Background The aim of this experiment was to investigate the effect of different levels of betaine(Bet)inclusion in the diet on the intestinal health of growing rabbits under summer heat.A total of 100 weaned Qixing m...Background The aim of this experiment was to investigate the effect of different levels of betaine(Bet)inclusion in the diet on the intestinal health of growing rabbits under summer heat.A total of 100 weaned Qixing meat rabbits aged 35 d with body weight of 748.61±38.59 g were randomly divided into 5 treatment groups:control group(basal diet)and Bet groups(basal diet+0.75,1.0,1.5 or 2.0 g/kg Bet).The average daily temperature in the rabbitry during the experiment was 30.48°C and the relative humidity was 69.44%.Results Dietary addition of Bet had no significant effect on growth performance and health status of growing rabbits(P>0.05),but it increased ileal secretory immunoglobulin A content compared to the control under summer heat(P<0.05).Addition of 0.75 g/kg Bet up-regulated jejunal IL-4,down-regulated ileal TNF-αexpression(P<0.05).The addition of 1.0 g/kg Bet increased the villi height(VH)in the jejunum(P<0.05).Serum glucose levels were reduced,and the expression of SLC6A20 was up-regulated in jejunum and ileum of rabbits fed with 1.5 g/kg Bet(P<0.05).When added at 2.0 g/kg,Bet reduced serum HSP70 content,increased jejunal VH,and up-regulated duodenal SLC7A6,SLC38A2,mTOR and 4EBP-2 expression(P<0.05).Correlation analysis revealed that intestinal mTOR expression was significantly and positively correlated with SLC7A6,SLC38A2,SLC36A1 and IL-4 expression(P<0.05).Conclusions Dietary addition of Bet can up-regulate the expression of anti-inflammatory factors through the AAT/mTOR pathway,improve the intestinal immune function,alleviate intestinal damage in growing rabbits caused by summer heat,and improve intestinal health.展开更多
Background Transmissible gastroenteritis virus(TGEV)is one of the main pathogens causing severe diarrhea of pig-lets.The pathogenesis of TGEV is closely related to intestinal inflammation.All-trans retinoic acid(ATRA)...Background Transmissible gastroenteritis virus(TGEV)is one of the main pathogens causing severe diarrhea of pig-lets.The pathogenesis of TGEV is closely related to intestinal inflammation.All-trans retinoic acid(ATRA)is the main active metabolite of vitamin A,which has immunomodulatory and anti-inflammatory properties.However,it is unclear whether ATRA can alleviate TGEV-induced intestinal inflammation and barrier dysfunction in piglets.This study aimed to investigate the effects of ATRA on growth performance,diarrhea,intestinal inflammation and intesti-nal barrier integrity of TGEV-challenged piglets.Methods In a 19-d study,32 weaned piglets were randomly divided into 4 treatments:Control group(basal diet),TGEV group(basal diet+TGEV challenge),TGEV+ATRA5 group(basal diet+5 mg/d ATRA+TGEV challenge)and TGEV+ATRA15 group(basal diet+15 mg/d ATRA+TGEV challenge).On d 14,piglets were orally administered TGEV or the sterile medium.Results Feeding piglets with 5 and 15 mg/d ATRA alleviated the growth inhibition and diarrhea induced by TGEV(P<0.05).Feeding piglets with 5 and 15 mg/d ATRA also inhibited the increase of serum diamine oxidase(DAO)activ-ity and the decrease of occludin and claudin-1 protein levels in jejunal mucosa induced by TGEV,and maintained intestinal barrier integrity(P<0.05).Meanwhile,5 mg/d ATRA feeding increased the sucrase activity and the expres-sions of nutrient transporter related genes(GLUT2 and SLC7A1)in jejunal mucosa of TGEV-challenged piglets(P<0.05).Furthermore,5 mg/d ATRA feeding attenuated TGEV-induced intestinal inflammatory response by inhibit-ing the release of interleukin(IL)-1β,IL-8 and tumor necrosis factor-α(TNF-α),and promoting the secretion of IL-10 and secretory immunoglobulin A(sIgA)(P<0.05).Feeding 5 mg/d ATRA also down-regulated the expressions of Toll-like receptors and RIG-I like receptors signaling pathway related genes(TLR3,TLR4,RIG-I,MyD88,TRIF and MAVS)and the phosphorylation level of nuclear factor-κB-p65(NF-κB p65),and up-regulated the inhibitor kappa B alpha(IκBα)protein level in jejunal mucosa of TGEV-challenged piglets(P<0.05).Conclusions ATRA alleviated TGEV-induced intestinal barrier damage by inhibiting inflammatory response,thus improving the growth performance and inhibiting diarrhea of piglets.The mechanism was associated with the inhibi-tion of NF-κB signaling pathway mediated by TLR3,TLR4 and RIG-I.展开更多
Background Intestinal inflammation is the main risk factor causing intestinal barrier dysfunction and lipopolysaccharide(LPS)can trigger inflammatory responses in various eukaryotic species.Yeast hydrolysate(YH)posses...Background Intestinal inflammation is the main risk factor causing intestinal barrier dysfunction and lipopolysaccharide(LPS)can trigger inflammatory responses in various eukaryotic species.Yeast hydrolysate(YH)possesses multibiological effects and is received remarkable attention as a functional ingredient for improving growth performance and promoting health in animals.However,there is still inconclusive on the protective effects of dietary YH supplementation on intestinal barrier of piglets.This study was conducted to investigate the attenuate effects of YH supplementation on inflammatory responses and intestinal barrier injury in piglets challenged with LPS.Methods Twenty-four piglets(with an average body weight of 7.42±0.34 kg)weaned at 21 days of age were randomly assigned to one of two dietary treatments(12 replications with one pig per pen):a basal diet or a basal diet containing YH(5 g/kg).On the 22nd d,6 piglets in each treatment were intraperitoneally injected with LPS at 150μg/kg BW,and the others were injected with the same amount of sterile normal saline.Four hours later,blood samples of each piglet were collected and then piglets were euthanized.Results Dietary YH supplementation increased average daily feed intake and average daily gain(P<0.01),decreased the ratio of feed intake to gain of piglets(P sponse,evidenced by the increase o=0.048).Lipopolysaccharide(LPS)injection induced systemic inflammatory ref serum concentrations of haptoglobin(HP),adrenocorticotropic hormone(ACTH),cortisol,and interleukin-1β(IL-1β).Furthermore,LPS challenge resulted in inflammatory intestinal damage,by up-regulation of the protein or mRNA abundances of tumor necrosis factor-α(TNF-α),IL-1β,toll-like receptors 4(TLR4)and phosphor-nuclear factor-κB-p65(p-NFκB-p65)(P<0.01),and down-regulation of the jejunal villus height,the protein and mRNA abundances of zonula occludens-1(ZO-1)and occludin(OCC;P<0.05)in jejunal mucosa.Dietary YH supplementation decreased the impaired effects of ACTH,cortisol,HP,IL-1βand diamine oxidase in serum(P<0.05).Moreover,YH supplementation also up-regulated the jejunal villus height,protein and mRNA abundances of ZO-1 and OCC(P<0.05),down-regulated the mRNA expressions of TNF-αand IL-1βand the protein abundances of TNF-α,IL-1β,TLR4 and p-NFκB-p65 in jejunal mucosa in LPS-challenged pigs(P<0.01).Conclusion Yeast hydrolysate could attenuate inflammatory response and intestinal barrier injury in weaned piglets challenged with LPS,which was associated with the inhibition of TLR4/NF-κB signaling pathway activation.展开更多
Background The skeletal muscle of pigs is vulnerable to oxidative damage,resulting in growth retardation.Selenoproteins are important components of antioxidant systems for animals,which are generally regulated by diet...Background The skeletal muscle of pigs is vulnerable to oxidative damage,resulting in growth retardation.Selenoproteins are important components of antioxidant systems for animals,which are generally regulated by dietary selenium(Se)level.Here,we developed the dietary oxidative stress(DOS)-inducing pig model to investigate the protective effects of selenoproteins on DOS-induced skeletal muscle growth retardation.Results Dietary oxidative stress caused porcine skeletal muscle oxidative damage and growth retardation,which is accompanied by mitochondrial dysfunction,endoplasmic reticulum(ER)stress,and protein and lipid metabolism disorders.Supplementation with Se(0.3,0.6 or 0.9 mg Se/kg)in form of hydroxy selenomethionine(OH-SeMet)linearly increased muscular Se deposition and exhibited protective effects via regulating the expression of selenotranscriptome and key selenoproteins,which was mainly reflected in lower ROS levels and higher antioxidant capacity in skeletal muscle,and the mitigation of mitochondrial dysfunction and ER stress.What’s more,selenoproteins inhibited DOS induced protein and lipid degradation and improved protein and lipid biosynthesis via regulating AKT/mTOR/S6K1 and AMPK/SREBP-1 signalling pathways in skeletal muscle.However,several parameters such as the activity of GSH-Px and T-SOD,the protein abundance of JNK2,CLPP,SELENOS and SELENOF did not show dose-dependent changes.Notably,several key selenoproteins such as MSRB1,SELENOW,SELENOM,SELENON and SELENOS play the unique roles during this protection.Conclusions Increased expression of selenoproteins by dietary OH-SeMet could synergistically alleviate mitochondrial dysfunction and ER stress,recover protein and lipid biosynthesis,thus alleviate skeletal muscle growth retardation.Our study provides preventive measure for OS-dependent skeletal muscle retardation in livestock husbandry.展开更多
Large-scale sand fracturing is a necessary means in the efficient exploitation of shale gas/oil.However,in the process of fracturing operation,the sand carrying fluid and proppant easily causes scouring and wear to pr...Large-scale sand fracturing is a necessary means in the efficient exploitation of shale gas/oil.However,in the process of fracturing operation,the sand carrying fluid and proppant easily causes scouring and wear to production strings,especially the casing perforation system,which damage the wellbore integrity and deformation to affect the subsequent fracturing.For this problem,taking the actual construction conditions and perforation technology of an oilfield in western China as an example,the structural parameters of the downhole string were measured and the wall thickness reduction model of casing perforation suitable for large-displacement sand fracturing in horizontal well section was established.With software ANSYS-FLUENT,the casing perforation erosion under the conditions of different displacements,sand content and perforation sand-passing quantity in the process of sand fracturing was simulated and calculated.The influences of three parameters on perforation erosion and expansion were analyzed and the prediction chart of the influences of three main control factors on perforation erosion and expansion was established.The perforation erosion images after fracturing construction were obtained with the downhole eagle perforation logging technology.The logging chart results were compared with the downhole eagle perforation data.The error between the established numerical simulation calculation charts and the real logging data was about 5%,indicating that the simulation charts were the valuable reference.展开更多

