The effects of seawater temperature on the physiological performance of three Halimeda species were studied for a period of 28 d.Five treatments were established for Halimeda cylindracea,Halimeda opuntia and Halimeda ...The effects of seawater temperature on the physiological performance of three Halimeda species were studied for a period of 28 d.Five treatments were established for Halimeda cylindracea,Halimeda opuntia and Halimeda lacunalis,in triplicate aquaria representing a factorial temperature with 24°C,28°C,32°C,34°C and 36°C,respectively.The average Fv/Fm of these species ranged from 0.732 to 0.756 between 24°C and 32°C but declined sharply between 34°C(0.457±0.035)and 36°C(0.122±0.014).Calcification was highest at 28°C,with net calcification rates(Gnet)of(20.082±2.482)mg/(g·d),(12.825±1.623)mg/(g·d)and(6.411±1.029)mg/(g·d)for H.cylindracea,H.opuntia and H.lacunalis,respectively.Between 24°C and 32°C,the specific growth rate(SGR)of H.lacunalis(0.079%–0.110%d–1)was lower than that of H.cylindracea(0.652%–1.644%d–1)and H.opuntia(0.360%–1.527%d–1).Three Halimeda species gradually bleached at 36°C during the study period.Malondialdehyde(MDA)and proline levels in tissues of the three Halimeda were higher in 34–36°C than those in 24–32°C.The results indicate that seawater temperature with range of 24–32°C could benefit the growth and calcification of these Halimeda species,however,extreme temperatures above 34°C have negative impacts.The measured physiological parameters also revealed that H.cylindracea and H.opuntia displayed broader temperature tolerance than H.lacunalis.展开更多

It is always interesting to search functional carbons with high heteroatom contents and desirable structure for ORR. Within this study, the surface modification of carbon nanotubes(CNTs) via hydrothermal carbonization(HTC) technique in the presence of glucose and urea was reported, where the surface of CNTs is successfully coated by nitrogen containing hydrothermal carbon layers. The resulting composite combines both advantages of the outstanding electrical conductivity of CNTs and the effective ORR active sites provided by doped nitrogen in the HTC carbon layers. By controlling the ratio of glucose and urea, the nitrogen contents coated on the surface of CNTs can reach up to 1.7 wt%. The resulting materials show outstanding electrochemical activity towards ORR in alkaline electrolyte, making it one of the valuable metal-free electrode materials and a competent alternative to the state-of-the-art Pt/C catalyst.展开更多
Lithophyllum okamurae is one of the important encrusting coralline algae,which plays important roles as primary producer,carbonate sediment builder,and habitat provider in the marine ecosystems.In this study,L.okamura...Lithophyllum okamurae is one of the important encrusting coralline algae,which plays important roles as primary producer,carbonate sediment builder,and habitat provider in the marine ecosystems.In this study,L.okamurae was collected from tropical coast of Sanya,and firstly described based on both detailed morph-anatomical characteristics and molecular studies of typic DNA sequences.The structure of the thalli of L.okamurae was pseudoparenchymatous construction with radially organized dimerous organizations in the crustose portion.The pseudoparenchymatous construction were composed of three parts,including 1 to 3 layers of epithelia cells which had flatten to round outermost walls,one layer of square or rectangular cells of the hypothallia and multiple layers of square or elongated rectangular peripheral cells.Palisade cells were observed,and the cells of the contiguous vegetative filaments were connected by secondary pit-connections with cell fusions absent.The carposporangial conceptacles,the spermatangial conceptacles,the bisporangial conceptacles and the tetrasporangial conceptacles were observed,and all these four kinds of conceptacles were uniporate.The spermatangial conceptacles were slightly convex and buried at shallow depths in the thalli tissues,and the carposporangial conceptacles and asexual conceptacles were protruding and conical.Phylogenetic studies based on DNA barcoding markers of 18 S rDNA,COI,rbc L and psb A revealed that L.okamurae clustered with the closest relation of L.atlanticum,and formed a distinct branch.Based on the comparative anatomical features and the molecular data,the detailed description of the valid species of L.okamurae was firstly given in this study to provide theoretical basis for algae resources utilization and conservation in marine ecosystems.展开更多
