Objective:To investigate the effect of the spinal cord extracts(SCE)after spinal cord injuries(SCIs)on the proliferation of rat embryonic neural stem cells(NSCs)and the expressions of mRNA of Notch1 as well as of Hes1...Objective:To investigate the effect of the spinal cord extracts(SCE)after spinal cord injuries(SCIs)on the proliferation of rat embryonic neural stem cells(NSCs)and the expressions of mRNA of Notch1 as well as of Hes1 in this process in vitro.Methods:The experiment was conducted in 4 different mediums:NSCs+PBS(Group A-blank control group),NSCs+SCE with healthy SD rats(Croup B-normal control group),NSCs+SCE with SD rats receiving sham-operation treatment(Croup C-sham-operation group)and NSCs+SCE with SCIs rats(Group D-paraplegic group).Proliferative abilities of 4 different groups were analyzed by MTT chromatometry after co-culture for 1,2,3,4 and 5 d,respectively.The expressions of Notch 1 and Hes1 mRNA were also detected with RT-PCR after co-culture for 24 and 48 h,respectively.Results:After co-culture for 1,2,3,4 and 5 d respectively,the MTT values of group D were significantly higher than those of group A,group B and group C(P<0.05).However,there were no significantly differences regarding MTT values between group A,group B and group C after co-culture for 1,2,3,4 and 5 d,respectively(P>0.05).Both the expressions of Notch1 and Hes1 mRNA of group D were significantly higher than those of other 3 groups after co-culture for 24 h and 48 h as well(P<0.05).But there was no difference oin expressions of Notch1 and Hes1 mRNA among group A,group B and group C after co-culture for 24 h and 48 h(P>0.05).There was no difference in expressions of Notch1and Hes1 mRNA between 24 h and 48 h treatment in group D.Conclusions:SCE could promote the proliferation of NSCs.It is demonstrated that the microenvironment of SCI may promote the proliferation of NSCs.Besides,SCE could increase the expression of Notch1 and Hes1 mRNA of NSC.It can be concluded that the Notch signaling pathway activation is one of the mechanisms that locally injured microenvironment contributes to the proliferation of ENSC after SCIs.This process may be performed by up-regulating the expressions of Notch1 and Hes1 gene.展开更多
Abstract—Focused crawlers (also known as subjectoriented crawlers), as the core part of vertical search engine, collect topic-specific web pages as many as they can to form a subject-oriented corpus for the latter ...Abstract—Focused crawlers (also known as subjectoriented crawlers), as the core part of vertical search engine, collect topic-specific web pages as many as they can to form a subject-oriented corpus for the latter data analyzing or user querying. This paper demonstrates that the popular algorithms utilized at the process of focused web crawling, basically refer to webpage analyzing algorithms and crawling strategies (prioritize the uniform resource locator (URLs) in the queue). Advantages and disadvantages of three crawling strategies are shown in the first experiment, which indicates that the best-first search with an appropriate heuristics is a smart choice for topic-oriented crawlingwhile the depth-first search is helpless in focused crawling. Besides, another experiment on comparison of improved ones (with a webpage analyzing algorithm added) is carried out to verify that crawling strategies alone are not quite efficient for focused crawling and in most cases their mutual efforts are taken into consideration. In light of the experiment results and recent researches, some points on the research tendency of focused crawler algorithms are suggested.展开更多
The traditional evaporator of water source heat pump system is easy to freeze when the water source temperature is too low in winter.A novel ice source heat pump system is proposed,which can use low temperature surfac...The traditional evaporator of water source heat pump system is easy to freeze when the water source temperature is too low in winter.A novel ice source heat pump system is proposed,which can use low temperature surface water to supply heat to buildings.One of the key problems for safe transportation is velocity range of ice slurry in ice outlet pipeline.In order to study the critical flow velocity of ice slurry in the pipe,an Euler model of horizontal straight pipe and 90°elbow pipe were established by Fluent software.The influence of five factors on the critical flow velocity of two kinds of pipes,including initial ice packing fraction,ice particle size,pipe diameter,length of straight pipe and radius curvature of elbow was studied.It found that the initial ice content should be controlled from 10%to 15%.The inlet flow rate was 20%higher than the critical flow rate under the most unfavorable condition.Ice particle size was less than 0.2 mm.The total length of ice discharge pipeline was less than 50 m.Curvature radius of the bend should be met the minimum requirements of the specification of the pipe used.展开更多