Background:This study was conducted to determine the effects of different dietary protein levels and amino acids supplementation patterns in low protein diets on the growth performance,carcass characteristics and nitr...Background:This study was conducted to determine the effects of different dietary protein levels and amino acids supplementation patterns in low protein diets on the growth performance,carcass characteristics and nitrogen excretion in growing-finishing pigs.Forty-two barrows(25.00±0.39 kg)were randomly assigned to 7 diets.Diet 1:the high crude protein diet with balanced for 10 essential amino acids(EAAs).Diet 2:the medium crude protein diet with 2%(approx)decreased protein level of Diet 1 and balanced 10 EAAs.Diet 3:the low crude protein diet with 4%decreased protein level of Diet 1 and balanced 10 EAAs.The protein levels of Diet 4,5,6 and 7 were the same as that of Diet 3.Diet 4 was only balanced for lysine(Lys),methionine(Met),threonine(Thr)and tryptophan(Trp);Diet 5 and 6 were further supplemented with extra isoleucine(Ile)or valine(Val),respectively;Diet 7 was further supplemented with extra Ile+Val.Results:Over the 112 days trial,the reduction of dietary protein by 2%or 4%with balanced10 EAAs significantly decreased nitrogen excretion(P<0.05),but had no effects on growth performance and carcass characteristics(P>0.05).In low protein diet,Val supplementation significantly increased body weight gain at 25–50 kg phase(P<0.05),while Ile supplementation at 75–100 kg phase and 100–125 kg phase significantly reduced the ratio of feed to gain(P<0.05).No effect of different dietary protein levels and amino acids supplementation patterns in low protein diets on carcass characteristics was observed(P>0.05).The total N excretion of pigs supplemented with only Lys,Met,Thr and Trp was numerically higher than that of pigs fed with extra Ile,or Val,or Ile+Val diets.Conclusion:In low protein diet,Val is more required than Ile in the early growing phage(25–50 kg),while Ile becomes more required in the late growing and finishing phage(75–125 kg).展开更多

