Plant AT-rich sequence and zinc binding(PLATZ)transcription factors are a class of plant specific zincdependent DNA-binding proteins that function in abiotic stress response and plant development.In this study,31 GmPL...Plant AT-rich sequence and zinc binding(PLATZ)transcription factors are a class of plant specific zincdependent DNA-binding proteins that function in abiotic stress response and plant development.In this study,31 GmPLATZ genes were identified in soybean.GmPLATZ17 was down-regulated by drought and exogenous abscisic acid.Transgenic Arabidopsis and soybean hairy roots overexpressing GmPLATZ17 showed drought sensitivity and inhibition of stress-associated gene transcription.In contrast,suppressed expression of GmPLATZ17 led to increased drought tolerance in transgenic soybean hairy roots.The GmPLATZ17 protein was verified to interact physically with the GmDREB5 transcription factor,and overexpression of GmDREB5 increased drought tolerance in soybean hairy roots.Interaction of GmPLATZ17 with GmDREB5 was shown to interfere with the DRE-binding activity of GmDREB5,suppressing downstream stress-associated gene expression.These results show that GmPLATZ17 inhibits drought tolerance by interacting with GmDREB5.This study sheds light on PLATZ transcription factors and the function of GmPLATZ17 in regulating drought sensitivity.展开更多

P1 antigen was detected both in vitro and in vivo by synthetic peptide-derived polyclonal antibody-based immunochemistry. The designed peptides were synthesized by solid-phase technique, purified by high performance liquid chromatography, coupled to Keyhole limpet hemocyanin, and injected into rabbits to prepare polyclonal antibody. The emergence of positive cells revealed that synthetic peptide could elicit antibodies against P1 and viral protein could be synthesized. The polyclonal peptide antibodies described here was successfully applied to immunochemical staining and proved helpful in diagnosing P1.展开更多

The indoor and field surveys w...In order to make clear the effects of water on yield and agronomic traits of wheat in Cangzhou,18 wheat cultivars from north part of Huanghuai Plain were collected for spring irrigation. The indoor and field surveys were used to analyze the effects of irrigation frequency on stem tiller situation,leaf area coefficient,yield and other traits of different wheat cultivars. The results indicated that the average yield of irrigation 1 increased by 68. 07 kg compared with that of irrigation 0,while the average yield of irrigation 2 increased by 60. 00 kg compared with that of irrigation 1. The survey of flag leaf area of wheat cultivars at the grain filling stage showed that the leaf area of irrigation 0 was the smallest( 18. 15 cm^2 on average),the leaf area of irrigation 1 was 20. 34 cm^2,and the leaf area of irrigation 2 was 24. 05 cm^2. With the increase in the irrigation times,the volume weight of wheat cultivars declined,the number of infertile spikelet dropped,and the plant height gradually increased. In conclusion,every decrease of one time of irrigation times,there will be loss of yield about 60. 00 kg.展开更多

Ursolic acid(UA)is a naturally occurring ursane triterpenoid,which exhibits a wide range of unique biological activities.To clarify its mechanism of action(MOA),a series of fluorescent derivatives of UA(5a-c)were desi...Ursolic acid(UA)is a naturally occurring ursane triterpenoid,which exhibits a wide range of unique biological activities.To clarify its mechanism of action(MOA),a series of fluorescent derivatives of UA(5a-c)were designed and synthesized by conjugation with 7-nitrobenzo-2-oxa-1,3-diazole(NBD)fluorophore.Among them,5c exhibited similar anti-proliferative activity with UA against HCT116 cells(half maximal inhibitory concentration(IC_(50))=9.21±0.50μmol/L).Cell imaging experiment indicated that 5c was rapidly taken up in HCT116 cells in a dose and time-dependent manner.Then,5c was found to localize in endoplasmic reticulum(ER),lysosomes,and mitochondria,but not in nucleus of HCT116 cells by confocal microscopy studies.Preliminary MOA proved that UA induced autophagy with a unique intracellular distribution mechanism involving ER and lysosome.In all,our work provides new clues for revealing the molecular mechanism of UA as an antitumor agent.展开更多
