS: Clinical and ophthalmological examinations were performed on the affected and unaffected family members. 文摘In this work, patterned macropores with a diameter larger than 100 μm were introduced to pristine three-dimensional (3D) nanofibrous bacterial cellulose (BC) scaffolds by using the infrared laser micromachining technique in an attempt to create an in vitro model for the culture of breast cancer cells. The morphology, pore structure, and mechanical performance of the obtained patterned macroporous BC (PM-BC) scaffolds were characterized by scanning electron microscopy (SEM), mercury intrusion porosimeter, and mechanical testing. A human breast cancer cell (MDA-MB-231) line was cultured onto the PM-BC scaffolds to investigate the role of macropores in the control of cancer cell behavior. MTT assay, SEM, and hematoxylin and eosin (H&E) staining were employed to determine cell adhesion, growth, proliferation, and infiltration. The PM-BC scaffolds were found to be able to promote cellular adhesion and proliferation on the scaffolds, and further to allow for cell infiltration into the PM-BC scaffolds. The results demonstrated that BC scaffolds with laser-patterned macropores were promising for the in vitro 3D culture of breast cancer cells.

文摘Potassium-ion capacitors(KICs)are promising for sustainable and eco-friendly energy storage technologies,yet their slow reaction kinetics and poor cyclability induced by large K-ion size are a major obstacle toward practical applications.Herein,by employing black phosphorus nanosheets(BPNSs)as a typical high-capacity anode material,we report that BPNS anodes armored with an ultrathin oriented-grown metal–organic-framework(MOF)interphase layer(BPNS@MOF)exhibit regulated potassium storage behavior for highperformance KICs.The MOF interphase layers as protective layer with ordered pores and high chemical/mechanical stability facilitate K ion diffusion and accommodate the volume change of electrode,beneficial for improved reaction kinetics and enhanced cyclability,as evidenced by substantial characterizations,kinetics analysis and DFT calculations.Consequently,the BPNS@MOF electrode as KIC anodes exhibits outstanding cycle performance outperforming most of the reported state-of-art KICs so far.

文摘BACKGROUND Waldenström macroglobulinemia(WM)is a distinct clinicopathologic entity characterized by the infiltration of the bone marrow by clonal lymphoplasmacytic cells that produce monoclonal immunoglobulin M(IgM)in the blood,and patients may present with symptoms related to the infiltration of the hematopoietic tissues or the effects of monoclonal IgM in the blood.Funduscopic abnormalities were noted in some of the patients due to hyperviscosity or other retinal lesions.Optical coherence tomography angiography(OCTA)as a noninvasive imaging tool can give qualitative and quantitative information about the status of retinal and choroidal vessels,which might be useful for diagnosing patients with WM-associated retinopathy.CASE SUMMARY The patient was a 67-year-old man who presented with sudden visual disturbance in both eyes.Ophthalmic tests showed that best corrected visual acuity(BCVA)for this patient was 20/100 in the right eye and 20/1000 in the left eye.Fundus examination,optical coherence tomography(OCT),and OCTA revealed substantial bilateral optic disc edema,dilated and tortuous retinal veins,and diffuse intraretinal blot hemorrhages and edema which were consistent with bilateral central retinal vein occlusion(CRVO).Meanwhile,remarkable bilateral serous macular detachments(SMD)were noticed on OCT.Systemic examinations showed that the patient had anemia and extremely high level of monoclonal IgM and infiltration of clonal lymphoplasmacytic cells in bone marrow.The diagnosis of WM with hyperviscosity and retinopathy was made based on the clinical manifestation and laboratory findings.He was subsequently treated with intravitreal ranibizumab injection,plasmapheresis,and bortezomib plus rituximab with dexamethasone.Six months after treatments,the central macular volume decreased by 16.1%in the right eye and 28.6%in the left eye on OCT,and the patient’s BCVA was improved to 20/60 in the right eye and 20/400 in the left eye.Very good partial response was obtained after systemic treatment.CONCLUSION WM may affect visual function and present as bilateral CRVO.OCTA can show characteristic changes in both retina and choroid vasculatures,which might be of great value for diagnosing or following patients with WM retinopathy.Intravitreal anti-vascular endothelial growth factor treatment combined with systemic therapy might be beneficial for WM patients with retinopathy(SMD and CRVO).