Rats receiving high-fat diet were randomly divided into two groups, the tel-...We sought to investigate the effects of telmisartan on high-fat diet-induced hypertension and to explore the possible underlying mechanisms. Rats receiving high-fat diet were randomly divided into two groups, the tel- misartan group (n = 9) and the high-fat diet group (n = 10). The control group consisted of age-matched rats on a regular diet (n = 10). At the end of the treatment, the body weight, blood pressure, insulin sensitivity and serum adiponectin levels of all rats were examined, and their visceral fat was extracted and weighed. Our results showed that telmisartan improved insulin resistance and dyslipidemia and increased serum adiponectin levels. Telmisar- tan also lowered both systolic blood pressure and diastolic blood pressure, and decreased the accumulation of perirenal fat associated with high-fat diet. Furthermore, telmisartan increased adiponectin mRNA expression in the perirenal fat. Correlation analysis showed that both systolic blood pressure and diastolic blood pressure were positively correlated with perirenal fat. These effects of telmisartan may be mediated through decreases in perirenal fat and contributed to the improvement of perirenal fat function. Our findings suggested a strong link between perirenal fat and high-fat diet-induced hypertension, and identified telmisartan as a potential drug for the treatment of obesity-related hypertension.展开更多
Background:Chronic heat stress(CHS)disrupts hepatic metabolic homeostasis and jeopardizes product quality of pigs.Selenium(Se)may regulate the metabolic state through affect selenoprotein.Thus,we investigate the prote...Background:Chronic heat stress(CHS)disrupts hepatic metabolic homeostasis and jeopardizes product quality of pigs.Selenium(Se)may regulate the metabolic state through affect selenoprotein.Thus,we investigate the protective effect of dietary hydroxy-4-methylselenobutanoic acid(HMSeBA)on CHS induced hepatic metabolic disorder in growing pigs,and the corresponding response of selenoprotein.Methods:Forty crossbreed growing pigs were randomly assigned to five groups:control group raised in the thermoneutral environment(22±2℃)with basal diet;four CHS groups raised in hyperthermal condition(33±2℃)with basal diet and supplied with 0.0,0.2,0.4,and 0.6 mg Se/kg HMSeBA,respectively.The trial lasted 28 d.The serum biochemical,hepatic metabolism related enzyme,protein and gene expression and 25 selenoproteins in liver tissue were determined by real-time PCR,ELISA and western blot.Results:CHS significantly increased the rectal temperature,respiration rate,serum aspartate aminotransferase(AST)and low-density lipoprotein cholesterol(LDL-C)of pigs,up-regulated hepatic heat shock protein 70(HSP70)and induced lower liver weight,glycogen content,hepatic glucokinase and glutathione peroxidase(GSH-Px).The CHSinduced liver metabolic disorder was associated with the aberrant expression of 6 metabolism-related gene and 11 selenoprotein encoding genes,and decreased the protein abundance of GCK,GPX4 and SELENOS.HMSeBA improved anti-oxidative capacity of liver.0.4 or 0.6 mg Se/kg HMSeBA supplementation recovered the liver weight,glycogen content and rescue of mRNA abundance of genes related to metabolism and protein levels of GCK.HMSeBA supplementation changed expressions of 15 selenoprotein encoding genes,and enhanced protein expression of GPX1,GPX4 and SELENOS in the liver affected by CHS.CHS alone showed no impact while HMSeBA supplementation increased protein levels of p-AMPKαin the liver.Conclusions:In summary,HMSeBA supplementation beyond nutrient requirement mitigates CHS-induced hepatic metabolic disorder,recovered the liver glycogen content and the processes that are associated with the activation of AMPK signal and regulation of selenoproteins in the liver of growing pigs.展开更多

In this research,we have performed a shear failure experiment on rock samples with prefabricated cracks to simulate the process of plate movement that forms strike-slip faults. We studied the evolution law of the deformation field to simulate the shear failure experiment, and these results gave us a comprehensive understanding of the elaborate strain distribution law and its formation process with which to identify actual fault zones. We performed uniaxial compression tests on marble slabs with prefabricated double shear cracks to study the distribution and evolution of the deformation field during shear failure. Analysis of the strain field at different loading stages showed that with an increase in the load, the shear strain field initially changed to a disordered-style distribution. Further, the strain field was partially concentrated and finally completely concentrated near the crack and then distributed in the shape of a strip along the crack. We also computed coefficients of variation(CVs) for the physical quantities u, v, and exy, which varied with the load. The CV curves were found to correspond to the different loading stages. We found that at the uniform deformation stage, the CV value was small and changed slowly,whereas at the later nonuniform deformation stage, the CV value increased sharply and changed abruptly. Therefore, the precursor to a rock sample breakdown can be predicted by observing the variation characteristics of CV statistics. The correlation we found between our experimental and theoretical results revealed that our crack evolution and sample deformation results showed good coupling with seismic distribution characteristics near the San Andreas Fault.展开更多