Although the adverse impacts of ocean acidification(OA)on marine calcifiers have been investigated extensively,the anti-stress capabilities regulated by increased light availability are unclear.Herein,the interactive ...Although the adverse impacts of ocean acidification(OA)on marine calcifiers have been investigated extensively,the anti-stress capabilities regulated by increased light availability are unclear.Herein,the interactive effects of three light levels(30μmol photons/(m^(2)·s),150μmol photons/(m^(2)·s),and 240μmol photons/(m^(2)·s)combined with two pCO_(2)concentrations(400 ppmv and 1400 ppmv)on the physiological acclimation of the calcifying macroalga Halimeda opuntia were investigated using a pCO_(2)-light coupling experiment.The OA negatively influenced algal growth,calcification,photosynthesis,and other physiological performances in H.opuntia.The relative growth rate under elevated pCO_(2)conditions significantly declined by 13.14%−41.29%,whereas net calcification rates decreased by nearly three-fold under OA conditions.Notably,increased light availability enhanced stress resistance through the accumulation of soluble organic molecules,especially soluble carbohydrate,soluble protein,and free amino acids,and in combination with metabolic enzyme-driven activities,OA stress was alleviated.The carotenoid content under low light conditions increased markedly,and the rapid light curve of the relative electron transport rate was enhanced significantly by increasing light intensities,indicating that this new organization of the photosynthetic machinery in H.opuntia accommodated light variations and elevated pCO_(2)conditions.Thus,the enhanced metabolic performance of the calcifying macroalga H.opuntia mitigated OA-related stress.展开更多
[Objectives]To evaluate the effects of Tongdu Tiaoshen Acupuncture method on improving learning and memory function and hippocampal nerve regeneration and repair after stroke.[Methods]Animal models of cognitive impair...[Objectives]To evaluate the effects of Tongdu Tiaoshen Acupuncture method on improving learning and memory function and hippocampal nerve regeneration and repair after stroke.[Methods]Animal models of cognitive impairment in the recovery phase of cerebral infarction in rats were prepared,and rats were randomly divided into control group,model group,donepezil group,and acupuncture group.Morris water maze experiment was carried out to evaluate learning and memory ability.Nissl staining,BrdU and DCX immunofluorescence staining were used to observe the number and morphological structure of neuronal pyramidal and granular cells in the dentate gyrus(DG)area of the hippocampus,as well as the proliferation and differentiation of neural progenitor cells.TUNEL method was used to detect cell apoptosis.[Results]Before intervention,the scores and ELP of the model group,donepezil group and acupuncture group were significantly higher than those of the blank group(P<0.05);after the intervention,the neurological scores and ELP of the blank group and the model group were not significantly different from those before the intervention(P<0.05);the donepezil group and acupuncture group were significantly reduced,there was no difference between the donepezil group and acupuncture group(P>0.05),but it was still significantly higher than the blank group(P<0.05).The results of BrdU and DCX immunofluorescence staining showed that there were only a few BrdU positive cells and DCX protein in the hippocampal DG area of mice in the blank group,and the positive cells in the model group were significantly increased(P<0.05);donepezil group and acupuncture group showed significant increase compared with the model group(P<0.05);compared with the blank group,the number of neurons in the hippocampal DG area of each group was significantly reduced(P<0.01),and the neuronal apoptosis index was significantly increased(P<0.01);compared with the blank group,the number of neurons in the hippocampal DG area of each group was significantly reduced(P<0.01),and the neuronal apoptosis index was significantly increased(P<0.01).The number of neurons in the hippocampal DG area of rats in the acupuncture group and donepezil group was significantly increased(P<0.01),and the neuronal apoptosis index was significantly decreased(P<0.01).[Conclusions]Tongdu Tiaoshen Acupuncture method can improve the learning and memory function of stroke model rats by promoting the regeneration and repair of hippocampal nerve in rats with cerebral infarction.展开更多