For the special geological features of the Takelamagan Desert area, we use the multi-channel analysis of surface wave (MASW) method to process multi-channel shallow surface wave records to determine the near surface velocity structure in the desert area. We also process, analyze, and compare the surface waves in many-trace records extracted from the oil exploration shot gathers in the area. We show that the MASW method can determine detailed shallow velocity structure in desert areas and the many-trace records can be used to get detailed deep geological structure. The combination of the two different datasets can obtain the exact velocity structure upper 60 m depth in the survey area.展开更多
Background:Consumption of resistant starch(RS)has been associated with various intestinal and systemic health benefits,but knowledge of its effects on intestinal health and inflammatory response in stressed birds is l...Background:Consumption of resistant starch(RS)has been associated with various intestinal and systemic health benefits,but knowledge of its effects on intestinal health and inflammatory response in stressed birds is limited.Here,we examined how dietary RS supplementation from 12%raw potato starch(RPS)modulated inflammatory severity induced by lipopolysaccharide(LPS)in meat ducks.Results:LPS administration at 14,16,and 18 d(chronic challenge)decreased body weight(BW)and glucagon-like peptide 1 receptor(GLP-1R)level with higher intestinal permeability and inflammation,evident by higher proinflammatory cytokine levels.Dietary 12%RPS supplementation enhanced Claudin-1 and GLP-1R expression,along with lower levels of inflammatory factors in both ileum and serum.Microbiome analysis showed that RS treatment shifted microbial structure reflected by enriched the proportion of Firmicutes,Bifidobacterium,Ruminococcus,etc.Dietary RS addition also significantly increased the concentrations of propionate and butyrate during chronic LPS challenge.Furthermore,response to acute challenge,the ducks received 2 mg/kg BW LPS at 14 d had higher concentrations of serum endotoxins and inflammatory cytokines,as well as upregulated transcription of toll like receptor 4(TLR4)in ileum when compared to control birds.Analogous to GLP-1 agonist liraglutide,dietary RS addition decreased endotoxins and inflammation cytokines,whereas it upregulated the GLP-1 synthesis related genes expression.Meanwhile,dietary RS supplementation suppressed the acute LPS challenge-induced TLR4 transcription.Conclusions:These data suggest that dietary 12%RPS supplementation could attenuate the LPS-induced inflammation as well as intestinal injury of meat ducks,which might involve in the alteration in gut microbiota,SCFAs production and the signaling pathways of TLR4 and GLP-1/GLP-1R.展开更多
Natural language semantic construction improves natural language comprehension ability and analytical skills of the machine.It is the basis for realizing the information exchange in the intelligent cloud-computing env...Natural language semantic construction improves natural language comprehension ability and analytical skills of the machine.It is the basis for realizing the information exchange in the intelligent cloud-computing environment.This paper proposes a natural language semantic construction method based on cloud database,mainly including two parts:natural language cloud database construction and natural language semantic construction.Natural Language cloud database is established on the CloudStack cloud-computing environment,which is composed by corpus,thesaurus,word vector library and ontology knowledge base.In this section,we concentrate on the pretreatment of corpus and the presentation of background knowledge ontology,and then put forward a TF-IDF and word vector distance based algorithm for duplicated webpages(TWDW).It raises the recognition efficiency of repeated web pages.The part of natural language semantic construction mainly introduces the dynamic process of semantic construction and proposes a mapping algorithm based on semantic similarity(MBSS),which is a bridge between Predicate-Argument(PA)structure and background knowledge ontology.Experiments show that compared with the relevant algorithms,the precision and recall of both algorithms we propose have been significantly improved.The work in this paper improves the understanding of natural language semantics,and provides effective data support for the natural language interaction function of the cloud service.展开更多
Ground short circuit faults in current transmission lines are common in the power systems.In order to prevent the power system from aggravating the accident caused by short-circuit faults of transmission lines,a novel...Ground short circuit faults in current transmission lines are common in the power systems.In order to prevent the power system from aggravating the accident caused by short-circuit faults of transmission lines,a novel convolutional neural network(CNN)model is constructed to identify the short-circuit fault of the transmission line in the power system.The CNN model is mainly consisted of five convolutional layers,three max-pooling layers,one concatenate layer,one dropout layer,one fully connected layer,and a Softmax classifier.This method uses a fixed time window to intercept system short-circuit fault data,extracts the deep features of these data from the training samples,and then corresponds the extracted features to labels one-to-one.Finally,in PSCAD/EMTDC,the new England 10 machine 39 nodes are taken as an example to realize the simulation.The experimental results show that the CNN model can quickly and accurately identify the short-circuit fault types,and the optimal model accuracy rate reaches 99.95%.The results of this manuscript-have positive effect on enhancing the disaster prevention capability and the operation stability of transmission lines.展开更多

The system is based on CIMISS (National Integrated Meteorological Information Sharing Platform) for expansion and construction, integrating key correction forecasts, weather model comparisons, flood predictions, etc. The forecasting technology, starting from the flood control and drought relief of the service basin and the comprehensive dispatch of the reservoir, has realized the monitoring of water and rain conditions, the production of key business products, the production and distribution of integrated service products, and the quality tracking assessment. Background: To determine responses of neonatal intestine to maternal increased consumption of DL-methionine(DLM) or DL-2-hydroxy-4-methylthiobutanoic acid(HMTBA), eighteen primiparous sows(Landrace × Yorksh...Background: To determine responses of neonatal intestine to maternal increased consumption of DL-methionine(DLM) or DL-2-hydroxy-4-methylthiobutanoic acid(HMTBA), eighteen primiparous sows(Landrace × Yorkshire) were allocated based on body weight and backfat thickness to the control, DLM and HMTBA groups(n = 6), with the nutritional treatments introduced from postpartum d0 to d14.Results: The DLM-fed sows showed negative energy balance manifested by lost bodyweight, lower plasma glucose, subdued tricarboxylic acid cycle, and increased plasma lipid metabolites levels. Both villus height and ratio of villus height to crypt depth averaged across the small intestine of piglets were higher in the DLM and HMTBA groups than in the control group. Piglet jejunal oxidized glutathione concentration and ratio of oxidized to reduced glutathione were lower in the HMTBA group than in the DLM and control groups. However, piglet jejunal aminopeptidase A, carnitine transporter 2 and IGF-II precursor m RNA abundances were higher in the DLM group than in the HMTBA and control groups.Conclusion: Increasing maternal consumption of methionine as DLM and HMTBA promoted neonatal intestinal growth by increasing morphological development or up-regulating expression of genes responsible for nutrient metabolism. And increasing maternal consumption of HMTBA promoted neonatal intestinal antioxidant capacity without compromising maternal energy homeostasis during early lactation.展开更多

We report a rare case o...Non-compaction cardiomyopathy is a rare form of cardiomyopathy;its most common clinical manifestations are heart failure (HF), ventricular arrhythmia, thromboembolism, and sudden cardiac death. We report a rare case of a 63-year-old man with chest tightness, worsening lower leg edema, dyspnea, and decreased exercise tolerance. He had a medical history of gastric cancer treated with subtotal gastrectomy and post-operative chemotherapy with paclitaxel and fluorouracil three years ago. At that time, he was diagnosed with non-compaction cardiomyopathy, and the thickened and reticulated trabecular muscle was exclusively confined to left ventricular apex. Five months ago, he was admitted to our hospital with heart failure and treated for dilated cardiomyopathy, echocardiography revealed severe trabecular noncompact myocardium in both ventricles, which was confirmed by cardiac magnetic resonance imaging (CMR). It is generally accepted that non-compacted myocardium forms in the early embryonic stage, which raises a question in our case whether acquired factors, such as antineoplastic drugs, potentially accelerate the pathological progression of non-compaction cardiomyopathy. Considering there are disparities between current screening tools such as echocardiography and CMR regarding diagnostic criteria, multi-detector CT may be an alternative examination method that could provide a new perspective for diagnosis.展开更多
文摘Background The aim of this experiment was to investigate the effect of different levels of betaine(Bet)inclusion in the diet on the intestinal health of growing rabbits under summer heat.A total of 100 weaned Qixing meat rabbits aged 35 d with body weight of 748.61±38.59 g were randomly divided into 5 treatment groups:control group(basal diet)and Bet groups(basal diet+0.75,1.0,1.5 or 2.0 g/kg Bet).The average daily temperature in the rabbitry during the experiment was 30.48°C and the relative humidity was 69.44%.Results Dietary addition of Bet had no significant effect on growth performance and health status of growing rabbits(P>0.05),but it increased ileal secretory immunoglobulin A content compared to the control under summer heat(P<0.05).Addition of 0.75 g/kg Bet up-regulated jejunal IL-4,down-regulated ileal TNF-αexpression(P<0.05).The addition of 1.0 g/kg Bet increased the villi height(VH)in the jejunum(P<0.05).Serum glucose levels were reduced,and the expression of SLC6A20 was up-regulated in jejunum and ileum of rabbits fed with 1.5 g/kg Bet(P<0.05).When added at 2.0 g/kg,Bet reduced serum HSP70 content,increased jejunal VH,and up-regulated duodenal SLC7A6,SLC38A2,mTOR and 4EBP-2 expression(P<0.05).Correlation analysis revealed that intestinal mTOR expression was significantly and positively correlated with SLC7A6,SLC38A2,SLC36A1 and IL-4 expression(P<0.05).Conclusions Dietary addition of Bet can up-regulate the expression of anti-inflammatory factors through the AAT/mTOR pathway,improve the intestinal immune function,alleviate intestinal damage in growing rabbits caused by summer heat,and improve intestinal health.

文摘Background Transmissible gastroenteritis virus(TGEV)is one of the main pathogens causing severe diarrhea of pig-lets.The pathogenesis of TGEV is closely related to intestinal inflammation.All-trans retinoic acid(ATRA)is the main active metabolite of vitamin A,which has immunomodulatory and anti-inflammatory properties.However,it is unclear whether ATRA can alleviate TGEV-induced intestinal inflammation and barrier dysfunction in piglets.This study aimed to investigate the effects of ATRA on growth performance,diarrhea,intestinal inflammation and intesti-nal barrier integrity of TGEV-challenged piglets.Methods In a 19-d study,32 weaned piglets were randomly divided into 4 treatments:Control group(basal diet),TGEV group(basal diet+TGEV challenge),TGEV+ATRA5 group(basal diet+5 mg/d ATRA+TGEV challenge)and TGEV+ATRA15 group(basal diet+15 mg/d ATRA+TGEV challenge).On d 14,piglets were orally administered TGEV or the sterile medium.Results Feeding piglets with 5 and 15 mg/d ATRA alleviated the growth inhibition and diarrhea induced by TGEV(P<0.05).Feeding piglets with 5 and 15 mg/d ATRA also inhibited the increase of serum diamine oxidase(DAO)activ-ity and the decrease of occludin and claudin-1 protein levels in jejunal mucosa induced by TGEV,and maintained intestinal barrier integrity(P<0.05).Meanwhile,5 mg/d ATRA feeding increased the sucrase activity and the expres-sions of nutrient transporter related genes(GLUT2 and SLC7A1)in jejunal mucosa of TGEV-challenged piglets(P<0.05).Furthermore,5 mg/d ATRA feeding attenuated TGEV-induced intestinal inflammatory response by inhibit-ing the release of interleukin(IL)-1β,IL-8 and tumor necrosis factor-α(TNF-α),and promoting the secretion of IL-10 and secretory immunoglobulin A(sIgA)(P<0.05).Feeding 5 mg/d ATRA also down-regulated the expressions of Toll-like receptors and RIG-I like receptors signaling pathway related genes(TLR3,TLR4,RIG-I,MyD88,TRIF and MAVS)and the phosphorylation level of nuclear factor-κB-p65(NF-κB p65),and up-regulated the inhibitor kappa B alpha(IκBα)protein level in jejunal mucosa of TGEV-challenged piglets(P<0.05).Conclusions ATRA alleviated TGEV-induced intestinal barrier damage by inhibiting inflammatory response,thus improving the growth performance and inhibiting diarrhea of piglets.The mechanism was associated with the inhibi-tion of NF-κB signaling pathway mediated by TLR3,TLR4 and RIG-I.

文摘Background Intestinal inflammation is the main risk factor causing intestinal barrier dysfunction and lipopolysaccharide(LPS)can trigger inflammatory responses in various eukaryotic species.Yeast hydrolysate(YH)possesses multibiological effects and is received remarkable attention as a functional ingredient for improving growth performance and promoting health in animals.However,there is still inconclusive on the protective effects of dietary YH supplementation on intestinal barrier of piglets.This study was conducted to investigate the attenuate effects of YH supplementation on inflammatory responses and intestinal barrier injury in piglets challenged with LPS.Methods Twenty-four piglets(with an average body weight of 7.42±0.34 kg)weaned at 21 days of age were randomly assigned to one of two dietary treatments(12 replications with one pig per pen):a basal diet or a basal diet containing YH(5 g/kg).On the 22nd d,6 piglets in each treatment were intraperitoneally injected with LPS at 150μg/kg BW,and the others were injected with the same amount of sterile normal saline.Four hours later,blood samples of each piglet were collected and then piglets were euthanized.Results Dietary YH supplementation increased average daily feed intake and average daily gain(P<0.01),decreased the ratio of feed intake to gain of piglets(P sponse,evidenced by the increase o=0.048).Lipopolysaccharide(LPS)injection induced systemic inflammatory ref serum concentrations of haptoglobin(HP),adrenocorticotropic hormone(ACTH),cortisol,and interleukin-1β(IL-1β).Furthermore,LPS challenge resulted in inflammatory intestinal damage,by up-regulation of the protein or mRNA abundances of tumor necrosis factor-α(TNF-α),IL-1β,toll-like receptors 4(TLR4)and phosphor-nuclear factor-κB-p65(p-NFκB-p65)(P<0.01),and down-regulation of the jejunal villus height,the protein and mRNA abundances of zonula occludens-1(ZO-1)and occludin(OCC;P<0.05)in jejunal mucosa.Dietary YH supplementation decreased the impaired effects of ACTH,cortisol,HP,IL-1βand diamine oxidase in serum(P<0.05).Moreover,YH supplementation also up-regulated the jejunal villus height,protein and mRNA abundances of ZO-1 and OCC(P<0.05),down-regulated the mRNA expressions of TNF-αand IL-1βand the protein abundances of TNF-α,IL-1β,TLR4 and p-NFκB-p65 in jejunal mucosa in LPS-challenged pigs(P<0.01).Conclusion Yeast hydrolysate could attenuate inflammatory response and intestinal barrier injury in weaned piglets challenged with LPS,which was associated with the inhibition of TLR4/NF-κB signaling pathway activation.