The aim of this study was to analyze the combination of three kinds of in-house IFN-γ ELISPOT using peptide A53 and peptide mixtures (E6 + E7 and E6 + E7 + C14) with tuberculin skin test (TST) to detect latent TB inf...The aim of this study was to analyze the combination of three kinds of in-house IFN-γ ELISPOT using peptide A53 and peptide mixtures (E6 + E7 and E6 + E7 + C14) with tuberculin skin test (TST) to detect latent TB infection (LTBI) in China. A total of 788 healthy people were recruited and analyzed by three kinds of IFN-g ELISPOT, 581 of them had TST results, of which 147 samples were also compared with the T- SPOT.TB test. The positive detection rates for T- SPOT.TB and three kinds of IFN-γ ELISPOT with A53, E6 + E7 and E6 + E7 + C14 were 14.28% (21/147), 29.43% (171/581), 23.24% (135/581) and 28.40% (165/581), respectively. These results were significantly lower than the positive TST results, which were positive in 82.99% (122/147) and 75.73% (440/ 581), respectively. The positive detection rates of three kinds of IFN-γ ELISPOT (31.60%, 26.65% and 32.11% in 788 cases, respectively) could better reflect over 40.00% of Mycobacterium tuberculosis (MTB) infection rate in China. Detection rates between contacts and non-contacts by three kinds of IFN-γ ELIS-POT were not significantly different (p > 0.05). It can be seen that the three kinds of in-house IFN-γ ELIS- POT might be used as a complementary tool of T- SPOT.TB for detecting LTBI in the healthy population of China.展开更多

Acetylcholinesterase(AChE) is a key enzyme used to detect organophosphorus pesticide residues by the enzyme inhibition method.An accidental discovery of a mutant strain with AChE activity was made in our laboratory du...Acetylcholinesterase(AChE) is a key enzyme used to detect organophosphorus pesticide residues by the enzyme inhibition method.An accidental discovery of a mutant strain with AChE activity was made in our laboratory during the process of AChE expression by Pichia pastoris.The pPIC9 K-Drosophila melanogaster acetylcholinesterase(DmAChE)-like expression vector was constructed by codon optimization of this mutant strain,which was transformed into P.pastoris GS115,and positive clones were selected on yeast peptone dextrose(YPD) plate with G418 at 4.0 mg/mL.The GS115-pPIC9 K-DmAChE-like strain was subjected to 0.5% methanol induction expression for 120 h,with a protein band at 4.3 kDa found by the tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) pattern of the fermentation supernatant.After preliminary purification by ammonium sulfate precipitation,the enzyme activity was detected to be 76.9 U/(mL·min).In addition,the pesticide sensitivity test proved that DmAChE-like is selective and sensitive to organophosphorus pesticides.展开更多
The aqueous foam template without any solvent and only using the particles stabilizer has attracted much attention for preparation of the porous adsorbents.Herein,a novel porous adsorbent was fabricated via thermal-in...The aqueous foam template without any solvent and only using the particles stabilizer has attracted much attention for preparation of the porous adsorbents.Herein,a novel porous adsorbent was fabricated via thermal-initiated polymerization of Pickering aqueous foams,which was stabilized by the natural sepiolite(Sep)and pine pollen,and utilized for the removal of antibiotic from aqueous solution.The stabilizing mechanism of Pickering aqueous foam of that the Sep was modified with the leaching substance from pine pollen and arranged orderly around the bubble to form a dense"shell"structure was revealed.The adsorbents possessed the hierarchical porous structure and excellent adsorption performance for antibiotic of chlorotetracycline hydrochloride(CTC)and tetracycline hydrochloride(TC).The equilibrium adsorption capacities of CTC and TC were achieved with 465.59 and 330.59 mg/g within 60 min at 25℃,respectively.The adsorption process obeyed Langmuir model and pseudo-second-order adsorption kinetic model.This work provided eco-friendly approach for fabricate porous adsorbents for wastewater treatment.展开更多
With increasing industrial activities,mercury has been largely discharged into environment and caused serious environmental problems.The growing level of mercury pollution has become a huge threat to human health due ...With increasing industrial activities,mercury has been largely discharged into environment and caused serious environmental problems.The growing level of mercury pollution has become a huge threat to human health due to its significant biotoxicity.Therefore,the simple and fast means for on-site monitoring discharged mercury pollution are highly necessary to protect human beings from its pernicious effects in time.Herein,a"turn off"fluorescent biosensor(mCherry L199 C)for sensing Hg2+was successfully designed based on direct modification of the chromophore environment of fluorescent protein mCherry.For rapid screening and characterization,the designed variant of mCherry(mCherry L199 C)was directly expressed on outer-membrane of Escherichia coli cells by cell surface display technique.The fluorescent biosensor was characterized to have favorable response to Hg2+at micromole level among other metal ions and over a broad pH range.Further,the cells of the fluorescent biosensor were encapsulated in alginate hydrogel to develop the cells-alginate hydrogel-based paper.The cells-alginate hydrogel-based paper could detect mercury pollution in 5 min with simple operation process and inexpensive equipment,and it could keep fluorescence and activity stable at 4℃ for 24 hr,which would be a high-throughput screening tool in preliminarily reporting the presence of mercury pollution in natural setting.展开更多

The biophysical pr...In this paper, size-controlled and surface-functionalized RhB-labeled and Cyclosporin A (CsA)-loaded polysty- rene (PS) nanospheres were successfully synthesized via miniemulsion polymerization. The biophysical properties of PEG functionalized PS nanospheres from protein adsorption, blood compatibility, cell compatibility and cell penetrability showed the nanoparticles with high biocompatibility. These results indicated that PEG modified PS nanospheres showed outstanding properties as low size distribution (0.164), high encapsulation efficiency (98.3%), long re-calcification time (50% than positive control), low hemolysis ratio (3.19%) and high cell viability (95.3%). This work could be used as a good drug delivery system for CsA.展开更多

文摘[Objectives]To evaluate the effects of Tongdu Tiaoshen Acupuncture method on improving learning and memory function and hippocampal nerve regeneration and repair after stroke.[Methods]Animal models of cognitive impairment in the recovery phase of cerebral infarction in rats were prepared,and rats were randomly divided into control group,model group,donepezil group,and acupuncture group.Morris water maze experiment was carried out to evaluate learning and memory ability.Nissl staining,BrdU and DCX immunofluorescence staining were used to observe the number and morphological structure of neuronal pyramidal and granular cells in the dentate gyrus(DG)area of the hippocampus,as well as the proliferation and differentiation of neural progenitor cells.TUNEL method was used to detect cell apoptosis.[Results]Before intervention,the scores and ELP of the model group,donepezil group and acupuncture group were significantly higher than those of the blank group(P<0.05);after the intervention,the neurological scores and ELP of the blank group and the model group were not significantly different from those before the intervention(P<0.05);the donepezil group and acupuncture group were significantly reduced,there was no difference between the donepezil group and acupuncture group(P>0.05),but it was still significantly higher than the blank group(P<0.05).The results of BrdU and DCX immunofluorescence staining showed that there were only a few BrdU positive cells and DCX protein in the hippocampal DG area of mice in the blank group,and the positive cells in the model group were significantly increased(P<0.05);donepezil group and acupuncture group showed significant increase compared with the model group(P<0.05);compared with the blank group,the number of neurons in the hippocampal DG area of each group was significantly reduced(P<0.01),and the neuronal apoptosis index was significantly increased(P<0.01);compared with the blank group,the number of neurons in the hippocampal DG area of each group was significantly reduced(P<0.01),and the neuronal apoptosis index was significantly increased(P<0.01).The number of neurons in the hippocampal DG area of rats in the acupuncture group and donepezil group was significantly increased(P<0.01),and the neuronal apoptosis index was significantly decreased(P<0.01).[Conclusions]Tongdu Tiaoshen Acupuncture method can improve the learning and memory function of stroke model rats by promoting the regeneration and repair of hippocampal nerve in rats with cerebral infarction.
文摘The aim of this study was to analyze the combination of three kinds of in-house IFN-γ ELISPOT using peptide A53 and peptide mixtures (E6 + E7 and E6 + E7 + C14) with tuberculin skin test (TST) to detect latent TB infection (LTBI) in China. A total of 788 healthy people were recruited and analyzed by three kinds of IFN-g ELISPOT, 581 of them had TST results, of which 147 samples were also compared with the T- SPOT.TB test. The positive detection rates for T- SPOT.TB and three kinds of IFN-γ ELISPOT with A53, E6 + E7 and E6 + E7 + C14 were 14.28% (21/147), 29.43% (171/581), 23.24% (135/581) and 28.40% (165/581), respectively. These results were significantly lower than the positive TST results, which were positive in 82.99% (122/147) and 75.73% (440/ 581), respectively. The positive detection rates of three kinds of IFN-γ ELISPOT (31.60%, 26.65% and 32.11% in 788 cases, respectively) could better reflect over 40.00% of Mycobacterium tuberculosis (MTB) infection rate in China. Detection rates between contacts and non-contacts by three kinds of IFN-γ ELIS-POT were not significantly different (p > 0.05). It can be seen that the three kinds of in-house IFN-γ ELIS- POT might be used as a complementary tool of T- SPOT.TB for detecting LTBI in the healthy population of China.