文摘Background The skeletal muscle of pigs is vulnerable to oxidative damage,resulting in growth retardation.Selenoproteins are important components of antioxidant systems for animals,which are generally regulated by dietary selenium(Se)level.Here,we developed the dietary oxidative stress(DOS)-inducing pig model to investigate the protective effects of selenoproteins on DOS-induced skeletal muscle growth retardation.Results Dietary oxidative stress caused porcine skeletal muscle oxidative damage and growth retardation,which is accompanied by mitochondrial dysfunction,endoplasmic reticulum(ER)stress,and protein and lipid metabolism disorders.Supplementation with Se(0.3,0.6 or 0.9 mg Se/kg)in form of hydroxy selenomethionine(OH-SeMet)linearly increased muscular Se deposition and exhibited protective effects via regulating the expression of selenotranscriptome and key selenoproteins,which was mainly reflected in lower ROS levels and higher antioxidant capacity in skeletal muscle,and the mitigation of mitochondrial dysfunction and ER stress.What’s more,selenoproteins inhibited DOS induced protein and lipid degradation and improved protein and lipid biosynthesis via regulating AKT/mTOR/S6K1 and AMPK/SREBP-1 signalling pathways in skeletal muscle.However,several parameters such as the activity of GSH-Px and T-SOD,the protein abundance of JNK2,CLPP,SELENOS and SELENOF did not show dose-dependent changes.Notably,several key selenoproteins such as MSRB1,SELENOW,SELENOM,SELENON and SELENOS play the unique roles during this protection.Conclusions Increased expression of selenoproteins by dietary OH-SeMet could synergistically alleviate mitochondrial dysfunction and ER stress,recover protein and lipid biosynthesis,thus alleviate skeletal muscle growth retardation.Our study provides preventive measure for OS-dependent skeletal muscle retardation in livestock husbandry.

文摘Large-scale sand fracturing is a necessary means in the efficient exploitation of shale gas/oil.However,in the process of fracturing operation,the sand carrying fluid and proppant easily causes scouring and wear to production strings,especially the casing perforation system,which damage the wellbore integrity and deformation to affect the subsequent fracturing.For this problem,taking the actual construction conditions and perforation technology of an oilfield in western China as an example,the structural parameters of the downhole string were measured and the wall thickness reduction model of casing perforation suitable for large-displacement sand fracturing in horizontal well section was established.With software ANSYS-FLUENT,the casing perforation erosion under the conditions of different displacements,sand content and perforation sand-passing quantity in the process of sand fracturing was simulated and calculated.The influences of three parameters on perforation erosion and expansion were analyzed and the prediction chart of the influences of three main control factors on perforation erosion and expansion was established.The perforation erosion images after fracturing construction were obtained with the downhole eagle perforation logging technology.The logging chart results were compared with the downhole eagle perforation data.The error between the established numerical simulation calculation charts and the real logging data was about 5%,indicating that the simulation charts were the valuable reference.

Due to the different stability of various forms of mercury in the environment, subsequent use of products containing desulfurization by-product additives will continue to be released into the environment, endangering human health. Therefore, it is very necessary to study the form and distribution of mercury in the by-products of desulfurization in coal-fired power plants to provide a theoretical basis for subsequent harmless treatment.</span></span></span><span><span><span style="font-family:""> </span></span></span><span style="font-family:Verdana;"><span style="font-family:Verdana;"><span style="font-family:Verdana;">For content and morphology of mercury analysis, 1 sample of dry FGD ash and 6 samples of wet FGD gypsum were analyzed. 文摘Background:This study was conducted to determine the effects of different dietary protein levels and amino acids supplementation patterns in low protein diets on the growth performance,carcass characteristics and nitrogen excretion in growing-finishing pigs.Forty-two barrows(25.00±0.39 kg)were randomly assigned to 7 diets.Diet 1:the high crude protein diet with balanced for 10 essential amino acids(EAAs).Diet 2:the medium crude protein diet with 2%(approx)decreased protein level of Diet 1 and balanced 10 EAAs.Diet 3:the low crude protein diet with 4%decreased protein level of Diet 1 and balanced 10 EAAs.The protein levels of Diet 4,5,6 and 7 were the same as that of Diet 3.Diet 4 was only balanced for lysine(Lys),methionine(Met),threonine(Thr)and tryptophan(Trp);Diet 5 and 6 were further supplemented with extra isoleucine(Ile)or valine(Val),respectively;Diet 7 was further supplemented with extra Ile+Val.Results:Over the 112 days trial,the reduction of dietary protein by 2%or 4%with balanced10 EAAs significantly decreased nitrogen excretion(P<0.05),but had no effects on growth performance and carcass characteristics(P>0.05).In low protein diet,Val supplementation significantly increased body weight gain at 25–50 kg phase(P<0.05),while Ile supplementation at 75–100 kg phase and 100–125 kg phase significantly reduced the ratio of feed to gain(P<0.05).No effect of different dietary protein levels and amino acids supplementation patterns in low protein diets on carcass characteristics was observed(P>0.05).The total N excretion of pigs supplemented with only Lys,Met,Thr and Trp was numerically higher than that of pigs fed with extra Ile,or Val,or Ile+Val diets.Conclusion:In low protein diet,Val is more required than Ile in the early growing phage(25–50 kg),while Ile becomes more required in the late growing and finishing phage(75–125 kg).