文摘Acetylcholinesterase(AChE) is a key enzyme used to detect organophosphorus pesticide residues by the enzyme inhibition method.An accidental discovery of a mutant strain with AChE activity was made in our laboratory during the process of AChE expression by Pichia pastoris.The pPIC9 K-Drosophila melanogaster acetylcholinesterase(DmAChE)-like expression vector was constructed by codon optimization of this mutant strain,which was transformed into P.pastoris GS115,and positive clones were selected on yeast peptone dextrose(YPD) plate with G418 at 4.0 mg/mL.The GS115-pPIC9 K-DmAChE-like strain was subjected to 0.5% methanol induction expression for 120 h,with a protein band at 4.3 kDa found by the tricine-sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) pattern of the fermentation supernatant.After preliminary purification by ammonium sulfate precipitation,the enzyme activity was detected to be 76.9 U/(mL·min).In addition,the pesticide sensitivity test proved that DmAChE-like is selective and sensitive to organophosphorus pesticides.

文摘The aqueous foam template without any solvent and only using the particles stabilizer has attracted much attention for preparation of the porous adsorbents.Herein,a novel porous adsorbent was fabricated via thermal-initiated polymerization of Pickering aqueous foams,which was stabilized by the natural sepiolite(Sep)and pine pollen,and utilized for the removal of antibiotic from aqueous solution.The stabilizing mechanism of Pickering aqueous foam of that the Sep was modified with the leaching substance from pine pollen and arranged orderly around the bubble to form a dense"shell"structure was revealed.The adsorbents possessed the hierarchical porous structure and excellent adsorption performance for antibiotic of chlorotetracycline hydrochloride(CTC)and tetracycline hydrochloride(TC).The equilibrium adsorption capacities of CTC and TC were achieved with 465.59 and 330.59 mg/g within 60 min at 25℃,respectively.The adsorption process obeyed Langmuir model and pseudo-second-order adsorption kinetic model.This work provided eco-friendly approach for fabricate porous adsorbents for wastewater treatment.

文摘With increasing industrial activities,mercury has been largely discharged into environment and caused serious environmental problems.The growing level of mercury pollution has become a huge threat to human health due to its significant biotoxicity.Therefore,the simple and fast means for on-site monitoring discharged mercury pollution are highly necessary to protect human beings from its pernicious effects in time.Herein,a"turn off"fluorescent biosensor(mCherry L199 C)for sensing Hg2+was successfully designed based on direct modification of the chromophore environment of fluorescent protein mCherry.For rapid screening and characterization,the designed variant of mCherry(mCherry L199 C)was directly expressed on outer-membrane of Escherichia coli cells by cell surface display technique.The fluorescent biosensor was characterized to have favorable response to Hg2+at micromole level among other metal ions and over a broad pH range.Further,the cells of the fluorescent biosensor were encapsulated in alginate hydrogel to develop the cells-alginate hydrogel-based paper.The cells-alginate hydrogel-based paper could detect mercury pollution in 5 min with simple operation process and inexpensive equipment,and it could keep fluorescence and activity stable at 4℃ for 24 hr,which would be a high-throughput screening tool in preliminarily reporting the presence of mercury pollution in natural setting.

文摘In this paper, size-controlled and surface-functionalized RhB-labeled and Cyclosporin A (CsA)-loaded polysty- rene (PS) nanospheres were successfully synthesized via miniemulsion polymerization. The biophysical properties of PEG functionalized PS nanospheres from protein adsorption, blood compatibility, cell compatibility and cell penetrability showed the nanoparticles with high biocompatibility. These results indicated that PEG modified PS nanospheres showed outstanding properties as low size distribution (0.164), high encapsulation efficiency (98.3%), long re-calcification time (50% than positive control), low hemolysis ratio (3.19%) and high cell viability (95.3%). This work could be used as a good drug delivery system for CsA.