文摘We sought to investigate the effects of telmisartan on high-fat diet-induced hypertension and to explore the possible underlying mechanisms. Rats receiving high-fat diet were randomly divided into two groups, the tel- misartan group (n = 9) and the high-fat diet group (n = 10). The control group consisted of age-matched rats on a regular diet (n = 10). At the end of the treatment, the body weight, blood pressure, insulin sensitivity and serum adiponectin levels of all rats were examined, and their visceral fat was extracted and weighed. Our results showed that telmisartan improved insulin resistance and dyslipidemia and increased serum adiponectin levels. Telmisar- tan also lowered both systolic blood pressure and diastolic blood pressure, and decreased the accumulation of perirenal fat associated with high-fat diet. Furthermore, telmisartan increased adiponectin mRNA expression in the perirenal fat. Correlation analysis showed that both systolic blood pressure and diastolic blood pressure were positively correlated with perirenal fat. These effects of telmisartan may be mediated through decreases in perirenal fat and contributed to the improvement of perirenal fat function. Our findings suggested a strong link between perirenal fat and high-fat diet-induced hypertension, and identified telmisartan as a potential drug for the treatment of obesity-related hypertension.

文摘Background:Chronic heat stress(CHS)disrupts hepatic metabolic homeostasis and jeopardizes product quality of pigs.Selenium(Se)may regulate the metabolic state through affect selenoprotein.Thus,we investigate the protective effect of dietary hydroxy-4-methylselenobutanoic acid(HMSeBA)on CHS induced hepatic metabolic disorder in growing pigs,and the corresponding response of selenoprotein.Methods:Forty crossbreed growing pigs were randomly assigned to five groups:control group raised in the thermoneutral environment(22±2℃)with basal diet;four CHS groups raised in hyperthermal condition(33±2℃)with basal diet and supplied with 0.0,0.2,0.4,and 0.6 mg Se/kg HMSeBA,respectively.The trial lasted 28 d.The serum biochemical,hepatic metabolism related enzyme,protein and gene expression and 25 selenoproteins in liver tissue were determined by real-time PCR,ELISA and western blot.Results:CHS significantly increased the rectal temperature,respiration rate,serum aspartate aminotransferase(AST)and low-density lipoprotein cholesterol(LDL-C)of pigs,up-regulated hepatic heat shock protein 70(HSP70)and induced lower liver weight,glycogen content,hepatic glucokinase and glutathione peroxidase(GSH-Px).The CHSinduced liver metabolic disorder was associated with the aberrant expression of 6 metabolism-related gene and 11 selenoprotein encoding genes,and decreased the protein abundance of GCK,GPX4 and SELENOS.HMSeBA improved anti-oxidative capacity of liver.0.4 or 0.6 mg Se/kg HMSeBA supplementation recovered the liver weight,glycogen content and rescue of mRNA abundance of genes related to metabolism and protein levels of GCK.HMSeBA supplementation changed expressions of 15 selenoprotein encoding genes,and enhanced protein expression of GPX1,GPX4 and SELENOS in the liver affected by CHS.CHS alone showed no impact while HMSeBA supplementation increased protein levels of p-AMPKαin the liver.Conclusions:In summary,HMSeBA supplementation beyond nutrient requirement mitigates CHS-induced hepatic metabolic disorder,recovered the liver glycogen content and the processes that are associated with the activation of AMPK signal and regulation of selenoproteins in the liver of growing pigs.

文摘Seismic hazard analysis is gaining increased attention in the present era because of the catastrophic effects of earthquakes.Scientists always have as a goal to develop new techniques that will help forecast earthquakes before their reoccurrence. In this research,we have performed a shear failure experiment on rock samples with prefabricated cracks to simulate the process of plate movement that forms strike-slip faults. We studied the evolution law of the deformation field to simulate the shear failure experiment, and these results gave us a comprehensive understanding of the elaborate strain distribution law and its formation process with which to identify actual fault zones. We performed uniaxial compression tests on marble slabs with prefabricated double shear cracks to study the distribution and evolution of the deformation field during shear failure. Analysis of the strain field at different loading stages showed that with an increase in the load, the shear strain field initially changed to a disordered-style distribution. Further, the strain field was partially concentrated and finally completely concentrated near the crack and then distributed in the shape of a strip along the crack. We also computed coefficients of variation(CVs) for the physical quantities u, v, and exy, which varied with the load. The CV curves were found to correspond to the different loading stages. We found that at the uniform deformation stage, the CV value was small and changed slowly,whereas at the later nonuniform deformation stage, the CV value increased sharply and changed abruptly. Therefore, the precursor to a rock sample breakdown can be predicted by observing the variation characteristics of CV statistics. The correlation we found between our experimental and theoretical results revealed that our crack evolution and sample deformation results showed good coupling with seismic distribution characteristics near the San Andreas Fault.
文摘Shallow surface wave methods are mostly used for investigation of the surface velocity structure in environmental and engineering geophysics in non-desert areas. For the special geological features of the Takelamagan Desert area, we use the multi-channel analysis of surface wave (MASW) method to process multi-channel shallow surface wave records to determine the near surface velocity structure in the desert area. We also process, analyze, and compare the surface waves in many-trace records extracted from the oil exploration shot gathers in the area. We show that the MASW method can determine detailed shallow velocity structure in desert areas and the many-trace records can be used to get detailed deep geological structure. The combination of the two different datasets can obtain the exact velocity structure upper 60 m depth in the survey area.

文摘Background:Consumption of resistant starch(RS)has been associated with various intestinal and systemic health benefits,but knowledge of its effects on intestinal health and inflammatory response in stressed birds is limited.Here,we examined how dietary RS supplementation from 12%raw potato starch(RPS)modulated inflammatory severity induced by lipopolysaccharide(LPS)in meat ducks.Results:LPS administration at 14,16,and 18 d(chronic challenge)decreased body weight(BW)and glucagon-like peptide 1 receptor(GLP-1R)level with higher intestinal permeability and inflammation,evident by higher proinflammatory cytokine levels.Dietary 12%RPS supplementation enhanced Claudin-1 and GLP-1R expression,along with lower levels of inflammatory factors in both ileum and serum.Microbiome analysis showed that RS treatment shifted microbial structure reflected by enriched the proportion of Firmicutes,Bifidobacterium,Ruminococcus,etc.Dietary RS addition also significantly increased the concentrations of propionate and butyrate during chronic LPS challenge.Furthermore,response to acute challenge,the ducks received 2 mg/kg BW LPS at 14 d had higher concentrations of serum endotoxins and inflammatory cytokines,as well as upregulated transcription of toll like receptor 4(TLR4)in ileum when compared to control birds.Analogous to GLP-1 agonist liraglutide,dietary RS addition decreased endotoxins and inflammation cytokines,whereas it upregulated the GLP-1 synthesis related genes expression.Meanwhile,dietary RS supplementation suppressed the acute LPS challenge-induced TLR4 transcription.Conclusions:These data suggest that dietary 12%RPS supplementation could attenuate the LPS-induced inflammation as well as intestinal injury of meat ducks,which might involve in the alteration in gut microbiota,SCFAs production and the signaling pathways of TLR4 and GLP-1/GLP-1R.

文摘Natural language semantic construction improves natural language comprehension ability and analytical skills of the machine.It is the basis for realizing the information exchange in the intelligent cloud-computing environment.This paper proposes a natural language semantic construction method based on cloud database,mainly including two parts:natural language cloud database construction and natural language semantic construction.Natural Language cloud database is established on the CloudStack cloud-computing environment,which is composed by corpus,thesaurus,word vector library and ontology knowledge base.In this section,we concentrate on the pretreatment of corpus and the presentation of background knowledge ontology,and then put forward a TF-IDF and word vector distance based algorithm for duplicated webpages(TWDW).It raises the recognition efficiency of repeated web pages.The part of natural language semantic construction mainly introduces the dynamic process of semantic construction and proposes a mapping algorithm based on semantic similarity(MBSS),which is a bridge between Predicate-Argument(PA)structure and background knowledge ontology.Experiments show that compared with the relevant algorithms,the precision and recall of both algorithms we propose have been significantly improved.The work in this paper improves the understanding of natural language semantics,and provides effective data support for the natural language interaction function of the cloud service.
文摘Ground short circuit faults in current transmission lines are common in the power systems.In order to prevent the power system from aggravating the accident caused by short-circuit faults of transmission lines,a novel convolutional neural network(CNN)model is constructed to identify the short-circuit fault of the transmission line in the power system.The CNN model is mainly consisted of five convolutional layers,three max-pooling layers,one concatenate layer,one dropout layer,one fully connected layer,and a Softmax classifier.This method uses a fixed time window to intercept system short-circuit fault data,extracts the deep features of these data from the training samples,and then corresponds the extracted features to labels one-to-one.Finally,in PSCAD/EMTDC,the new England 10 machine 39 nodes are taken as an example to realize the simulation.The experimental results show that the CNN model can quickly and accurately identify the short-circuit fault types,and the optimal model accuracy rate reaches 99.95%.The results of this manuscript-have positive effect on enhancing the disaster prevention capability and the operation stability of transmission lines.
文摘The overall design and system functions of the meteorological service integrated service system of the Yangtze River Basin are introduced. The system is based on CIMISS (National Integrated Meteorological Information Sharing Platform) for expansion and construction, integrating key correction forecasts, weather model comparisons, flood predictions, etc. The forecasting technology, starting from the flood control and drought relief of the service basin and the comprehensive dispatch of the reservoir, has realized the monitoring of water and rain conditions, the production of key business products, the production and distribution of integrated service products, and the quality tracking assessment. The application shows that the system can make the forecaster effectively grasp the service focus, improve the timeliness and pertinence of the service, and have a good promotion and application prospect in the basin meteorological service.

文摘Background: To determine responses of neonatal intestine to maternal increased consumption of DL-methionine(DLM) or DL-2-hydroxy-4-methylthiobutanoic acid(HMTBA), eighteen primiparous sows(Landrace × Yorkshire) were allocated based on body weight and backfat thickness to the control, DLM and HMTBA groups(n = 6), with the nutritional treatments introduced from postpartum d0 to d14.Results: The DLM-fed sows showed negative energy balance manifested by lost bodyweight, lower plasma glucose, subdued tricarboxylic acid cycle, and increased plasma lipid metabolites levels. Both villus height and ratio of villus height to crypt depth averaged across the small intestine of piglets were higher in the DLM and HMTBA groups than in the control group. Piglet jejunal oxidized glutathione concentration and ratio of oxidized to reduced glutathione were lower in the HMTBA group than in the DLM and control groups. However, piglet jejunal aminopeptidase A, carnitine transporter 2 and IGF-II precursor m RNA abundances were higher in the DLM group than in the HMTBA and control groups.Conclusion: Increasing maternal consumption of methionine as DLM and HMTBA promoted neonatal intestinal growth by increasing morphological development or up-regulating expression of genes responsible for nutrient metabolism. And increasing maternal consumption of HMTBA promoted neonatal intestinal antioxidant capacity without compromising maternal energy homeostasis during early lactation.
文摘Non-compaction cardiomyopathy is a rare form of cardiomyopathy;its most common clinical manifestations are heart failure (HF), ventricular arrhythmia, thromboembolism, and sudden cardiac death. We report a rare case of a 63-year-old man with chest tightness, worsening lower leg edema, dyspnea, and decreased exercise tolerance. He had a medical history of gastric cancer treated with subtotal gastrectomy and post-operative chemotherapy with paclitaxel and fluorouracil three years ago. At that time, he was diagnosed with non-compaction cardiomyopathy, and the thickened and reticulated trabecular muscle was exclusively confined to left ventricular apex. Five months ago, he was admitted to our hospital with heart failure and treated for dilated cardiomyopathy, echocardiography revealed severe trabecular noncompact myocardium in both ventricles, which was confirmed by cardiac magnetic resonance imaging (CMR). It is generally accepted that non-compacted myocardium forms in the early embryonic stage, which raises a question in our case whether acquired factors, such as antineoplastic drugs, potentially accelerate the pathological progression of non-compaction cardiomyopathy. Considering there are disparities between current screening tools such as echocardiography and CMR regarding diagnostic criteria, multi-detector CT may be an alternative examination method that could provide a new perspective for diagnosis.
